Deamidation at Asparagine and Glutamine As a Major Modification upon Deterioration/Aging of Proteinaceous Binders in MuralPaintings
Proteomic strategies are herein proved to be a
complementary approach to the well established amino acid
composition analysis for the characterization of the aging and
deterioration phenomena occurring to proteinaceous materials
in works-of-art. Amino acid analyses on several samples demonstrated
that proteins in the frescoes from the Camposanto
Monumentale in Pisa are deteriorated as revealed by the
decrease in Met, Lys, and Tyr content and by the presence in
all the samples of amino malonic acid as a result of Ser, Phe, and
Cys oxidation. Proteomic analysis identified deamidation at Asn
and Gln as a further major event occurred. This work paves the
way to the exploitation of proteomic strategies for the investigation
of the molecular effects of aging and deterioration in
historical objects. Results show that proteomic searches for
deamidation by liquid chromatography-tandem mass spectrometry
(LC-MS/MS) could constitute a routine analysis for paintings or any artistic and historic objects where proteins are present.

Neuro-hormonal effects of physical activity in the elderly.
Thanks to diagnostic and therapeutic advances, the elderly population is continuously increasing in the western countries. Accordingly, the prevalence of most chronic age-related diseases will increase considerably in the next decades, thus it will be necessary to implement effective preventive measures to face this epidemiological challenge. Among those, physical activity exerts a crucial role, since it has been proven to reduce the risk of cardiovascular diseases, diabetes, obesity, cognitive impairment and cancer. The favorable effects of exercise on cardiovascular homeostasis can be at least in part ascribed to the modulation of the neuro-hormonal systems implicated in cardiovascular pathophysiology. In the elderly, exercise has been shown to affect catecholamine secretion and biosynthesis, to positively modulate the renin-angiotensin-aldosterone system and to reduce the levels of plasma brain natriuretic peptides. Moreover, drugs modulating the neuro-hormonal systems may favorably affect physical capacity in the elderly. THE CAPO ROSSELLO BORE-HOLE (AGRIGENTO, SICILY) CYCLOSTRATIGRAPHIC AND PALEOCEANOGRAPHIC RECONSTRUCTIONS FROM QUANTITATIVE ANALYSES OF THE ZANCLEAN FORAMINIFERAL ASSEMBLAGES
Cyclostratigraphy based on the quantitative analyses of planktonic foraminifera in the Zanclean segment (M Pl 1 base of M Pl 3 biozones) of the bore-hole drilled in the Capo Rossello area (Sicily, Southern Italy) allows us to recognize that the calcareous plankton biostratigraphic events occur in the same cycles as identified by previous authors. Exceptional thick cycles which are thought to represent two precession cycles show two fluctuations is greater than the number of lithologic cycles. Between the base of the Pliocene and the first occurrence of Globorotalia puncticulata thirty-five lithologic cycles and thirty eight planktonic foraminifera fluctuations are present. By comparison with relative abundance fluctuations in the benthic foraminifera and the astronomical record is recognized. In particular, the BN>125 µm and the Planulina ariminensis quantitative distributions are related to the long eccentricity. Conversely, the relative abundance fluctuations of Siphonina reticulata, Cibicidoides bradyi, Pullenia bulloides and Uvigerina pygmaea appear to be generally forced by the precession periodicity, but in the intervals coincident with the eccentricity minima such a relationship is less clear. The very abundant and widespread distribution in several Mediterranean sites of S. reticulata, which we consider a quasi-endemic species of this basin during the Early and Middle Pliocene is anticovariant with the quantitative distribution of Cibicidoides brady-robertsonianus, which is considered a characteristic species of the North Atlantic Deep Water (NADW). This relationship suggest that a peculiar, intermediate water mass was present in the Mediterranean during this time interval. We propose for this water mass the name of Early Pliocene Mediterranenan Intermediate Water (EPMIW). According to bibliographic data that report the sharp decrease in abundance of S. reticulata during the M Pl 5 biozone, EPMIW was present in the Mediterranean basin until about 2.5 Ma.

Validation of "(fr)AGILE": A quick tool to identify multidimensional frailty in the elderly
Background Several tools have been proposed and validated to operationally define frailty. Recently, the Italian Frailty index (IFi), an Italian modified version of Frailty index, has been validated but its use in clinical practice is limited by long time of administration. Therefore, the aim of this study was to create and validate a quick version of the IFi (AGILE). Methods Validation study was performed by administering IFi and AGILE, after a Comprehensive Geriatric Assessment (CGA) in 401 subjects aged 65 or over (77 +/- 7 years). AGILE was a 10-items tool created starting from the more predictive items of the four domains of frailty investigated by IFi (mental, physical, socioeconomic and nutritional). AGILE scores were stratified in light, moderate and severe frailty. At 24 months of follow-up, death, disability (taking into account an increase in ADL lost >= 1 from the baseline) and hospitalization were considered. Area under curve (AUC) was evaluated for both IFi and AGILE. Results Administration time was 9.5 +/- 3.8 min for IFi administered after a CGA, and 2.4 +/- 1.2 min for AGILE, regardless of CGA (p < 0.001). With increasing degree of frailty, prevalence of mortality increased progressively from 6.5 to 41.8% and from 9.0 to 33.3%, disability from 16.1 to 64.2% and from 22.1 to 59.8% and hospitalization from 17.2 to 58.7% and from 27.0 to 52.2% with AGILE and IFi, respectively (p = NS). Relative Risk for each unit of increase in AGILE was 56, 44 and 24% for mortality, disability and hospitalization, respectively and was lower for IFi (8, 7 and 4% for mortality, disability and hospitalization, respectively). The AUC was higher in AGILE vs. IFi for mortality (0.729 vs. 0.698), disability (0.715 vs. 0.682) and hospitalization (0.645 vs. 0.630). Conclusions Our study shows that AGILE is a rapid and effective tool for screening multidimensional frailty, able to predict mortality, disability and hospitalization, especially useful in care settings that require reliable assessment instruments with short administration time
Contribution of future wide-swath altimetry missions to ocean analysis and forecasting
The impact of forthcoming wide-swath altimetry missions on the ocean analysis
and forecasting system was investigated by means of OSSEs (observing system
simulation experiments). These experiments were performed with a regional data assimilation system,
implemented in the Iberian–Biscay–Ireland (IBI) region, at 1∕12°
resolution using simulated observations derived from a fully eddy-resolving
free simulation at 1∕36° resolution over the same region. The
objective of the experiments was to assess the ability of different satellite
constellations to constrain the ocean analyses and forecasts, considering
both along-track altimeters and future wide-swath missions; consequently,
the capability of the data assimilation techniques used in the Mercator Ocean
operational system to effectively combine the different kinds of
measurements was also investigated. These assessments were carried out as part of a European Space Agency (ESA)
study on the potential role of wide-swath altimetry in future versions of the
European Union Copernicus programme. The impact of future wide-swath
altimetry data is evident for investigating the reliability of sea level values in
OSSEs. The most significant results were obtained when looking at the sensitivity
of the system to wide-swath instrumental error: considering a constellation
of three nadir and two accurate (small instrumental error) wide-swath
altimeters, the error in ocean analysis was reduced by up to 50 %
compared to conventional altimeters. Investigating the impact of the
repetitivity of the future measurements, the results showed that two
wide-swath missions had a major impact on sea-level forecasting – increasing
the accuracy over the entire time window of the 5-day forecasts – compared
with a single wide-swath instrument. A spectral analysis underlined that the
contributions of wide-swath altimetry data observed in ocean analyses and
forecast statistics were mainly due to the more accurate resolution,
compared with along-track data, of ocean variability at spatial scales
smaller than 100 km. Considering the ocean currents, the results confirmed
that the information provided by wide-swath measurements at the surface is
propagated down the water column and has a considerable impact (30 %) on
ocean currents (up to a depth of 300 m), compared with the present
constellation of altimeters. The ocean analysis and forecasting systems used
here are those currently used by the Copernicus Marine Environment and
Monitoring Service (CMEMS) to provide operational services and ocean
reanalysis. The results obtained in the OSSEs considering along-track
altimeters were consistent with those derived from real data (observing
system experiments, OSEs). OSSEs can also be used to assess the potential of
new observing systems, and in this study the results showed that future
constellations of altimeters will have a major impact on constraining the
CMEMS ocean analysis and forecasting systems and their applications.</p
Effects of losartan treatment on cardiac autonomic control during volume loading in patients with DCM
This study evaluated the effect of angiotensin II receptor blockade on cardiac autonomic control adaptation and urine output in response to acute isotonic volume load in patients with idiopathic dilated cardiomyopathy (DCM) and asymptomatic to mildly symptomatic heart failure. Left ventricular volumes and heart rate variability measurements were assessed at baseline and during intravenous saline load in 14 patients before and after 2 mo of losartan treatment. After losartan treatment, blood pressure values were lower, whereas left ventricular ejection fraction was higher (F = 79, P50 ms) decreased during saline load in untreated patients (F = 3.1, P< 0.05 and F = 6.5, P< 0.01, respectively), but not after losartan. Similarly, a decrease in very low frequency (F = 3.2, P< 0.05), low-frequency (F = 2.9, P< 0.05), and high-frequency power (F = 6.1, P< 0.01) after saline load was observed only in untreated patients. In patients with DCM, losartan treatment improves the cardiac autonomic adaptation and increases urine output in response to volume overload
Shape oscillations of an oil drop rising in water: effect of surface contamination
Inertial shape oscillations of heptane drops rising in water are investigated experimentally. Diameters from 0.59 to 3.52 mm are considered, corresponding to a regime where the rising motion should not affect shape oscillations for pure immiscible fluids. The interface, however, turns out to be contaminated. The drag coefficient is considerably increased compared to that of a clean drop due to the well-known. Marangoni effect resulting from a gradient of surfactant concentration generated by the fluid motion along the interface. Thanks to the decomposition of the shape into spherical harmonics, the eigenfrequencies and the damping rates of oscillation modes n = 2, 3, 4 and 5 have been measured. Frequencies are not affected by contamination, while damping rates are increased by a considerable amount that depends neither on drop instantaneous velocity nor on diameter. This augmentation, however, depends on the mode number: it is maximum for mode two (multiplied by 2.4) and then relaxes towards the value of a clean drop as n increases. A previous similar investigation of a drop attached to a capillary has not revealed such an increase of the damping rates, indicating that the coupling between rising motion and surface contamination is responsible for this effect
Elaia, Pergamon's maritime satellite:The rise and fall of an ancient harbour city shaped by shoreline migration
Throughout human history, communication and trade have been key to society. Because maritime trade facilitated the rapid transportation of passengers and freight at relatively low cost, harbours became hubs for traffic, trade and exchange. This general statement holds true for the Pergamenian kingdom, which ruled wide parts of today's western Turkey during Hellenistic times. Its harbour, located at the city of Elaia on the eastern Aegean shore, was used extensively for commercial and military purposes. This study reconstructs the coastal evolution in and around the ancient harbour of Elaia and compares the observed environmental modifications with archaeological and historical findings. We use micropalaeontological, sedimentological and geochemical proxies to reconstruct the palaeoenvironmental dynamics and evolution of the ancient harbour. The geoarchaeological results confirm the archaeological and historical evidence for Elaia's primacy during Hellenistic and early Roman times, and the city's gradual decline during the late Roman period. Furthermore, our study demonstrates that Elaia holds a unique position as a harbour city during ancient times in the eastern Aegean region, because it was not greatly influenced by the high sediment supply associated with river deltas. Consequently, no dredging of the harbour basins is documented, creating exceptional geo-bioarchives for palaeoenvironmental reconstructions
