Travel Blogs in Marketing Research: Stories of Stratford, Canada
This paper provides a case study for the exploration of travel blogs for market research. The growth and popularity of travel blogs over the Internet influenced few tourism researchers to see the online narratives as valuable sources of information for destination marketing. This research analyzed the content of travel blogs related to trips to Stratford, Canada to establish the most talked about topics and the experiences surrounding these. Three phases of data analysis were completed to achieve both quantifiable and qualitative results reflecting the travel experiences. The findings suggest that Stratford is primarily seen as a cultural and culinary destination. Methodological and practical implications are discussed
Assessing Visitors' Satisfaction at Parks Canada Sites
This study addresses the measurement of satisfaction. In doing so, it proposes a hypothetical conceptual framework for examining visitors’ satisfaction with their experiences in nature-based settings. Visitors’ overall satisfaction with their experiences was examined in terms of its relationships to visitors’ satisfaction with various site attributes, to visitors’ perceived importance of interpretive programs to learning, and how it is affected by visitors’ age, gender (male or female) and visitation pattern. The responses of 1309 Visitor Information Program (VIP) surveys returned by visitors to two national sites in Nova Scotia (Port Royal National Historic Site and Kejimkujik National Park and National Historic Site) provided the data for this study’s investigation. Secondary data analyses revealed that visitors’ satisfaction with the site attributes has the strongest effect on their overall satisfaction with their experiences. While visitors’ perceived importance to learning was also found to be positively correlated to overall experience satisfaction, when other variable are taken into consideration, its effect on overall satisfaction was found not to be statistically significant. Amongst these variables, gender was found to explain a significant amount of variance in visitors’ overall satisfaction with experiences in nature-based settings. Moreover, visitors of different age groups differ significantly in their perceptions of importance of interpretation to learning, and in their satisfaction levels. Although the findings show high levels of satisfaction at both sites, these are taken with caution. A discussion of the issues related to the measurement of satisfaction is provided, along with recommendations for a more discriminant, valid and reliable satisfaction measurement instrument
The Image of Tourism Destinations: A Case of Dracula Tourism
Destination image is a concept created by different demand and supply agents. The balance between what is expected and what is being delivered is essential in promoting the destination. The main goal of this study is to describe the different destination images related to Dracula tourism in Transylvania. This is accomplished by looking at the Western tourists’ expectations and demands as expressed in their travel blogs, and by analyzing the supply side through the elicitation of information from locals and Bran Castle tour guides through qualitative interviews.
Qualitative content analysis was used to analyze the content, visual and audio information on sampled blogs and interviews. This method identified 11 different themes which captured the overall destination image. The themes focused primarily on historical and fictional images of the destination, on the type of tourists visiting Bran Castle, but also on the locals’ image and response to Dracula tourism.
The findings of this study reveal that the majority of Western bloggers visit Bran in search for Count Dracula; however, the Bran Castle tour guides focus on presenting the historical truth. Local residents, although seeing Dracula as a national hero, sometimes adopt the mythical image as a marketing or product development tactic in selling to visitors. Nevertheless, the stakeholders involved in the creation of the image each fit into different destination image frameworks, allowing for the creation of an overall destination image based on historical and fictional elements
Impact of safety-related dose reductions or discontinuations on sustained virologic response in HCV-infected patients: Results from the GUARD-C Cohort
BACKGROUND:
Despite the introduction of direct-acting antiviral agents for chronic hepatitis C virus (HCV) infection, peginterferon alfa/ribavirin remains relevant in many resource-constrained settings. The non-randomized GUARD-C cohort investigated baseline predictors of safety-related dose reductions or discontinuations (sr-RD) and their impact on sustained virologic response (SVR) in patients receiving peginterferon alfa/ribavirin in routine practice.
METHODS:
A total of 3181 HCV-mono-infected treatment-naive patients were assigned to 24 or 48 weeks of peginterferon alfa/ribavirin by their physician. Patients were categorized by time-to-first sr-RD (Week 4/12). Detailed analyses of the impact of sr-RD on SVR24 (HCV RNA <50 IU/mL) were conducted in 951 Caucasian, noncirrhotic genotype (G)1 patients assigned to peginterferon alfa-2a/ribavirin for 48 weeks. The probability of SVR24 was identified by a baseline scoring system (range: 0-9 points) on which scores of 5 to 9 and <5 represent high and low probability of SVR24, respectively.
RESULTS:
SVR24 rates were 46.1% (754/1634), 77.1% (279/362), 68.0% (514/756), and 51.3% (203/396), respectively, in G1, 2, 3, and 4 patients. Overall, 16.9% and 21.8% patients experienced 651 sr-RD for peginterferon alfa and ribavirin, respectively. Among Caucasian noncirrhotic G1 patients: female sex, lower body mass index, pre-existing cardiovascular/pulmonary disease, and low hematological indices were prognostic factors of sr-RD; SVR24 was lower in patients with 651 vs. no sr-RD by Week 4 (37.9% vs. 54.4%; P = 0.0046) and Week 12 (41.7% vs. 55.3%; P = 0.0016); sr-RD by Week 4/12 significantly reduced SVR24 in patients with scores <5 but not 655.
CONCLUSIONS:
In conclusion, sr-RD to peginterferon alfa-2a/ribavirin significantly impacts on SVR24 rates in treatment-naive G1 noncirrhotic Caucasian patients. Baseline characteristics can help select patients with a high probability of SVR24 and a low probability of sr-RD with peginterferon alfa-2a/ribavirin
The invertebrate lysozyme effector ILYS-3 is systemically activated in response to danger signals and confers antimicrobial protection in C. elegans
Little is known about the relative contributions and importance of antibacterial effectors in the nematode C. elegans, despite extensive work on the innate immune responses in this organism. We report an investigation of the expression, function and regulation of the six ilys (invertebrate-type lysozyme) genes of C. elegans. These genes exhibited a surprising variety of tissue-specific expression patterns and responses to starvation or bacterial infection. The most strongly expressed, ilys-3, was investigated in detail. ILYS-3 protein was expressed constitutively in the pharynx and coelomocytes, and dynamically in the intestine. Analysis of mutants showed that ILYS-3 was required for pharyngeal grinding (disruption of bacterial cells) during normal growth and consequently it contributes to longevity, as well as being protective against bacterial pathogens. Both starvation and challenge with Gram-positive pathogens resulted in ERK-MAPK-dependent up-regulation of ilys-3 in the intestine. The intestinal induction by pathogens, but not starvation, was found to be dependent on MPK-1 activity in the pharynx rather than in the intestine, demonstrating unexpected communication between these two tissues. The coelomocyte expression appeared to contribute little to normal growth or immunity. World of Crayfishâ„¢: a web platform towards real-time global mapping of freshwater crayfish and their pathogens
Freshwater crayfish are amongst the largest macroinvertebrates and play a keystone role in the ecosystems they occupy. Understanding the global distribution of these animals is often hindered due to a paucity of distributional data. Additionally, non-native crayfish introductions are becoming more frequent, which can cause severe environmental and economic impacts. Management decisions related to crayfish and their habitats require accurate, up-to-date distribution data and mapping tools. Such data are currently patchily distributed with limited accessibility and are rarely up-to-date. To address these challenges, we developed a versatile e-portal to host distributional data of freshwater crayfish and their pathogens (using Aphanomyces astaci, the causative agent of the crayfish plague, as the most prominent example). Populated with expert data and operating in near real-time, World of Crayfishâ„¢ is a living, publicly available database providing worldwide distributional data sourced by experts in the field. The database offers open access to the data through specialized standard geospatial services (Web Map Service, Web Feature Service) enabling users to view, embed, and download customizable outputs for various applications. The platform is designed to support technical enhancements in the future, with the potential to eventually incorporate various additional features. This tool serves as a step forward towards a modern era of conservation planning and management of freshwater biodiversity

NMR Structure of the Netrin-like Domain (NTR) of Human Type I Procollagen C-Proteinase Enhancer Defines Structural Consensus of NTR Domains and Assesses Potential Proteinase Inhibitory Activity and Ligand Binding
Procollagen C-proteinase enhancer (PCOLCE) proteins are extracellular matrix proteins that enhance the activities of procollagen C-proteinases by binding to the C-propeptide of procollagen I. PCOLCE proteins are built of three structural modules, consisting of two CUB domains followed by a C-terminal netrin-like (NTR) domain. While the enhancement of proteinase activity can be ascribed solely to the CUB domains, sequence homology of the NTR domain with tissue inhibitors of metalloproteinases suggest proteinase inhibitory activity for the NTR domain. Here we present the three-dimensional structure of the NTR domain of human PCOLCE1 as the first example of a structural domain with the canonical features of an NTR module. The structure rules out a binding mode to metalloproteinases similar to that of tissue inhibitors of metalloproteinases but suggests possible inhibitory function toward specific serine proteinases. Sequence conservation between 13 PCOLCE proteins from different organisms suggests a conserved binding surface for other protein partners
Genetic heterogeneity and recombination in type-3 human astroviruses
Human astroviruses (HAstVs) are important enteric pathogens and can be classified genetically and antigenically into eight types. During molecular surveillance for HAstVs in Italy, sequence analysis of the diagnostic region C (about 400 nucleotide in length), located on the capsid (ORF2) gene, identified a novel type-3 strain. Upon sequencing of the full-length ORF2, the type-3 HAstV strain was characterized as a novel ORF2 genetic lineage, designated as 3c. By converse, in the ORF1b the virus was more similar to type-1 HAstVs, rather than to type-3 strains, suggesting a recombination nature, with the crossover site being mapped to the ORF1b/ORF2 junction region. Region C sequences of similar type-3 HAstV identified from European and extra-European countries were retrieved in the databases, suggesting the global distribution of this novel type-3 lineage
