    Maternal undernutrition and the ovine acute phase response to vaccination

    Background: The acute phase response is the immediate host response to infection, inflammation and trauma and can be monitored by measuring the acute phase proteins (APP) such as haptoglobin ( Hp) or serum amyloid A (SAA). The plane of nutrition during pregnancy is known to affect many mechanisms including the neuroendocrine and neuroimmune systems in neonatal animals but effects on the APP are unknown. To investigate this phenomenon the serum concentration of Hp and SAA was initially determined in non-stimulated lambs from 3 groups (n = 10/group). The dams of the lambs of the respective groups were fed 100% of requirements throughout gestation (High/High; HH); 100% of requirements for the first 65 d of gestation followed by 70% of requirements until 125 d from when they were fed 100% of requirements (High/Low; HL); 65% of liveweight maintenance requirements for the first 65 d gestation followed by 100% of requirements for the remainder of pregnancy ( Low/High; LH). The dynamic APP response in the lambs was estimated by measuring the concentration of Hp and SAA following routine vaccination with a multivalent clostridial vaccine with a Pasteurella component, Heptavac P (TM) following primary and secondary vaccination. Results: The Hp and SAA concentrations were significantly lower at the time of vaccination ( day 8-14) than on the day of birth. Vaccination stimulated the acute phase response in lambs with increases found in both Hp and SAA. Maternal undernutrition led to the SAA response to vaccination being significantly lower in the HL group than in the HH group. The LH group did not differ significantly from either the HH or HL groups. No significant effects of maternal undernutrition were found on the Hp concentrations. A significant reduction was found in all groups in the response of SAA following the second vaccination compared to the response after the primary vaccination but no change occurred in the Hp response. Conclusion: Decreased SAA concentrations, post-vaccination, in lambs born to ewes on the HL diet shows that maternal undernutrition prior to parturition affects the innate immune system of the offspring. The differences in response of Hp and SAA to primary and secondary vaccinations indicate that the cytokine driven APP response mechanisms vary with individual AP

    Acute phase protein response in an experimental model of ovine caseous lymphadenitis

    <p>Abstract</p> <p>Background</p> <p>Caseous lymphadenitis (CLA) is a disease of small ruminants caused by <it>Corynebacterium pseudotuberculosis</it>. The pathogenesis of CLA is a slow process, and produces a chronic rather than an acute disease state. Acute phase proteins (APP) such as haptoglobin (Hp) serum amyloid A (SAA) and α<sub>1 </sub>acid glycoprotein (AGP) are produced by the liver and released into the circulation in response to pro-inflammatory cytokines. The concentration of Hp in serum increases in experimental CLA but it is not known if SAA and AGP respond in parallel or have differing response profiles.</p> <p>Results</p> <p>The concentration in serum of Hp, SAA and AGP in 6 sheep challenged with 2 × 10<sup>5 </sup>cells of <it>C. pseudotuberculosis </it>showed significant increases (P < 0.05) compared to 3 unchallenged control sheep. By day 7 post infection. (p.i.) the Hp and SAA concentrations reached mean (± SEM) values of 1.65 ± 0.21 g/L and 18.1 ± 5.2 mg/L respectively. Thereafter, their concentrations fell with no significant difference to those of the control sheep by day 18 p.i.. In contrast, the serum AGP concentration in infected sheep continued to rise to a peak of 0.38 ± 0.05 g/L on day 13 p.i., after which a slow decline occurred, although the mean concentration remained significantly higher (P < 0.05) than the control group up to 29 days p.i.. Specific IgG to phospholidase D of <it>C. pseudotuberculosis </it>became detectable at 11 days p.i. and continued to rise throughout the experiment.</p> <p>Conclusion</p> <p>The serum concentrations of Hp, SAA and AGP were raised in sheep in an experimental model of CLA. An extended response was found for AGP which occurred at a point when the infection was likely to have been transforming from an acute to a chronic phase. The results suggest that AGP could have a role as a marker for chronic conditions in sheep.</p

    Essential Oil Headspace Volatiles Prevent Invasive Box Tree Moth (Cydalima perspectalis) Oviposition—Insights from Electrophysiology and Behaviour

    The box tree moth (Cydalima perspectalis Walker) is an invasive species in Europe causing severe damage both in natural and ornamental boxwood (Buxus spp.) vegetation. Pest management tactics are often based on the use of chemical insecticides, whereas environmentally-friendly control solutions are not available against this insect. The application of essential oils may provide effective protection against oviposition and subsequent larval damage. Oviposition deterrence of cinnamon, eucalyptus and lavender essential oils was tested on female C. perspectalis in behavioural bioassays. Our results indicate that all the studied essential oils may be adequate deterrents; however, cinnamon oil exhibited the strongest effect. To determine the physiologically active compounds in the headspace of the essential oils, gas chromatography coupled with electroantennography recordings were performed in parallel with gas chromatography-mass spectrometry to identify the volatile constituents. In addition, the release rates of various components from vial-wick dispensers were measured during the oviposition bioassay. These results may serve as a basis for the development of a practical and insecticide-free plant protection method against this invasive moth species

    Early modern human settlement of Europe north of the Alps occurred 43,500 years ago in a cold steppe-type environment.

    The first settlement of Europe by modern humans is thought to have occurred between 50,000 and 40,000 calendar years ago (cal B.P.). In Europe, modern human remains of this time period are scarce and often are not associated with archaeology or originate from old excavations with no contextual information. Hence, the behavior of the first modern humans in Europe is still unknown. Aurignacian assemblages--demonstrably made by modern humans--are commonly used as proxies for the presence of fully behaviorally and anatomically modern humans. The site of Willendorf II (Austria) is well known for its Early Upper Paleolithic horizons, which are among the oldest in Europe. However, their age and attribution to the Aurignacian remain an issue of debate. Here, we show that archaeological horizon 3 (AH 3) consists of faunal remains and Early Aurignacian lithic artifacts. By using stratigraphic, paleoenvironmental, and chronological data, AH 3 is ascribed to the onset of Greenland Interstadial 11, around 43,500 cal B.P., and thus is older than any other Aurignacian assemblage. Furthermore, the AH 3 assemblage overlaps with the latest directly radiocarbon-dated Neanderthal remains, suggesting that Neanderthal and modern human presence overlapped in Europe for some millennia, possibly at rather close geographical range.     Bisurea-Based Supramolecular Polymers for Tunable Biomaterials

    Water-soluble supramolecular polymers show great potential to develop dynamic biomaterials with tailored properties. Here, we elucidate the morphology, stability and dynamicity of supramolecular polymers derived from bisurea-based monomers. An accessible synthetic approach from 2,4-toluene diisocyanate (TDI) as the starting material is developed. TDI has two isocyanates that differ in intrinsic reactivity, which allows to obtain functional, desymmetrized monomers in a one-step procedure. We explore how the hydrophobic/hydrophilic ratio affects the properties of the formed supramolecular polymers by increasing the number of methylene units from 10 to 12 keeping the hydrophilic hexa(ethylene glycol) constant. All bisurea-based monomers form long, fibrous structures with 3-5 monomers in the cross-section in water, indicating a proper hydrophobic\hydrophilic balance. The stability of the supramolecular polymers increases with an increasing amount of methylene units, whereas the dynamic nature of the monomers decreases. The introduction of one Cy3 dye affords modified supramolecular monomers, which co-assemble with the unmodified monomers into fibrous structures. All systems show excellent water-compatibility and no toxicity for different cell-lines. Importantly, in cell culture media, the fibrous structures remain present, highlighting the stability of these supramolecular polymers in physiological conditions. The results obtained here motivate further investigation of these bisurea-based building blocks as dynamic biomaterial.</p

    The NANOGrav 15 yr Data Set: Constraints on Supermassive Black Hole Binaries from the Gravitational-wave Background

    El conjunto de datos de 15 años de NANOGrav muestra evidencias de la presencia de un fondo de ondas gravitacionales (GWB) de baja frecuencia. Aunque muchos procesos físicos pueden originar estas ondas gravitacionales de baja frecuencia, aquí analizamos la señal como procedente de una población de agujeros negros binarios supermasivos (SMBH) distribuidos por todo el Universo. Demostramos que los modelos astrofísicos de poblaciones binarias SMBH son capaces de reproducir tanto la amplitud como la forma del espectro de ondas gravitacionales de baja frecuencia observado. Aunque múltiples variaciones del modelo son capaces de reproducir el espectro GWB con nuestra precisión de medida actual, nuestros resultados subrayan la importancia de modelar con precisión la evolución binaria para producir espectros GWB realistas. Además, aunque unos parámetros razonables son capaces de reproducir las observaciones de 15 años, la amplitud implícita del GWB requiere que un gran número de parámetros se sitúen en los límites de los valores esperados o que un pequeño número de parámetros difieran notablemente de las expectativas estándar. While many physical processes can source such low-frequency gravitational waves, here we analyze the signal as coming from a population of supermassive black hole (SMBH) binaries distributed throughout the Universe. We show that astrophysically motivated models of SMBH binary populations are able to reproduce both the amplitude and shape of the observed low-frequency gravitational-wave spectrum. While multiple model variations are able to reproduce the GWB spectrum at our current measurement precision, our results highlight the importance of accurately modeling binary evolution for producing realistic GWB spectra. Additionally, while reasonable parameters are able to reproduce the 15 yr observations, the implied GWB amplitude necessitates either a large number of parameters to be at the edges of expected values or a small number of parameters to be notably different from standard expectations. While we are not yet able to definitively establish the origin of the inferred GWB signal, the consistency of the signal with astrophysical expectations offers a tantalizing prospect for confirming that SMBH binaries are able to form, reach subparsec separations, and eventually coalesce. As the significance grows over time, higher-order features of the GWB spectrum will definitively determine the nature of the GWB and allow for novel constraints on SMBH populations. © 2023.     Fear learning and aversive stimuli differentially change excitatory synaptic transmission in perisomatic inhibitory cells of the basal amygdala

    Inhibitory circuits in the basal amygdala (BA) have been shown to play a crucial role in associative fear learning. How the excitatory synaptic inputs received by BA GABAergic interneurons are influenced by memory formation, a network parameter that may contribute to learning processes, is still largely unknown. Here, we investigated the features of excitatory synaptic transmission received by the three types of perisomatic inhibitory interneurons upon cue-dependent fear conditioning and aversive stimulus and tone presentations without association. Acute slices were prepared from transgenic mice: one group received tone presentation only (conditioned stimulus, CS group), the second group was challenged by mild electrical shocks unpaired with the CS (unsigned unconditioned stimulus, unsigned US group) and the third group was presented with the CS paired with the US (signed US group). We found that excitatory synaptic inputs (miniature excitatory postsynaptic currents, mEPSCs) recorded in distinct interneuron types in the BA showed plastic changes with different patterns. Parvalbumin (PV) basket cells in the unsigned US and signed US group received mEPSCs with reduced amplitude and rate in comparison to the only CS group. Coupling the US and CS in the signed US group caused a slight increase in the amplitude of the events in comparison to the unsigned US group, where the association of CS and US does not take place. Excitatory synaptic inputs onto cholecystokinin (CCK) basket cells showed a markedly different change from PV basket cells in these behavioral paradigms: only the decay time was significantly faster in the unsigned US group compared to the only CS group, whereas the amplitude of mEPSCs increased in the signed US group compared to the only CS group. Excitatory synaptic inputs received by PV axo-axonic cells showed the least difference in the three behavioral paradigm: the only significant change was that the rate of mEPSCs increased in the signed US group when compared to the only CS group. These results collectively show that associative learning and aversive stimuli unpaired with CS cause different changes in excitatory synaptic transmission in BA perisomatic interneuron types, supporting the hypothesis that they play distinct roles in the BA network operations upon pain information processing and fear memory formation

    Mint a legtöbb sportágnak, így a versenyszerű kézilabdázásnak is megvannak a maga árnyoldalai. Kontaktsport révén gyakoriak a különböző sportsérülések és a krónikus sport okozta betegségek. A Pécsi Egyetemi Atlétikai Club férfi kézilabda csapata 2017 szeptembere óta a harmadosztályú férfi kézilabda bajnokságban szerepel. Ezért fontosnak tartottuk a körükben egy átfogó kutatást végezni, különös tekintettel a domináns és nem domináns felső végtag közötti különbségekre, a törzsizomzat állapotára, a testtartásukra, a gerincoszlop állapotára és a derékfájdalomra. Kutatásunkban 30 főt vizsgáltunk, antropometriai és szomatometriai vizsgálatokkal mértük fel a játékosokat a speciális tesztek és saját szerkesztésű önkitöltős kérdőív mellett. A mérések kimutatták, hogy domináns és nem domináns felső végtag között szignifikáns különbség van, ugyanígy a jobb és bal oldali vállízület mozgástartománya között. Azok a feltevések azonban, miszerint ez az erős dominancia a gerincoszlopot a domináns oldal irányába elhúzhatja, nem igazolódott. A derékfájással kapcsolatban a panaszok előfordulása nem gyakoribb a fiziológiástól eltérő testtartással rendelkezőknél, mint helyesebb testtartással élő társaiknál. Kutatásunk azonban kimutatta, hogy a fiatalabb játékosok körében gyakoribb a gerincferdülés és a rossz testtartás előfordulása

    Folyadékbiopszia-vizsgálatok alkalmazási lehetőségei az onkohematológiában

    A molekuláris biológiai technikák utóbbi évtizedekben látott fejlődésének köszönhetően a vérplazmában keringő biomolekuláknak, így például a tumor eredetű sejtmentes DNS-fragmentumoknak is korábban elképzelhetetlen mélységű vizsgálata vált elérhetővé. A minimális invazivitással járó folyadékbiopsziás mintavétel metasztatikus tumorok esetében lehetővé teszi eltérő anatómiai lokalizációval megjelenő genetikai aberrációk egyidejű feltérképezését, azok nyomon követését, illetve a klonális evolúció következtében megjelenő további abnormalitások kimutatását is. Ebből következően számos, szolid tumorokat vizsgáló, folyadékbiopsziás mintavételt alkalmazó tanulmány jelent meg az utóbbi évtizedben, néhány entitás vonatkozásában pedig az eljárás már a rutindiagnosztikában is alkalmazásra került. Onkohematológiában az ebből a szempontból legintenzívebben kutatott megbetegedések közé a diff úz nagy B-sejtes limfóma, a Hodgkin-limfóma, illetve a plazmasejtes mielóma tartozik. Saját, illetve irodalmi adatok alapján a tumor eredetű sejtmentes DNS vizsgálata hasznos lehet a kezelést megelőző prognózisbecslésben, a célozható molekuláris eltérések azonosításában, a terápiás hatékonyság és a minimális reziduális betegség monitorozásában, valamint a terápiarezisztens klónok kimutatásában.
