    Integrated Bioinformatic Analysis of a Competing Endogenous RNA Network Reveals a Prognostic Signature in Endometrial Cancer

    In endometrial carcinoma, the clinical outcome directly correlates with the TNM stage, but the lack of sufficient information prevents accurate prediction. The molecular mechanism underlying the competing endogenous RNA (ceRNA) hypothesis has not been investigated in endometrial cancer. Multi-bioinformatic analyses, including differentially expressed gene analysis, ceRNA network construction, Cox regression analysis, function enrichment analysis, and protein-protein network analysis, were performed on the sequence data acquired from The Cancer Genome Atlas (TCGA) data bank. A ceRNA network comprising 366 mRNAs, 27 microRNAs (miRNAs), and 66 long non-coding RNAs (lncRNAs) was established. Survival analysis performed with the univariate Cox regression analysis revealed nine lncRNAs with prognostic power in endometrial carcinoma. In multivariate Cox regression analysis, a signature comprising LINC00491, LINC00483, ADARB2-AS1, and C8orf49 showed remarkable prognostic power. Risk score and neoplasm status, but not TNM stage, were independent prognostic factors of endometrial carcinoma. A ceRNA network comprising differentially expressed mRNAs, miRNAs, and lncRNAs may reveal the molecular events involved in the progression of endometrial carcinoma. In addition, the signature with prognostic value may discriminate patients with increased risk for poor outcome, which may allow physicians to take accurate decisions

    Evolved Massive Stars at Low-metallicity V. Mass-Loss Rate of Red Supergiant Stars in the Small Magellanic Cloud

    We assemble the most complete and clean red supergiant (RSG) sample (2,121 targets) so far in the Small Magellanic Cloud (SMC) with 53 different bands of data to study the MLR of RSGs. In order to match the observed spectral energy distributions (SEDs), a theoretical grid of 17,820 Oxygen-rich models (``normal'' and ``dusty'' grids are half-and-half) is created by the radiatively-driven wind model of the DUSTY code, covering a wide range of dust parameters. We select the best model for each target by calculating the minimal modified chi-square and visual inspection. The resulting MLRs from DUSTY are converted to real MLRs based on the scaling relation, for which a total MLR of 6.16×1036.16\times10^{-3} MM_\odot yr1^{-1} is measured (corresponding to a dust-production rate of 6×106\sim6\times10^{-6} MM_\odot yr1^{-1}), with a typical MLR of 106\sim10^{-6} MM_\odot yr1^{-1} for the general population of the RSGs. The complexity of mass-loss estimation based on the SED is fully discussed for the first time, indicating large uncertainties based on the photometric data (potentially up to one order of magnitude or more). The Hertzsprung-Russell and luminosity versus median absolute deviation diagrams of the sample indicate the positive relation between luminosity and MLR. Meanwhile, the luminosity versus MLR diagrams show a ``knee-like'' shape with enhanced mass-loss occurring above log10(L/L)4.6\log_{10}(L/L_\odot)\approx4.6, which may be due to the degeneracy of luminosity, pulsation, low surface gravity, convection, and other factors. We derive our MLR relation by using a third-order polynomial to fit the sample and compare our result with previous empirical MLR prescriptions. Given that our MLR prescription is based on a much larger sample than previous determinations, it provides a more accurate relation at the cool and luminous region of the H-R diagram at low-metallicity compared to previous studies.Comment: 16 pages, 19 figures, accepted by A&

    Gaussian quantum steering under the influence of a dilaton black hole

    We study the dynamics of Gaussian quantum steering in the background of a Garfinkle–Horowitz–Strominger dilaton black hole. It is found that the gravity induced by dilaton field will destroy the quantum steerability between the inertial observer Alice and the observer Bob who hovers outside the event horizon, while it generates steering-type quantum correlations between the causally disconnected regions. Therefore, the observers can steer each other’s state by local measurements even though they are separated by the event horizon. Unlike quantum entanglement in the dilaton spacetime, the quantum steering experiences catastrophic behaviors such as “sudden death” and “sudden birth” with increasing dilaton charge. In addition, the dilaton gravity destroys the symmetry of Gaussian steering and the latter is always asymmetric in the dilation spacetime. Interestingly, the attainment of maximal steering asymmetry indicates the critical point between one-way and two-way steering for the two-mode Gaussian state in the dilaton spacetime

    The integration of advanced sensor technologies has significantly propelled the dynamic development of robotics, thus inaugurating a new era in automation and artificial intelligence. Given the rapid advancements in robotics technology, its core area—robot control technology—has attracted increasing attention. Notably, sensors and sensor fusion technologies, which are considered essential for enhancing robot control technologies, have been widely and successfully applied in the field of robotics. Therefore, the integration of sensors and sensor fusion techniques with robot control technologies, which enables adaptation to various tasks in new situations, is emerging as a promising approach. This review seeks to delineate how sensors and sensor fusion technologies are combined with robot control technologies. It presents nine types of sensors used in robot control, discusses representative control methods, and summarizes their applications across various domains. Finally, this survey discusses existing challenges and potential future directions

    Characteristic of Clinical Studies on Baduanjin during 2000–2019: A Comprehensive Review

    To date, a growing number of clinical studies have demonstrated the safety and health benefits from Baduanjin intervention. Based on this, our objective is to systematically retrieve and summarize the clinical studies on Baduanjin, with a view to providing more evidence-based evidence in support of the application of Baduanjin for healthcare, and to identify the shortcomings of existing research and provide feasibility suggest for further clinical research. Both four English language and four Chinese language electronic databases were used to search articles related to Baduanjin during 2000–2019. SPSS 22.0 software was used to analyze the data, and the risk of bias tool in the RevMan 5.3.5 software was used to evaluate the methodological quality of randomized controlled trials. A total of 810 publications were identified, including 43 (5.3%) systematic reviews, 614 (75.8%) randomized controlled trials, 66 (8.1%) nonrandomized controlled clinical studies, 84 (10.4%) case series, and 3 (0.4%) case reports. The top 10 diseases/conditions included diabetes, chronic obstructive pulmonary disease, hypertension, low back pain, neck pain, stroke, coronary heart disease, cognitive impairment, insomnia, and osteoporosis or osteopenia. The style of State General Administration of Sport of China in 2003 was the most commonly used version of Baduanjin, and Baduanjin was practiced with an average of 35 minutes, 1 or 2 times a day, 3–5 days per week, and a 18-week average duration. It is also worth noting that there were no serious adverse events related to Baduanjin intervention. Most studies were small sample size research, and the methodological quality of randomized controlled trials is generally low. The clinical studies of Baduanjin have a substantial quantity and evidence base. However, there are significant differences among different studies in the specific intervention measures such as style, intensity, duration, learning, and practice methods, which need to be further standardized and unified. Further high-quality designed and reporting studies are recommended to further validate the clinical benefits of Baduanjin

    Prediction of Toxoplasma gondii virulence factor ROP18 competitive inhibitors by virtual screening

    Abstract Background Rhoptry protein 18 (ROP18) is a key virulence factor of Toxoplasma gondii. The host’s immune responses mediated by immune-related GTPases (IRGs) could be blocked by ROP18’s kinase activity. ROP18 also interacts with various substrates, such as activating transcription factor 6 beta (ATF6β) and affects multiple physiological functions within host cells, thereby inducing intense virulence. In this study, competitive inhibitors targeted to ROP18 were subjected to virtual screening based on the principle of structure-based drug design (SBDD). Methods The preparation of the ROP18 structure was conducted using the “Structure Prepare” function of Molecular Operating Environment (MOE) software. The ATP-binding pocket was selected as the starting point for virtual screening. Construction of the pharmacophore model used Extended Hückel Theory (EHT) half-quantitative measurement and construction, as well as the characteristics of Type I kinase inhibitors. The pharmacophore model of ROP18 was imported into the Specs database for small molecule similarity screening using EHT pharmacophore measurement. Hit compounds were selected using the functions of London dG and generalized-born volume integral/weighted surface area (GBVI/WSA) scoring. The top 100 hits were analyzed by molecular docking and structure activity relationships (SAR) analysis. Results The final pharmacophore comprised three typical characteristics: three hydrogen bond acceptors/donors, two ring aromatic features occupying the hydrophobic core, and one cation group feature targeted to the terminus of ATP. A total of 1314 hit compounds analogous to ROP18 pharmacophore were passed through the Specs. After two rounds of docking, 25 out of 100 hits were identified as belonging to two main scaffold types: phthalimide ring structure, thiazole ring and styrene structure. Additionally, the screen also identified 13 inhibitors with distinct scaffold types. The docking models and SAR analysis demonstrated that these hits could engage in multiple hydrogen bonds, salt bridges halogen bonds, and hydrophobic interactions with ROP18, and para-position halo substituents on the benzene ring may enhance their affinity scoring. Conclusions A pharmacophore against the ROP18 ATP-binding pocket was successfully constructed, and 25 representative inhibitors from 15 scaffold clusters were screened using the Specs database. Our results provide useful scaffold types for the chemical inhibition of ROP18 or alternative conformations to develop new anti-toxoplasmosis drug leads
