    FLOW INJECTION WITH CHEMILUMINESCENCE DETECTION FOR THE DETERMINATION OF IRON IN SURFACE ATLANTIC WATERS

    This thesis describes the design, optimisation and shipboard deployment of a flow injection - chemiluminescence (FI-CL) technique for the determination of iron (Fe) in seawater. Chapter One presents an overview of the marine biogeochemistry of Fe, including its speciation, sources and sinks, abundance and limitation for phytoplankton growth in the Wodd*s oceans. Current analytical methods for the determination of Fe in natural waters are also reviewed. Chapter Two reports the instrumental development of the FI - CL method. Each component is described and its suitability to the flow manifold discussed. Different CL detection systems are evaluated and a charge coupled device used to investigate the spectral profile of the Fe-catalysed luminol reaction. Automation of the FI manifold is also detailed along with acquisition of CL signals. Chapter Three details the optimisation of a FI - CL procedure for the determination of Fe in seawater. Reagent clean-up techniques, blank procedures and a standard addition operating routine are detailed. Fe(III) reduction using sulphite is treated theoretically. Matrix effects are investigated and the synthesis of an 8-hydroxyquinoline resin used for in-line matrix elimination and preconcentration is reported. The optimised method is selective to Fe(II+IIl) in the linear range 0.04-10 nM, with a precision of 3.2% (n=5) for a LO nM standard and a limit of detection (3s) of 40 pM for a load time of 1 min. Chapter Four presents the results of an investigation into the kinetic effect of Fe on luminol CL using the continuous addition of reagent (CAR) technique. Instrumental and chemical parameters are optimised, interferences investigated and the CAR-CL technique compared with alternative flow configurations. In Chapter Five, the application of the F - CL method to the shipboard determination of Fe in the surface North and South Atlantic (SO'^N to 50°S) is presented. Data are reported for samples collected from the upper water column (<200 m) in eight different biogeochemical provinces, which represent coastal, upwelling and oligotrophic regions of the Adbntic Ocean. Total dissolvable iron (unfiltered, TD-Fe) levels range ftom <0.1 to 6.1 nM and indicate that high and spatially variable TD-Fe (>2 nM) concentrations exist in Equatorial and tropical North Atlantic regions influenced by atmospheric deposition from the West African continent. Away from strong input mechanisms, TD-Fe concentrations in the upper water column average 0.6 nM. Input sources are fingerprinted via correlation with other trace metals (Al, Co, Ni), nutrients and hydrography, whilst active biological uptake is shown to be the dominant sink. TD-Fe vertical distributions through the upper mixed layer display strong relationships with chlorophyll a concentrations, and measurements in the deep chlorophyll maximum of the South Atlantic oligotrophic gyre show that, despite elevated nitrate at such depths, Fe concentrations are at a minimum (ca. 0.1 nM) and may be low enough to (co-)limit phytoplankton growth.Centre for Coastal and Marine Sciences - Plymouth Marine Laborator

    Dissolved iron in the North Atlantic Ocean and Labrador Sea along the GEOVIDE section (GEOTRACES section GA01)

    Dissolved Fe (DFe) samples from the GEOVIDE voyage (GEOTRACES GA01, May–June 2014) in the North Atlantic Ocean were analysed using a SeaFAST-picoTM coupled to an Element XR HR-ICP-MS and provided interesting insights on the Fe sources in this area. Overall, DFe concentrations ranged from 0.09 ± 0.01 nmol L−1 to 7.8 ± 0.5 nmol L−1. Elevated DFe concentrations were observed above the Iberian, Greenland and Newfoundland Margins likely due to riverine inputs from the Tagus River, meteoric water inputs and sedimentary inputs. Air-sea interactions were suspected to be responsible for the increase in DFe concentrations within subsurface waters of the Irminger Sea due to deep convection occurring the previous winter, that provided iron-to-nitrate ratios sufficient to sustain phytoplankton growth. Increasing DFe concentrations along the flow path of the Labrador Sea Water were attributed to sedimentary inputs from the Newfoundland Margin. Bottom waters from the Irminger Sea displayed high DFe concentrations likely due to the dissolution of Fe-rich particles from the Denmark Strait Overflow Water and the Polar Intermediate Water. Finally, the nepheloid layers were found to act as either a source or a sink of DFe depending on the nature of particles

    Non-canonical inflammasome activation mediates the adjuvanticity of nanoparticles

    The non-canonical inflammasome sensor caspase-11 and gasdermin D (GSDMD) drive inflammation and pyroptosis, a type of immunogenic cell death that favors cell-mediated immunity (CMI) in cancer, infection, and autoimmunity. Here we show that caspase-11 and GSDMD are required for CD8+ and Th1 responses induced by nanoparticulate vaccine adjuvants. We demonstrate that nanoparticle-induced reactive oxygen species (ROS) are size dependent and essential for CMI, and we identify 50- to 60-nm nanoparticles as optimal inducers of ROS, GSDMD activation, and Th1 and CD8+ responses. We reveal a division of labor for IL-1 and IL-18, where IL-1 supports Th1 and IL-18 promotes CD8+ responses. Exploiting size as a key attribute, we demonstrate that biodegradable poly-lactic co-glycolic acid nanoparticles are potent CMI-inducing adjuvants. Our work implicates ROS and the non-canonical inflammasome in the mode of action of polymeric nanoparticulate adjuvants and establishes adjuvant size as a key design principle for vaccines against cancer and intracellular pathogens

    The most common reaction to suggesting that we could learn valuable lessons from the way the current pandemic has been/ is being handled, is to discourage the attempt; as it is suggested that it can all be done more accurately and authoritatively after the inevitable Public Inquiry (Slater, 2019). On the other hand, a more constructive approach, is to capture and understand the work that was actually done.This would include normal activities, as well as positive adaptations to challenges and failures that may have occurred. Such an approach aimed at improving what worked, rather than blaming people for what went wrong, has the potential to contribute more successfully to controlling the consequences of the current crisis. Such an approach should thus be aimed at detecting and feeding back lessons from emerging and probably unexpected behaviours and helping to design the system to adapt better to counter the effects. The science and discipline of Human Factors (HF) promotes system resilience. This can be defined as an organisation's ability to adjust its functioning before, during or after significant disturbances (such as a pandemic), enabling adaptation and operation under both anticipated and unanticipated circumstances. A “functional” approach methodology enables the identification of where the system and its various interdependent functions (an activity or set of activities that are required to give a certain output), could be improved and strengthened; if not immediately, at least for the future. Along these lines, suggestions for adding key resilience functions are additionally identified and outlined. The application and insights gained from this functional approach to the 2015 MERS-Cov pandemic in South Korea has been seen as contributing substantially to the effective response to the current crisis in that country (Min, submitted for publication).     Is the “never event” concept a useful safety management strategy in complex primary healthcare systems?

    Why is the area important?: A sub-group of rare but serious patient safety incidents, known as ‘never events,’ is judged to be ‘avoidable.’ There is growing interest in this concept in international care settings, including UK primary care. However, issues have been raised regarding the well-intentioned coupling of ‘preventable harm’ with zero tolerance ‘never events,’ especially around the lack of evidence for such harm ever being totally preventable. What is already known and gaps in knowledge?: We consider whether the ideal of reducing preventable harm to ‘never’ is better for patient safety than, for example, the goal of managing risk materializing into harm to ‘as low as reasonably practicable,’ which is well-established in other complex socio-technical systems and is demonstrably achievable. We reflect on the ‘never event’ concept in the primary care context specifically, although the issues and the polarized opinion highlighted are widely applicable. Recent developments to validate primary care ‘never event’ lists are summarized and alternative safety management strategies considered, e.g. Safety-I and Safety-II. Future areas for advancing research and practice: Despite their rarity, if there is to be a policy focus on ‘never events,’ then specialist training for key workforce members is necessary to enable examination of the complex system interactions and design issues, which contribute to such events. The ‘never event’ term is well intentioned but largely aspirational—however, it is important to question prevailing assumptions about how patient safety can be understood and improved by offering alternative ways of thinking about related complexities

    Patient safety incidents occur across all healthcare settings worldwide. Patients, families and carers can be physically and psychologically traumatised and often experience additional and prolonged harms due to a lack of apology, openness and transparency. Healthcare professionals can also be emotionally impacted and subject to embarrassment, guilt, complaints, regulatory investigations and medico-legal action. Despite significant healthcare policy and professional attention, evidence of related learning and successful risk reductions at all levels are severely limited. In this article, rather than focussing on the individual “failings” of professionals, we take a Human Factors systems perspective in explaining how and why highly complex systems generally fail. We introduce a series of Systems Thinking principles for potentially guiding more meaningful discussions and learning from when things go wrong in highly complex sociotechnical systems, such as much of healthcare. We suggest to the medico-legal community whether a debate is needed around the need for the judiciary, expert witnesses, regulators and legal professionals to be better informed in the Human Factors “systems approach” to patient safety investigations as part of the medico-legal process

    Biogeochemical cycling of cadmium isotopes in the Southern Ocean along the Zero Meridian

    We present depth profiles of Cd isotopes and concentrations from the Southern Ocean at four stations in the Atlantic sector along the Greenwich Meridian (47°S to 68°S) located across the main Antarctic frontal zones and productivity belt. The vertical profiles of Cd concentration typically show low values in surface waters, elevated values at intermediate depths, reflecting remineralization of sinking particulate organic matter, and constant values in deep waters. The surface-to-deep isotopic gradient shows “heavy” Cd isotope signatures in the mixed surface layer, becoming more pronounced northward, with values up to ɛ112/110Cd of around +4.1 in the Subantarctic sector of the Southern Ocean. Deep Antarctic waters display a uniform and “light” ɛ112/110Cd of +1.18 ± 0.38 and Cd concentrations of 0.761 ± 0.101 nmol/kg (n = 23, 2SD). Intermediate waters are characterized by ɛ112/110Cd lying between those of surface and deep waters, with a constant value of about +0.8 in the High Nutrient Low Chlorophyll sector and a notably higher value of +2.3 in the Subantarctic sector. The Cd isotope fractionation in the Southern Ocean closely follows a simple closed-system Rayleigh model, in which biological uptake of Cd imparts the ɛ112/110Cd signature to the surface layer while that of deep waters is determined by the flux of regenerated isotopically-light Cd from sinking organic matter from the surface ocean and the degree of mixing of distinct water masses. The vertical gradient documented for Cd isotopes and nutrient ratios, along with the meridional gradient in surface waters, highlights the important role played by upwelling in the Southern Ocean in closing the meridional overturning circulation via the export of Antarctic intermediate and mode waters which have a distinctive chemical (low Cd:P) and Cd isotope (“heavy”) signature. The combined Cd–Zn isotope systematics provide evidence for a strong link between the magnitude of biological Cd stable isotope fractionation and Zn availability in the contrasted nutrient and ecological regimes of the Southern Ocean. Substitution of Cd for Zn in the enzyme carbonic anhydrase appears to be the driving mechanism for Cd isotope fractionation in the Antarctic Circumpolar Current, while an “excess-uptake” mechanism seems to predominate in the Weddell Gyre. Our study highlights some of the complexities of the biogeochemical cycling of Cd in the oceans. Nevertheless, systematic variations in Cd isotopic compositions with water mass distribution in the Southern Ocean suggest that Cd isotopes could, with some caveats, be useful tracers of changes in past nutrient utilization and deep water circulation.
