    Spontaneous physical activity down-regulates Pax7 in cancer cachexia

    Emerging evidence suggests that the muscle microenvironment plays a prominent role in cancer cachexia. We recently showed that NF-kB - induced Pax7 overexpression impairs the myogenic potential of muscle precursors in cachectic mice, suggesting that lowering Pax7 expression may be beneficial in cancer cachexia. We evaluated the muscle regenerative potential after acute injury in C26 colon carcinoma tumor-bearing mice and healthy controls. Our analyses confirmed that the delayed muscle regeneration observed in muscles form tumor-bearing mice was associated with a persistent local inflammation and Pax7 overexpression. Physical activity is known to exert positive effects on cachectic muscles. However, the mechanism by which a moderate voluntary exercise ameliorates muscle wasting is not fully elucidated. To verify if physical activity affects Pax7 expression, we hosted control and C26-bearing mice in wheel-equipped cages and we found that voluntary wheel running down-regulated Pax7 expression in muscles from tumor-bearing mice. As expected, down-regulation of Pax7 expression was associated with a rescue of muscle mass and fiber size. Our findings shed light on the molecular basis of the beneficial effect exerted by a moderate physical exercise on muscle stem cells in cancer cachexia. Furthermore, we propose voluntary exercise as a physiological tool to counteract the over-expression of Pax7observed in cancer cachexia

    Nell\u2019ambito delle attivit\ue0 di un pi\uf9 ampio progetto di ricerca relativo alla tutela e salvaguardia del patrimonio storico- culturale soggetto a fenomeni franosi, la memoria propone un caso studio riferito alla chiesa romanica di S.Nicol\uf2 di Capodimonte situata sul promontorio di Portofino (Liguria), complesso inserito nella lista dei monumenti nazionali italiani. Lo studio \ue8 stato condotto innanzitutto raccogliendo dati generali (idrogeologici, geotecnici, strutturali, sui danni, ecc.), quindi proseguendo nella loro rielaborazione. Sono state identificate dettagliate sezioni di interesse che sono state utilizzate per effettuare le analisi numeriche FEM, finalizzate a comprendere i fenomeni in atto. Sono state eseguite diverse analisi, sia in condizioni free-field, sia per tenere conto della presenza della struttura, cambiando scenari di pericolosit\ue0. Gli andamenti degli spostamenti sotto il piano di fondazione sono stati confrontati con i danni presenti nella chiesa, consentendo la lettura del quadro fessurativo anche in base alla direzione dei movimenti di versante. Infine, \ue8 stata eseguita una valutazione di massima del livello di danno, che ha permesso di identificare il contesto critico per il monumento in esame

    Orthotopic Heart Transplantation: Bicaval Versus Biatrial Surgical Technique

    In 1967, the first cardiac transplantation was performed in South Africa by Christiaan Barnard, becoming one of the most pioneering events of the human history, comparable to the first step on the moon, 2 years later. Even if Barnard became extremely famous because of this outstanding operation, behind this event there were years and years of studies, experimentations and hard work done by others, in particular by Lower and Shumway. The initial technique, still called ‘standard technique’ is the biatrial one. In the late 1980s, alternatives like the ‘bicaval technique’ were developed in order to get a more anatomical result. In the present chapter, we will throw the reader into the early years of the cardiac transplantation era, describing all the efforts made by the “fathers” of the cardiac surgery in order to standardize techniques inherited by the modern surgeons. Afterwards, we will present a review of the literature to answer the question if the biatrial technique should still be called “standard technique”

    Metabolic risk factor profile associated with use of second generation antipsychotics: a cross-sectional study in a community mental health centre

    open9noBACKGROUND: Second generation antipsychotics (SGA) have demonstrated several advantages over first generation antipsychotics (FGA) in terms of positive, negative, cognitive, and affective symptoms and a lower propensity for extrapyramidal side effects. Despite these undeniable advantages, SGA have been associated with causing and exacerbating metabolic disorders, such as obesity, diabetes, and hyperlipidemia. This cross sectional study aimed to evaluate the metabolic risk factor profile associated with use of SGAs in comparison with non -treated control patients. METHODS: The study was carried out at a Community Mental Health Centre (CMHC) in Bologna. The study subjects were outpatients with serious mental disorders treated with SGA (clozapine, olanzapine, risperidone, quetiapine). A sample of adult men and women suffering from idiopathic hyperhydrosis, without psychiatric history or antipsychotic treatment, were randomly selected from outpatients of the Department of Neurology in Bologna as a reference group. We investigated differences among the treatment and reference groups for glycaemia, cholesterolaemia and triglyceridaemia levels. RESULTS: The study sample was composed of 76 patients, 38 males and 38 females. The reference group was composed of 36 subjects, 19 females and 17 males. All patients treated with SGAs had higher mean glycaemia and triglyceridaemia and a significantly higher risk of receiving a diagnosis of hyperglycaemia and hypertriglyceridaemia than the reference group. We did not find any differences in mean glycaemia or mean triglyceridaemia levels among treatment groups. Patients with clozapine had a significantly higher mean BMI value and rate of obesity than patients treated with other SGAs. CONCLUSION: The rate of obesity and metabolic disorders observed in this study were higher than the prevalence in the control group and similar to that previously reported in psychiatric samples; these findings imply per se that more attention should be paid to the metabolic condition of psychiatric patients.     In this paper we study automatic composition synthesis of e-Services, based on automated reasoning. We represent the behavior of an e-Service in terms of a deterministic transition syst (or a finite state machine), in which for each action the role of the e-Service, either as initiator or as servant, is highlighted. In this setting we present an algorithm based on satisfiability in a variant of Propositional Dynamic Logic that solves the automatic composition probl. Specifically, given (i) a possibly incomplete specification of the sequences of actions that a client would like to realize, and (ii) a set of available e-Services, our technique synthesizes a composite e-Service that (i) uses only the available e-Services and (ii) interacts with the client "in accordance" to the given specification.     Ultra late onset group B streptococcal sepsis with acute renal failure in a child with urethral obstruction: a case report

    <p>Abstract</p> <p>Introduction</p> <p>Group B streptococci are a well-known cause of early and late onset sepsis. In neonates and older children gram-negative bacteria are mostly found in urinary tract infections and urosepsis. In adults predisposing factors for group B streptococci urinary tract infection may include diabetes mellitus and chronic renal failure.</p> <p>Case presentation</p> <p>We present a rare case of a five-month-old Caucasian boy with ultra late onset urosepsis and acute renal failure caused by group B streptococci serotype V. Excretion urography showed a subvesical obstruction that consequently was surgically corrected after antibiotic treatment of the acute infection.</p> <p>Conclusions</p> <p>Group B streptococci serotype V, urogenitary tract malformations, previous hospitalization and medical interventions may be important risk factors for the development of ultra late onset Group B streptococci sepsis in non-neonates.</p

    Epha3 acts as proangiogenic factor in multiple myeloma

    This study investigates the role of ephrin receptor A3 (EphA3) in the angiogenesis of Multiple Myeloma (MM) and the effects of a selective target of EphA3 by a specific monoclonal antibody on primary bone marrow endothelial cells (ECs) of MM patients.EphA3 mRNA and protein were evaluated in ECs of MM patients (MMECs), in ECs of patients with monoclonal gammopathies of undetermined significance (MGECs) and in ECs of healthy subjects (control ECs). The effects of EphA3 targeting by mRNA silencing (siRNA) or by the anti EphA3 antibody on the angiogenesis were evaluated. We found that EphA3 is highly expressed in MMECs compared to the other EC types. Loss of function of EphA3 by siRNA significantly inhibited the ability of MMECs to adhere to fibronectin, to migrate and to form tube like structures in vitro, without affecting cell proliferation or viability. In addition, gene expression profiling showed that knockdown of EphA3 down modulated some molecules that regulate adhesion, migration and invasion processes. Interestingly, EphA3 targeting by an anti EphA3 antibody reduced all the MMEC angiogenesis-related functions in vitro. In conclusion, our findings suggest that EphA3 plays an important role in MM angiogenesis
