    <p>Abstract</p> <p>Objective</p> <p>The present study was carried out to determine the effect of cadmium exposure on Urinary N-acetyl-beta -D-glucosaminidase and its isoenzymes A and B in workers exposed at cadmium plating.</p> <p>Methods</p> <p>50 subjects using cadmium during cadmium plating formed the study group. An equal number of age-sex matched subjects working in administrative section formed the control group. Urinary cadmium levels were determined by using a flameless atomic absorption spectrophotometer. Urinary N-acetyl-beta -D-glucosaminidase and its isoenzymes A and B were determined by using spectrophotmetric method.</p> <p>Results</p> <p>A significant increase of urinary total N-acetyl-beta -D-glucosaminidase and its isoenzymes A and B profiles were noted in study as compared to controls. The levels of urinary N-acetyl-beta -D-glucosaminidase and its isoenzymes A and B profiles were positively and significantly correlated with cadmium levels in urine. Multiple regression analysis was used to assess the effect of urinary cadmium or life style confounding factors (age, BMI, smoking and alcohol consumption) on urinary N-acetyl-beta -D-glucosaminidase and its isoenzymes A and B. The analysis showed that the study subjects who had urine cadmium levels greater than 5 μg/g of creatinine, work duration >15 years, smoking and body mass index variables were significantly associated with urinary total N-acetyl-beta -D-glucosaminidase but not on isoenzymes A&B.</p> <p>Conclusion</p> <p>The results presented in this study shows that the increased levels of urinary N-acetyl-beta -D-glucosaminidase observed in cadmium-exposed workers could be used as biomarkers for suggesting preventive measure.</p

    Association of dopaminergic pathway gene polymorphisms with chronic renal insufficiency among Asian Indians with type-2 diabetes

    <p>Abstract</p> <p>Background</p> <p>Genetic markers conferring susceptibility to diabetes specific renal disease remains to be identified for early prediction and development of effective drugs and therapies. Inconsistent results obtained from analysis of genes from classical pathways generate need for examination of unconventional genetic markers having role in regulation of renal function. Experimental and clinical evidences suggest that dopamine is an important natriuretic hormone. Therefore, various genes involved in regulation of dopamine bioavailability could play a role in diabetic chronic renal insufficiency (CRI). We investigated the contribution of 12 polymorphisms from five Dopaminergic pathway genes to CRI among type-2 diabetic Asian Indian subjects.</p> <p>Methods</p> <p>Genetic association of 12 polymorphisms (SNPs) from five genes namely-dopamine receptor-1 (<it>DRD1</it>), <it>DRD2</it>, <it>DRD3</it>, <it>DRD4</it>, andcatechol-O-methyltransferase (<it>COMT</it>) with diabetic CRI was investigated using a case-control approach. Logistic regression analysis was carried out to correlate various clinical parameters with genotypes, and to study pair wise interactions between SNPs of different genes.</p> <p>Results</p> <p>SNPs <it>-141 ins/del C and G>A </it>(1 kb upstream from exon 2) in DRD2 gene showed significant allelic and genotypic association. Allele <it>-141 insC </it>and genotype <it>-141 insC/insC </it>of <it>-141 ins/del C </it>polymorphism, and allele A of <it>G>A </it>SNP were found to be predisposing to CRI. Our result of allelic and genotypic association of -<it>141 insC/delC </it>SNP was also reflected in the haplotypic association. Heterozygous genotype of polymorphism <it>900 ins/del C in COMT </it>gene was predisposing towards CRI.</p> <p>Conclusion</p> <p>Some polymorphisms in <it>DRD2 </it>and <it>COMT </it>genes are significantly associated with susceptibility to CRI in the Asian Indian population which, if confirmed would be consistent with a suggested role of dopamine metabolism in disease occurrence.</p

    IS BACTERIOLOGY A CONTRIBUTING FACTOR IN UNSALVAGEABLE NATURE OF DIABETIC FOOT INFECTIONS?–A STUDY IN A DISTRICT HOSPITAL IN MALAYSIA

    Objective: The objective is to study bacterial pathogens isolated in diabetic foot infection (DFI) and their sensitivity pattern to antibiotics commonly used in the management of DFI in the salvageable and unsalvageable groups of patients in a district hospital.Methods: 122 patients with diabetic limb infections treated at the Orthopedic Department of Hospital Sultan Abdul Halim, Sungai Petani, Kedah State in Malaysia. Clinically, limb infections were classified as salvageable and unsalvageable infections. Salvageable-mild, superficial/deep, localized ulcer with no systemic derangements necessitating conservative treatment or surgical procedures with minor amputations of limb (toe/ray amputation). Unsalvageable-deep seated extensive or spreading ulcers threatening the integrity of limb with or without toxic symptoms or metabolic derangement and could result in major limb amputation. A standard questionnaire was used to collect demographic, clinical and microbiological details of patients in both groups. Co-morbid illnesses, type/severity of limb infection during presentation and results of routine blood investigations were recorded. Details of nature of each specimen, species of isolate pathogen and sensitivity pattern to antibiotic of each clinical isolates were recorded.Results: 62 and 60 patients respectively belonged to the salvageable and unsalvageable groups. Only 11.66% presented with evidence of toxemia in the unsalvageable group. ESBL was the commonest nosocomial organisms. Percentage of organism sensitivity was most to vancomycin, ceftazidime, and gentamicin.Conclusion: All severe infections do not present with toxemia in diabetic patients. Gram-negative organisms were predominant in both groups although Staphylococcal organisms were the single largest group in the unsalvageable group. 3rd generation antibiotics are more useful in its control.Â

    Water-deficit stress - Induced physio-biochemical changes in cotton (Gossypium hirsutum L.) Cultivars

    Water stress is a serious global issue regarding growth of agricultural crops and sustainable food production for the large population. In the present situation due to low rainfall and unavailability of advanced irrigation methods, water deficit stress is the most limiting factor decreasing crop production in many regions of the world. In this study, to assess the drought tolerance mechanism in cotton cultivars was monitored by drought induced physio-biochemical changes. To assess the tolerance in cotton cultivars, a field experiment was conducted in split plot design in which the main plot consists of irrigated and complete rainfed conditions as a stress and cotton cultivars arranged in the main plot as a subplot. The overall comparative analysis revealed that hybrid was superior over their parents under well-watered as well as in water deficit conditions in terms of chlorophyll content, wax content, accumulation of compatible solutes, photosynthesis rate, stomatal conductance, transpiration rate and yield parameters. The findings from the results indicate that under water deficit conditions plants having a different adaptive mechanisms for coping with the stress situation. So, some of the adaptive mechanisms such as accumulation of sugars, polyphenols, amino acids, non-enzymatic antioxidants, and wax deposition helps to maintain osmotic balance, to protect cellular macromolecules, to detoxify the cells, and to scavenge free radicals under water deficit condition

    High frequency (HF) radar detection of the weak 2012 Indonesian tsunamis

    We report here on the observation and offline detection of the weak tsunamis generated by earthquakes near Indonesia on 11 April 2012 using radar systems and tide gauges on the coasts of Sumatra and the Andaman Islands. This work extends the previous observations of the much stronger 2011 Japan tsunami. The distance offshore at which the tsunami can be detected, and hence the warning time provided, depends primarily on the bathymetry: the wider the shallow continental shelf, the greater this time. The weak Indonesia tsunamis were detected successfully in spite of the narrow shallow-water shelf offshore from the radar systems. Larger tsunamis could obviously be detected further from the coast. This paper provides further confirmation that radar is an important tool to aid in tsunami observation and warning

    Phenolic and antioxidant capacity retention of potato peel waste as a function of cultivar, pretreatment and drying procedure

    Drying procedures employed for potato peels (both raw and boiled) may adversely affect the useful bioactivecomponents present in them. This study envisaged the identification of a feasible drying procedure for handlingbulk potato peel waste for maximising the retention of phytochemicals in the peel powder. The total phenols (TP), flavonoids (TF) and antioxidant capacity (TAC) were assessed in peels of three commercial and one newly developed anthocyanin rich Indian potato cultivars in response to boiling pretreatment and varying drying procedures. Microwave drying (600W) was best in terms of drying rate for both raw and boiled peels. It yielded the greatest amount of TP and TF in the dried raw peel, irrespective of cultivar. Dried raw peels of an thocyanin rich Kufri Neelkanth cultivar exhibited maximum TAC. Retention of TF, metal scavenging activity and reducing power followed almost a similar pattern as TP irrespective of cultivar, pretreatment and drying procedure. Our study shows that potato peel from Kufri Neelkanth (raw) and Kufri Frysona (both raw and boiled) are best source of phenolics and flavonoids and can serve as a suitable matrix for extraction of bioactive compounds which holds promise for use in the food industry

    Fermentation process for alcoholic beverage production from mahua (Madhuca indica J. F. Mel.) flowers

    Mahua flowers are rich in sugar (68-72%), in addition to a number of minerals and one of the most important raw materials for alcohol fermentation. The present investigation was for the development of a non-distilled alcoholic beverage from Mahua flowers. Eighteen (18) treatment combinations consisting of two temperatures (25 and 30°C), three pH (4.0, 4.5 and 5.0) and three period of fermentation (7, 14 and 21 days) were used in the fermentation conditions. The maximum yield of ethanol (9.51 %) occurred at 25°C with pH 4.5 after 14 days of  fermentation of Mahua flower juice. The fermented non-distilled alcoholic beverage contained total sugar (8.83 mg/ml), reducing sugar (0.82 mg/ml), total soluble solids (6.37°Brix) titrable acidity (0.65 %), and volatile acidity (0.086%). Methanol was not detected at any stage of fermentation.     Genetic characterization of mango anthracnose pathogen Colletotrichum gloeosporioides Penz. by random amplified polymorphic DNA analysis

    Twenty-five isolates of Colletotrichum gloeosporioides causing mango anthracnose were collected from different agroclimatic zones of India. The isolates were evaluated for their pathogenic variability on mango seedlings and genetic characterization using random amplified polymorphic DNA (RAPD molecular techniques). The random primers OPA-1, 3, 5, 9, 11, 15, 16 and 18 were used and the twenty five isolates were grouped into two. The amplified DNA fragments (amplicons) obtained was comparedby agarose gel electrophoresis. Isolate specific RAPD fingerprints were obtained. Out of eight primers in RAPD, OPA-1, 3 and 18 were able to produce reproducible banding pattern. Each of these primers generated a short spectrum of amplicons, located between 661 and 2291-bp markers, indicative of genetic polymorphism. Dendogram revealed more than 75% level of similarity. 4.36% polymorphism was also found in individual isolates that was not statistically significant (P &gt; 0.05) among the sample, it also indicates that all the isolates tested had approximately same genetic identity. The data suggest that RAPD may be of value by virtue of its rapidity, efficiency and reproducibility in generating genetic fingerprints of C. gloeosporioides isolates
