    Co-targeting of Bcl-2 and mTOR pathway triggers synergistic apoptosis in BH3 mimetics resistant acute lymphoblastic leukemia

    Several chemo-resistance mechanisms including the Bcl-2 protein family overexpression and constitutive activation of the PI3K/Akt/mTOR signaling have been documented in acute lymphoblastic leukemia (ALL), encouraging targeted approaches to circumvent this clinical problem. Here we analyzed the activity of the BH3 mimetic ABT-737 in ALL, exploring the synergistic effects with the mTOR inhibitor CCI-779 on ABT-737 resistant cells. We showed that a low Mcl-1/Bcl-2 plus Bcl-xL protein ratio determined ABT-737 responsiveness. ABT-737 exposure further decreased Mcl-1, inducing apoptosis on sensitive models and primary samples, while not affecting resistant cells. Co-inhibition of Bcl-2 and the mTOR pathway resulted cytotoxic on ABT-737 resistant models, by downregulating mTORC1 activity and Mcl-1 in a proteasome-independent manner. Although Mcl-1 seemed to be critical, ectopic modulation did not correlate with apoptosis changes. Importantly, dual targeting proved effective on ABT-737 resistant samples, showing additive/synergistic effects. Together, our results show the efficacy of BH3 mimetics as single agent in the majority of the ALL samples and demonstrate that resistance to ABT-737 mostly correlated with Mcl-1 overexpression.     On distributed strains in a CFA pile via DFOSs measurements and numerical analysis

    Fibre Optic Sensors (FOSs) offer unprecedented possibilities for the monitoring of engineering structures, such as foundation systems. Notably, the type of FOSs known as Distributed Fibre Optic Sensors (DFOSs) has the capability of monitoring variations in the observed physical field, such as strain and temperature, with spatial continuity along the fibre. This paper presents and discusses the distributed strain measurements collected along a continuous flight auger (CFA) pile, belonging to the foundation raft of a new bridge subjected to an acceptance static load test. The monitoring was performed using a DFOS system, which works according to the optical frequency domain reflectometry (OFDR) method and provides a spatial resolution of 10 mm and a strain resolution of 1 \u3bc\u3b5. The in-situ monitoring results were used to calibrate a 3D Finite Element Model of the foundation system.     Os laboratórios desempenham um papel de extrema relevância nos centros de pesquisa, com toda a gama de serviços que prestam ao processo de PD&I. Entretanto, a qualidade desses serviços depende, em grande parte, da aquisição de reagentes e materiais utilizados nos experimentos e nas análises laboratoriais. Especificações incompletas e consolidação manual de informações contribuem para a morosidade do processo de compra. Para minimizar esse problema, construiu-se um modelo experimental embasado no aplicativo Excel, que padroniza e automatiza as etapas desse processo. A estruturação do modelo foi feita da seguinte forma: i) planilha, por solicitante, com especificação completa de reagentes e materiais para laboratórios (baseada em histórico de compras), com campos para seleção dos itens desejados e inserção de quantitativos por projeto; ii) planilha de controle orçamentário com indicação de valores financeiros solicitados por projeto, campos para inserção de valores disponíveis no Sistema de Acompanhamento Orçamentário (SAO) e saldo por projeto; iii) planilha de consolidação automática das demandas, com identificação dos demandantes e projetos correlacionados; iv) planilha para coleta de preço, separada por lote; v) planilha de acompanhamento do pedido, com indicação do número de propostas recebidas dos fornecedores; vi) planilha com as especificações que serão usadas no Edital de compra. A simulação do modelo com um número pequeno de variáveis de entrada (materiais, solicitantes, etc.) mostrou que ele automatiza uma série de cálculos, classifica e disponibiliza as informações do pedido de compra numa formatação que facilita a coleta de preço e as especificações dos itens no Edital de licitação, acelerando a realização de compras destinadas aos laboratórios.     Randomised phase II trial of CAPTEM or FOLFIRI as SEcond-line therapy in NEuroendocrine CArcinomas and exploratory analysis of predictive role of PET/CT imaging and biological markers (SENECA trial): A study protocol

    Introduction Patients with metastatic or locally advanced, non-resectable, grade 3 poorly differentiated gastroenteropancreatic (GEP) and lung neuroendocrine carcinomas (NECs) are usually treated with in first-line platinum compounds. There is no standard second-line treatment on progression. Accurate biomarkers are needed to facilitate diagnosis and prognostic assessment of patients with NEC. Methods and analysis The SEcond-line therapy in NEuroendocrine CArcinomas (SENECA) study is a randomised, non-comparative, multicentre phase II trial designed to evaluate the efficacy and safety of folinic acid, 5-fluorouracil and irinotecan (FOLFIRI) or capecitabine plus temozolomide (CAPTEM) regimens after failure of first-line chemotherapy in patients with lung NEC and GEP-NEC. Secondary aims are to correlate the serum miRNA profile and primary mutational status of MEN1, DAXX, ATRX and RB-1 with prognosis and outcome and to investigate the prognostic and predictive role of the Ki-67 score and 18-fluorodeoxyglucose positron emission tomography/computed tomography (18 F-FDG PET/CT) or 68 Ga-PET/CT. The main eligibility criteria are age ≥18 years; metastatic or locally advanced, non-resectable, grade 3 lung or GEP-NECs; progression to first-line platinum-based chemotherapy. A Bryant and Day design taking into account treatment activity and toxicity was used to estimate the sample size. All analyses will be performed separately for each treatment group in the intention-to-treat population. A total of 112 patients (56/arm) will be randomly assigned (1:1) to receive FOLFIRI every 14 days or CAPTEM every 28 days until disease progression or unacceptable toxicity or for a maximum of 6 months. Patients undergo testing for specific biomarkers in primary tumour tissue and for miRNA in blood samples. MiRNA profiling will be performed in the first 20 patients who agree to participate in the biological substudy. Ethics and dissemination The SENECA trial, supported by Istituto Scientifico Romagnolo per lo Studio e la Cura dei Tumori (IRST), was authorised by the locals Ethics Committee and the Italian Medicines Agency (AIFA). Results will be widely disseminated via peer-reviewed manuscripts, conference presentations and reports to relevant authorities. The study is currently open in Italy.     Reliability of an isometric and isokinetic strength testing protocol of the knee and ankle in young adults

    Background: Isokinetic dynamometers are becoming very common in assessing muscle strength and pathology, both in research and clinical practice, but for most of those devices reliability studies are still needed to support their extended use. The aim of this study is to assess the test-retest reliability also in health adults. Methods: Thirty adults (13 male and 17 females; mean age 25.4 ± 2.7 years) were recruited among University students. They participated to two testing sessions (7 day apart) in which they performed isokinetic and isometric strength assessment of the knee and ankle flexion and extension. Results: All variable showed an Intra-class correlation coefficient higher than 0.7 (isometric knee extension 0.96; isokinetic knee extension 0.96; isokinetic knee flexion 0.97; isometric ankle right flexion pl and flexion do 0.75-0.96; isometric ankle left flexion pl and flexion do 0.78-0.97; isokinetic ankle right flexion pl and flexion do 0.88-0.73; isokinetic ankle right flexion pl and flexion do 0.88-0.85) and paired-sample t-test showed no significant difference. Moreover, most of the recorded values were included within the upper and lower limits of agreement. Conclusion: Multi-joint evaluation system is a reliable device to assess knee and ankle isokinetic and isometric strength among healthy adults
