    Vascular Endothelial Growth Factor Expression and its Application in Vascular Tissue Engineering

    This paper deals with the expression of vascular endothelial growth factor (vascular endothelial growth factor, VEGF) and its use in tissue engineering of vascular wall. During the work interaction of endothelial cells with the modified fibrin-based biomaterial into which vascular endothelial growth factor (VEGF-A121) has been incorporated was monitored. This modification supported the adhesion and growth of endothelial cells. Vascular endothelial growth factor VEGF-A121 is signal glycoprotein that activates transmembrane receptors on endothelial cells. VEGF-A121 is a key regulator in vasculogenesis, angiogenesis, proliferation, migration and survival of endothelial cells. In this work, this protein was heterologously expressed at a thioredoxin fusion partner in an expression system of E. coli Origami B (DE3). Recombinant VEGF-A121 was additionally coexpressed with bacterial chaperones GroEL/GroES for potential increase of its solubility and biological activity. In the next part of this work thin fibrin network was prepared by catalytic action of thrombin on the polystyrene-bound monolayer of fibrinogen. This network has been further enriched by vascular endothelial growth factor (VEGF-A121), which was covalently incorporated in it by enzyme activity of transglutaminase (factor XIIIa). The last..

    Preparation of expression vectors for NKp65 and KACL, new members of human NK cell receptor family

    Natural killer cells create an important part of innate immune system. Their importance lies in their ability to recognize and kill abnormal cells, especially tumour cells and virally infected ones, without previous activation. To recognize their targets, NK cells use a wide variety of surface receptors, both activating and inhibitory. If a ligand binds to an NK receptor, immune response is triggered. Examples of such ligand-receptor pairs are NKp80-AICL and NKRP1-LLT1 on human lymphocytes. Another ligand-receptor system of this kind is NKp65 and KACL, two recently discovered lectin receptors on human immunocytes. KACL is the last and most recently characterized member of CLEC2 receptor family in humans. Its expression is almost exclusively restricted to skin. NKp65, a close relative of NKp80, is a glycoprotein which stimulates NK92MI cell cytotoxicity upon KACL engagement. NKp65 has been shown to bind to KACL with a fairly high affinity by surface plasmon resonance measurement.     Ojeté pneumatiky jsou velkoobjemový odpad, který je buď skladován ve velkých hromadách, nebo používán k nejrůznějším účelům, například jako výplňový nebo překryvový materiál pro zakrytí odkališť s jemným materiálem, palivo pro cementářské pece nebo zdroj energie nebo uhlovodíků. Velká nahromadění pneumatik na skládkách vytváří podmínky pro vznik požárů, které je velmi obtížné uhasit a navíc při nich dochází ke kontaminaci životního prostředí toxickými zplodinami a pyrolýzními produkty. Předložený příspěvek rozebírá tuto problematiku a poskytuje základní informace k prevenci a represi při požárech velkých objemů pneumatik. Dále jsou v příspěvku uvedeny základní informace o pyrolýze pneumatik, jako jedné z možností recyklace.Waste scrap tyres are bulk volume waste and are landfill in big stack. On the other hand, waste scrap tyres can be used for several purpose, e.g. for restorative or overlay material for covering of pond with fine material, as a fuel for cement kiln or source of energy and hydrocarbons. Accumulation of large number of scrap tyres in landfill generates conditions for fire. Fire of scrap tyres is (i) very bad subdual and (ii) contamination of environment with toxic fumes and pyrolytic products can take place. This article deals with study of this problem and provides basic information of prevention and repression during fi re of bulk volume of scrap tyres. Further, basic information of pyrolysis of scrap tyres is described as a one of the options of recyclation

    MorfFlex CZ 2.0 is the Czech morphological dictionary developed originally by Jan Hajič as a spelling checker and lemmatization dictionary. MorfFlex is a flat list of lemma-tag-wordform triples. For each wordform, full inflectional information is coded in a positional tag. Wordforms are organized into entries (paradigm instances or paradigms in short) according to their formal morphological behavior. The paradigm (set of wordforms) is identified by a unique lemma. Apart from traditional morphological categories, the description also contains some semantic, stylistic and derivational information.
