    New and Simple Approach for Preventing Postoperative Peritoneal Adhesions: Do not Touch the Peritoneum without Viscous Liquid—A Multivariate Analysis

    Background. Postoperative peritoneal adhesions (PPAs) are an unsolved and serious problem in abdominal surgery. Method. Viscous liquids of soybean oil, octyl methoxycinnamate, flax oil, aloe vera gel, and glycerol were used in five experiments, using the same methodology for each. Liquids were applied in the peritoneal cavity before and after mechanical peritoneal trauma. Results were evaluated by multivariate analysis. Results. Compared with the control group, macroscopic and microscopic adhesion values before (P < .001) and after (P < .05) application of viscous liquids significantly reduced PPAs. Values were significantly lower when liquids were applied before rather than after peritoneal trauma (P < .0001). Discussion. Viscous liquids injected into the peritoneal cavity before or after mechanical peritoneal trauma decrease PPA. Injection before trauma was more effective than after trauma. In surgical practice, PPA formation may be prevented or decreased by covering the peritoneal cavity with an appropriate viscous liquid before abdominal surgery

    Efficacy of adalimumab therapy in experimental rat sclerosing encapsulated peritonitis model

    Aim To investigate the efficacy of adalimumab treatment in an experimental rat sclerosing encapsulated peritonitis (SEP) model. Methods The study involved 40 Wistar albino rats divided into four groups: chlorhexidine (CH) group, control group, CH + adalimumab group, and CH + resting group. The control group received normal saline intraperitoneally (i.p.). Other groups received 0.1% CH gluconate, 15% ethanol, and normal saline mixture i.p. for three weeks in order to induce SEP. CH + adalimumab group received 5 mg/kg adalimumab i.p. at the beginning of week 4 and week 6, while CH + resting group was followed-up for three weeks without applying any procedure after the onset of SEP. Rats in groups CH and control group were sacrificed on day 21, and rats in group CH + adalimumab and CH + resting were sacrificed on day 42. All groups were evaluated for peritoneal thickness, inflammation, vascularization, and fibrosis. Results CH + adalimumab group showed a significant decrease in peritoneal thickness, fibrosis score, and vascular score compared with CH group and CH + resting group. Conclusion Adalimumab can prevent SEP development

    ASR suppression by FA, SF, EPP, FA in combination with SF and EPP were evaluated by both mortar bar and concrete prism test. Mortar bars were made based on JIS A 1146, meanwhile concrete prism bars were casted in accordance with Rilem AAR-3. Both specimens were stored in 40°C 100% R.H. controlled room. Mortar and concrete mixtures used reactive aggregate in pessimum proportion. The results indicated that FA in combination with SF and EPP showed smaller expansion compared to FA. The best of concrete mixtures in reducing expansion is combination of FA with SF (FA15SF10)

    A Pilot Study on the Isolation of Dental Pulp Stem Cells, Potential of Forming Colonies and Defining the Content of Stem Cells

    Objective: Dental pulp has been used as a source in various stem cells experiments in the recent years. The aim of this study was to isolate dental pulp stem cells in sterilized conditions, to define the content level of stem cells and to optimize the methods that were used in obtaining single cell suspension for cell culture experiments. Material and Methods: In this study, a third molar tooth was used. Dental pulp tissue was digested with collagenase Type 1 (3 mg/mL) in alpha-MEM containing 10% fetal calf serum to obtain single cell suspension. The cells were incubated in 6-well-plates with culture medium at 5% CO(2) for 72 hours at moist conditions. After 72 hours, the cells were examined by inverted microscopy for cell morphology and development of colonies and the ratio of CD34 positive cells were defined with flow cytometry. Results: The development of the cell colonies was observed in culture. Three different cell populations were defined from the culture and the ratios of these populations that expressed CD34 were 10.99%, 12.99% and 2.99%. Conclusion: Considering the high rate of CD34 positive cells and the capability of forming colonies in in vitro culture, dental pulp is a good stem cell source

    Infliximab Alleviates The Mortality, Mesenteric Hypoperfusion, Aortic Dysfunction, And Multiple Organ Damage In Septic Rats

    Tumor necrosis factor-alpha (TNF-alpha) is a pivotal mediator that triggers inflammatory process, oxidative stress, and multiple organ injury in sepsis. We investigated the effects of infliximab on survival, mesenteric artery blood flow (MBF), vascular reactivity, and oxidative and inflammatory injuries in cecal ligation and puncture (CLP)-induced sepsis. Wistar rats were divided into Sham, CLP, Sham+infliximab, and CLP+infliximab subgroups. Twenty-four hours before the operations, rats were injected intraperitoneally with infliximab (7 mg/kg) or vehicle (saline; 1 mL/kg). Twenty hours after the operations, MBF and phenylephrine responses of isolated aortic rings were measured. Tissue damages were examined biochemically and histopathologically. Furthermore, survival rates were monitored throughout 96 h. Infliximab improved survival, mesenteric perfusion, and aortic function after CLP. Increases of serum AST, ALT, LDH, BUN, Cr, and inflammatory cytokines (tumor necrosis factor-alpha, interleukin-1 beta, and interleukin-6) induced by CLP were blocked by infliximab. Infliximab prevented malondialdehyde elevations in septic liver, lung, spleen, and kidney tissues, as well as glutathione reductions in septic liver, spleen, and kidney tissues. Protective effects of infliximab on multiple organ damage were also observed histopathologically. Infliximab showed protective effects in sepsis due to its improvement effects on mesenteric perfusion, aortic function, and its anti-inflammatory and antioxidative effects.Wo

    Microwave-Assisted Synthesis Of Condensed 1,4-Dihydropyridines As Potential Calcium Channel Modulators

    This study reports the design, synthesis, and calcium channel modulatory activity evaluation of a series of 14 novel fused 1,4-dihydropyridine derivatives. The molecular design of the compounds was based on modifications of nifedipine, which is a calcium channel blocker. The compounds were achieved by one-pot microwave-assisted reaction of 4,4-dimethyl-1,3-cyclohexanedione, 5-chlorosalicylaldehyde/3,5-dichlorosalicylaldehyde, an appropriate alkyl acetoacetate, and ammonium acetate in ethanol according to a modified Hantzsch reaction. The structures of the compounds were confirmed by spectral methods and elemental analysis. To evaluate their relaxant activities, the maximum relaxant response (E-max) and pD(2) values of the compounds and nifedipine were determined on isolated rat aorta rings. The obtained results indicated that all compounds produced concentration-dependent relaxation on the rings possibly due to the blockade of calcium channels. The E-max values (a measure of efficacy) of five compounds were higher than those of nifedipine.WoSScopu
