    Scavenging capacity of strawberry tree (Arbutus unedo L.) leaves on free radicals

    Despite strawberry tree (Arbutus unedo L.) leaves had a long use in traditional medicine due to its antiseptic, diuretic, astringent and depurative properties, the potential of their antioxidant activity are still lacking. Our study goals to assess the antioxidant and free radical scavenging potential of water, ethanol, methanol and diethyl ether extracts of A. unedo leaves. Total phenols content was achieved spectrophotometrically using Folin–Ciocalteau reagent with gallic acid as standard. Antioxidant activity was evaluated using three different methods: reducing power of iron (III)/ferricyanide complex assay, scavenging effect on DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals and scavenging effect on superoxide radicals by using the PMS–NADH–nitroblue tetrazolium system. Ethanol extracts of A. unedo leaves were the highest in reducing power (IC50 232.7 lg/mL) and DPPH scavenging effect (IC50 63.2 lg/mL) followed by water extracts (with IC50 of 287.7 and 73.7 lg/mL, respectively); whereas diethyl ether extracts were the lowest. In the scavenging on superoxide radical assay, methanol extracts obtained the best results (IC50 6.9 lg/mL). For all the methods tested the antioxidant activity was concentration dependent. In accordance with antioxidant activity, highest total phenols content were found in ethanol, followed by water, methanol and diethyl ether extract. The results indicated that A. unedo leaves are a potential source of natural antioxidants

    Formation of a Massive Black Hole at the Center of the Superbubble in M82

    We performed 12CO(1-0), 13CO(1-0), and HCN(1-0) interferometric observations of the central region (about 450 pc in radius) of M82 with the Nobeyama Millimeter Array, and have successfully imaged a molecular superbubble and spurs. The center of the superbubble is clearly shifted from the nucleus by 140 pc. This position is close to that of the massive black hole (BH) of >460 Mo and the 2.2 micron secondary peak (a luminous supergiant dominated cluster), which strongly suggests that these objects may be related to the formation of the superbubble. Consideration of star formation in the cluster based on the infrared data indicates that (1) energy release from supernovae can account for the kinetic energy of the superbubble, (2) the total mass of stellar-mass BHs available for building-up the massive BH may be much higher than 460 Mo, and (3) it is possible to form the middle-mass BH of 100-1000 Mo within the timescale of the superbubble.     Lentiviral Vector-Mediated Correction of a Mouse Model of Leukocyte Adhesion Deficiency Type I

    Leukocyte adhesion deficiency type I (LAD-I) is a primary immunodeficiency caused by mutations in the ITGB2 gene and is characterized by recurrent and life-threatening bacterial infections. These mutations lead to defective or absent expression of β2 integrins on the leukocyte surface, compromising adhesion and extravasation at sites of infection. Three different lentiviral vectors (LVs) conferring ubiquitous or preferential expression of CD18 in myeloid cells were constructed and tested in human and mouse LAD-I cells. All three hCD18-LVs restored CD18 and CD11a membrane expression in LAD-I patient-derived lymphoblastoid cells. Corrected cells recovered the ability to aggregate and bind to sICAM-1 after stimulation. All vectors induced stable hCD18 expression in hematopoietic cells from mice with a hypomorphic Itgb2 mutation (CD18(HYP)), both in vitro and in vivo after transplantation of corrected cells into primary and secondary CD18(HYP) recipients. hCD18(+) hematopoietic cells from transplanted CD18(HYP) mice also showed restoration of mCD11a surface co-expression. The analysis of in vivo neutrophil migration in CD18(HYP) mice subjected to two different inflammation models demonstrated that the LV-mediated gene therapy completely restored neutrophil extravasation in response to inflammatory stimuli. Finally, these vectors were able to correct the phenotype of human myeloid cells derived from CD34(+) progenitors defective in ITGB2 expression. These results support for the first time the use of hCD18-LVs for the treatment of LAD-I patients in clinical trials

    Dense Molecular Gas Associated with the Circumnuclear Star Forming Ring in the Barred Spiral Galaxy NGC 6951

    We present high resolution (3" - 5") observations of CO(1-0) and HCN(1-0) emission from the circumnuclear star forming ring in the barred spiral galaxy NGC 6951, a host of a type-2 Seyfert, using the Nobeyama Millimeter Array and 45 m telescope. We find that most of the HCN emission is associated with the circumnuclear ring, where vigorous star formation occurs. The HCN to CO integrated intensity ratio is also enhanced in the star forming ring; the peak value of HCN/CO ratio is 0.18, which is comparable to the ratio in the starbursts NGC 253 and M82. The formation mechanism of dense molecular gas has been investigated. We find that the shocks along the orbit crowding do not promote the formation of the dense molecular gas effectively but enhance the presence of low density GMCs. Instead, gravitational instabilities of the gas can account for the dense molecular gas formation. The HCN/CO ratio toward the Seyfert nucleus of NGC 6951 is a rather normal value (0.086), in contrast with other Seyferts NGC 1068 and M51 where extremely high HCN/CO value of ~ 0.5 have been reported.Comment: 33 pages, 17 figures, to appear in the Astrophysical Journa

    Acquiring 21st Century Skills: gaining insight in the design and applicability of a serious game with 4C-ID

    Despite the growth of interest in serious games, there is little systematic guidance on how to assure a game fits the instruction required. Game design frameworks are still under development and do not help to articulate the educational merits of a game to a teacher nor fit with their background. In this paper we discuss the results of a GaLA workshop which examined how a widely applied instructional design model, 4C-ID, can ease the uptake of serious games by offering teachers a model fitting their background to assess games on the applicability for their learning contexts. The paper will introduce the 4C-ID model and its use in the CHERMUG project with the design of mini-games for research methods and statistics. Next, we will discuss how workshop participants used the 4C-ID model to evaluate two games on their applicability for a given learning context. Începând din 1997 am folosit o sutură sero-seroasă invaginantă în peste 1200 de LSG izolate sau asociate cu Duodenal Switch (laparotomic sau laparoscopic) pentru a acoperi zona de sutură. Gagner a descris procedeul în anul 2000.Background: Laparoscopic Sleeve Gastrectomy (LSG) is, even if popular, a non-standardized bariatric technique. The results varied and there are controversial issues such as: 1) Size of the sleeve; 2) Protection against esophageal-gastric junction (EGJ) leaks; 3) The use of partial antrectomy. We have always used, since 1997, in more than 1200 isolated or combined open or laparoscopic Duodenal Switch (LDS) a continue inverting Lembert-type sero-serosa suture to cover the staple-line.
