    Becoming a European Homegrown Jihadist

    How and why do people become involved in European homegrown jihadism? This book addresses this question through an in-depth study of the Dutch Hofstadgroup, infamous for containing the murderer of filmmaker Theo van Gogh, who was killed in November 2004 in Amsterdam, and for plotting numerous other terrorist attacks. The Hofstadgroup offers a window into the broader phenomenon of homegrown jihadism that arose in Europe in 2004 and is still with us today. Utilizing interviews with former Hofstadgroup participants and the extensive police files on the group, Becoming a European Homegrown Jihadist overcomes the scarcity of high-quality data that has hampered the study of terrorism for decades. The book advances a multicausal and multilevel understanding of involvement in European homegrown jihadism that is critical of the currently prevalent 'radicalization'-based explanatory frameworks. It stresses that the factors that initiate involvement are separate from those that sustain it, which in turn are again likely to differ from those that bring some individuals to actual acts of terrorism. This is a key resource for scholars of terrorism and all those interested in understanding the pathways that can lead to involvement in European homegrown jihadism

    Background and Preparatory Behaviours of Right-Wing Extremist Lone Actors: A Comparative Study

    The threat posed by lone actors ranks high on the list of terrorism-related security concerns. In recent years especially, discussions about these perpetrators have focused primarily on those associated with, or inspired by, Islamic State and other jihadist entities. However, a significant portion of lone actors actually hail from right-wing extremist milieus. This article serves to draw attention to this subcategory of lone-actor terrorists, with a particular focus on their backgrounds and pre-attack behaviours. To that end, two datasets are presented that allow a comparison to be made between right-wing extremist lone actors and other ideologically-motivated lone actors. While several differences are noted, perhaps the most surprising finding is the degree of similarity between right-wing extremist lone actors and those adhering to different ideological currents. The results contribute to a knowledge-base that can inform discussion about whether risk assessment tools and protocols should differentiate between ideological categories of lone actor terrorists

    Reintegrating Terrorists in the Netherlands: Evaluating the Dutch approach

    This article presents an in-depth evaluation of a specialized reintegration initiative within the Dutch Probation Service focused on individuals convicted or suspected of involvement in terrorism. Using 72 interviews with program staff as well as several of their clients, the authors assess the initiative’s program theory, its day-to-day implementation and provide a qualified assessment of its overall effectiveness in the 2016 to 2018 period. The results suggest that the initiative is based on a sound understanding of how and why individuals may deradicalize or disengage from terrorism behaviorally, but that it continues to face serious challenges in terms of accurately defining success and systematically gathering objective indicators of its attainment. As terrorism remains a key challenge for societies across the globe, the relevance of these findings extends beyond the Netherlands to all academics, policymakers and practitioners working to design, implement and assess terrorist reintegration programs

    Deep Brain Stimulation for Obsessive-Compulsive Disorder: A Meta-Analysis of Treatment Outcome and Predictors of Response

    Background Deep brain stimulation (DBS) has been proposed as an alternative to ablative neurosurgery for severe treatment-resistant Obsessive-Compulsive Disorder (OCD), although with partially discrepant results probably related to differences in anatomical targetting and stimulation conditions. We sought to determine the efficacy and tolerability of DBS in OCD and the existence of clinical predictors of response using meta-analysis. Methods We searched the literature on DBS for OCD from 1999 through January 2014 using PubMed/MEDLINE and PsycINFO. We performed fixed and random-effect meta-analysis with score changes (pre-post DBS) on the Yale-Brown Obsessive Compulsive Scale (Y-BOCS) as the primary-outcome measure, and the number of responders to treatment, quality of life and acceptability as secondary measures. Findings Thirty-one studies involving 116 subjects were identified. Eighty-three subjects were implanted in striatal areas anterior limb of the internal capsule, ventral capsule and ventral striatum, nucleus accumbens and ventral caudate 27 in the subthalamic nucleus and six in the inferior thalamic peduncle. Global percentage of Y-BOCS reduction was estimated at 45.1% and global percentage of responders at 60.0%. Better response was associated with older age at OCD onset and presence of sexual/religious obsessions and compulsions. No significant differences were detected in efficacy between targets. Five patients dropped out, but adverse effects were generally reported as mild, transient and reversible. Conclusions Our analysis confirms that DBS constitutes a valid alternative to lesional surgery for severe, therapy-refractory OCD patients. Well-controlled, randomized studies with larger samples are needed to establish the optimal targeting and stimulation conditions and to extend the analysis of clinical predictors of outcome

    PAXIP1 and STAG2 converge to maintain 3D genome architecture and facilitate promoter/enhancer contacts to enable stress hormone-dependent transcription

    How steroid hormone receptors (SHRs) regulate transcriptional activity remains partly understood. Upon activation, SHRs bind the genome together with a co-regulator repertoire, crucial to induce gene expression. However, it remains unknown which components of the SHR-recruited co-regulator complex are essential to drive transcription following hormonal stimuli. Through a FACS-based genome-wide CRISPR screen, we functionally dissected the Glucocorticoid Receptor (GR) complex. We describe a functional cross-talk between PAXIP1 and the cohesin subunit STAG2, critical for regulation of gene expression by GR. Without altering the GR cistrome, PAXIP1 and STAG2 depletion alter the GR transcriptome, by impairing the recruitment of 3D-genome organization proteins to the GR complex. Importantly, we demonstrate that PAXIP1 is required for stability of cohesin on chromatin, its localization to GR-occupied sites, and maintenance of enhancer-promoter interactions. In lung cancer, where GR acts as tumor suppressor, PAXIP1/STAG2 loss enhances GR-mediated tumor suppressor activity by modifying local chromatin interactions. All together, we introduce PAXIP1 and STAG2 as novel co-regulators of GR, required to maintain 3D-genome architecture and drive the GR transcriptional programme following hormonal stimuli.</p

    High quality of SARS-CoV-2 molecular diagnostics in a diverse laboratory landscape through supported benchmark testing and External Quality Assessment

    A two-step strategy combining assisted benchmark testing (entry controls) and External Quality Assessments (EQAs) with blinded simulated clinical specimens to enhance and maintain the quality of nucleic acid amplification testing was developed. This strategy was successfully applied to 71 diagnostic laboratories in The Netherlands when upscaling the national diagnostic capacity during the SARS-CoV-2 pandemic. The availability of benchmark testing in combination with advice for improvement substantially enhanced the quality of the laboratory testing procedures for SARS-CoV-2 detection. The three subsequent EQA rounds demonstrated high quality testing with regard to specificity (99.6% correctly identified) and sensitivity (93.3% correctly identified). Even with the implementation of novel assays, changing workflows using diverse equipment and a high degree of assay heterogeneity, the overall high quality was maintained using this two-step strategy. We show that in contrast to the limited value of Cq value for absolute proxies of viral load, these Cq values can, in combination with metadata on strategies and techniques, provide valuable information for laboratories to improve their procedures. In conclusion, our two-step strategy (preparation phase followed by a series of EQAs) is a rapid and flexible system capable of scaling, improving, and maintaining high quality diagnostics even in a rapidly evolving (e.g. pandemic) situation.</p
