    Genetic analysis for sooty mold resistance and heart of palm yield in Archontophoenix.

    Palmeiras do gênero Archontophoenix, utilizadas tanto como ornamentais quanto produtoras de palmito de qualidade, são susceptíveis à fumagina, doença associada à infestação por pulgões, que afeta a fotossíntese, o crescimento e a aparência das plantas. Foram avaliados a campo a resistência à fumagina conjuntamente com três caracteres associados ao crescimento em 24 famílias de meios-irmãos, 28 meses após o plantio, a fim de identificar a variabilidade genética para os caracteres resistência à fumagina, altura, diâmetro e número de folhas; estimar as correlações genotípicas e fenotípicas envolvendo esses quatro caracteres; e aplicar a estratégia de seleção usando o índice multiefeitos. Houve diferenças entre as famílias para os caracteres avaliados, sugerindo a possibilidade de seleção. O baixo coeficiente de variação observado para resistência à fumagina (9,48%) indica que o método de avaliação adotado, baseado em escala de notas após observação visual, foi eficiente e prático para comparar níveis de infestação do complexo fungo+pulgão em palmeiras do gênero Archontophoenix. As estimativas da herdabilidade no sentido restrito foram baixas a médias para os caracteres relacionados ao crescimento (0,10, 0,26 e 0,26 para número de folhas, diâmetro e altura da planta, respectivamente) e muito altas (0,91) para resistência à fumagina. Correlação genética positiva foi observada entre resistência à fumagina e altura da planta, indicando que a eliminação de plantas muito susceptíveis pode ser feita sem interferência na seleção indireta para produção de palmito. A estratégia de seleção pelo índice multiefeitos (com ganhos genéticos esperados variando de 6,23 a 11,83%) mostrou-se adequada para melhorar simultaneamente caracteres relacionados ao crescimento e à produção de palmito

    Stealth silica nanoparticles for theranostic applications

    Silica nanoparticles (SNPs) are a promising carrier system for medical applications. They can be conveniently tailored with different moieties in order to obtain peculiar capabilities. SNPs surface is successfully modified in order to obtain a specific targeting ability through Boletus Edulis Lectin (BEL) bioconjugation, a lectin that specifically recognize overexpressed tumor markers. Moreover, stealthing abilities are imparted with a one-step cocondensation of silanated poly(ethylene) glycol monomethyl ether. Herein we present the preliminary results of these studies

    Preparation and characterization of silica nanoparticles conjugated with a protein that specifically recognizes human cancer cells

    BEL \u3b2-trefoil is a novel lectin purified from the fruiting bodies of king bolete mushrooms [1]. The lectin has potent anti-proliferative effects on human epithelial cancer cells which confers to it an interesting therapeutic potential as an antineoplastic agent. All the three potential binding sites present in the \u3b2-trefoil fold are able to selectively bind the T-antigen disaccharide (Thomsen Friedenreich antigen;Gal\u3b21-3GalNAc) witch is specifically exposed in human carcinomas and other neoplastic tissues. Therefore BEL \u3b2-trefoil linked to nanoparticles could enable them to discriminate between healthy and neoplastic cells targeting nanoparticles specifically towards cancer cells. BEL \u3b2-trefoil is also available recombinantly expressed in E.coli fused with the GFP (Green Fluorescence Protein) that can be useful to immediately detect the presence and activity of the protein after the coupling reaction to nanoparticle

    Effect of a freeze on some technological characteristics of several sugar cane varieties

    Em 18 de julho de 1975, ocorreu geada no Estado de São Paulo; em Piracicaba, a temperatura atingiu -1ºC. Aproveitou-se a ocorrência da geada para investigar seus efeitos sobre características tecnológicas de algumas variedades. Foram tomadas amostras de dez colmos cada uma, a partir de doze dias após a geada, e depois, aproximadamente, a cada duas semanas. Analisaram-se brix % caldo, pol % caldo e, por inferência, produção de açúcar provável (em kg/t de cana). Dadas as condições do meio ambiente e das próprias plantas, pode-se dizer que estas não sofreram alterações dignas de nota nas sete amostras quinzenais efetuadas após a geada, nas características analisadas. Aplicando-se análise de regressão, verificou-se que os dados de brix % caldo ajustaram-se melhor às curvas de 2º grau. O teste F para regressão foi significativo em nove dos quinze casos estudados.A freeze occurred in the State of São Paulo, on July, 18th, 1975 with the temperature decreasing to -1ºC in Piracicaba. Advantage was taken of this occurrence to study the effect of freeze on the technological characteristics of some commercial sugar cane varieties. Ten stalk samples were taken for the experiment, beginning at twelve days after the freeze and thereafter every two weeks. Brix % juice, pol % juice and, by inference, theoretical sugar yield (kg/t of cane) were studied. Due to the environmental and plant conditions in the field, it could be concluded that the varieties did not show significant alterations caused by the freeze in none of the seven fortnight samples. Regression analysis showed that brix % juice data fitted 2nd degree curves better than the other data, with F test significant in nine out of fifteen cases

    Purpose: Coronavirus disease 19 (COVID-19) has, to date, been diagnosed in over 130 million persons worldwide and is caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Several variants of concern have emerged including those in the United Kingdom, South Africa, and Brazil. SARS-CoV-2 can cause a dysregulated inflammatory response known as a cytokine storm, which can progress rapidly to acute respiratory distress syndrome (ARDS), multi-organ failure, and death. Suppressing these cytokine elevations may be key to improving outcomes. Remote ischemic conditioning (RIC) is a simple, non-invasive procedure whereby a blood pressure cuff is inflated and deflated on the upper arm for several cycles. “RIC in COVID-19” is a pilot, multi-center, randomized clinical trial, designed to ascertain whether RIC suppresses inflammatory cytokine production. Methods: A minimum of 55 adult patients with diagnosed COVID-19, but not of critical status, will be enrolled from centers in the United Kingdom, Brazil, and South Africa. RIC will be administered daily for up to 15 days. The primary outcome is the level of inflammatory cytokines that are involved in the cytokine storm that can occur following SARS-CoV-2 infection. The secondary endpoint is the time between admission and until intensive care admission or death. The in vitro cytotoxicity of patient blood will also be assessed using primary human cardiac endothelial cells. Conclusions: The results of this pilot study will provide initial evidence on the ability of RIC to suppress the production of inflammatory cytokines in the setting of COVID-19.     Connective tissue anomalies in patients with spontaneous cervical artery dissection.

    OBJECTIVE: To investigate the prevalence of connective tissue abnormalities in patients with spontaneous cervical artery dissections (sCeAD). METHODS: We systematically assessed clinically detectable signs of connective tissue aberration in a series of consecutive patients with sCeAD and of age- and sex-matched patients with ischemic stroke unrelated to CeAD (non-CeAD IS) by a standard examination protocol including 68 items, and performed extensive molecular investigation for hereditary connective tissue disorders in all patients with sCeAD. RESULTS: The study group included 84 patients with sCeAD (mean age, 44.5 ± 7.8 years; 66.7% men) and 84 patients with non-CeAD IS. None of the patients with sCeAD met clinical or molecular diagnostic criteria for established hereditary connective tissue disorder. Connective tissue abnormalities were detected more frequently in the group of patients with sCeAD than in the group of those with non-CeAD IS (mean number of pathologic findings, 4.5 ± 3.5 vs 1.9 ± 2.3; p < 0.001). Eighty-one patients (96.4%) in the sCeAD group had at least one detectable sign compared with 55 patients (66.7%) in the group with non-CeAD IS (p < 0.001). Skeletal, ocular, and skin abnormalities, as well as craniofacial dysmorphisms, were the clinical signs more strongly associated with sCeAD. Signs suggesting connective tissue abnormality were also more frequently represented in patients with sCeAD than in patients with traumatic CeAD (28.6%, p < 0.001; mean number of pathologic findings, 1.7 ± 3.7, p = 0.045). CONCLUSIONS: Connective tissue abnormalities are frequent in patients with sCeAD. This reinforces the hypothesis that systemic aberrations of the connective tissue might be implicated in the pathogenesis of the disease

    Association of pretreatment hippocampal volume with neurocognitive function in patients treated with hippocampal avoidance whole brain radiation therapy for brain metastases: Secondary analysis of NRG Oncology/RTOG 0933

    PURPOSE: Hippocampal volume (HV) is an established predicting factor for neurocognitive function (NCF) in neurodegenerative disease. Whether the same phenomenon exists with hippocampal-avoidant whole brain radiation therapy is not known; therefore, we assessed the association of baseline HV with NCF among patients enrolled on RTOG 0933. METHODS AND MATERIALS: Hippocampal volume and total brain volume were calculated from the radiation therapy plan. Hippocampal volume was correlated with baseline and 4-month NCF scores (Hopkins Verbal Learning Test-Revised [HVLT-R] Total Recall [TR], Immediate Recognition, and Delayed Recall [DR]) using Pearson correlation. Deterioration in NCF was defined per the primary endpoint of RTOG 0933(mean 4-month relative decline in HVLT-R DR). Comparisons between patients with deteriorated and nondeteriorated NCF were made using the Wilcoxon test. RESULTS: Forty-two patients were evaluable. The median age was 56.5 years (range, 28-83 years), and 81% had a class II recursive partitioning analysis. The median total, right, and left HVs were 5.4 cm CONCLUSIONS: Larger HV was positively associated with improved performance on baseline and 4-month HVLT-R TR and DR scores in patients with brain metastases undergoing hippocampal-avoidant whole brain radiation therapy but was not associated with a change in NCF
