    Mean platelet volume-to-platelet count ratio after elective cardiac surgical procedures is superior in reflecting platelets metabolic hyperactivity compared to other routine morphological platelet indices: A preliminary report

    Background: Excessive metabolic excitation of platelets after cardiac procedures may be related to some adverse events but assessment of their metabolic activity is not routine. The purpose of this study was to evaluate which of the basic platelet morphological parameters best reflects their metabolic status. Methods: The blood samples of 22cardiac surgical patients (mean age of 62.3 ± 10.3 years) were taken before surgery (BS), and 1, 24 and 48 hours after the operation. Correlations between morphological platelet parameters (platelet count [PLT], mean platelet volume [MPV], platelet distribution width [PDW] and MPV/PLT) and their metabolic activity (total concentration of malondialdehyde [MDA] and MDA/PLT) were estimated. Results: Significant decline in PLT after operation (from 223 ± 44 × 1012/L to 166 ± 57 × 1012/L) was accompanied by marked increase in MPV (from 8.4 ± 0.9 fL to 9.1 ± 1.2 fL) and no change of PDW. Consequently, MPV/PLT index increased significantly after procedures from (median with IQR) 0.038 (0.030–0.043) to 0.053 (0.043–0.078). Simultaneously, a significant increase in total platelet MDA content and MDA/PLT was noted reaching peak levels soon after operation. The strongest correlation was observed between MPV/PLT and MDA/PLT (r = 0.56; p < 0.001), although the others were also found to be significant (MDA/PLT vs. MPV; r = 0.35; MDA/PLT vs. PDW; r = 0.34). Conclusions: Among basic morphological parameters and indices, the MPV-to-PLT ratio reflects the best metabolic status of platelets in cardiac surgical patients

    Could Life Be based on Arsenic? Mistakes in GFAJ-1 Bacteria Investigation

    Life constitutes of six essentials elements: carbon, hydrogen, carbon, nitrogen, sulphur, phosphorus. However, “shadow biosphere” theory assumes the existence of alternative life pathway that uses radically different biochemical and molecular processes than currently known life. Paper by Wolfe-Simon et al. (2011) published in high-profiled journal Science described bacterial species GFAJ-1 with ability not only to maintain life processes in high extracellular concentrations of arsenic but also to substitute phosphorus with arsenic in DNA. This report caused a number controversies related to applied methodology and interpretation of obtained results. Case of Wolfe-Simon et al. publication demonstrates the important role which is played by criticism against obtained results as well as concern for correction of applied methods.Funkcjonowanie wszystkich poznanych organizmów żywych zależne jest od sześciu podstawowych pierwiastków: węgla, wodoru, tlenu, azotu, siarki i fosforu. Teoria „biosfery cieni” zakłada jednak możliwość istnienia alternatywnych ścieżek rozwoju życia, w którym procesy i biorące w nich udział molekuły biochemiczne radykalnie różnią się od obecnie znanych. Opublikowana w 2011 roku na łamach jednego z najbardziej prestiżowych czasopism naukowych, Science, praca Wolfe-Simon i wsp. opisywała gatunek bakterii, zdolny nie tylko do zachowania funkcji życiowych w warunkach wysokiego stężenia arsenu, ale również do substytuowania tym pierwiastkiem fosforu w cząsteczkach DNA. Doniesienie to wywołało szereg dyskusji dotyczących zastosowanej metodyki interpretacji otrzymanych danych. Przypadek publikacji Wolfe-Simon i wsp. pokazuje jak istotną rolę w badaniach naukowych pełni krytycyzm wobec otrzymywanych wyników, a także dbałość o poprawność zastosowanych metod

    Willingness to Receive the Booster COVID-19 Vaccine Dose in Poland

    COVID-19 vaccinations are essential to mitigate the pandemic and prevent severe SARS-CoV-2 infections. However, the serum antibody levels in vaccinated individuals gradually decrease over time, while SARS-CoV-2 is undergoing an evolution toward more transmissible variants, such as B.1.617.2, ultimately increasing the risk of breakthrough infections and further virus spread. This cross-sectional online study of adult Poles (n = 2427) was conducted in September 2021 (before a general recommendation to administer a booster COVID-19 vaccine dose in Poland was issued) to assess the attitude of individuals who completed the current vaccination regime toward a potential booster dose of the COVID-19 vaccine and identify potential factors that may influence it. Overall, 71% of participants declared willingness to receive a booster COVID-19 dose, with a low median level of fear of receiving it of 1.0 (measured by the 10-point Likert-type scale), which was increased particularly in those having a worse experience (in terms of severity of side effects and associated fear) with past COVID-19 vaccination. The lowest frequency of willingness to receive a booster dose (26.7%) was seen in the group previously vaccinated with Ad26.COV2.S. The majority of individuals vaccinated previously with mRNA vaccines wished to receive the same vaccine, while in the case of AZD1222, such accordance was observed only in 9.1%. The main reasons against accepting a booster COVID-19 dose included the side effects experienced after previous doses, the opinion that further vaccination is unnecessary, and safety uncertainties. Women, older individuals (≥50 years), subjects with obesity, chronic diseases, and pre-vaccination and post-vaccination SARS-CoV-2 infections, and those with a history of vaccination against influenza were significantly more frequently willing to receive a booster COVID-19 dose. Moreover, the majority of immunosuppressed individuals (88%) were willing to receive an additional dose. The results emphasize some hesitancy toward potential further COVID-19 vaccination in the studied group of Poles and indicate the main groups to be targeted with effective science communication regarding the booster doses

    Flow cytometry as a valuable tool to study cyanobacteria:A mini-review

    Flow cytometry (FCM) is routinely used in medical and veterinary diagnostics although it is also widely applied in environmental studies, including phytoplankton investigations. Cyanobacteria are wide-spread photosynthetic microorganisms that attract attention due to their ecology and potential toxicity. Therefore, novel research tools are being applied in their investigation. This paper characterizes FCM as a technique that enables photopigments (chlorophylls and phycocyanin) expressed by cyanobacteria to be excited and their emission to be subsequently detected. This feature not only allows cells to be counted in a rapid manner but also enables a wide range of potential applications in ecological and biochemical studies. The main advantages of FCM, such as rapid, automatic and precise measurements requiring small sample volumes, are also discussed in this paper along with challenges including analyses of filamentous cyanobacteria and signal overlapping. It is expected that FCM will continue to be used in some fields of cyanobacterial studies

    Abstract In this paper we have reviewed dermatotoxins produced by blue-green algae. Dermatotoxins are harmful compounds that target human skin. Blue-green algae (Cyanobacteria) are prokaryotic microorganisms, mainly found in the water environment -marine and freshwater. A number of species are able to synthesize several toxic substances that affect human health upon exposure to lyngbya-, aplysia-and debromoaplysiatoxin. Lipopolysaccharides present in cyanobacteria cell walls can be irritating to human skin. Therefore, surface waters for recreational use should be monitored for the presence of toxic species of blue-green algae

    Platelets in Fetal Growth Restriction: Role of Reactive Oxygen Species, Oxygen Metabolism, and Aggregation

    Fetal growth restriction (FGR) is mainly caused by failure of the uteroplacental unit. The exact pathogenesis remains unclear. The cause is thought to be related to abnormal platelet activation, which may result in microthrombus formation in the small vessels of the placenta. Reactive oxygen species (ROS) may initiate the pathological process of platelet activation. This study aimed to evaluate selected platelet parameters in pregnancy complicated by FGR and relate them to the severity of hemodynamic abnormalities. A total of 135 women (pregnant with FGR, with an uncomplicated pregnancy, and non-pregnant) were enrolled to study different platelet parameters: count (PLT), mean volume (MPV), ROS levels, intracellular oxygen level, oxygen consumption, and aggregation indices. No abnormalities in PLT and MPV were found in the FGR group, although it revealed increased ROS levels in platelets, lower platelet oxygen consumption, and intraplatelet deprivation. Aggregation parameters were similar as in uncomplicated pregnancy. No significant relationships were observed between hemodynamic abnormalities and the studied parameters. Platelets in pregnancies complicated by FGR may reveal an impaired oxidative metabolism, which may, in turn, lead to oxidative stress and, consequently, to an impaired platelet function. This study adds to the understanding of the role of platelets in the etiology of FGR

    Frequency and Nuisance Level of Adverse Events in Individuals Receiving Homologous and Heterologous COVID-19 Booster Vaccine

    This study aimed to compare the occurrence and nuisance of adverse events following administration of each COVID-19 vaccine dose between two groups: individuals given three doses of mRNA vaccine (homologous group, 3 × mRNA, n = 252) and those given two doses of adenoviral vector vaccine further boosted with mRNA vaccine (heterologous group, 2 × AZ + 1 × mRNA, n = 205). Although the studied groups differed significantly in the frequency and number of side effects after the first and second vaccine dose, no relevant differences were seen following the booster administration. Arm pain and fatigue were the most common effects, regardless of the vaccination group and vaccine dose. In the homologous group, female sex, lower BMI, and no history of regular influenza vaccination were associated with a higher frequency of side effects of a booster dose. In the heterologous group, the history of COVID-19 was associated with an increased number of side effects seen after a booster. In both groups, the number of side effects related to the first and second dose correlated with the number observed after administration of a booster dose. Individuals receiving a homologous booster reported a higher nuisance of side effects than the heterologous group. It was similar to the level reported after the second dose in both groups. The use of pharmaceuticals to counteract the side effects was more frequent after a first dose in the 2 × AZ + 1 × mRNA group, but higher after second dose in individuals receiving the 3 × mRNA vaccination scheme. The frequency of pharmaceutical use after a booster dose was similar in both groups (approx. 60%). Paracetamol was most frequently chosen, regardless of the group and vaccine dose. In addition, the vast majority of participants (93%) declared to accept future doses of the COVID-19 vaccine if their administration would be recommended. This study provides an overview of the response to homologous and heterologous mRNA vaccine booster dose that may be valuable in shaping accurate and honest communication with vaccinated individuals, especially in those regions which are yet to pursue booster strategies
