    The neuroanatomy of autism – a developmental perspective

    Autism Spectrum Disorders (ASDs) are a heterogeneous group of neurodevelopmental disorders that are diagnosed solely on the basis of behaviour. A large body of work has reported neuroanatomical differences between individuals with ASD and neurotypical controls. Despite the huge clinical and genetic heterogeneity that typifies autism, some of these anatomical features appear to be either present in most cases or so dramatically altered in some that their presence is now reasonably well replicated in a number of studies. One such finding is the tendency towards overgrowth of the frontal cortex during the early postnatal period. Although these reports have been focused primarily on the presumed pathological anatomy, they are providing us with important insights into normal brain anatomy and are stimulating new ideas and hypotheses about the normal trajectory of brain development and the function of specific anatomical brain structures. The use of model systems that include genetic model organisms such as the mouse and, more recently, human induced pluripotent stem cell‐derived brain organoids to model normal and pathological human cortical development, is proving particularly informative. Here we review some of the neuroanatomical alterations reported in autism, with a particular focus on well‐validated findings and recent advances in the field, and ask what these observations can tell us about normal and abnormal brain development

    Erectile dysfunction and relationships : views of men with erectile dysfunction and their partners

    There has been limited previous research that has examined the views of both men with erectile dysfunction (ED) and their partners on the impact of ED on their sexuality, relationship and general functioning. The current study was designed to evaluate the above variables among men with ED and their partners. Participants for the current study were 40 heterosexual men with ED and their partners. All participants completed a questionnaire that evaluated their reaction to ED, their past and current sexual activity, their sexual and relationship satisfaction as well as their levels of self-esteem and quality of life (QOL). The results demonstrated that both men with ED and their partners reported a reduction in their levels of sexual activity since the development of ED and that they wanted to seek a solution to the problem. Men with ED demonstrated lower levels of self-esteem, QOL and sexual satisfaction than their partners but there were no differences between the partners in their level of relationship satisfaction. These findings demonstrate that ED has an impact on both the man and his partner. They also indicate the importance of including the man\u27s partner in the assessment and treatment of ED. <br /

    Self-driven soft imaging in liquid by means of photothermal excitation

    The use of a laser to induce oscillations of an atomic force microscopy cantilever provides a way to excite the dynamics of the system in a very controlled manner. This excitation scheme has been used to obtain reliable self-sustained oscillations, in air and in liquid environments, and to implement an additional control loop leading to a soft, low-interaction, working mode. The dynamics of the oscillating cantilever has been characterized, both theoretically and experimentally, and the effectiveness of the approach has been verified on a test sample. © 2011 American Institute of Physics.Peer Reviewe

    Sprouty1 regulates reversible quiescence of a self-renewing adult muscle stem cell pool during regeneration.

    Satellite cells are skeletal muscle stem cells capable of self-renewal and differentiation after transplantation, but whether they contribute to endogenous muscle fiber repair has been unclear. The transcription factor Pax7 marks satellite cells and is critical for establishing the adult satellite cell pool. By using a lineage tracing approach, we show that after injury, quiescent adult Pax7(+) cells enter the cell cycle; a subpopulation returns to quiescence to replenish the satellite cell compartment, while others contribute to muscle fiber formation. We demonstrate that Sprouty1 (Spry1), a receptor tyrosine kinase signaling inhibitor, is expressed in quiescent Pax7(+) satellite cells in uninjured muscle, downregulated in proliferating myogenic cells after injury, and reinduced as Pax7(+) cells re-enter quiescence. We show that Spry1 is required for the return to quiescence and homeostasis of the satellite cell pool during repair. Our results therefore define a role for Spry1 in adult muscle stem cell biology and tissue repair

    Electricity supply industry modeling for multiple objectives under demand growth uncertainty

    Appropriate energy–environment–economic (E3) modelling provides key information for policy makers in the electricity supply industry (ESI) faced with navigating a sustainable development path. Key challenges include engaging with stakeholder values and preferences, and exploring trade-offs between competing objectives in the face of underlying uncertainty. As a case study we represent the South African ESI using a partial equilibrium E3 modelling approach, and extend the approach to include multiple objectives under selected future uncertainties. This extension is achieved by assigning cost penalties to non-cost attributes to force the model's least-cost objective function to better satisfy non-cost criteria. This paper incorporates aspects of flexibility to demand growth uncertainty into each future expansion alternative by introducing stochastic programming with recourse into the model. Technology lead times are taken into account by the inclusion of a decision node along the time horizon where aspects of real options theory are considered within the planning process. Hedging in the recourse programming is automatically translated from being purely financial, to include the other attributes that the cost penalties represent. From a retrospective analysis of the cost penalties, the correct market signals, can be derived to meet policy goal, with due regard to demand uncertainty

    An FGFR1-SPRY2 Signaling Axis Limits Basal Cell Proliferation in the Steady-State Airway Epithelium.

    The steady-state airway epithelium has a low rate of stem cell turnover but can nevertheless mount a rapid proliferative response following injury. This suggests a mechanism to restrain proliferation at steady state. One such mechanism has been identified in skeletal muscle in which pro-proliferative FGFR1 signaling is antagonized by SPRY1 to maintain satellite cell quiescence. Surprisingly, we found that deletion of Fgfr1 or Spry2 in basal cells of the adult mouse trachea caused an increase in steady-state proliferation. We show that in airway basal cells, SPRY2 is post-translationally modified in response to FGFR1 signaling. This allows SPRY2 to inhibit intracellular signaling downstream of other receptor tyrosine kinases and restrain basal cell proliferation. An FGFR1-SPRY2 signaling axis has previously been characterized in cell lines in vitro. We now demonstrate an in vivo biological function of this interaction and thus identify an active signaling mechanism that maintains quiescence in the airway epithelium.This study was supported by the Medical Research Council (G0900424 to ER); Wellcome Trust clinical PhD fellowship (JJ).     Pharmacodynamic and cytotoxicity effects of Syzygium cordatum {S Ncik, 48 (UZ)} fruit-pulp extract in gastrointestinal tract infections

    Purpose: To evaluate the pharmacodynamic effect and cytotoxicity of S. cordatum pulp extract in the treatment of gastrointestinal tract infections.Methods: The air-dried fruit pulps were ground, extracted with 100 % methanol and screened for phytochemicals. Serial microdilution method was used to determine the antibacterial activity of the extract against Bacillus cereus (ATCC 10102), Staphylococcus aureus (ATCC 25925), Enterococcus hirae (ATCC 8043), Escherichia coli (ATCC 25922), Salmonella typhimurium (ATCC 700030), Klebsiella pneumonia (ATCC 4352), Pseudomonas aeruginosa (ATCC 7700), Vibrio fluvialis (AL 019) and Vibrio vulnificus (AL 042). The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay was used to assess the cytotoxicity of the extract. Antidiarrheal and antimotility activities were evaluated using castor oil-induced diarrhoea model in rats.Results: The extract revealed the presence of alkaloids, cardiac glycosides, flavonoids, saponins and terpenoids. Total phenolic content was 16.4 ± 1.8 μg/mg. The extract exhibited antibacterial activity with minimum inhibitory concentration (MIC) of as low as 3.13 mg/ml against B. cereus (ATCC 10102), S. aureus (ATCC 25925), E. hirae (ATCC 8043), P. aeruginosa (ATCC 7700) and K. pneumonia (ATCC 4352). Median inhibitory concentration (IC50) of 92 μg/ml and therapeutic index of 0.1 - 0.3 were exerted by the extract. In vivo antidiarrheal activity was 49 % at extract dose of 400 mg/kg, which was much higher than that of the control (0 %).Conclusion: The fruit-pulp extract of Syzyhium cordatum has both antibacterial, antimotility and antidiarrheal activities, and may therefore be clinically safe for use at low concentrations as an antidiarrheal agent.Keywords: Syzyhium cordatum, Antibacterial, Antidiarrheal, Antimotility, Cytotoxicit
