    The unfulfilled gene is required for the development of mushroom body neuropil in Drosophila

    <p>Abstract</p> <p>Background</p> <p>The mushroom bodies (MBs) of <it>Drosophila </it>are required for complex behaviors and consist of three types of neurons, γ, α'/β' and α/β. Previously, roles for transcription factors in MB neuronal differentiation have only been described for a subset of MB neurons. We are investigating the roles of <it>unfulfilled </it>(<it>unf</it>; <it>HR51</it>, CG16801) in MB development. <it>unf </it>encodes a nuclear receptor that is orthologous to the nuclear receptors fasciculation of axons defective 1 (FAX-1) of the nematode and photoreceptor specific nuclear receptor (PNR) of mammals. Based on our previous observations that <it>unf </it>transcripts accumulate in MB neurons at all developmental stages and the presence of axon pathfinding defects in <it>fax-1 </it>mutants, we hypothesized that <it>unf </it>regulates MB axon growth and pathfinding.</p> <p>Results</p> <p>We show that <it>unf </it>mutants exhibit a range of highly penetrant axon stalling phenotypes affecting all neurons of the larval and adult MBs. Phenotypic analysis of <it>unf</it><sup><it>X1 </it></sup>mutants revealed that α'/β' and α/β neurons initially project axons but stall prior to the formation of medial or dorsal MB lobes. <it>unf</it><sup><it>Z0001 </it></sup>mutants form medial lobes, although these axons fail to branch, which results in a failure to form the α or α' dorsal lobes. In either mutant background, γ neurons fail to develop larval-specific dorsal projections. These mutant γ neurons undergo normal pruning, but fail to re-extend axons medially during pupal development. <it>unf</it><sup><it>RNAi </it></sup>animals displayed phenotypes similar to those seen in <it>unf</it><sup><it>Z0001 </it></sup>mutants. Unique asymmetrical phenotypes were observed in <it>unf</it><sup><it>X1</it></sup>/<it>unf</it><sup><it>Z0001 </it></sup>compound heterozygotes. Expression of <it>UAS-unf </it>transgenes in MB neurons rescues the larval and adult <it>unf </it>mutant phenotypes.</p> <p>Conclusions</p> <p>These data support the hypothesis that <it>unf </it>plays a common role in the development of all types of MB neurons. Our data indicate that <it>unf </it>is necessary for MB axon extension and branching and that the formation of dorsal collaterals is more sensitive to the loss of <it>unf </it>function than medial projections. The asymmetrical phenotypes observed in compound heterozygotes support the hypothesis that the earliest MB axons may serve as pioneers for the later-born MB neurons, providing evidence for pioneer MB axon guidance in post-embryonic development.</p

    A Black Deer at Black Cave: New Pictograph Radiocarbon Date for the Lower Pecos, Texas

    A Pecos River style painting of a black deer from Black Cave Annex (41VV76a) in southwest Texas was radiocarbon dated. Using plasma oxidation and accelerator mass spectrometry, we obtained an age of 1465 ± 40 RCYBP (2 sigma calibrated age range of A.D. 470-660). This age is younger than the accepted age range for Pecos River style paintings, which is approximately 4000-3000 years B.P. This new measurement in association with other younger dates prompts us to question whether the Pecos River style endured for a longer time period than previously thought. More radiocarbon research is needed in order to understand how this anomalous result might fit within the Lower Pecos Canyonlands rock art chronology

    Are Killer Bees Good for Coffee? The Contribution of a Paper\u27s Title and Other Factors to Its Future Citations

    How can the title of a paper affect its subsequent number of citations? We compared the citation rate of 5941 papers published in the journal Biological Conservation from 1968 to 2012 in relation to: paper length; title length; number of authors; paper age; presence of punctuation (colons, commas or question marks); geographic and taxonomic breadth; the word ‘method’; and the type of manuscript (article, review). The total number of citations increased in more recently published papers and thus we corrected citation rate (average number of citations per year since publication) by publication age. As expected, review papers had, on average, twice the number of citations compared to other types of articles. Papers with the greatest geographic or taxonomic breadth were cited up to twice as frequently as narrowly focused papers. Titles phrased as questions, shorter titles, and papers with more authors had slightly higher numbers of citations. However, overall, we found that the included parameters explained only 12% of the variability in citation rate. This suggests that finding a good title is necessary, but that other factors are more important to construct a well-cited paper. We suggest that to become highly cited, a primary requirement is that papers need to advance the science significantly and be useful to readers

    Annual sea-air CO2fluxes in the Bering Sea: insights from new autumn and winter observations of a seasonally ice-covered continental shelf

    High-resolution data collected from several programs have greatly increased the spatiotemporal resolution of pCO2(sw) data in the Bering Sea, and provided the first autumn and winter observations. Using data from 2008 to 2012, monthly climatologies of sea-air CO2 fluxes for the Bering Sea shelf area from April to December were calculated, and contributions of physical and biological processes to observed monthly sea-air pCO2 gradients (?pCO2) were investigated. Net efflux of CO2 was observed during November, December, and April, despite the impact of sea surface cooling on ?pCO2. Although the Bering Sea was believed to be a moderate to strong atmospheric CO2 sink, we found that autumn and winter CO2 effluxes balanced 65% of spring and summer CO2 uptake. Ice cover reduced sea-air CO2 fluxes in December, April, and May. Our estimate for ice-cover corrected fluxes suggests the mechanical inhibition of CO2 flux by sea-ice cover has only a small impact on the annual scale (&lt;2%). An important data gap still exists for January to March, the period of peak ice cover and the highest expected retardation of the fluxes. By interpolating between December and April using assumptions of the described autumn and winter conditions, we estimate the Bering Sea shelf area is an annual CO2 sink of ?6.8 Tg C yr?1. With changing climate, we expect warming sea surface temperatures, reduced ice cover, and greater wind speeds with enhanced gas exchange to decrease the size of this CO2 sink by augmenting conditions favorable for greater wintertime outgassing

    Bodyweight Perceptions among Texas Women: The Effects of Religion, Race/Ethnicity, and Citizenship Status

    Despite previous work exploring linkages between religious participation and health, little research has looked at the role of religion in affecting bodyweight perceptions. Using the theoretical model developed by Levin et al. (Sociol Q 36(1):157–173, 1995) on the multidimensionality of religious participation, we develop several hypotheses and test them by using data from the 2004 Survey of Texas Adults. We estimate multinomial logistic regression models to determine the relative risk of women perceiving themselves as overweight. Results indicate that religious attendance lowers risk of women perceiving themselves as very overweight. Citizenship status was an important factor for Latinas, with noncitizens being less likely to see themselves as overweight. We also test interaction effects between religion and race. Religious attendance and prayer have a moderating effect among Latina non-citizens so that among these women, attendance and prayer intensify perceptions of feeling less overweight when compared to their white counterparts. Among African American women, the effect of increased church attendance leads to perceptions of being overweight. Prayer is also a correlate of overweight perceptions but only among African American women. We close with a discussion that highlights key implications from our findings, note study limitations, and several promising avenues for future research

    Different paths to the modern state in Europe: the interaction between domestic political economy and interstate competition

    Theoretical work on state formation and capacity has focused mostly on early modern Europe and on the experience of western European states during this period. While a number of European states monopolized domestic tax collection and achieved gains in state capacity during the early modern era, for others revenues stagnated or even declined, and these variations motivated alternative hypotheses for determinants of fiscal and state capacity. In this study we test the basic hypotheses in the existing literature making use of the large date set we have compiled for all of the leading states across the continent. We find strong empirical support for two prevailing threads in the literature, arguing respectively that interstate wars and changes in economic structure towards an urbanized economy had positive fiscal impact. Regarding the main point of contention in the theoretical literature, whether it was representative or authoritarian political regimes that facilitated the gains in fiscal capacity, we do not find conclusive evidence that one performed better than the other. Instead, the empirical evidence we have gathered lends supports to the hypothesis that when under pressure of war, the fiscal performance of representative regimes was better in the more urbanized-commercial economies and the fiscal performance of authoritarian regimes was better in rural-agrarian economie

    Mitochondrial ATP fuels ABC transporter-mediated drug efflux in cancer chemoresistance

    Chemotherapy remains the standard of care for most cancers worldwide, however development of chemoresistance due to the presence of the drug-effluxing ATP binding cassette (ABC) transporters remains a significant problem. The development of safe and effective means to overcome chemoresistance is critical for achieving durable remissions in many cancer patients. We have investigated the energetic demands of ABC transporters in the context of the metabolic adaptations of chemoresistant cancer cells. Here we show that ABC transporters use mitochondrial-derived ATP as a source of energy to efflux drugs out of cancer cells. We further demonstrate that the loss of methylation-controlled J protein (MCJ) (also named DnaJC15), an endogenous negative regulator of mitochondrial respiration, in chemoresistant cancer cells boosts their ability to produce ATP from mitochondria and fuel ABC transporters. We have developed MCJ mimetics that can attenuate mitochondrial respiration and safely overcome chemoresistance in vitro and in vivo. Administration of MCJ mimetics in combination with standard chemotherapeutic drugs could therefore become an alternative strategy for treatment of multiple cancers

    Thaptomys Thomas 1915 (Rodentia, Sigmodontinae, Akodontini) with karyotypes 2n = 50, FN = 48, and 2n = 52, FN = 52: Two monophyletic lineages recovered by molecular phylogeny

    A novel karyotype with 2n = 50, FN = 48, was described for specimens of Thaptomys collected at Una, State of Bahia, Brazil, which are morphologically indistinguishable from Thaptomys nigrita, 2n = 52, FN = 52, found in other localities. It was hence proposed that the 2n = 50 karyotype could belong to a distinct species, cryptic of Thaptomys nigrita, once chromosomal rearrangements observed, along with the geographic distance, might represent a reproductive barrier between both forms. Phylogenetic analyses using maximum parsimony and maximum likelihood based on partial cytochrome b sequences with 1077 bp were performed, attempting to establish the relationships among the individuals with distinct karyotypes along the geographic distribution of the genus; the sample comprised 18 karyotyped specimens of Thaptomys, encompassing 15 haplotypes, from eight different localities of the Atlantic Rainforest. The intra-generic relationships corroborated the distinct diploid numbers, once both phylogenetic reconstructions recovered two monophyletic lineages, a northeastern clade grouping the 2n = 50 and a southeastern clade with three subclades, grouping the 2n = 52 karyotype. The sequence divergence observed between their individuals ranged from 1.9% to 3.5%

    Citizen science reveals widespread negative effects of roads on amphibian distributions

    Landscape structure is important for shaping the abundance and distribution of amphibians, but prior studies of landscape effects have been species or ecosystem-specific. Using a large-scale, citizen science-generated database, we examined the effects of habitat composition, road disturbance, and habitat split (i.e. the isolation of wetland from forest by intervening land use) on the distribution and richness of frogs and toads in the eastern and central United States. Undergraduates from nine biology and environmental science courses collated occupancy data and characterized landscape structure at 1617 sampling locations from the North American Amphibian Monitoring Program. Our analysis revealed that anuran species richness and individual species distributions were consistently constrained by both road density and traffic volume. In contrast, developed land around wetlands had small, or even positive effects on anuran species richness and distributions after controlling for road effects. Effects of upland habitat composition varied among species, and habitat split had only weak effects on species richness or individual species distributions. Mechanisms underlying road effects on amphibians involve direct mortality, behavioral barriers to movement, and reduction in the quality of roadside habitats. Our results suggest that the negative effects of roads on amphibians occur across broad geographic regions, affecting even common species, and they underscore the importance of developing effective strategies to mitigate the impacts of roads on amphibian populations
