    Fatty acid based microemulsions to combat ophthalmia neonatorum caused by 'Neisseria gonorrhoeae' and 'Staphylococcus aureus'

    The bacterial species Neisseria gonorrhoeae (N. gonorrhoeae) and Staphylococcus aureus (S. aureus) are amongst the main microorganisms that cause ophthalmia neonatorum. The current treatment involves the use of various antibiotics such as ciprofloxacin, cephalosporin, ceftriaxone and cefotaxime. However, this treatment strategy is becoming more ineffective due to the antibiotic resistance in N. gonorrhoeae. The current study explores the potential use of fatty acid based microemulsions (ME) to prevent N. gonorrhoeae and S. aureus infections in new-borns’ eyes without harmful side effects such as corneal or conjunctiva irritation. Pseudo-ternary phase diagrams were constructed to evaluate microemulsion regions and six different α-linolenic acid based microemulsions were prepared. The prepared formulations were characterized for α-linolenic acid content, size, transparency, zeta potential, Polarized light Microscopy, antimicrobial activity and ex vivo ocular toxicity. The mean droplet size of the ME formulations was in the range of 190.4 to 350.5 nm and polydispersity index (PDI) values were in the range of 0.102 to 0.561. All formulations were found stable upon storage for at least 8 weeks. In addition, self-diffusion coefficients determined by nuclear magnetic resonance (NMR) reflected that the diffusability of water increased at higher than 30% w/w water, while that of fatty acids and surfactants was in reverse. The antimicrobial efficacy of microemulsions was determined against N. gonorrhoeae and S. aureus. It was concluded that all microemulsions have strong antimicrobial effects against N. gonorrhoeae and S. aureus. Finally, bovine corneal opacity permeability (BCOP) and hen’s egg chorioallantoic (HET-CAM) tests results showed that all microemulsion formulations were not strong ocular irritants

    Tunable diode laser measurements of hydrothermal/volcanic CO2 and implications for the global CO2 budget

    Quantifying the CO2 flux sustained by low-temperature fumarolic fields in hydrothermal/volcanic environments has remained a challenge, to date. Here, we explored the potential of a commercial infrared tunable laser unit for quantifying such fumarolic volcanic/hydrothermal CO2 fluxes. Our field tests were conducted between April 2013 and March 2014 at Nea Kameni (Santorini, Greece), Hekla and KrĂœsuvĂ­k (Iceland) and Vulcano (Aeolian Islands, Italy). At these sites, the tunable laser was used to measure the path-integrated CO2 mixing ratios along cross sections of the fumaroles' atmospheric plumes. By using a tomographic post-processing routine, we then obtained, for each manifestation, the contour maps of CO2 mixing ratios in the plumes and, from their integration, the CO2 fluxes. The calculated CO2 fluxes range from low (5.7 ± 0.9 t d−1; KrĂœsuvĂ­k) to moderate (524 ± 108 t d−1; La Fossa crater, Vulcano). Overall, we suggest that the cumulative CO2 contribution from weakly degassing volcanoes in the hydrothermal stage of activity may be significant at the global scale

    Perspectives for sustainable development of Nordic aquaculture:The Paban-Report

    Aquaculture is one of the fastest growing productions and the value of aquaculture is now about to pass the value of capture fisheries. Among the Nordic countries, Norway in particular has been able to create a large aquaculture industry with high growth rates over a number of years. The other Nordic countries have only had limited growth, but have a high unutilised potential. There is a need to identify opportunities and limitations for increased growth with due care for sustainability and environment. This report identifies seven focus areas with special potential for creation of a sustainable, competitive Nordic aquaculture sector. These areas are new feeds, better use and reuse of nutrients (nitrogen, phosphorus and carbon), value adding of by-products, technological and regional development, domestication of new species and reduction of energy consumption. The aquaculture sector with its strengths and weaknesses is described for each of the Nordic countries.The report is compiled on basis of contributions from a Nordic group of experts. It was presented at a seminar on Green Growth at the annual meeting of the Nordic Ministers of Fisheries in Trondheim, July 2012

    Respiratory Physiology of European Plaice (<i>Pleuronectes platessa</i>) Exposed to <i>Prymnesium parvum</i>

    During the last century, the blooms of the toxic haptophyte Prymnesium parvum have been responsible for massive fish kills in both aquaculture and wild populations. Despite decades of research, the ichthyotoxic properties of P. parvum, and how this alga affects fish, is still debated. Using a novel device to measure the respirometry, ventilation volume, ventilation frequency, oxygen extraction, and oxygen consumption of undisturbed European plaice (Pleuronectes platessa) were acquired during exposure to two algal species as well as hypoxia. Fourteen fish (258 &#177; 44 g) were initially exposed to severe hypoxia and left to recover for at least 48 h. Half of these fish were then exposed to known harmful concentrations of P. parvum (median &#177; standard deviation (SD); 2.6 &#215; 105 &#177; 0.6 &#215; 105 cells mL&#8722;1), while the remaining half were exposed to the non-toxic alga Rhodomonas salina (median &#177; SD; 3.2 &#215; 105 &#177; 0.7 &#215; 105 cells mL&#8722;1). During exposure to severe hypoxia, all of the fish were able to maintain oxygen consumption by increasing the ventilation volume. The results from fish that were exposed to P. parvum showed a significant decrease in oxygen extraction (median &#177; SD; 52.6 &#177; 6.9 percentage points) from pre-exposure to the end of the experiment, as opposed to fish exposed to R. salina, which were unaffected. These results indicate that suffocation affects the European plaice when exposed to P. parvum. The observed severe decrease in oxygen extraction can be ascribed to either damage of the gill epithelia or increased mucus secretion on the gills, as both would limit the transfer of oxygen, and both have been observed

    Micrometre-scale deformation observations reveal fundamental controls on geological rifting

    Many of the world’s largest volcanic eruptions are associated with geological rifting where major fractures open at the Earth’s surface, yet fundamental controls on the near-surface response to the rifting process are lacking. New high resolution observations gleaned from seismometer data during the 2014 Bárðarbunga basaltic dyke intrusion in Iceland allow us unprecedented access to the associated graben formation process on both sub-second and micrometre scales. We find that what appears as quasi steady-state near-surface rifting on lower resolution GPS observation comprises discrete staccatolike deformation steps as the upper crust unzips through repetitive low magnitude (MW < 0) failures on fracture patches estimated between 300 m2 and 1200 m2 in size. Stress drops for these events are one to two orders of magnitude smaller than expected for tectonic earthquakes, demonstrating that the uppermost crust in the rift zone is exceptionally weak

    Prevention of ophthalmia neonatorum caused by 'Neisseria gonorrhoea' using a fatty acid-based formulation

    Ophthalmia neonatorum, also called neonatal conjunctivitis, acquired during delivery can occur in the first 28 days of life. Commonly caused by the bacterial pathogen Neisseria gonorrhoeae, infection can lead to corneal scarring, perforation of the eye, and blindness. One approach that can be taken to prevent the disease is the use of an ophthalmic prophylaxis, which kills the bacteria on the surface of the eye shortly after birth. Current prophylaxes are based on antibiotic ointments. However, N. gonorrhoeae is resistant to many antibiotics and alternative treatments must be developed before the condition becomes untreatable. This study focused on developing a fatty acid-based prophylaxis. For this, 37 fatty acids or fatty acid derivatives were screened in vitro for fast antigonococcal activity. Seven candidates were identified as bactericidal at 1 mM. These seven were subjected to irritation testing using three separate methods: the bovine corneal opacity and permeability (BCOP) test; the hen’s egg test—chorioallantoic membrane (HET-CAM); and the red blood cell (RBC) lysis assay. The candidates were also tested in artificial tear fluid to determine whether they were effective in this environment. Four of the candidates remained effective. Among these, two lead candidates, monocaprin and myristoleic acid, displayed the best potential as active compounds in the development of a fatty acid-based prophylaxis for prevention of ophthalmia neonatorum
