    Clinical and pathological aspects and cerebellar lectin binding in cattle poisoned with Solanum fastigiatum var. fastigiatum and Solanum bonariense

    Microscopic and lectin histochemical studies were performed using the cerebella of 33 natural cases of Solanum fastigiatum var. fastigiatum intoxication in the cattle from southern Brazil and 2 natural and 4 experimental cases of Solanum bonariense from Uruguay. The following biotinylated lectins were used in both cases: WGA, sWGA, BS-I, Con-A, RCA-I, DBA, and UEA-I, with the addition of LCA in S. fastigiatum poisoning cases. Histologically, the lesions consisted of fine vacuolization, distention of portions of the Purkinje cells, axonal spheroids measuring 14-50 m in the granular cell layer and adjacent white matter and, proliferation of the Bergmann?s glia. Lectin histochemistry revealed strong reactivity of stored material in Purkinje neurons with the lectins sWGA, Con-A, and LCA in S. fastigiatum cases. A similar pattern was found in S. bonariense cases with a most intense reactions to WGA, and less intense reaction to Con-A, whereas BS-I and RCA-I binding was absent to poor in these neurons in all the cases studied.Lectin reactivity in Purkinje cells between cases was independent of cell damage (from mild to severe loss of neurons). Both S. fastigiatum and S. bonariense have similar lectin binding,  suggesting a similar pathogenesis. Since comparable binding patterns have been described in animals poisoned with swainsonine-containing plants, perhaps the toxins in these plants contain related glycosidaseinhibiting toxins or inhibit glycoprotein and lysosomal metabolism through some related mechanism. The results of this study showed that in spontaneous poisoning by S. fastigiatum and S. bonariense in the cattle, the pattern of lectin binding is similar to those observed in S. fastigiatum experimental conditions.Fil: Sant’Ana, Fabiano J.F..     Protein-energy malnutrition halts hemopoietic progenitor cells in the G0/G1 cell cycle stage, thereby altering cell production rates

    Protein energy malnutrition (PEM) is a syndrome that often results in immunodeficiency coupled with pancytopenia. Hemopoietic tissue requires a high nutrient supply and the proliferation, differentiation and maturation of cells occur in a constant and balanced manner, sensitive to the demands of specific cell lineages and dependent on the stem cell population. In the present study, we evaluated the effect of PEM on some aspects of hemopoiesis, analyzing the cell cycle of bone marrow cells and the percentage of progenitor cells in the bone marrow. Two-month-old male Swiss mice (N = 7-9 per group) were submitted to PEM with a low-protein diet (4%) or were fed a control diet (20% protein) ad libitum. When the experimental group had lost about 20% of their original body weight after 14 days, we collected blood and bone marrow cells to determine the percentage of progenitor cells and the number of cells in each phase of the cell cycle. Animals of both groups were stimulated with 5-fluorouracil. Blood analysis, bone marrow cell composition and cell cycle evaluation was performed after 10 days. Malnourished animals presented anemia, reticulocytopenia and leukopenia. Their bone marrow was hypocellular and depleted of progenitor cells. Malnourished animals also presented more cells than normal in phases G0 and G1 of the cell cycle. Thus, we conclude that PEM leads to the depletion of progenitor hemopoietic populations and changes in cellular development. We suggest that these changes are some of the primary causes of pancytopenia in cases of PEM.FAPESPCNP

    Cerebellar Cortical Degeneration in Cattle Poisoned With Solanum spp. in South America: An Epidemiological, Clinicopathological, Pathological, and Toxicological Review

    Cattle that consume Solanum bonariense L (= Solanum fastigiatum Willd.) or Solanum paniculatum L. develop a typical cerebellar cortical degeneration characterized by periodic episodes of ataxia, hypermetria, hyperesthesia, head and thoracic limb extension, opisthotonus, nystagmus, and falling to the side or backward. Histological lesions include vacuolation, degeneration, and loss of Purkinje cells. Axonal spheroids, microcavitations, and other changes of Wallerian degeneration in cerebellar granular layer and white matter are also observed. Neurotoxic compounds in Solanum spp. causing neurologic dysfunction in ruminants were not definitively elucidated. The same Solanaceae species are extensively used with culinary purposes or for the treatment of liver and gastrointestinal disorders as hangovers in humans. In the present paper, we review the epidemiology, clinical signs, and pathological hallmarks of poisoning by Solanum —S. bonariense L. (=S. fastigiatum Willd.) and S. paniculatum—with emphasis in histopathology, ultrastructural, and lectin- and immuno-histochemical changes in spontaneous and experimentally poisoned cattle in South America. The current knowledge of the pathogenesis of these bovine cerebellar cortical degenerations is discussed, and some advances in botanical and toxicological aspects of these Solanaceae species are presented, taking into account the potential risk of human poisoning.Fil: Verdes, Jose M..     Transforming growth factor-β (TGF-β) maintains follicular ultrastructure and stimulates preantral follicle growth in caprine ovarian tissue cultured in vitro

    O objetivo desse estudo foi investigar se o TGF-β afeta a sobrevivência, ativação e crescimento de folículos primordiais caprinos inclusos no córtex ovariano após o cultivo in vitro. Ovários de cabras foram coletados em abatedouro e fragmentos de tecido ovariano foram cultivados por um e sete dias em meio essencial mínimo alfa (α-MEM+) sozinho ou suplementado com TGF-β (1, 5, 10 ou 50ng/mL). Fragmentos ovarianos não cultivados e cultivados foram processados para análise histológica e ultraestrutural. Os resultados mostraram que, comparado ao controle fresco, houve diminuição no percentual de folículos morfologicamente normais em todos os tratamentos somente após sete dias de cultivo. O TGF-β não afetou a ativação folicular independente da concentração testada, contudo, o diâmetro folicular foi superior (P<0.05) no tratamento com 10ng/mL de TGF-β quando comparado ao controle fresco e aos demais tratamentos. Além disso, essa mesma concentração manteve a ultraestrutura normal dos folículos após sete dias de cultivo. Em conclusão, o TGF-β apresentou efeito adicional no crescimento folicular e na manutenção da integridade ultraestrutural de folículos pré-antrais caprinos inclusos no tecido ovariano quando utilizado na concentração de 10ng/mL durante sete dias de cultivo.The objectives of this study were to investigate whether TGF-β affect the survival, activation and further growth of goat primordial follicles enclosed in ovarian cortex after in vitro culture. Goat ovaries were collected from an abattoir and pieces of ovarian tissues were cultured for one or seven days in a supplemented alpha Minimum Essential Medium, alone or containing TGF-β (1, 5, 10 or 50ng/mL). Ovarian tissues from the fresh control as well as those cultured were processed for histological and ultrastructural studies. The results showed that when compared with fresh control, there was decrease in the percentages of histologically normal follicles in all treatments only after seven days culture. TGF-β did not affect the activation of preantral follicles regardless of its concentration, however, larger follicles diameter (P<0.05) was observed using 10ng/mL TGF-β than in the fresh control and other treatments. Moreover, this concentration maintained the normal ultrastructure after seven days of culture. In conclusion, TGF-β showed additional effect on the follicle growth and the maintenance of ultrastructural integrity of goat preantral follicles enclosed in ovarian tissue when used at 10ng/mL during seven days of culture

    Adequacies And Inadequacies In The Anthropometric And Dietetic Profiles Of Preschool Children

    Nutritional conditions and food patterns in preschool children are elements that emphasise the importance of health monitoring in this period of nutritional transition, both to ensure nutritional adequacy and how much to intervene in identified inadequacies. Hence, it may also constitute a strategy for public programs and school health services to make decisions. Objective: To analyse the anthropometric and dietary profiles of pre-schoolers of a pole city in Northeastern Brazil. Methods: This is a cross-sectional evaluative study with 114 children aged 2-5 years, of both genders, in three municipal centres of early childhood education. Anthropometry was used to measure weight and height, and the nutritional condition was assessed using the indexes Height for Age (H/A), Weight for Age (W/A), Weight for Height (W/H) and Body Mass Index for Age (BMI/A) in z-score values with classifications established by the World Health Organization. The food intake record was done by direct weighing of the food menu offered during a week in the three institutions. This procedure allowed for the evaluation of the nutritional composition of menus, from the estimates in percentages, average and standard deviations of total calories, macronutrients (proteins, carbohydrates and lipids) and micronutrients (calcium, iron, vitamins A and C), to the adjustments and comparisons to the reference values of the Recommended Dietary Allowances, National Research Council and the PNAE recommendations, by age stages, in full-time units, 1-3 years, 700 kcal; 4-5 years, 950 kcal (70% coverage prediction of daily nutritional requirements) and the part-time units, these same ranges of age, respectively, 200 and 270 kcal (20% coverage for forecasting nutritional needs daily), considering adequate consumption to that with a variation of up to 10% above or below 100% of these recommendations. Results: Most of the preschool children had adequate nutritional conditions, especially those of full-time units, with relative frequencies (W/A: 94.5%; W/H: 89.5%, BMI/A: 81.7%). There were registered also overweight percentages higher than the deficits, in the indices W/A (22.2%), P/E (33.3%) to municipal early childhood centre CMEI-A girls; the indices W/A, W/H, BMI/A (23.8% each) in CMEI B-boys. In relation to the adequacy of the food profile, there was only convergence between the offer and the recommendation of 70% coverage of the daily energy needs, for pre-schoolers of 4-5 years from the centre of full-time units (mean: 951.2 ± 172.3 kcal). As for nutrients, inadequacies had a trend in the coverage of the daily needs on the menu offered in part-time units. Conclusion: Although most children do not present indicative of nutritional risk, requires attention to food portion that was with inadequacies of nutritional condition, with emphasis on weight surplus, while the readjustments in per capita and in portions of the menu implemented in those locations.26223424
