    Canopy Changes of Brachiaria Managed Under Continuous Stocking in the Dry-Water Transition

    Pasture supplementation is an alternative to take advantage of the forage mass deferred in the rainy season and maintain or provide weight gain in periods of rain absence. The objective was to compare the structural characteristics, mass production, density and population dynamics of tillers of Urochloa brizantha cv. ‘Paiaguas’ and U. spp. cv. ‘Convert’ under fixed and continuous stocking with steers supplemented in the trough or on the ground in the water/dry transition period. The experiment was carried out at Jatai Federal University, from March to June, in a completely randomized design and a 2x2 factorial scheme, using six paddocks/treatment. The completely randomized design was adopted and variance analysis was performed with software SAS following the GLM procedure, were compared using repeated-measures. There was a significant interaction between Brachiaria and the method of supplementation for basal, aerial, and total tiller density. Convert had an average of 551 basal and 577 total tiller/m2. Paiaguas grass presented higher tiller density (1,03 vs. 582 tiller/m2) and higher tiller birth rate (12.92 vs. 9.14%) than Convert. No significant difference was observed between brachiarias. The average height of Paiaguas was 62.34 and 50.70 cm for Convert. The average height was 57.83 and on the ground it was 54.90 cm. Supplementation offer method changed Paiaguas canopy. The Convert showed higher leaf and dead mass production but lower weeds despite its smaller tillering

    Evaluation of Cardiac Autonomic Modulation Using Symbolic Dynamics after Cardiac Transplantation

    Objective: To characterize the behavior of cardiac autonomic modulation in individuals with different times after orthotopic heart transplantation (HTx) using symbolic dynamics analysis. Methods: Sixty patients were evaluated after HTx. We recorded their instantaneous R-R intervals (RRi) by cardiac monitor Polar (R) RS800CX (TM) (Polar Electro Oy, Kempele, Finland) for 10 minutes. The same sequence of RRi with 256 consecutive beats was used to perform spectral analysis and symbolic dynamics analysis. We used hierarchical clustering to form groups. One-way analysis of variance (ANOVA) (with Holm-Sidak method) or one-way Kruskal-Wallis test (with Dunn's post-hoc test) was used to analyze the difference between groups. Linear correlation analysis between variables was performed using Pearson's or Spearman's tests. P-value < 0.05 was considered statistically significant. Results: The 0V% index increased, the 2UV% index and the normalized complexity index decreased with an increase of HTx postoperative time. There were a negative correlation between complexity indexes and 0V% and a positive correlation between complexity indexes and 2UV%. Conclusion: Symbolic dynamics indexes were able to show a specific cardiac autonomic modulation pattern for HTx recipients with different postoperative times

    Dynamic and cell-specific DACH1 expression in human neocortical and striatal development

    DACH1 is the human homolog of the Drosophila dachshund gene, which is involved in the development of the eye, nervous system, and limbs in the fly. Here, we systematically investigate DACH1 expression patterns during human neurodevelopment, from 5 to 21 postconceptional weeks. By immunodetection analysis, we found that DACH1 is highly expressed in the proliferating neuroprogenitors of the developing cortical ventricular and subventricular regions, while it is absent in the more differentiated cortical plate. Single-cell global transcriptional analysis revealed that DACH1 is specifically enriched in neuroepithelial and ventricular radial glia cells of the developing human neocortex. Moreover, we describe a previously unreported DACH1 expression in the human striatum, in particular in the striatal medium spiny neurons. This finding qualifies DACH1 as a new striatal projection neuron marker, together with PPP1R1B, BCL11B, and EBF1. We finally compared DACH1 expression profile in human and mouse forebrain, where we observed spatio-temporal similarities in its expression pattern thus providing a precise developmental description of DACH1 in the 2 mammalian species

    Cobalt oxide nanoparticles induce oxidative stress and alter electromechanical function in rat ventricular myocytes

    Background: Nanotoxicology is an increasingly relevant field and sound paradigms on how inhaled nanoparticles (NPs) interact with organs at the cellular level, causing harmful conditions, have yet to be established. This is particularly true in the case of the cardiovascular system, where experimental and clinical evidence shows morphological and functional damage associated with NP exposure. Giving the increasing interest on cobalt oxide (Co3O4) NPs applications in industrial and bio-medical fields, a detailed knowledge of the involved toxicological effects is required, in view of assessing health risk for subjects/workers daily exposed to nanomaterials. Specifically, it is of interest to evaluate whether NPs enter cardiac cells and interact with cell function. We addressed this issue by investigating the effect of acute exposure to Co3O4-NPs on excitation-contraction coupling in freshly isolated rat ventricular myocytes. Results: Patch clamp analysis showed instability of resting membrane potential, decrease in membrane electrical capacitance, and dose-dependent decrease in action potential duration in cardiomyocytes acutely exposed to Co3O4-NPs. Motion detection and intracellular calcium fluorescence highlighted a parallel impairment of cell contractility in comparison with controls. Specifically, NP-treated cardiomyocytes exhibited a dose-dependent decrease in the fraction of shortening and in the maximal rate of shortening and re-lengthening, as well as a less efficient cytosolic calcium clearing and an increased tendency to develop spontaneous twitches. In addition, treatment with Co3O4-NPs strongly increased ROS accumulation and induced nuclear DNA damage in a dose dependent manner. Finally, transmission electron microscopy analysis demonstrated that acute exposure did lead to cellular internalization of NPs. Conclusions: Taken together, our observations indicate that Co3O4-NPs alter cardiomyocyte electromechanical efficiency and intracellular calcium handling, and induce ROS production resulting in oxidative stress that can be related to DNA damage and adverse effects on cardiomyocyte functionality
