Osteoclast-like activity of U937 cells, peripheral blood mononuclear cells, and periodontal ligament fibroblasts subjected to mechanical stress by centrifugal force

Several in vitro models have investigated the consequences of mechanical stimuli on osteoclasts (OCs). However, the mechanisms whereby mechanical forces trigger osteoclast responses remain poorly understood, and the generation of reactive oxygen species (ROS) and their relationship with bone resorption in OCs under the influence of mechanical forces require investigation. The present work examined the role of centrifugal force application on ROS production and its effect on osteoclast activity and differentiation. Human U937 macrophage cells, peripheral blood mononuclear cells (PBMCs), and periodontal ligament (PDL) fibroblasts treated with polyethylene glycol (PEG) or N-acetylcysteine (NAC) were subject to centrifugal force. Osteoclast markers such as tartrate-resistant acid phosphatase (TRAP) and bone resorption activities were measured. ROS levels and actin ring formation were determined. Also, U937 cell responses to centrifugal force and PEG-induced fusion were studied. Individual cells subjected to centrifugal exertion increased their ROS levels, formed actin-like rings, revealed TRAP expression and bone resorption activities, and expressed typical osteoclast markers. Control PEG-fused U937 cells also showed these effects, and cell treatment with NAC stopped all these responses. Centrifugal force, as well as PEG-induced cell fusion, can promote osteoclast-like features, including oxidative stress. The present experimental model allowed us to understand the mechanisms underlying the osteoclast differentiation associated with ROS production stimulated by mechanical compressive force, where NAC can contribute to reducing this oxidative stress condition

The Research Journey as a Challenge Towards New Trends

The academic community of the department of Risaralda, in its permanent interest in evidencing the results of the research processes that are carried out from the Higher Education Institutions and as a product of the VI meeting of researchers of the department of Risaralda held in November 2021 presents its work: “The journey of research as a challenge towards new trends”, which reflects the result of the latest research and advances in different lines of knowledge in Agricultural Sciences, Health Sciences, Social Sciences and Technology and Information Sciences, which seek to solve and meet the demands of the different sectors. Texto digital.Presentamos una nueva edición del Boletín del Observatorio de Internacionalización Empresarial y Regional (OIER), una herramienta para la presentación de datos, contactos, casos y herramientas que resultan útiles para el tejido empresarial y el fortalecimiento de sus capacidades empresariales, para la comunidad académica y empresarial de la región. Esta edición, correspondiente al mes de octubre, es una edición especial en convenio con la Cámara de Comercio Aburrá Sur. Además de esto, es una edición concentrada en el continente africano, dada la importancia que se le ha dado a esta región en la estrategia de la política exterior del gobierno colombiano. Este boletín se divide en seis secciones de análisis de la internacionalización. La primera sección está dedicada al análisis del comercio internacional de Colombia con el mundo, en este número se analizó el comportamiento de las exportaciones e importaciones del país con el continente africano en el periodo enero-julio de 2023. Esto con la intensión de determinar el nivel de intercambio comercial con este continente de 54 países, en el que se presentan más 1500 dialectos y con el que Colombia mantiene una enorme distancia cultural. Nonsurgical periodontal therapy

ABSTRACT: In the treatment of periodontal diseases, we can rely on nonsurgical periodontal therapy, which has been scientifically proven as its effectiveness has been recognized. The main objective of this article is to demonstrate the usefulness of nonsurgical periodontal therapy, as evidenced in several studies that serve as guidelines, as well as microbiological aspects, effects on tissues and the importance of maintenance therapy after treatment completion. Methods: a review was conducted in order to revise this topic considering key periodontal concepts as a guideline. Results: nonsurgical periodontal therapy (NSPT) is a procedure that cannot and should not be carried out in a few sessions; the time required for its completion is extended as necessary in order to achieve cleaning and adjustment of the roots as completely as possible. Conclusions: Several authors have reported that reduction of microbiota remains between 14 and 180 days after therapy. This explains the regular periodontal maintenance appointments. They also indicate that the critical aspect of this therapy is not the choice between a surgical or a nonsurgical procedure, but a detailed and thorough cleaning by the dental professional and the good level of oral hygiene achieved by the patient.RESUMEN: En el tratamiento de las enfermedades periodontales contamos con la terapia periodontal no quirúrgica, la cual ha sido avalada científicamente mostrando su efectividad. El principal objetivo de este artículo es demostrar la efectividad de la terapia periodontal no quirúrgica evidenciada en múltiples estudios con relación a las indicaciones, aspectos microbiológicos, efectos en los tejidos y la importancia de la terapia de mantenimiento una vez finalizado el tratamiento. Métodos: se hizo una revisión con relación al tema en los últimos años teniendo como patrón los conceptos clave periodontales. Resultados: la terapia periodontal no quirúrgica (TPNQ) no es un procedimiento que pueda y deba realizarse en un corto tiempo y en pocas citas, el tiempo para su ejecución se amplía lo necesario en especial para lograr una limpieza y regularización de las raíces lo más completa posible. Conclusión: varios autores reportan que la reducción de la microbiota se mantuvo entre 14 y 180 días, luego de la terapia, esto justifica las citas periódicas de mantenimiento periodontal y estos señalan que el aspecto crítico de la terapia no es la escogencia entre un procedimiento quirúrgico o no quirúrgico, sino la limpieza detallada y completa por el profesional y el buen nivel de higiene bucal por parte del paciente
