    Retreatment of a 6-Canalled Mandibular First Molar with Four Mesial Canals: A Case Report

    Successful root canal treatment requires adequate knowledge regarding morphologic variations in root canal system of teeth. This report describes a six-canalled mandibular first molar with four mesial root canals requiring endodontic retreatment. The two additional canals in the mesial root were found during retreatment with the aid of illumination and magnification.     Protective Effects of Crocin Against Streptozotocin-Induced Oxidative Damage in Rat Striatum

    The study aimed to investigate the protective effects of crocin (Cro) against streptozotocin (STZ)-induced oxidative damage in rat striatum. Animals were randomly divided into four groups (five each). Group 1 (sham) were treated with normal saline (2 ml/kg, p.o.). Group 2 (STZ-lesioned or lesion) were injected with ICV-STZ (3 mg/kg bilaterally, on day 1 and 3) and treated with normal saline (2 ml/kg, p.o.) respectively, for 21 days. Group 3 (sham+Cro) were injected ICV on day 1 and 3 with artificial CSF and treated with crocin (100 mg/kg, p.o.) for 21 days. Group 4 (lesion+Cro) were injected with ICV STZ (3 mg/kg bilaterally, on day 1 and 3) and treated with crocin (100 mg/kg, p.o.) for 21 days. The homogenized striatum was used for measuring malondialdehyde (MDA), and total thiol contents besides glutathione peroxidase (GPx) activity. Crocin treatment resulted in a significant reduction in MDA concentration as compared to the STZ-lesioned rats. Moreover, crocin produced a significant elevation in total thiol content and GPx activity, as compared with STZ-lesioned group. The present findings provide evidence that crocin may have a therapeutic significance for neurodegenerative diseases such as Alzheimer's disease (AD)

    Potentiation of Indomethacin-Induced Anti-Inflammatory Response by Montelukast in Formalin-Induced Inflammation in Rats

    The leukotrienes and prostaglandins are biologically active metabolites derived from arachidonic acid. The leukotrienes have a role in inflammatory diseases such as allergic rhinitis, inflammatory bowel disease and asthma. Montelukast, a cysteinyl leukotriene receptor antagonist, is claimed to be effective in asthma. The present study aimed to assess the role of cysteinyl leukotriene receptor antagonist on peripheral inflammation and whether montelukast treatment enhances the anti-inflammatory effect of indomethacin. Anti-inflammatory response was measured using a plethysmometer. Histopathologic examination for leukocyte accumulation was done. Montelukast (0.5-2mg/kg, i.p.) produced a significant anti-inflammatory effect in dose dependent manner against formalin-induced rat paw oedema at 1h but not in 3 and 5 h. When indomethacin (5 mg/kg, i.p) was co-administered with montelukast (1 mg/kg, i.p), the anti-inflammatory effects of indomethacin were significantly increased as compared to the per se effect at 3 and 5 hour after formalin challenge. In histopathology it has been found that combination therapy significantly decreased migration of leucocytes into the site of inflammation. These results show that montelukast has anti-inflammatory properties in peripheral tissue and markedly potentiates the anti-inflammatory activity of indomethacin at 3 and 5 h. It is expected that combination of montelukast with cyclooxygenase inhibitor would prove to be a novel approach to manage complex inflammatory conditions

    Mandibular First and Second Molars with Three Mesial Canals: A Case Series

    Adequate cleaning, shaping and filling of the root canal system are mandatory for successful root canal treatment. Thorough knowledge of root canal morphology and unusual anatomy of the tooth is critical for the practitioner. The occurrence and location of the third mesial canal (Middle Mesial Canal) in mandibular first and second molars in relation to other two mesial canals that were treated in private practice were studied. In 27 clinical cases, the presence of a middle mesial canal was demonstrated. The third canal was located in the middle of the distance between the mesiobuccal and mesiolingual canals. This canal configuration was found in six second lower molars and twenty one first molars. Middle mesial canal in all of our cases joined to mesiobuccal or mesiolingual canals. None of the teeth consisted of three independent canals with three apical foramina. In conclusion, every attempt should be made to find and treat all root canals of a tooth

    Memantine Enhances the Effect of Olanzapine in Patients With Schizophrenia: A Randomized, Placebo-Controlled Study

    Glutamate dysregulation may be involved in the neuropathology of schizophrenia. Memantine, a drug approved by the FDA for the treatment of moderate to severe Alzheimer's disease, acts as a partial uncompetitive NMDA receptor antagonist. The aim of this study was to examine the efficacy of memantine as an adjunctive treatment to olanzapine in patients with schizophrenia. In this double-blind, placebo-controlled studies, patients with schizophrenia according to DSM-IV clinical criteria were selected. Patients were randomly assigned to receive either memantine (week 1:10 mg/day; weeks 2-6:20 mg/day) plus olanzapine (15-20 mg/day) or olanzapine plus placebo. At baseline, no statistically significant difference regarding the mean total PANSS scores between treatment groups was found. Results showed that memantine significantly improved the positive and negative PANSS score in patients maintained on olanzapine after six weeks compared to olanzapine alone (P<0.001). Furthermore, female patients showed significantly better response than males, especially in positive PANSS score. No significant changes in extrapyramidal symptoms were observed.These findings indicate that olanzapine efficacy might be augmented with memantine. Furthermore, this effect is more remarkable in female patients with schizophrenia

    Coronal Microleakage in Root Canals Obturated with Lateral Compaction, Warm Vertical Compaction and Guttaflow System

    INTRODUCTION: Root canal obturation seals the root canal system to prevent re-entry and/or growth of microorganisms. The provision of an appropriate restoration to coronally seal the access cavity affects the success of endodontic treatment. The purpose of this study was to evaluate the coronal microbial leakage in root canals that were either filled by lateral compaction, GuttaFlow or warm vertical compaction. MATERIALS AND METHODS: In this ex vivo study, 80 single-rooted human extracted teeth were randomly divided into three experimental groups (n=20) and two positive and negative control groups (n=10). The teeth in experimental groups were obturated with cold lateral compaction, GuttaFlow system or warm vertical compaction techniques. After sterilization of the whole system with gamma-ray, saliva leakage was tested using a split-chamber model. Specimens were monitored every 24 hours for 30 days. The data were analyzed using log-rank and Kaplan-Meier survival analysis tests. RESULTS: There were no significant differences in impeding saliva leakage between the three experimental groups (P&gt;0.05). CONCLUSION: Under the conditions of this ex vivo study, it can be concluded that the sealing ability of cold lateral compaction, warm vertical compaction and GuttaFlow system was comparable
