    Fluoxetine effects on periodontogenesis: histomorphometrical and immunohistochemical analyses in rats

    Reports have indicated that serotonin plays an important role in cell migration and differentiation during the organogenesis of several tissues, including the oral types. Administration of selective serotonin reuptake inhibitor (SSRI) drugs during pregnancy could affect the delivery of serotonin to embryonic tissues altering its development. Objective This study aimed to assess the effects of fluoxetine, a selective serotonin reuptake inhibitor, on the formation of the periodontal ligament during pregnancy and lactation in rat pups. Material and Methods Twelve pregnant rats of Wistar lineage were divided into four study groups. In the control group, 0.9% sodium chloride solution was administered orally, throughout the entire period of the 21 days of pregnancy (CG group) and in the CGL group, it was administrated during pregnancy and lactation (from day 1 of pregnancy to the 21st day after birth). Fluoxetine was administered orally at the dose of 20 mg/kg in a group treated during pregnancy only (FG group), and during pregnancy and lactation (FGL group). Histometrical, histochemical and immunohistochemical analysis of the maxillary first molar periodontium region of the 24 rat pups was made under light microscopy, and periodontal ligament collagen was qualitatively evaluated under a polarizing light microscope. Results The quantity of fibroblasts (p=0.006), osteoblasts (p=0.027) and cementoblasts (p=0.001) was reduced in pups from the rats that received fluoxetine during pregnancy and lactation. No alterations were seen in the collagen fibers. Conclusion These findings suggest that periodontal tissue may be sensitive to fluoxetine, and its interference in reducing periodontal cells depends on exposure time during lactation

    Neurofibromatosis type 1 (NF1) is a dominantly inherited multi-system disorder. Major features include pigmentary abnormalities, benign tumors of the nerve sheath (neurofibromas), malignant tumors, learning disabilities, and skeletal dysplasia. The NF1 gene functions as a tumor suppressor, but haploinsuffiency probably accounts for some aspects of the non-tumor phenotype. The protein product, neurofibromin, is a Ras GTPase-activating protein, and various Ras pathway inhibitors are being tested in preclinical models and clinical trials for effectiveness in treating NF1 complications. This month in BMC Medicine, a paper by Kolanczyk et al describes a preclinical mouse model for tibial dysplasia and provides evidence that the drug lovastatin – in use to treat cardiovascular disease – may be beneficial, opening the door to clinical trials in humans

    Intersectional Cre Driver Lines Generated Using Split-Intein Mediated Split-Cre Reconstitution

    Tissue and cell type highly specific Cre drivers are very rare due to the fact that most genes or promoters used to direct Cre expressions are generally expressed in more than one tissues and/or in multiple cell types. We developed a split-intein based split-Cre system for highly efficient Cre-reconstitution through protein splicing. This split-intein-split-Cre system can be used to intersect the expression patterns of two genes or promoters to restrict full-length Cre reconstitution in their overlapping domains. To test this system in vivo, we selected several conserved human enhancers to drive the expression of either Cre-N-intein-N, or intein-C-Cre-C transgene in different brain regions. In all paired CreN/CreC transgenic mice, Cre-dependent reporter was efficiently induced specifically in the intersectional expression domains of two enhancers. This split-intein based method is simpler to implement compared with other strategies for generating highly-restricted intersectional Cre drivers to study complex tissues such as the nervous system

    Electronic and magnetic properties of SnO2/CrO2 thin superlattices

    In this article, using first-principles electronic structure calculations within the spin density functional theory, alternated magnetic and non-magnetic layers of rutile-CrO2 and rutile-SnO2 respectively, in a (CrO2)n(SnO2)n superlattice (SL) configuration, with n being the number of monolayers which are considered equal to 1, 2, ..., 10 are studied. A half-metallic behavior is observed for the (CrO2)n(SnO2)n SLs for all values of n. The ground state is found to be FM with a magnetic moment of 2 μB per chromium atom, and this result does not depend on the number of monolayers n. As the FM rutile-CrO2 is unstable at ambient temperature, and known to be stabilized when on top of SnO2, the authors suggest that (CrO2)n(SnO2)n SLs may be applied to spintronic technologies since they provide efficient spin-polarized carriers

    Antileishmanial activity of Handroanthus serratifolius (Vahl) S. Grose (Bignoniaceae).

    This study aimed to evaluate the leishmanicidal activity of ethanol extract, fractions, and isolated substance from Handroanthus serratifolius against Leishmania amazonensis. Furthermore, this activity was related to cytotoxicity, and the selectivity index was determined.The ethanol extract was obtained by maceration of the stem powder, and the extract was subjected to fractionation on chromatographic column. The lapachol was obtained by acid base extraction followed by purification in chromatographic column. The antipromastigote activity and cytotoxicity tests were carried out by the cell viabilitymethod (MTT).Modified THP-1 cells were infected with L. amazonensis promastigotes and treated for 24 h with different concentrations of the extract, fractions, and lapachol. Theethanol extract, dichloromethane, and ethyl acetate fractions were not active against promastigotes (IC50 > 200 ?g/mL) or cytotoxic (CC50 > 500 ?g/mL), and the selectivity index (SI)was greater than 2.5.The ethyl acetate fractionwas active only in promastigotes; it is not cytotoxic (CC50 > 500 ?g/mL, SI > 5).The lapachol was selectively active only against amastigote (IS > 2.5, CC50 > 500 ?g/mL). In summary, lapachol and ethyl acetate fraction are promising against amastigote and promastigote forms, respectively

    Assessment of workload in the postoperative period of cardiac surgery according to the Nursing Activities Score

    Objetivo Identificar los factores asociados con la carga de trabajo de enfermería en el cuidado de los pacientes después de la cirugía cardíaca. Método Estudio prospectivo de cohorte, realizado con 187 pacientes de la Unidad Quirúrgica de Cuidados Intensivos (UCI) del Instituto do Coração. Los datos fueron recogidos en las primeras 24 y 72 horas el paciente en la UCI. La variable dependiente fue la carga de trabajo calculada por el Nursing Activities Score (NAS) y eran independientes de la naturaleza y de mortalidad puntajes demográficas y clínicas. Para el análisis de los datos se utilizó la prueba de Wilcoxon-Mann-Whitney y Spearman correlación y de regresión lineal con el modelo de efectos mixtos. Resultados La mayoría de los pacientes eran varones (59,4%) con una edad media de 61 años (±12,7) y 43,9% desarrollaron algún tipo de complicación en el postoperatorio. Dentro de 24 horas, la carga de trabajo fue 82,4% (±3,4) y 58,1% (±3,4) en 72 horas. Los factores asociados con el aumento de NAS fueron: longitud del paciente de la estancia en la UCI (p=0,036) y la presencia de complicaciones (pObjective Identify factors associated with the workload of nursing care for patients in the postoperative period of cardiac surgery. Results The majority of patients were male (59.4%), with a mean age of 61 years (±12.7), and 43.9% developed some kind of complication in the postoperative period. In the first 24 hours, the workload was 82.4% (±3.4), and 58.1% (±3.4) in 72 hours.
