    Simple methodology for the quantitative analysis of fatty acids in human red blood cells

    In the last years, there has been an increasing interest in evaluating possible relations between fatty acid (FA) patterns and the risk for chronic diseases. Due to the long life span (120 days) of red blood cells (RBCs), their FA profile reflects a longer term dietary intake and was recently suggested to be used as an appropriate biomarker to investigate correlations between FA metabolism and diseases. Therefore, the aim of this work was to develop and validate a simple and fast methodology for the quantification of a broad range of FAs in RBCs using gas chromatography with flame ionization detector, as a more common and affordable equipment suitable for biomedical and nutritional studies including a large number of samples. For this purpose, different sample preparation protocols were tested and compared, including a classic two-step method (Folch method) with modifications and different one-step methods, in which lipid extraction and derivatization were performed simultaneously. For the one-step methods, different methylation periods and the inclusion of a saponification reaction were evaluated. Differences in absolute FA concentrations were observed among the tested methods, in particular for some metabolically relevant FAs such as trans elaidic acid and eicosapentaenoic acid. The one-step method with saponification and 60 min of methylation time was selected since it allowed the identification of a higher number of FAs, and was further submitted to in-house validation.     OBJETIVO: Compreender a experiência de uma equipe de enfermagem na implantação da Iniciativa Hospital Amigo da Criança (IHAC). METODOS: Trata-se de uma pesquisa qualitativa, cujo cenário foi o Alojamento Conjunto do Hospital Universitário da Universidade de São Paulo. Participaram cinco enfermeiras e seis técnicas e auxiliares de enfermagem. A coleta de dados foi realizada através de entrevistas semi-estruturadas e as narrativas foram analisadas na perspectiva de Janesick. RESULTADOS: Das narrativas foram extraídas quatro categorias: o impacto resultante da implantação das medidas preconizadas pela IHAC; o processo de sensibilização para a implantação das medidas da IHAC; os fatores intervenientes na implantação da IHAC e a concretização dos Dez Passos da IHAC. A análise dessas categorias ocorreu mediante autores que investigam o tema mudança. CONCLUSÕES: Acredita-se que este estudo possa contribuir para a avaliação da implantação da referida estratégia de intervenção, fornecendo subsídios para a reorganização das atividades assistenciais e gerenciais.<br>OBJETIVO: Comprender la experiencia de un equipo de enfermería en la implantación de la Iniciativa Hospital Amigo del niño (IHAN). METODOS: Se trata de una investigación cualitativa, cuyo escenario fue el Alojamiento Conjunto del Hospital Universitario de la Universidad de Sao Paulo. Participaron cinco enfermeras y seis técnicas y auxiliares de enfermería. La recolección de datos fue realizada a través de entrevistas semi-estructuradas y las narrativas fueron analizadas en la perspectiva de Janesick. RESULTADOS: De las narrativas fueron extraídas cuatro categorías: el impacto resultante de la implantación de las medidas preconizadas por la IHAN; el proceso de sensibilización para la implantación de las medidas de la IHAN; los factores intervinientes en la implantación de la IHAN y la concretización de los Diez Pasos de la IHAN. El análisis de esas categorías ocurrió mediante autores que investigan el tema de cambio. CONCLUSIONES: Se acredita que este estudio puede contribuir para la evaluación de la implantación de la referida estrategia de intervención, ofreciendo subsidios para la reorganización de las actividades asistenciales y gerenciales.<br>OBJECTIVE: To understand the experience of a nursing team in the implantation of the Child-Friendly Hospital Initiative (CFHI). METHODS: This qualitative research was carried out at the Rooming-in unit of the University Hospital at the University of São Paulo, Brazil. Five nurses and six technicians and nursing assistants took part. Data were collected by means of semi-structured interviews and the analysis of the reports was based on Janesick's perspective. RESULTS: Four categories were extracted from the reports: the impact of putting in practice the measures recommended by the CFHI; the awareness process for putting CFHI measures in practice; intervening factors when putting CFHI in practice and concretization of the "Ten Steps" of the CFHI. Authors who researched on change were used to analyze these categories. CONCLUSIONS: This study can contribute to evaluate this intervention strategy in practice, supporting the reorganization of care and management activities
