Protocolo de coleta e preparação de amostras de macroinvertebrados bentônicos em riachos.
O biomonitoramento de corpos hÃdricos através do uso de macroinvertebrados bentônicos é cada vez mais usado e aceito como uma importante ferramenta na avaliação da qualidade da água. Embora seja uma ferramenta utilizada desde o inÃcio do século XX na Europa e na América do Norte, no Brasil esta técnica tem apenas algumas décadas e não existem publicações de protocolos especÃficos para a coleta de macroinvertebrados em rios tropicais. Na América do Norte, em particular, a publicação de Rosenberg & Resh (1993) constitui uma das principais obras sobre o tema, sendo uma boa fonte de consulta para os iniciantes no estudo do biomonitoramento da qualidade das águas. Por ser uma metodologia de baixo custo e de aparato técnico simples, é interessante que se estabeleça um protocolo padrão para utilização em riachos nos paÃses em desenvolvimento, a fim de que estudos futuros possam ser comparados, desde que desenvolvidos em áreas de clima e geografia semelhantes. Assessment of immune function and prediction of survival and infection in patients with severe alcoholic hepatitis: An exploratory study
Background and Aim: Alcoholic hepatitis (AH), a severe complication of long-term alcohol misuse, has a 30% 90-day mortality. Infections are common and associated with higher mortality. There is currently no accurate method to predict infection in these patients. We aimed to test a measure of immune function, the QuantiFERON Monitor (QFM), in predicting clinical outcomes in patients with severe AH. Methods: Peripheral blood was taken at baseline, and QFM performed according to the manufacturer's instructions. In parallel, QFM samples were analyzed with a cytokine multiplex. Clinical outcomes of mortality at 28 and 90 days and development of infection were recorded prospectively. Results: Forty-nine patients were recruited (mean age 51, 59% male and mean discriminant function 57.8). Interferon (IFN)-γ release measured by standard QFM was significantly higher in survivors compared to non-survivors at 28 (102 vs 16 IU/mL, P = 0.02) and 90 days (115 vs 32 IU/mL; P = 0.046). The area under the receiver operating characteristic curve (AUROC) was 0.79 for 28-day mortality. IFN-γ, IL-10, and IL-23 release measured by multiplex were significantly lower in patients who developed a subsequent infection compared to those who did not (115 vs 27 IU/mL, P = 0.037; 457 vs 202 pg/mL, P = 0.008; and 1039 vs 663 pg/mL, p = 0.01, respectively). Conclusion: Immune dysfunction is associated with poorer outcomes in patients with severe AH. Measurement of IFN-γ release by standard QFM accurately predicts early mortality, which can be applied to clinical practice as a biomarker of survival. Adaptation of the test to measure IL-10 could be used as a biomarker of subsequent infection to guide clinical treatment decisions
Comparação de métodos para determinação de cálcio, magnésio e potássio em resÃduos agropecuários.
O uso de resÃduos orgânicos na agricultura contribui para a reciclagem da energia (matéria orgânica) e dos nutrientes neles contidos, ao mesmo tempo em que previne seu descarte inadequado que poderia resultar em contaminação ambiental. Diversos desses resÃduos contêm teores expressivos de nutrientes, e podem, portanto, substituir parcial ou totalmente as adubações minerais. No entanto, o conhecimento das caracterÃsticas quÃmicas, fÃsicas e biológicas de resÃduos potencialmente úteis como fonte de nutrientes é limitado pela falta de métodos de análise especÃficos, padronizados e amplamente aceitos. Independentemente de sua natureza, a análise quÃmica de resÃduos agropecuários é efetuada conforme métodos usados na análise de tecidos vegetais e também de materiais inorgânicos. Neste trabalho, avaliou-se a eficiência de decomposição de três misturas de digestão via úmida: ácido sulfúrico + água oxigenada, ácido nÃtrico + ácido perclórico e ácido nÃtrico + ácido clorÃdrico (água régia).Concomitantemente, foi avaliada a precisão dos resultados obtidos, em função do uso de três diferentes massas de amostras no processo de digestão: 0,050, 0,100 e 0,200 g. Foram utilizados cinco resÃduos, três deles in natura (bagaço de cana, esterco caprino verde, esterco caprino curtido) e dois transformados (um produto derivado de cama de frango e um de origem indefinida). As digestões foram efetuadas em sistema aberto, em blocos digestores. O método de digestão com a mistura [ácido sulfúrico + água oxigenada]utilizando-se massa de amostra de 0,200 g de resÃduos orgânicos apresentou maior taxa de recuperação de Ca, Mg e K do que os demais métodos
Acute Strenuous Exercise Induces an Imbalance on Histone H4 Acetylation/Histone Deacetylase 2 and Increases the Proinflammatory Profile of PBMC of Obese Individuals
This study evaluated the response of global histone H4 acetylation (H4ac), histone deacetylase 2 (HDAC2) activity, as well as the production of proinflammatory cytokines and monocyte phenotypes of lean and obese males after exercise. Ten lean and ten obese sedentary men were submitted to one session of strenuous exercise, and peripheral blood mononuclear cells (PBMC) were stimulated in vitro with lipopolysaccharide (LPS). Global H4ac levels, HDAC2 activity in PBMC, and IL-6, IL-8, and TNF-α production were analyzed. Monocyte phenotype was determined in accordance with the expression of CD14 and CD16. At rest, obese individuals presented higher frequency of proinflammatory CD14+CD16+ monocytes. LPS induced a significant augment in global H4ac and in the production of IL-6, IL-8, and TNF-α mainly in obese individuals. After exercise, the increased production of IL-8 and TNF-α and peripheral frequency of CD14+CD16+ were observed in both groups. In addition, exercise also induced a significant hyperacetylation of histone H4 and decreased HDAC2 activity in both nonstimulated and LPS-stimulated PBMC of obese individuals. Our data indicate that the obesity impacts on H4ac levels and that strenuous exercise leads to an enhanced chronic low-grade inflammation profile in obesity via an imbalance on H4ac/HDAC2
Potencial de utilização de resÃduos orgânicos da região nordeste como fertilizantes.
A utilização racional de resÃduos orgânicos como fertilizantes na agricultura depende do conhecimento da composição quÃmica desses materiais. Na fruticultura comercial, especialmente naquela voltada à exportação, essa informação assume grande importância pelo fato da adubação orgânica representar parte dos custos de produção, os quais devem ser otimizados para permitir maior competitividade no mercado. Um dos principais entraves à intensificação do uso dos resÃduos orgânicos na agricultura reside na falta de métodos de análise especÃficos, padronizados e amplamente aceitos, sendo em geral utilizados os métodos comuns à análise foliar para sua caracterização quÃmica. Krug (1996) indica métodos de digestão usados na análise de tecidos vegetais que podem ser aplicados na decomposição de resÃduos orgânicos, embora a composição quÃmica de muitos desses materiais possa diferir marcadamente da composição foliar. Outro fator que deve ser levado em conta na análise quÃmica desses resÃduos é a massa de amostra usada na digestão, pois o grau de heterogeneidade de muitos deles não pode ser compensado na fase de pré-tratamento (principalmente nas etapas mais crÃticas, de trituração e peneiramento). Tal heterogeneidade das amostras pode influenciar simultaneamente a exatidão e a precisão dos resultados. Neste trabalho, teve-se como objetivo avaliar métodos de digestão utilizando-se as misturas [ácido sulfúrico + água oxigenada], [ácido nÃtrico + ácido perclórico] e água régia como extratores, avaliando-se também diferentes massas de amostra. Utilizaram-se amostras de resÃduos orgânicos que podem ser usados como fertilizantes na fruticultura irrigada praticada na Região Nordeste do Brasil, visando-se obter um procedimento de fácil aplicação em laboratórios de análise de solos e de plantas, disponibilizando-se dados para as tabelas de adubação de culturas frutÃcolas, pela avaliação de teores de alguns nutrientes essenciais

Contribution of ocean physics and dynamics at different scales to heat uptake in low-resolution AOGCMs
Using an ensemble of atmosphere-ocean general circulation models (AOGCMs) in an idealized climate change experiment, this study quantifies the contributions to ocean heat uptake (OHU) from ocean physical parameterizations and resolved dynamical processes operating at different scales. Analysis of heat budget diagnostics reveals a leading-order global heat balance in the sub-surface upper ocean in a steady state between the large-scale circulation warming it and mesoscale processes cooling it, and shows that there are positive contributions from processes on all scales to the subsurface OHU during climate change. There is better agreement among the AOGCMs in the net OHU than in the individual scales/processes contributing to it. In the upper ocean and at high latitudes, OHU is dominated by small-scale diapycnal processes. Below 400 m, OHU is dominated by the super-residual transport, representing large-scale ocean dynamics combined with all parameterized mesoscale and submesoscale eddy effects. Weakening of the AMOC leads to less heat convergence in the subpolar North Atlantic and less heat divergence at lower latitudes, with a small overall effect on the net Atlantic heat content. At low latitudes, the dominance of advective heat redistribution is contrary to the diffusive OHU mechanism assumed by the commonly used upwelling-diffusion model. Using a density watermass framework, it is found that most of the OHU occurs along isopycnal directions. This feature of OHU is used to accurately reconstruct the global vertical ocean warming profile from the surface heat flux anomalies, supporting advective (rather than diffusive) models of OHU and sea-level rise
Exploring the diverse definitions of ‘evidence’: a scoping review
OBJECTIVES:
To systematically collect and analyse diverse definitions of ‘evidence’ in both health and social sciences, and help users to correctly use the term ‘evidence’ and rethink what is the definition of ‘evidence’ in scientific research.
DESIGN:
Scoping review.
METHODS:
Definitions of evidence in the health sciences and social sciences were included. We have excluded the definition of evidence applied in the legal field, abstracts without full text, documents not published in either Chinese or English and so on. We established a multidisciplinary working group and systematically searched five electronic databases including Medline, Web of Science, EBSCO, the Chinese Social Sciences Citation Index and the Chinese Science Citation Database from their inception to 26 February 2022. We also searched websites and reviewed the reference lists of the identified studies. Six reviewers working in pairs, independently, selected studies according to the inclusion and exclusion criteria, and extracted information. Any differences were discussed in pairs, and if there was disagreement, it was resolved via discussion or with the help of a third reviewer. Reviewers extracted document characteristics, the original content for the definitions of ‘evidence’, assessed definitions as either intensional or extensional, and any citations for the given definition.
RESULTS:
Forty-nine documents were finally included after screening, and 68 definitions were obtained. After excluding duplicates, a total of 54 different definitions of ‘evidence’ were identified. There were 42 intensional definitions and 12 extensional definitions. The top three definiens were ‘information’, ‘fact’ and ‘research/study’. The definition of ‘evidence’ differed between health and social sciences. The term ‘research’ appeared most frequently in the definitions.
CONCLUSIONS:
The definition of ‘evidence’ has gradually attracted the attention of many scholars and decision-makers in health and social sciences. Nevertheless, there is no widely recognised and accepted definition in scientific research. Given the wide use of the term, we need to think about whether, or under what circumstances, a standardised, clear, meaningful and widely applicable definition of ‘evidence’ might be helpful
