Direct observation of local xylem embolisms induced by soil drying in intact Zea mays leaves
The vulnerability of vascular plants to xylem embolism is closely related to their stable long-distance water transport, growth, and survival. Direct measurements of xylem embolism are required to understand what causes embolism and what strategies plants employ against it. In this study, synchrotron X-ray microscopy was used to non-destructively investigate both the anatomical structures of xylem vessels and embolism occurrence in the leaves of intact Zea mays (maize) plants. Xylem embolism was induced by water stress at various soil drying periods and soil water contents. X-ray images of dehydrated maize leaves showed that the ratio of gas-filled vessels to all xylem vessels increased with decreased soil water content and reached approximately 30% under severe water stress. Embolism occurred in some but not all vessels. Embolism in maize leaves was not strongly correlated with xylem diameter but was more likely to occur in the peripheral veins. The rate of embolism formation in metaxylem vessels was higher than in protoxylem vessels. This work has demonstrated that xylem embolism remains low in maize leaves under water stress and that there xylem has characteristic spatial traits of vulnerability to embolism.117Ysciescopu
Quantum spin liquid states in the two dimensional kagome antiferromagnets, ZnxCu4-x(OD)6Cl2
A three-dimensional system of interacting spins typically develops static
long-range order when it is cooled. If the spins are quantum (S = 1/2),
however, novel quantum paramagnetic states may appear. The most highly sought
state among them is the resonating valence bond (RVB) state in which every pair
of neighboring quantum spins form entangled spin singlets (valence bonds) and
the singlets are quantum mechanically resonating amongst all the possible
highly degenerate pairing states. Here we provide experimental evidence for
such quantum paramagnetic states existing in frustrated antiferromagnets,
ZnxCu4-x(OD)6Cl2, where the S = 1/2 magnetic Cu2+ moments form layers of a
two-dimensional kagome lattice. We find that in Cu4(OD)6Cl2, where distorted
kagome planes are weakly coupled to each other, a dispersionless excitation
mode appears in the magnetic excitation spectrum below ~ 20 K, whose
characteristics resemble those of quantum spin singlets in a solid state, known
as a valence bond solid (VBS), that breaks translational symmetry. Doping
nonmagnetic Zn2+ ions reduces the distortion of the kagome lattice, and weakens
the interplane coupling but also dilutes the magnetic occupancy of the kagome
lattice. The VBS state is suppressed and for ZnCu3(OD)6Cl2 where the kagome
planes are undistorted and 90% occupied by the Cu2+ ions, the low energy spin
fluctuations in the spin liquid phase become featureless

Generalized Flows around Neutron Stars
In this chapter, we present a brief and non-exhaustive review of the
developments of theoretical models for accretion flows around neutron stars. A
somewhat chronological summary of crucial observations and modelling of timing
and spectral properties are given in sections 2 and 3. In section 4, we argue
why and how the Two-Component Advective Flow (TCAF) solution can be applied to
the cases of neutron stars when suitable modifications are made for the NSs. We
showcase some of our findings from Monte Carlo and Smoothed Particle
Hydrodynamic simulations which further strengthens the points raised in section
4. In summary, we remark on the possibility of future works using TCAF for both

Identification and characterization of seed-specific transcription factors regulating anthocyanin biosynthesis in black rice
Black rice is rich in anthocyanin and is expected to have more healthful dietary potential than white rice. We assessed expression of anthocyanin in black rice cultivars using a newly designed 135 K Oryza sativa microarray. A total of 12,673 genes exhibited greater than 2.0-fold up- or down-regulation in comparisons between three rice cultivars and three seed developmental stages. The 137 transcription factor genes found to be associated with production of anthocyanin pigment were classified into 10 groups. In addition, 17 unknown and hypothetical genes were identified from comparisons between the rice cultivars. Finally, 15 out of the 17 candidate genes were verified by RT-PCR analysis. Among the genes, nine were up-regulated and six exhibited down-regulation. These genes likely play either a regulatory role in anthocyanin biosynthesis or are related to anthocyanin metabolism during flavonoid biosynthesis. While these genes require further validation, the results here underline the potential use of the new microarray and provide valuable insight into anthocyanin pigment production in rice
Targeting of highly conserved Dengue virus sequences with anti-Dengue virus trans-splicing group I introns
<p>Abstract</p> <p>Background</p> <p>Dengue viruses (DENV) are one of the most important viral diseases in the world with approximately 100 million infections and 200,000 deaths each year. The current lack of an approved tetravalent vaccine and ineffective insecticide control measures warrant a search for alternatives to effectively combat DENV. The <it>trans</it>-splicing variant of the <it>Tetrahymena thermophila </it>group I intron catalytic RNA, or ribozyme, is a powerful tool for post-transcriptional RNA modification. The nature of the ribozyme and the predictability with which it can be directed makes it a powerful tool for modifying RNA in nearly any cell type without the need for genome-altering gene therapy techniques or dependence on native cofactors.</p> <p>Results</p> <p>Several anti-DENV Group I <it>trans</it>-splicing introns (αDENV-GrpIs) were designed and tested for their ability to target DENV-2 NGC genomes <it>in situ</it>. We have successfully targeted two different uracil bases on the positive sense genomic strand within the highly conserved 5'-3' cyclization sequence (CS) region common to all serotypes of DENV with our αDENV-GrpIs. Our ribozymes have demonstrated ability to specifically <it>trans</it>-splice a new RNA sequence downstream of the targeted site <it>in vitro </it>and in transfected insect cells as analyzed by firefly luciferase and RT-PCR assays. The effectiveness of these αDENV-GrpIs to target infecting DENV genomes is also validated in transfected or transformed Aedes mosquito cell lines upon infection with unattenuated DENV-2 NGC.</p> <p>Conclusions</p> <p>Analysis shows that our αDENV-GrpIs have the ability to effectively <it>trans</it>-splice the DENV genome <it>in situ</it>. Notably, these results show that the αDENV-GrpI 9v1, designed to be active against all forms of Dengue virus, effectively targeted the DENV-2 NGC genome in a sequence specific manner. These novel αDENV-GrpI introns provide a striking alternative to other RNA based approaches for the transgenic suppression of DENV in transformed mosquito cells and tissues.</p
Novel functions and regulation of cryptic cellobiose operons in Escherichia coli
Presence of cellobiose as a sole carbon source induces mutations in the chb and asc operons of Escherichia coli and allows it to grow on cellobiose. We previously engineered these two operons with synthetic constitutive promoters and achieved efficient cellobiose metabolism through adaptive evolution. In this study, we characterized two mutations observed in the efficient cellobiose metabolizing strain: duplication of RBS of ascB gene, (beta-glucosidase of asc operon) and nonsense mutation in yebK, (an uncharacterized transcription factor). Mutations in yebK play a dominant role by modulating the length of lag phase, relative to the growth rate of the strain when transferred from a rich medium to minimal cellobiose medium. Mutations in ascB, on the other hand, are specific for cellobiose and help in enhancing the specific growth rate. Taken together, our results show that ascB of the asc operon is controlled by an internal putative promoter in addition to the native cryptic promoter, and the transcription factor yebK helps to remodel the host physiology for cellobiose metabolism. While previous studies characterized the stress-induced mutations that allowed growth on cellobiose, here, we characterize the adaptation-induced mutations that help in enhancing cellobiose metabolic ability. Study of the decays B->D_s1(2536)+ anti-D(*)
We report a study of the decays B -> D_s1(2536)+ anti-D(*), where anti-D(*)
is anti-D0, D- or D*-, using a sample of 657 x 10^6 B anti-B pairs collected at
the Upsilon(4S) resonance with the Belle detector at the KEKB asymmetric-energy
e+e- collider. The branching fractions of the decays B+ -> D_s1(2536)+ anti-D0,
B0 -> D_s1(2536)+ D- and B0 -> D_s1(2536)+ D*- multiplied by that of
D_s1(2536)+ -> (D*0K+ + D*+K0) are found to be (3.97+-0.85+-0.56) x 10^-4,

Evidence for B- -> tau- nu_bar with a Semileptonic Tagging Method
We present a measurement of the decay B- -> tau- nu_bar using a data sample
containing 657 million BB_bar pairs collected at the Upsilon(4S) resonance with
the Belle detector at the KEKB asymmetric-energy e+e- collider. A sample of
BB_bar pairs are tagged by reconstructing one B meson decaying
semileptonically. We detect the B- -> tau- nu_bar candidate in the recoil. We
obtain a signal with a significance of 3.6 standard deviations including
systematic uncertainties, and measure the branching fraction to be Br(B- ->
tau- nu_bar) = [1.54+0.38-0.37(stat)+0.29-0.31(syst)]*10^-4. This result
confirms the evidence for B- -> tau- nu_bar obtained in a previous Belle
