Trends in Integration of Knowledge and Large Language Models: A Survey and Taxonomy of Methods, Benchmarks, and Applications
Large language models (LLMs) exhibit superior performance on various natural
language tasks, but they are susceptible to issues stemming from outdated data
and domain-specific limitations. In order to address these challenges,
researchers have pursued two primary strategies, knowledge editing and
retrieval augmentation, to enhance LLMs by incorporating external information
from different aspects. Nevertheless, there is still a notable absence of a
comprehensive survey. In this paper, we propose a review to discuss the trends
in integration of knowledge and large language models, including taxonomy of
methods, benchmarks, and applications. In addition, we conduct an in-depth
analysis of different methods and point out potential research directions in
the future. We hope this survey offers the community quick access and a
comprehensive overview of this research area, with the intention of inspiring
future research endeavors.Comment: Work in progress; 22 pages. This work has been submitted to the IEEE

A Search for Double-peaked narrow emission line Galaxies and AGNs in the LAMOST DR1
LAMOST has released more than two million spectra, which provide the
opportunity to search for double-peaked narrow emission line (NEL) galaxies and
AGNs. The double-peaked narrow-line profiles can be well modeled by two
velocity components, respectively blueshifted and redshifted with respect to
the systemic recession velocity. This paper presents 20 double-peaked NEL
galaxies and AGNs found from LAMOST DR1 using a search method based on
multi-gaussian fit of the narrow emission lines. Among them, 10 have already
been published by other authors, either listed as genuine double-peaked NEL
objects or as asymmetric NEL objects, the remaining 10 being first discoveries.
We discuss some possible origins for double-peaked narrow-line features, as
interaction between jet and narrow line regions, interaction with companion
galaxies and black hole binaries. Spatially resolved optical imaging and/or
follow-up observations in other spectral bands are needed to further discuss
the physical mechanisms at work.Comment: 17 pages, 5figures, 4 tables, accepted by RA
In-situ tunable giant electrical anisotropy in a grating gated AlGaN/GaN two-dimensional electron gas
Materials with in-plane electrical anisotropy have great potential for
designing artificial synaptic devices. However, natural materials with strong
intrinsic in-plane electrical anisotropy are rare. We introduce a simple
strategy to produce extremely large electrical anisotropy via grating gating of
a semiconductor two-dimensional electron gas (2DEG) of AlGaN/GaN. We show that
periodically modulated electric potential in the 2DEG induces in-plane
electrical anisotropy, which is significantly enhanced in a magnetic field,
leading to an ultra large electrical anisotropy. This is induced by a giant
positive magnetoresistance and a giant negative magnetoresistance under two
orthogonally oriented in-plane current flows, respectively. This giant
electrical anisotropy is in-situ tunable by tailoring both the grating gate
voltage and the magnetic field. Our semiconductor device with controllable
giant electrical anisotropy will stimulate new device applications, such as
multi-terminal memtransistors and bionic synapses
Dynamic diffusion tensor imaging reveals structural changes in the bilateral pyramidal tracts after brain stem hemorrhage in rats
Background and Purpose: Few studies have concentrated on pyramidal tract (PY) changes after brain stem hemorrhage (BSH). In this study, we used a diffusion tensor imaging (DTI) technique and histologic identification to investigate longitudinal PY changes on both the contralateral and ipsilateral sides after experimental BSH. Methods: BSH was induced in 61 Sprague-Dawley rats by infusing 30 μl of autogenous tail blood into each rat’s right pons. DTI and motor function examinations were performed repeatedly on days 1, 3, 7, 14, and 28 after surgery. Fractional anisotropy, mean diffusivity, axial diffusivity, and radial diffusivity were measured in the bilateral PYs. The axon and myelin injury in the PY were evaluated by histologic study. Results: As compared with normal controls, the bilateral PYs in rats with induced BSH showed an early decrease and a late increase in fractional anisotropy and an early increase and a late decrease in mean diffusivity. A progressive decrease in axial diffusivity with dramatic axon loss from day 1 to day 28 after BSH was found bilaterally. The bilateral PYs showed an early increase and a late decrease in radial diffusivity. Early myelin injury and late repair were also detected pathologically in the bilateral PYs of rats with BSH. Thus, the early motor function deficits of rats with BSH began to improve on day 14 and had almost completely disappeared by day 28. Conclusions: DTI revealed dynamic changes in the bilateral PYs after BSH, which was confirmed by histologic findings and which correlated with motor function alteration. These findings support the idea that quantitative DTI can track structural changes in the bilateral PYs and that DTI may serve as a noninvasive tool to predict the prognoses of patients with BSH
MEIS2C and MEIS2D promote tumor progression via Wnt/β-catenin and hippo/YAP signaling in hepatocellular carcinoma
Abstract
Background
MEIS2 has been identified as one of the key transcription factors in the gene regulatory network in the development and pathogenesis of human cancers. Our study aims to identify the regulatory mechanisms of MEIS2 in hepatocellular carcinoma (HCC), which could be targeted to develop new therapeutic strategies.
Methods
The variation of MEIS2 levels were assayed in a cohort of HCC patients. The proliferation, clone-formation, migration, and invasion abilities of HCC cells were measured to analyze the effects of MEIS2C and MEIS2D (MEIS2C/D) knockdown with small hairpin RNAs in vitro and in vivo. Chromatin immunoprecipitation (ChIP) was performed to identify MEIS2 binding site. Immunoprecipitation and immunofluorescence assays were employed to detect proteins regulated by MEIS2.
Results
The expression of MEIS2C/D was increased in the HCC specimens when compared with the adjacent noncancerous liver (ANL) tissues. Moreover, MEIS2C/D expression negatively correlated with the prognosis of HCC patients. On the other hand, knockdown of MEIS2C/D could inhibit proliferation and diminish migration and invasion of hepatoma cells in vitro and in vivo. Mechanistically, MESI2C activated Wnt/β-catenin pathway in cooperation with Parafibromin (CDC73), while MEIS2D suppressed Hippo pathway by promoting YAP nuclear translocation via miR-1307-3p/LATS1 axis. Notably, CDC73 could directly either interact with MEIS2C/β-catenin or MEIS2D/YAP complex, depending on its tyrosine-phosphorylation status.
Conclusions
Our studies indicate that MEISC/D promote HCC development via Wnt/β-catenin and Hippo/YAP signaling pathways, highlighting the complex molecular network of MEIS2C/D in HCC pathogenesis. These results suggest that MEISC/D may serve as a potential novel therapeutic target for HCC.https://deepblue.lib.umich.edu/bitstream/2027.42/152244/1/13046_2019_Article_1417.pd
Continuous Glucose Monitoring in Newly Diagnosed Type 2 Diabetes Patients Reveals a Potential Risk of Hypoglycemia in Older Men
Objectives. We performed continuous glucose monitoring (CGM) to define the features of patients with newly diagnosed type 2 diabetes (T2D) before and after Continuous Subcutaneous Insulin Infusion (CSII) therapy. Methods. This was a retrospective analysis. Newly diagnosed T2D patients (106) were admitted from eight centers in China. They were divided into a younger patient group (<60 years) and an older patient group (≥60 years). Each group was further divided into male and female patients. CSII therapy was maintained for 3 weeks after the glycemic target was reached. CGM was performed 2 times before and after completion of insulin treatment. Results. CGM data showed the expected significant improvement of mean amplitude glycemic excursion (MAGE) with CSII therapy. The older patients had lower hourly glucose concentrations from 0200 to 0700 o’clock compared to the younger patients at baseline. Surprisingly, in the older patient group, the male patients had a potential risk of hypoglycemia after CSII therapy, especially during periods from 2300 to 2400 and 0400 to 0600. Conclusions. Our data suggested that older male patients with newly diagnosed T2D may have lower nocturnal glucose concentrations. This may potentially increase the risk of nocturnal hypoglycemia during CSII therapy. This study was registered with Chinese Clinical Trial Registry, number CliCTR-TRC-11001218
Post-marketing risk analysis of bendamustine: a real-world approach based on the FAERS database
Objective: Bendamustine was approved for treating chronic lymphocytic leukemia and indolent B-cell non-Hodgkin lymphoma. Despite its therapeutic benefits, the long-term safety of bendamustine in a large population remains inadequately understood. This study evaluates the adverse events (AEs) associated with bendamustine, using a real-world pharmacovigilance database to support its clinical application. Methods: We conducted a post-marketing risk analysis to assess the association between bendamustine and its AEs. Data were extracted from the US FDA’s Adverse Event Reporting System (FAERS), covering the period from January 2017 to September 2023. The characteristics of bendamustine-associated AEs and the onset time were further analyzed. Statistical analysis was performed using MYSQL 8.0, Navicat Premium 15, Microsoft EXCEL 2016, and Minitab 21.0.Results: 9,461,874 reports were collected from the FAERS database, 9,131 identified bendamustine as the “primary suspected” drug. We identified 331 significant disproportionality preferred terms (PTs). Common AEs included pyrexia, neutropenia, infusion site reaction, progressive multifocal leukoencephalopathy (PML), injection site vasculitis, and pneumonia—all documented on bendamustine’s label. Notably, 16 unexpected and significant AEs were discovered, including hypogammaglobulinemia, which is concerning due to its potential to increase infection susceptibility following bendamustine treatment. Other significant findings were anaphylactic reactions, PML, and cutaneous malignancies, suggesting updates to the drug’s label may be necessary. Physicians should monitor for neurological and skin changes in patients and discontinue treatment if PML is suspected. Moreover, the median onset time for bendamustine-associated AEs was 13 days, with an interquartile range [IQR] of 0–59 days, predominantly occurring on the first day post-initiation. The β of bendamustine-related AEs suggested risk reduction over time. Conclusion: Our study uncovered some potential pharmacovigilance signals for bendamustine, providing important insights for its safe and effective clinical use
