CONGELABILIDADE DO SÊMEN SUÍNO DE ACORDO COM O PERÍODO DE EQUILÍBRIO PRÉ-CONGELAMENTO E DA SENSIBILIDADE AO RESFRIAMENTO
O objetivo deste estudo foi avaliar a influência do período de equilíbrio pré-congelamento
e da sensibilidade ao resfriamento sobre a qualidade do sêmen suíno congelado. Foram utilizados dez
ejaculados de cada um dos 8 machos doadores. A motilidade de doses de sêmen preparadas a partir
dos mesmos ejaculados utilizados no congelamento, foi avaliada simultaneamente, durante o resfriamento
a 15ºC, a cada 24h. A porção de cada ejaculado destinada ao congelamento foi separada em duas
partes que foram submetidas a equilíbrio prévio, a 20ºC, curto (1,5h) ou longo (20h). As palhetas foram
descongeladas a 37ºC por 20 segundos e seu conteúdo (0,5mL) foi diluído (1:6) em Beltsville Thawing
Solution (BTS). Foram avaliados a motilidade (MOT), acrossomos normais após o descongelamento
(NARPD) e após o teste de termoresistência (NARTTR), e membranas íntegras (MI). Para a análise
estatística, os ejaculados foram separados, de acordo com sua sensibilidade ao resfriamento, em dois
grupos: motilidade 0,05). O equilíbrio por 20h não apresentou diferenças na MOT e MI (P>0,05), mas o
NARPD e NARTTR foram superiores (
PRODUTIVIDADE DE LEITOAS ALOJADAS EM GAIOLAS INDIVIDUAIS OU BAIAS COLETIVAS DURANTE A GESTAÇÃO
O objetivo do presente estudo foi determinar os efeitos do sistema de alojamento, em
gaiolas individuais ou baias coletivas, durante a metade da gestação, no desempenho de fêmeas
suínas e de suas leitegadas. Após terem sido inseminadas com 220 dias de idade, em média, as
leitoas foram alojadas em gaiolas, até aproximadamente 55 dias de gestação, quando parte delas
permaneceram em gaiolas (n=94) e outra parte foi transferida para baias coletivas (n=113), com seis
fêmeas/baia. Não houve efeito dos tratamentos na taxa de parto ajustada (P>0,05). As leitoas que
permaneceram em gaiolas foram mais pesadas aos 110 dias de gestação e após o parto (P0,05), quando comparadas às
transferidas para baias coletivas. Os sistemas de alojamento não afetaram o total de leitões nascidos
e nascidos vivos, o peso médio dos leitões ao nascer e o intervalo desmame-estro (P>0,05). No
entanto, verificou-se que as fêmeas alojadas em baias consumiram mais ração na lactação (P0.05). However, it was observed

VOLUME DE MEIO E NÚMERO DE OÓCITOS PARA FECUNDACÃO NA PRODUÇÃO IN VITRO DE EMBRIÕES BOVINOS
A origem dos Complexos Cumulus-Oócitos (CCO) bovinos e os diferentes protocolos entre
equipes geram variações nos índices de produção in vitro (PIV). O processo pode ser influenciado pelo número
de CCO e volume das gotas de maturação, fecundação e cultivo. Neste estudo, avaliou-se a densidade de
CCO e o volume de meio de fecundação sobre a PIV com 672 oócitos bovinos. A maturação in vitro (MIV) foi
conduzida com 20 CCO em 200µL de TCM-199+rFSHh+10% de soro de vaca em estro (SVE), por 24h, em
estufa a 39ºC, 5% CO2 em ar e umidade saturada. Na fecundação (Dia 0 =D0) utilizou-se 5, 10 ou 20 CCO e 2
volumes de meio (1:5 e 1:10), num fatorial 3x2, em 6 repetições. O sêmen foi selecionado por migração
ascendente e para a fertilização (1x106 espermatozóides/mL) os gametas foram incubados em Fert-Talp, por
18h. O cultivo foi em grupos de 20 zigotos, em 200µL de fluído sintético do oviduto (SOF) aaci+5%SVE, por 8
dias e as avaliações em D2 (clivagem), D7 (Blastocistos), D9 (Blastocistos expandidos, em eclosão e eclodidos).
As taxas de clivagem (80, 85 e 87%), embriões em D9 (23, 27 e 28%) e de eclosão (42, 37 e 48%) foram
semelhantes (P>0,05) para 5, 10 ou 20 CCO, respectivamente. Não houve diferença (P>0,05) na clivagem (83
e 85%), embriões em D9 (24 e 28%) e eclosão (41 e 44%) entre as proporções 1:5 e 1:10µL. A produção de
blastocistos e taxa de eclosão em D9 não são influenciados pelo número de CCO, nem pela proporção de
CCO por volume de meio na fecundação.
Volume of medium and number of oocytes for fertilization
in the in vitro production of bovine embryos
Abstract
The origin of the bovine Cumulus Oocytes Complexes (COC) and the different protocols among
teems are responsible for the variation rates of the in vitro production (IVP). The process is also influenced by
COC number and by the volume of the drop in the maturation, fertilization and culture. In this study, the effect of
bovine COC density per drop medium and the fertilization medium volume were evaluated during the bovine IVP
using 672 COC. The in vitro maturation was performed with 20 COC in 200µL of TCM-199+rFSHh+10% estrus
cow serum (ECS), for 24h in an incubator with 5%CO2 in air, saturated humidity, at 39oC. For the fertilization (Day
0 = D0) 5, 10 or 20 COC and 2 volumes ratio of medium (1:5 and 1:10) were used in a factorial outline 3x2, in 6
replications. Thawed semen was prepared by a swim up process and for the fertilization (1x106 spermatozoa/
ml) the gametes were maintained in Fert-Talp for 18h. Groups of twenty presumed zygotes were cultured in
200µL synthetic oviduct fluid (SOFaaci)+5% ECS, for 8 days. The evaluations were carried out in D2 (cleavage),
D7 (blastocysts) and D9 (expanded and hatched blastocyts). The rates of cleavage (80, 85 and 87%), embryos
in D9/ D2 (23, 27 and 28%) and hatched blastocstys in D9/D7 (42, 37 and 48%) were similar (P>0.05) for 5, 10
and 20 COC, respectively. No difference was observed in cleavage (83 and 85%), embryos in D9 (24 and 28%)
and hatching rates (41 and 44%) for 1:5 and 1:10µL of fertilization medium. Blastocysts production and the
hatching rate in D9 were neither influenced by the COC number, nor by the proportion of COC per medium
volume used during fertilization
Oxygen equilibrium curve of normal human blood and its evaluation by Adair's equation
Oxygen equilibrium curves of fresh, normal human blood have been measured by new methods which allow the control of pH, pCO2, and 2,3-diphosphoglycerate and which yield higher accuracy at the extremes of saturation than was possible previously. The curve determined by these techniques lies slightly to the right of the standard curve of Roughton et al. (Roughton, F.J.W., Deland, E.C., Kernohan, J.C., and Severinghaus, J.W. (1972) in Oxygen Affinity of Hemoglobin and Red Cell Acid Base Status (Astrup, P., and R\uf8rth, M., eds) pp. 73-83, Academic Press, New York). The greatest difference is at low oxygen saturation, probably owing to the fact that the latter data were obtained under conditions which would lead to depletion of cellular 2,3-diphosphoglycerate. The range of p50 (oxygen pressure at half-saturation) values for four normal subjects was 28.3 mm Hg to 29.0 mm Hg. Adair's stepwise oxygenation scheme has been used to analyze the curves with the result that a1 = 0.1514 X 10(-1) (+/- 10%) mm-1; a2 = 0.9723 X 10(-3) (+/- 8%) mm-2; a3 = 0.1703 X 10(-3) (+/- 50%) mm-3; a4 = 0.1671 X 10(-5) (+/- 2%) mm-4 for the best of four data sets. Because these constants are very sensitive to changes in the shape of the oxygenation curve, this analysis is much more useful than p50 measurements in the investigation of the various allosteric effectors of the function of hemoglobin within the red cell
LÍQUIDO FOLICULAR EQÜINO NA MATURAÇÃO IN VITRO DE OÓCITOS BOVINOS
O desenvolvimento embrionário de oócitos bovinos maturados in vitro (MIV) foi
avaliado em meio suplementado com líquido folicular eqüino (Lfe). Foram distribuídos 1045 oócitos
em 11 repetições formando três grupos tratamentos (T1, T2, T3) e um controle (C). O meio de
maturação utilizado foi o TCM-199 acrescido de piruvato de sódio, hormônio folículo estimulante
recombinante (rFSHh) e hormônio luteinizante equino (LHe). Suplementou-se esse meio com
10% de soro de égua em estro para o grupo controle e para T1, T2 e T3, o meio foi suplementado
com 5, 10, e 20% de LFe, respectivamente. Os oócitos foram maturados in vitro (MIV) por 24h.
A fecundação in vitro (FIV) foi realizada em meio Talp-Fert. A MIV e a FIV foram realizadas em
estufa a 39ºC com 5% de CO2 em ar e umidade saturada. Os zigotos foram cultivados em meio
SOFaaci, sob óleo mineral no interior de bolsas plásticas gaseificadas. As taxas de clivagem e
de blastocistos foram observadas diariamente (D), e em D7, foram superiores (P0,05) às do grupo controle. Em D9, a taxa
de blastocistos do T2 foi superior (P0,05). O LFe, na concentração de 10%
pode ser utilizado, em substituição ao soro de égua em estro para suplementar o meio de MIV
de oócitos bovinos.
Equine follicular fluid on in vitro maturation of bovine oocytes
Abstract
Embryo development of bovine oocytes was evaluated using maturation medium
supplemented with equine follicular fluid (eFF). One thousand and forty five (1045) oocytes were distributed
in 11 replications forming three treatment groups (T1, T2 e T3) and one Control (C). TCM-199 added with
sodium pyruvate, rFSHh and LHe was used as maturation medium. This medium was supplemented with
10% estrous mare serum for Control group, and 5, 10, and 20% eFF, respectively, for T1, T2 e T3
groups. In vitro maturation (IVM) of all groups was performed during 24h. In vitro fertilization (IVF) was
performed in TALP-FERT medium. IVM and IVF were carried out in an incubator at 39ºC with 5% CO2 in
air and saturated humidity. Zygotes were cultured in SOFaaci medium, under mineral oil in gasified
bags. Cleavage and blastocyst rates were daily observed (D), and at D7, were higher (P0.05) for those from control group. At D9, blastocyst rate of T2
was higher (P0.05). The eFF, at a 10% concentration, can replace the use of
estrous mare serum to supplement the IVM medium of bovine oocytes
MONITORAMENTO OVARIANO AO ABATE DE LEITOAS DESCARTADAS POR ANESTRO OU ESTRO ATÍPICO
Granjas , manejadas dentro de intenso fluxo de produção, trabalham com taxas anuais
de descarte relativamente elevadas (35-50%). Muitas fêmeas são descartadas por problemas irreais.
Esses podem ser oriundos de falhas humanas durante sua determinação, levando a um aumento
na taxa de renovação do plantel, diminuição da produtividade e, conseqüentemente, aumento dos
dias não produtivos e elevados custos. O objetivo do presente trabalho foi realizar uma análise
observacional dos ovários de leitoas descartadas por anestro ou estro atípico. O experimento foi
realizado em dois frigoríficos de abate de suínos, onde foram avaliados macroscopicamente os
ovários de 145 leitoas descartadas por anestro ou estro atípico. Os ovários foram classificados em
lisos ou com folículos; com corpos lúteos, e com corpos albicans e corpos lúteos. Das 83 leitoas
descartadas por anestro, 13% apresentaram ovários lisos ou com folículos, indicando que essas
não haviam ciclado na granja. Por outro lado, 83% das fêmeas apresentaram ovários com corpos
lúteos e/ou corpos albicans, indicando que já haviam ciclado na granja. Algumas fêmeas (4%)
apresentaram ovários policísticos. Das 62 fêmeas descartadas por estro atípico na granja, 2, 24%
não haviam ciclado na granja e 76% haviam ciclado, segundo a avaliação de seus ovários. Os
resultados mostram que o acompanhamento ao abate, de fêmeas descartadas por falhas
reprodutivas, permite verificar se as razões do descarte condizem ou não com o status fisiológico do
trato reprodutivo.
Ovarian monitorament at slaughter of gilts culled by anoestrus or atypic oestrus
Abstract
Intensively managed pig farms work with relatively high annual culling rate, between
35-50%. Many females are culled by causes unrelated to the suspected diagnosis. The problems
can arise from human failure, leading to an increased herd replacement rate, decrease on productivity
and consequent increase in non-productive days and raised costs. The objective of the present work
was to perform an observational analysis of the ovaries of gilts culled by anoestrus or atypical oestrus.
The experiment was carried out in two swine slaughterhouses where 145 ovaries of gilts, culled by
anoestrus or atypical oestrus, were macroscopically evaluated. The ovaries were classified in: inactive
or with follicles; with corpora lutea; and with corpora albicantia and corpora lutea. From 83 gilts culled
by anoestrus, 13% presented inactive ovaries or with follicles, indicating that they had not cycled on
the farm. On the other hand, 83% presented ovaries with corpora lutea and/or corpora albicantia,
indicating that they had previously cycled. Some gilts (4%) presented policystic ovaries. From 62
females culled by atypical oestrus on farm 2, 24% did not cycle on the farm and 76% cycled, according
to the ovary examination. The results show that a slaughter examination of gilts culled by reproductive
causes, allow to assess if culling reasons correspond to the physiological status of the reproductive
tract

A manutenção dos complexos cumulus-oócitos (CCO) em líquido folicular (LF) antes da
sua maturação, além de visar a capacitação, viabiliza o transporte até o laboratório por ser de baixo
custo, de fácil aquisição e o congelamento do LF permite seu armazenamento para futura utilização.
Neste experimento avaliou-se o efeito do congelamento do LF obtido de folículos de 2-8mm e de
folículos >8mm, sobre a taxa de produção embrionária. Oócitos foram aspirados de folículos de 2 a
8mm de ovários provenientes de abatedouro. No grupo controle (n=295) os CCO foram maturados
por 24h. Nos tratamentos GF (n=297) e GC (n=282), os CCO foram mantidos por 6h a 30ºC em LF
fresco ou congelado, respectivamente, de folículos >8mm. Já no tratamento PF(n=278) e PC (n=281),
os CCO foram mantidos em LF fresco ou congelado, respectivamente, de folículos de 2-8mm.
Posteriormente, os CCO dos tratamentos GF, GC, PF e PC foram maturados por 18h. Não houve
efeito negativo do congelamento do líquido folicular e nem do tamanho dos folículos sobre as taxas
de clivagem e produção embrionária em D7 e D9 (P>0,05). No entanto, o congelamento do LF de
folículos de 2 a 8mm resultou em redução da taxa de eclosão e do número de células dos blastocistos.
A manutenção de oócitos bovinos por 6h a 30ºC, antes da maturação, pode ser efetuada em líquido
folicular de folículos >8mm, fresco ou congelado.

In addition to the capacitation, the maintenance of cumulus-oocyte complex (COC) in
follicular fluid (FF) before maturation, allows the transport to the laboratory, being a practical and less
expensive media. The FF can be stored after freezing to future use. Oocytes aspirated from bovine
slaughterhouse ovaries, were used to evaluate the effect of maintaining the oocytes in fresh or frozen
bovine FF (from 2-8mm and >8mm follicles) on the blastocyst rate. In the control group (n=259) the
COC were matured for 24h. On treatments GF (n=297) and GC (n=282) the COC were held for 6h at
30°C in fresh or frozen FF from >8mm follicles, respectively. In treatments PF (n=278) and PC (n=281)
the COC were held in fresh or frozen FF from 2-8mm follicles, respectively. Later, the COC from GF,
GC, PF and PC were matured for 18h. The freezing process as well as the follicle size had no effect
on the cleavage, D7 or D9 blastocyst rates (P>0,05). Nevertheless, the frozen FF from 2-8mm follicles
resulted in a reduced hatching rate and lower ICM cells. Fresh or frozen follicular fluid of >8mm
follicles could be used for a 6h transport of bovine oocytes before maturation for 18h

Constraining the expansion rate of the Universe using low-redshift ellipticals as cosmic chronometers
We present a new methodology to determine the expansion history of the
Universe analyzing the spectral properties of early type galaxies (ETG). We
found that for these galaxies the 4000\AA break is a spectral feature that
correlates with the relative ages of ETGs. In this paper we describe the
method, explore its robustness using theoretical synthetic stellar population
models, and apply it using a SDSS sample of 14 000 ETGs. Our motivation
to look for a new technique has been to minimise the dependence of the cosmic
chronometer method on systematic errors. In particular, as a test of our
method, we derive the value of the Hubble constant (stat)
(syst) (68% confidence), which is not only fully compatible with the
value derived from the Hubble key project, but also with a comparable error
budget. Using the SDSS, we also derive, assuming w=constant, a value for the
dark energy equation of state parameter (stat)
(syst). Given the fact that the SDSS ETG sample only reaches , this
result shows the potential of the method. In future papers we will present
results using the high-redshift universe, to yield a determination of H(z) up

Phylogenetic convergence and multiple shell shape optima for gliding scallops (Bivalvia: Pectinidae)
An important question in evolutionary biology is how often, and to what extent, do similar ecologies elicit distantly related taxa to evolve towards the same phenotype? In some scenarios, the repeated evolution of particular phenotypes may be expected, for instance when species are exposed to common selective forces that result from strong functional demands. In bivalved scallops (Pectinidae), some species exhibit a distinct swimming behaviour (gliding), which requires specific biomechanical attributes to generate lift and reduce drag during locomotive events. Further, a phylogenetic analysis revealed that gliding behaviour has independently evolved at least four times, which raises the question as to whether these independent lineages have also converged on a similar phenotype. Here, we test the hypothesis that gliding scallops display shell shape convergence using a combination of geometric morphometrics and phylogenetic comparative methods that evaluate patterns of multivariate trait evolution. Our findings reveal that the gliding species display less morphological disparity and significant evolutionary convergence in morphospace, relative to expectations under a neutral model of Brownian motion for evolutionary phenotypic change. Intriguingly, the phylomorphospace patterns indicate that gliding lineages follow similar evolutionary trajectories to not one, but two regions of morphological space, and subsequent analyses identified significant differences in their biomechanical parameters, suggesting that these two groups of scallops accomplish gliding in different ways. Thus, whereas there is a clear gliding morphotype that has evolved convergently across the phylogeny, functionally distinct morphological subforms are apparent, suggesting that there may be two optima for the gliding phenotype in the Pectinidae.J.
