    Genomic innovation for crop improvement

    Crop production needs to increase to secure future food supplies, while reducing its impact on ecosystems. Detailed characterization of plant genomes and genetic diversity is crucial for meeting these challenges. Advances in genome sequencing and assembly are being used to access the large and complex genomes of crops and their wild relatives. These have helped to identify a wide spectrum of genetic variation and permitted the association of genetic diversity with diverse agronomic phenotypes. In combination with improved and automated phenotyping assays and functional genomic studies, genomics is providing new foundations for crop-breeding systems

    Intercultural competence among early childhood educators

    The purpose of this study was to assess and develop intercultural competence among early childhood educators. Intercultural competence is an integral part of creating a welcoming environment for all students in a classroom. It is not only the acknowledgment of individual differences but the acceptance and celebration of what makes each person an individual and member of a cultural group. This project assessed the intercultural competence of 24 early childhood educators and staff at a child development center in the mid-south. Participants completed the Intercultural Development Inventory (IDI) as pre- and post-assessments. In between assessments, our team conducted a cultural competence training workshop to advance the educators’ intercultural development skills. Based on the training and conversations with the educators, diversity books and toys were purchased and implemented into the classroom. On average, intercultural competence increased significantly over time. Overall, most educators scored in the Minimization orientation, which is the most common orientation among adults. In order to advance beyond the Minimization orientation to the Acceptance orientation, educators need additional educational opportunities, which may aid them in understanding concepts regarding power and privilege, as well as other crucial differences between cultures

    A transmission spectrum of the sub-Earth planet L98-59~b in 1.1-1.7 μ\mum

    With the increasing number of planets discovered by TESS, the atmospheric characterization of small exoplanets is accelerating. L98-59 is a M-dwarf hosting a multi-planet system, and so far, four small planets have been confirmed. The innermost planet b is ∼15%\sim15\% smaller and ∼60%\sim60\% lighter than Earth, and should thus have a predominantly rocky composition. The Hubble Space Telescope observed five primary transits of L98-59b in 1.1−1.7 μ1.1-1.7\ \mum, and here we report the data analysis and the resulting transmission spectrum of the planet. We measure the transit depths for each of the five transits and, by combination, we obtain a transmission spectrum with an overall precision of ∼20\sim20 ppm in for each of the 18 spectrophotometric channels. With this level of precision, the transmission spectrum does not show significant modulation, and is thus consistent with a planet without any atmosphere or a planet having an atmosphere and high-altitude clouds or haze. The scenarios involving an aerosol-free, H2_2-dominated atmosphere with H2_2O or CH4_4 are inconsistent with the data. The transmission spectrum also disfavors, but does not rules out, an H2_2O-dominated atmosphere without clouds. A spectral retrieval process suggests that an H2_2-dominated atmosphere with HCN and clouds or haze may be the preferred solution, but this indication is non-conclusive. Future James Webb Space Telescope observations may find out the nature of the planet among the remaining viable scenarios.Comment: 17 pages, 5 figures, 7 tables, accepted for publication in A

    The L 98-59 System: Three Transiting, Terrestrial-size Planets Orbiting a Nearby M Dwarf

    We report the Transiting Exoplanet Survey Satellite (TESS) discovery of three terrestrial-size planets transiting L 98-59 (TOI-175, TIC 307210830)-a bright M dwarf at a distance of 10.6 pc. Using the Gaia-measured distance and broadband photometry, we find that the host star is an M3 dwarf. Combined with the TESS transits from three sectors, the corresponding stellar parameters yield planet radii ranging from 0.8 R ⊕ to 1.6 R ⊕. All three planets have short orbital periods, ranging from 2.25 to 7.45 days with the outer pair just wide of a 2:1 period resonance. Diagnostic tests produced by the TESS Data Validation Report and the vetting package DAVE rule out common false-positive sources. These analyses, along with dedicated follow-up and the multiplicity of the system, lend confidence that the observed signals are caused by planets transiting L 98-59 and are not associated with other sources in the field. The L 98-59 system is interesting for a number of reasons: the host star is bright (V = 11.7 mag, K = 7.1 mag) and the planets are prime targets for further follow-up observations including precision radial-velocity mass measurements and future transit spectroscopy with the James Webb Space Telescope; the near-resonant configuration makes the system a laboratory to study planetary system dynamical evolution; and three planets of relatively similar size in the same system present an opportunity to study terrestrial planets where other variables (age, metallicity, etc.) can be held constant. L 98-59 will be observed in four more TESS sectors, which will provide a wealth of information on the three currently known planets and have the potential to reveal additional planets in the system.Funding for the TESS mission is provided by NASA’s Science Mission directorate.     A phase I study of bendamustine hydrochloride administered day 1+2 every 3 weeks in patients with solid tumours

    The aim of the study was to determine the maximum tolerated dose (MTD), the dose limiting toxicity (DLT), and the pharmacokinetic profile (Pk) of bendamustine (BM) on a day 1 and 2 every 3 weeks schedule and to recommend a safe phase II dose for further testing. Patients with solid tumours beyond standard therapy were eligible. A 30-min intravenous infusion of BM was administered d1+d2 q 3 weeks. The starting dose was 120 mg m−2 per day and dose increments of 20 mg m−2 were used. Plasma and urine samples were analysed using validated high-performance liquid chromatography/fluorescence assays. Fifteen patients were enrolled. They received a median of two cycles (range 1–8). The MTD was reached at the fourth dose level. Thrombocytopaenia (grade 4) was dose limiting in two of three patients at 180 mg m−2. One patient also experienced febrile neutropaenia. Lymphocytopaenia (grade 4) was present in every patient. Nonhaematologic toxicity including cardiac toxicity was not dose limiting with this schedule. Mean plasma Pk values of BM were tmax 35 min, t1/2 49.1 min, Vd 18.3 l m−2, and clearance 265 ml min−1 m−2. The mean total amount of BM and its metabolites recovered in the first micturition was 8.3% (range 2.7–26%). The MTD of BM in the present dose schedule was 180 mg m−2 on day 1+2. Thrombocytopaenia was dose limiting. The recommended dose for future phase II trials with this schedule is 160 mg m−2 per day

    Phase II trial of oral S-1 combined with gemcitabine in metastatic pancreatic cancer

    We conducted a phase II trial of gemcitabine with S-1, oral fluorouracil (5-FU) prodrug tegafur combined with two modulators, 5-chloro-2, 4-dihydroxypyridine and potassium oxonate, to evaluate the activity and toxicity of such a combination in metastatic pancreatic cancer (MPC) patients. Patients who had pathologically proven pancreatic cancer with metastatic lesions were eligible candidates for entry into the study. S-1 was given orally (30 mg m−2) b.i.d. for 14 consecutive days and gemcitabine (1000 mg m−2) was given on days 8 and 15. The cycle was repeated every 21 days. We enrolled 33 MPC patients. The median number of cycles was eight (range 1–20). Grade 3–4 toxicities were leucopenia (33%), neutropenia (55%), anaemia (9%), thrombocytopenia (15%), anorexia (6%), fever (9%), and interstitial pneumonia (6%). Objective responses were obtained in 16 patients (one complete response and 15 partial responses; response rate, 48%; 95% confidence interval (CI), 33–65). Median survival and 1-year survival rate were 12.5 months (95% CI, 5.9–19.1) and 54% (95% CI, 36–72), respectively. Combination chemotherapy with GEM and S-1 was well tolerated and yielded a significantly high response rate

    A modified sequence capture approach allowing standard and methylation analyses of the same enriched genomic DNA sample

    Background: Bread wheat has a large complex genome that makes whole genome resequencing costly. Therefore, genome complexity reduction techniques such as sequence capture make re-sequencing cost effective. With a high-quality draft wheat genome now available it is possible to design capture probe sets and to use them to accurately genotype and anchor SNPs to the genome. Furthermore, in addition to genetic variation, epigenetic variation provides a source of natural variation contributing to changes in gene expression and phenotype that can be profiled at the base pair level using sequence capture coupled with bisulphite treatment. Here, we present a new 12 Mbp wheat capture probe set, that allows both the profiling of genotype and methylation from the same DNA sample. Furthermore, we present a method, based on Agilent SureSelect Methyl-Seq, that will use a single capture assay as a starting point to allow both DNA sequencing and methyl-seq. Results: Our method uses a single capture assay that is sequentially split and used for both DNA sequencing and methyl-seq. The resultant genotype and epi-type data is highly comparable in terms of coverage and SNP/methylation site identification to that generated from separate captures for DNA sequencing and methyl-seq. Furthermore, by defining SNP frequencies in a diverse landrace from the Watkins collection we highlight the importance of having genotype data to prevent false positive methylation calls. Finally, we present the design of a new 12 Mbp wheat capture and demonstrate its successful application to re-sequence wheat. Conclusions: We present a cost-effective method for performing both DNA sequencing and methyl-seq from a single capture reaction thus reducing reagent costs, sample preparation time and DNA requirements for these complementary analyses

    Subnational climate entrepreneurship: innovative climate action in California and São Paulo

    The distinct role of subnational governments such as states and provinces in addressing climate change has been increasingly acknowledged. But while most studies investigate the causes and consequences of particular governments’ actions and networking activities, this article argues that subnational governments can develop climate action as a collective entrepreneurial activity. Addressing many elements explored in this special issue, it focuses on the second question and identifies climate entrepreneurship in two subnational governments—the states of California (USA) and São Paulo (Brazil). Examining internal action, as well as interaction with local authorities, national governments and the international regime, entrepreneurial activities are identified in the invention, diffusion and evaluation of subnational climate policy in each case. The article draws from the recent scholarship on policy innovation, entrepreneurship and climate governance. It contributes to the literature by exploring entrepreneurial subnational government activity in addressing climate change and expanding the understanding of the effects of policy innovation at the subnational level
