    Gene expression profile of cervical and skin tissues from human papillomavirus type 16 E6 transgenic mice

    <p>Abstract</p> <p>Background</p> <p>Although K14E6 transgenic mice develop spontaneous tumors of the skin epithelium, no spontaneous reproductive tract malignancies arise, unless the transgenic mice were treated chronically with 17β-estradiol. These findings suggest that E6 performs critical functions in normal adult cervix and skin, highlighting the need to define E6-controlled transcriptional programs in these tissues.</p> <p>Methods</p> <p>We evaluated the expression profile of 14,000 genes in skin or cervix from young K14E6 transgenic mice compared with nontransgenic. To identify differentially expressed genes a linear model was implemented using R and the LIMMA package. Two criteria were used to select the set of relevant genes. First a set of genes with a Log-odds ≥ 3 were selected. Then, a hierarchical search of genes was based on Log Fold Changes.</p> <p>Results</p> <p>Microarray analysis identified a total of 676 and 1154 genes that were significantly up and down-regulated, respectively, in skin from K14E6 transgenic mice. On the other hand, in the cervix from K14E6 transgenic mice we found that only 97 and 252 genes were significantly up and down-regulated, respectively. One of the most affected processes in the skin from K14E6 transgenic mice was the cell cycle. We also found that skin from transgenic mice showed down-regulation of pro-apoptotic genes and genes related to the immune response. In the cervix of K14E6 transgenic mice, we could not find affected any gene related to the cell cycle and apoptosis pathways but did observe alterations in the expression of immune response genes. Pathways such as angiogenesis, cell junction and epidermis development, also were altered in their gene expression profiles in both tissues.</p> <p>Conclusion</p> <p>Expression of the HPV16 E6 oncoprotein in our model alters expression of genes that fell into several functional groups providing insights into pathways by which E6 deregulate cell cycle progression, apoptosis, the host resistance to infection and immune function, providing new opportunities for early diagnostic markers and therapeutic drug targets.</p

    Essential Medicines at the National Level : The Global Asthma Network's Essential Asthma Medicines Survey 2014

    Patients with asthma need uninterrupted supplies of affordable, quality-assured essential medicines. However, access in many low- and middle-income countries (LMICs) is limited. The World Health Organization (WHO) Non-Communicable Disease (NCD) Global Action Plan 2013-2020 sets an 80% target for essential NCD medicines' availability. Poor access is partly due to medicines not being included on the national Essential Medicines Lists (EML) and/or National Reimbursement Lists (NRL) which guide the provision of free/subsidised medicines. We aimed to determine how many countries have essential asthma medicines on their EML and NRL, which essential asthma medicines, and whether surveys might monitor progress. A cross-sectional survey in 2013-2015 of Global Asthma Network principal investigators generated 111/120 (93%) responses41 high-income countries and territories (HICs); 70 LMICs. Patients in HICs with NRL are best served (91% HICs included ICS (inhaled corticosteroids) and salbutamol). Patients in the 24 (34%) LMICs with no NRL and the 14 (30%) LMICs with an NRL, however no ICS are likely to have very poor access to affordable, quality-assured ICS. Many LMICs do not have essential asthma medicines on their EML or NRL. Technical guidance and advocacy for policy change is required. Improving access to these medicines will improve the health system's capacity to address NCDs.Peer reviewe

    DAMPAK PERNIKAHAN ANAK PADA KEHARMONISAN RUMAH TANGGA DI DESA KANREPIA KECAMATAN TOMBOLO PAO KABUPATEN GOWA

    Tujuan penelitian ini yaitu untuk mengetahui bagaimana faktor penyebab terjadinya pernikahan Anak di Desa Kanreapia Kecamatan tombolo Pao Kabupaten Gowa, dan bagaimana dampak pernikahan usia muda pada keharmonisan rumah tangga di  Desa tersebut. Jenis penelitian ini adalah penelitian kualitatif dengan pendekatan sosiologi dan kesejahteraan sosial. Sumber data yaitu sumber data primer dan sekunder. Selanjutnya, metode pengumpulan data yang digunakan adalah observasi, wawancara dan dokumentasi. Teknik pengolahan dan analisis data dilakukan melalui empat tahapan, yaitu reduksi data, penyajian data, teknis analisis perbandingan dan penarikan kesimpulan. Hasil penelitian ini menunjukkan bahwa pernikahan anak terjadi disebabkan karena beberapa faktor, yaitu: 1) pendidikan, 2) ekonomi, 3) Budaya. Adapun dampak pernikahan anak terhadap keharmonisan rumaha tangga terdiri atas dampak positi dan negatif, dampak positif berupa memperbaiki silaturahim dan perbaikan ekonomi, sementara dampak negatifnya cenderun terjadinya percekcokan yang berujung pada perceraian. Implikasi dari penelitian ini adalah: Diharapkan perhatian dari pemerintah dalam hal ini DP3A Kabupaten Gowa untuk memberikan arahan kepada perempuan atau anak yang melakukan pernikahan anak, dan upaya pemerintah desa untuk mengembangkan penguatan sumber perlindungan, agar memiliki fikiran untuk tidak menikah diusia anak dalam meningkatkan pola hidup yang lebih baik. Diharapkan bagi para remaja agar mementingkan pendidikan dan juga lebih bijak dalam mengambil keputusan

    Grazing impacts of the invasive bivalve Limnoperna fortunei (Dunker, 1857) on single-celled, colonial and filamentous cyanobacteria

    Feeding behavior of the invasive bivalve Limnoperna fortunei in the presence of single-celled, colonial, and filamentous cyanobacteria was tested in laboratory experiments to evaluate the effects of size and shape on mussel feeding. The first hypothesis holds that golden mussel filters more efficiently smaller particles, such as single cells of Microcystis, which could be more easily assimilated by its filtering apparatus. The second hypothesis sustains that L. fortunei filters more efficiently rounded colonies, such as Microcystis, which would be more easily ingested than lengthy filamentous, such as Planktothrix. Filtration rates of golden mussel in the presence of single-celled, colonial and filamentous cyanobacteria were similar. Nevertheless, there was a great difference in the ingestion and pseudofeces production rates. Single cells were widely accepted as food, while filamentous and colonial cyanobacteria were massively expelled as pseudofeces. The results confirmed the first hypothesis that golden mussel prefers to ingest smaller particles. The second hypothesis was rejected since filamentous were preferentially ingested than colonial cyanobacteria. Golden mussel has the potential to remove toxic cells (Microcystis), however this potential would be reduced in cyanobacteria blooms, where colonial forms which are preferentially rejected by L. fortunei, are predominant. In this case, the presence of this invasive bivalve could also enhance the occurrence of blooms by rejecting colonial and filamentous cyanobacteria in pseudofeces
