    Didymosphenia geminata: Algal blooms in oligotrophic streams and rivers

    In recent decades, the diatom Didymosphenia geminata has emerged as nuisance species in river systems around the world. This periphytic alga forms large “blooms” in temperate streams, presenting a counterintuitive result: the blooms occur primarily in oligotrophic streams and rivers, where phosphorus (P) availability typically limits primary production. The goal of this study is to examine how high algal biomass is formed under low P conditions. We reveal a biogeochemical process by which D. geminata mats concentrate P from flowing waters. First, the mucopolysaccaride stalks of D. geminata adsorb both iron (Fe) and P. Second, enzymatic and bacterial processes interact with Fe to increase the biological availability of P. We propose that a positive feedback between total stalk biomass and high growth rate is created, which results in abundant P for cell division. The affinity of stalks for Fe in association with iron-phosphorus biogeochemistry suggest a resolution to the paradox of algal blooms in oliogotrophic streams and rivers

    Metabolic alteration of urinary steroids in pre- and post-menopausal women, and men with papillary thyroid carcinoma

    <p>Abstract</p> <p>Background</p> <p>To evaluate the metabolic changes in urinary steroids in pre- and post-menopausal women and men with papillary thyroid carcinoma (PTC).</p> <p>Methods</p> <p>Quantitative steroid profiling combined with gas chromatography-mass spectrometry was used to measure the urinary concentrations of 84 steroids in both pre- (n = 21, age: 36.95 ± 7.19 yr) and post-menopausal female (n = 19, age: 52.79 ± 7.66 yr), and male (n = 16, age: 41.88 ± 8.48 yr) patients with PTC. After comparing the quantitative data of the patients with their corresponding controls (pre-menopause women: n = 24, age: 33.21 ± 10.48 yr, post-menopause women: n = 16, age: 49.67 ± 8.94 yr, male: n = 20, age: 42.75 ± 4.22 yr), the levels of steroids in the patients were normalized to the mean concentration of the controls to exclude gender and menopausal variations.</p> <p>Results</p> <p>Many urinary steroids were up-regulated in all PTC patients compared to the controls. Among them, the levels of three active androgens, androstenedione, androstenediol and 16α-hydroxy DHEA, were significantly higher in the pre-menopausal women and men with PTC. The corticoid levels were increased slightly in the PTC men, while progestins were not altered in the post-menopausal PTC women. Estrogens were up-regulated in all PTC patients but 2-hydroxyestrone and 2-hydroxy-17β-estradiol were remarkably changed in both pre-menopausal women and men with PTC. For both menopausal and gender differences, the 2-hydroxylation, 4-hydroxylation, 2-methoxylation, and 4-methoxylation of estrogens and 16α-hydroxylation of DHEA were differentiated between pre- and post-menopausal PTC women (<it>P </it>< 0.001). In particular, the metabolic ratio of 2-hydroxyestrone to 2-hydroxy-17β-estradiol, which could reveal the enzyme activity of 17β-hydroxysteroid dehydrogenase, showed gender differences in PTC patients (<it>P </it>< 1 × 10<sup>-7</sup>).</p> <p>Conclusions</p> <p>These results are expected be helpful for better understanding the pathogenic differences in PTC according to gender and menopausal conditions.</p

    Genome-Wide Analysis of Glucocorticoid Receptor Binding Regions in Adipocytes Reveal Gene Network Involved in Triglyceride Homeostasis

    Glucocorticoids play important roles in the regulation of distinct aspects of adipocyte biology. Excess glucocorticoids in adipocytes are associated with metabolic disorders, including central obesity, insulin resistance and dyslipidemia. To understand the mechanisms underlying the glucocorticoid action in adipocytes, we used chromatin immunoprecipitation sequencing to isolate genome-wide glucocorticoid receptor (GR) binding regions (GBRs) in 3T3-L1 adipocytes. Furthermore, gene expression analyses were used to identify genes that were regulated by glucocorticoids. Overall, 274 glucocorticoid-regulated genes contain or locate nearby GBR. We found that many GBRs were located in or nearby genes involved in triglyceride (TG) synthesis (Scd-1, 2, 3, GPAT3, GPAT4, Agpat2, Lpin1), lipolysis (Lipe, Mgll), lipid transport (Cd36, Lrp-1, Vldlr, Slc27a2) and storage (S3-12). Gene expression analysis showed that except for Scd-3, the other 13 genes were induced in mouse inguinal fat upon 4-day glucocorticoid treatment. Reporter gene assays showed that except Agpat2, the other 12 glucocorticoid-regulated genes contain at least one GBR that can mediate hormone response. In agreement with the fact that glucocorticoids activated genes in both TG biosynthetic and lipolytic pathways, we confirmed that 4-day glucocorticoid treatment increased TG synthesis and lipolysis concomitantly in inguinal fat. Notably, we found that 9 of these 12 genes were induced in transgenic mice that have constant elevated plasma glucocorticoid levels. These results suggested that a similar mechanism was used to regulate TG homeostasis during chronic glucocorticoid treatment. In summary, our studies have identified molecular components in a glucocorticoid-controlled gene network involved in the regulation of TG homeostasis in adipocytes. Understanding the regulation of this gene network should provide important insight for future therapeutic developments for metabolic diseases

    An Exploratory Study Mapping Approaches to Teaching Writing in Engineering

    Approaches to teaching writing to engineers vary widely across institutions, from standalone courses housed in English departments to fully integrated writing-in-the-disciplines programs, as well as a range of models in between that represent partnerships between writing and technical faculty. But little is known about how widely each approach is used, who is responsible for instruction, or what the overall goals are. In this exploratory study, we present an initial meta-analysis to map current approaches to engineering writing instruction by analyzing papers presented at American Society for Engineering Education (ASEE) conferences between 2012 and 2022. This exploratory study is a first step in a larger mapping project designed to better understand not only how engineering writing is taught and, perhaps more importantly, what approaches to engineering writing instruction are effective in what contexts and why. Such mapping can facilitate a deeper dialogues locally, nationally, and globally about the strengths and limitations of various approaches
