    Influence of seed treatment and packaging materials on seed longevity of cluster bean [Cyamopsistetra gonoloba (L.) Taub.]

    An experiment was conducted to investigate the influence of packaging materials and seed treatments on storability of cluster bean under ambient conditions of Bengaluru. The experiment consisted of six treatments viz. control (T1), bavistin @ 2 g kg-1 (T2), spinosad @ 0.04 ml/kg (T3), neem leaf powder @ 1:20 ratio (T4), Acorus calamus@ 10 g kg-1 (T5) and cow dung powder @ 10 g kg-1 (T6) and three packaging materials viz., cloth bag (C1) super grain bag (C2) and poly lined cloth bag (C3). Treated seed samples were stored in three containers under ambient storage conditions up to the duration of which seeds maintain minimum seed certification standards and samples were drawn at bimonthly intervals for ascertaining the seed quality parameters. The study suggested that seed treat-ment could be useful to prolong the storage life of cluster bean seeds. The seeds treated with spinosad (0.04 ml/kg) and stored in super-grain bag were better for maintenance of higher seed quality parameters [germination (80.00%), root length (11.70 cm), shoot length (13.60 cm), mean seedling dry weight (152 mg), seedling vigour index I and II (2024&12140) and TDH activity (1.224) with low electrical conductivity (0.368 dSm-1)] up to 18 months under ambient conditions of Bengaluru (room temperature). Super-grain bag proved to be better storage container with higher seed quality attributes viz., germination (72.38 %), seedling vigour index-I (1726), total dehydrogenase activity (1.201) and other seed quality parameters compared to cloth bag. The study suggested that use of appropriate packaging material and seed treatment could be useful to prolong the storage life of cluster bean seeds

    Video sharing has been a gradually more popular application in OSNs facilitating users to share their personal videos or interesting videos they found with their friends. However OSN’s additional progress is strictly caught up by the inherent limits of the conventional client/server architecture of its video sharing system which is not only costly in terms of server storage and bandwidth but also not scalable with the high amount of users and video content in OSNs. The efforts have been dedicated to perk up the client/server architecture for video sharing with the peer-to-peer (P2P) architecture being the most promising. P2P-based video sharing has been used in on demand video streaming.The dimension reveals that mainly of the viewers of a user’s videos are the user’s close friends, most video views are driven by social relationships and the rest are driven by interests and viewers of the same video tend to live in the same location. Based on our observations we propose Social Tube a system that discover the social relationship interest resemblance and location to improve the presentation of video sharing in OSNs. Specifically an OSN has a social network (SN)-based P2P overlay construction algorithm that come together peers based on their social relationships and interests.

    Effect of Temperature and Period of Storage on Breaking Dormancy in Gladiolus (Gladiolus grandiflorus Hort.) Corms

    An experiment was conducted in 2010-2011 at Indian Institute of Horticultural Research, Bengaluru, on three gladiolus cultivars viz., 'Arka Amar', 'Darshan' and 'Kum Kum' to study effects of storage temperature (4°C and room temperature 27±2°C) and length of storage (50, 70 and 90 days) on dormancy of corms. Cv. 'Kum Kum' registered minimum number of days for sprouting (42.71 days), spike emergence (116 days) and days to opening of first floret (128 days). Corms stored at 4°C resulted in lowest number of days for-sprouting (45.24 days), days to spike emergence (114.63 days) and days to opening of first floret (126.60 days) and resulted in highest sprouting percentage (58.7%). Interaction effects revealed that cv. 'Kum Kum' stored at 4°C for 90 days after harvest took minimum number of days to sprouting (25.07 days), days to spike emergence (90.38 days) and days to opening of first floret (102.38 days) resulting in 100% sprouting

    An International Reference Consensus Genetic Map with 897 Marker Loci Based on 11 Mapping Populations for Tetraploid Groundnut (Arachis hypogaea L.)

    Only a few genetic maps based on recombinant inbred line (RIL) and backcross (BC) populations have been developed for tetraploid groundnut. The marker density, however, is not very satisfactory especially in the context of large genome size (2800 Mb/1C) and 20 linkage groups (LGs). Therefore, using marker segregation data for 10 RILs and one BC population from the international groundnut community, with the help of common markers across different populations, a reference consensus genetic map has been developed. This map is comprised of 897 marker loci including 895 simple sequence repeat (SSR) and 2 cleaved amplified polymorphic sequence (CAPS) loci distributed on 20 LGs (a01–a10 and b01–b10) spanning a map distance of 3, 863.6 cM with an average map density of 4.4 cM. The highest numbers of markers (70) were integrated on a01 and the least number of markers (21) on b09. The marker density, however, was lowest (6.4 cM) on a08 and highest (2.5 cM) on a01. The reference consensus map has been divided into 20 cM long 203 BINs. These BINs carry 1 (a10_02, a10_08 and a10_09) to 20 (a10_04) loci with an average of 4 marker loci per BIN. Although the polymorphism information content (PIC) value was available for 526 markers in 190 BINs, 36 and 111 BINs have at least one marker with >0.70 and >0.50 PIC values, respectively. This information will be useful for selecting highly informative and uniformly distributed markers for developing new genetic maps, background selection and diversity analysis...

    Large-scale transcriptome analysis in chickpea (Cicer arietinum L.), an orphan legume crop of the semi-arid tropics of Asia and Africa

    Chickpea (Cicer arietinum L.) is an important legume crop in the semi-arid regions of Asia and Africa. Gains in crop productivity have been low however, particularly because of biotic and abiotic stresses. To help enhance crop productivity using molecular breeding techniques, next generation sequencing technologies such as Roche/454 and Illumina/Solexa were used to determine the sequence of most gene transcripts and to identify drought-responsive genes and gene-based molecular markers. A total of 103 215 tentative unique sequences (TUSs) have been produced from 435 018 Roche/454 reads and 21 491 Sanger expressed sequence tags (ESTs). Putative functions were determined for 49 437 (47.8%) of the TUSs, and gene ontology assignments were determined for 20 634 (41.7%) of the TUSs. Comparison of the chickpea TUSs with the Medicago truncatula genome assembly (Mt 3.5.1 build) resulted in 42 141 aligned TUSs with putative gene structures (including 39 281 predicted intron/splice junctions). Alignment of ∼37 million Illumina/Solexa tags generated from drought-challenged root tissues of two chickpea genotypes against the TUSs identified 44 639 differentially expressed TUSs. The TUSs were also used to identify a diverse set of markers, including 728 simple sequence repeats (SSRs), 495 single nucleotide polymorphisms (SNPs), 387 conserved orthologous sequence (COS) markers, and 2088 intron-spanning region (ISR) markers.     Delineation of the population genetic structure of Culicoides imicola in East and South Africa

    BACKGROUND: Culicoides imicola Kieffer, 1913 is the main vector of bluetongue virus (BTV) and African horse sickness virus (AHSV) in Sub-Saharan Africa. Understanding the population genetic structure of this midge and the nature of barriers to gene flow will lead to a deeper understanding of bluetongue epidemiology and more effective vector control in this region. METHODS: A panel of 12 DNA microsatellite markers isolated de novo and mitochondrial DNA were utilized in a study of C. imicola populations from Africa and an outlier population from the Balearic Islands. The DNA microsatellite markers and mitochondrial DNA were also used to examine a population of closely related C. bolitinos Meiswinkel midges. RESULTS: The microsatellite data suggest gene flow between Kenya and south-west Indian Ocean Islands exist while a restricted gene flow between Kenya and South Africa C. imicola populations occurs. Genetic distance correlated with geographic distance by Mantel test. The mitochondrial DNA analysis results imply that the C. imicola populations from Kenya and south-west Indian Ocean Islands (Madagascar and Mauritius) shared haplotypes while C. imicola population from South Africa possessed private haplotypes and the highest nucleotide diversity among the African populations. The Bayesian skyline plot suggested a population growth. CONCLUSIONS: The gene flow demonstrated by this study indicates a potential risk of introduction of new BTV serotypes by wind-borne infected Culicoides into the Islands. Genetic similarity between Mauritius and South Africa may be due to translocation as a result of human-induced activities; this could impact negatively on the livestock industry. The microsatellite markers isolated in this study may be utilised to study C. bolitinos, an important vector of BTV and AHSV in Africa and identify sources of future incursions.     Identification of groundwater potential zones using remote sensing and GIS techniques, in Muguru Addahalla watershed, Mysore and Chamarajnagar districts, Karnataka, India

    Groundwater recharge from surface water sources is an essential aspect in any watershed or a hydrological unit. For any small scale groundwater assessment, experimental approaches are usually adopted. However, recharge into aquifers depends on several hydrogeological factors on a regional scale analysis; groundwater assessment is conducted in a comprehensive approach considering all the geospatial data of different contributing factors. These factors include geomorphological, geological and structural elements like lineaments, drainage frequency, drainage density, lithology, land use and land cover. This study has been conducted with an integrated approach to identify the groundwater potential of the area, and the factors influencing groundwater recharge. The study has shown that the prevailing groundwater conditions are very good along the stream networks and the remaining zones show moderate to poor groundwater resources. It is also found that for such an integrated study, the tools and techniques of remote sensing and GIS are more useful
