Exome sequencing from nanogram amounts of starting DNA: comparing three approaches
Hybridization-based target enrichment protocols require relatively large starting amounts of genomic DNA, which is not always available. Here, we tested three approaches to pre-capture library preparation starting from 10 ng of genomic DNA: (i and ii) whole-genome amplification of DNA samples with REPLI-g (Qiagen) and GenomePlex (Sigma) kits followed by standard library preparation, and (iii) library construction with a low input oriented ThruPLEX kit (Rubicon Genomics). Exome capture with Agilent SureSelectXT2 Human AllExon v4+UTRs capture probes, and HiSeq2000 sequencing were performed for test libraries along with the control library prepared from 1 µg of starting DNA. Tested protocols were characterized in terms of mapping efficiency, enrichment ratio, coverage of the target region, and reliability of SNP genotyping. REPLI-g- and ThruPLEX-FD-based protocols seem to be adequate solutions for exome sequencing of low input samples
High power diode laser surface glazing of concrete
This present work describes the utilisation of the relatively novel high power diode laser
(HPDL) to generate a surface glaze on the ordinary Portland cement (OPC) surface of
concrete. The value of such an investigation would be to facilitate the hitherto impossible
task of generating a durable and long-lasting surface seal on the concrete, thereby extending
the life and applications base of the concrete. The basic process phenomena are investigated
and the laser effects in terms of glaze morphology, composition and microstructure
are presented. Also, the resultant heat affects are analysed and described, as well as
the effects of the shield gases, O2 and Ar, during laser processing. HPDL glazing of OPC
was successfully demonstrated with power densities as low as 750 W cm-2 and at scanning
rates up to 480 mm min-1. The work showed that the generation of the surface glaze resulted
in improved mechanical and chemical properties over the untreated OPC surface of concrete.
Both untreated and HPDL glazed OPC were tested for pull-off strength, rupture strength,
water absorption, wear resistance and corrosion resistance. The OPC laser glaze exhibited
clear improvements in wear, water sorptivity, and resistance (up to 80% concentration) to
nitric acid, sodium hydroxide and detergent. Life assessment testing revealed that the OPC
laser glaze had an increase in actual wear life of 1.3 to 14.8 times over the untreated OPC
surface of concrete, depending upon the corrosive environment
Integrated modeling of food, energy and water security management for sustainable social, economic and environmental developments
This activity aims to investigation of coordinated robust sustainable developments derived accounting for complex linkages and differences in spatial and temporal scales between agriculture, energy and water security, potential systemic risks and new coherent feasible policies at a country level taking into account interregional and international trades, global pollution, related risks, insurance markets, direct and indirect exchange of resources. In contrast to numerous existing isolated studies using different research principles and data treatments from diverse regions at different time intervals, this project focuses on new integrated multidisciplinary approaches by a research team from six non-profit institutes of the National Academy of Sciences, Ukraine, working in collaboration with International Institute of Systems Analysis.
The research develops a harmonized strategic approach for integrated modeling secure sustainable supply of food, energy and water under natural and human-related intentional and unintentional threats
The HypHI project: Hypernuclear spectroscopy with stable heavy ion beams and rare isotope beams at GSI and FAIR
The HypHI collaboration aims to perform a precise hypernuclear spectroscopy
with stable heavy ion beams and rare isotope beams at GSI and fAIR in order to
study hypernuclei at extreme isospin, especially neutron rich hypernuclei to
look insight hyperon-nucleon interactions in the neutron rich medium, and
hypernuclear magnetic moments to investigate baryon properties in the nuclei.
We are currently preparing for the first experiment with Li and C
beams at 2 AGeV to demonstrate the feasibility of a precise hypernuclear
spectroscopy by identifying H, H and
He. The first physics experiment on these hypernuclei is
planned for 2009. In the present document, an overview of the HypHI project and
the details of this first experiment will be discussed.Comment: 5 pages, 2 figures, French-Japanese symposium 2008, Paris (France
Synthesis of Layered Organic-Inorganic Nanocomposites of Zinc and Copper by Laser Ablation in Liquid
The experimental data of studies of layered organic-inorganic nanocomposites (()-Zn(OH)2+DS) and (Cu2(OH)3+DS) which were produced in the result ablation of zinc and copper in aqueous solutions of surfactants—dodecyl sodium sulfate (SDS) and sodium bis-ethylhexyl succinate (AOT)—are presented. Dependence of the formation dynamics of these composites on an exposure time of radiation and on an aging time of colloids was studied by the absorption spectroscopy, by X-ray diffraction, by scanning electron (SEM), and atomic force microscopy (AFM). Composite (Cu2(OH)3+DS) with bilayered structure was produced by method of laser ablation of the copper metal target in liquid for the first time
High precision measurement of the associated strangeness production in proton proton interactions
A new high precision measurement of the reaction pp -> pK+Lambda at a beam
momentum of 2.95 GeV/c with more than 200,000 analyzed events allows a detailed
analysis of differential observables and their inter-dependencies. Correlations
of the angular distributions with momenta are examined. The invariant mass
distributions are compared for different regions in the Dalitz plots. The cusp
structure at the N Sigma threshold is described with the Flatt\'e formalism and
its variation in the Dalitz plot is analyzed.Comment: accepted for publication in Eur. Phys. J.
First Model-Independent Measurement of the Spin Triplet Scattering Length from Final State Interaction in the Reaction
The reaction has been measured with the
COSY-TOF detector at a beam momentum of . The polarized
proton beam enables the measurement of the beam analyzing power by the
asymmetry of the produced kaon (). This observable allows the
spin triplet scattering length to be extracted for the first time
model independently from the final-state interaction in the reaction. The
obtained value is . This value is
compatible with theoretical predictions and results from model-dependent
analyses.Comment: Revised version as accepted for publication in PR
