    SUB-ACUTE TOXICITY STUDY OF PEGAGAN EMBUN (HYDROCOTYLE SIBTHORPIOIDES LAM.) EXTRACT ON THE SGPT AND SGOT LEVEL OF WISTAR WHITE MALE RATS

    Objective: Pegagan embun (Hydrocotyle sibthorpioides Lam.) is one of the herbs used in ethnomedicines as an immunostimulant during the COVID-19 pandemic. This present study aims to discover the potential toxicity effect of pegagan embun extract through sub-acute administration on the SGPT and SGOT levels of Wistar white male rats. Methods: Thirty-six test animals were divided into four groups: the control group was given Na CMC 0.5%, and the treatment groups were treated with ethanol extract of pegagan embun at doses of 7, 35, and 150 mg/kgBW. All groups were treated orally for 7, 14, and 21 d once daily. On the 8th, 15th, and 22nd day, the SGPT and SGOT of the test animal level were measured. The data were analyzed by two-way ANOVA followed by Duncan’s multiple range test (p<0.05). Results: The study revealed that administration of pegagan embun extract did not cause any harmful effect on the liver but significantly decreased the level of SGPT and SGOT influenced by the variety of doses and duration of administration (p<0.05). Significant reductions in SGPT and SGOT levels are seen after extract administration at dosages of 7 mg/kgBW for 21 d. Conclusion: This study showed that pegagan embun (Hydrocotyle sibthorpioides Lam.) extract sub-acute administration at doses of 7, 35, and 150 mg/kgBW is relatively non-toxic and safe to be used as an immunostimulant. There was no sign of damage showed in the liver of treated rats based on the levels of SGOT and SGPT

    Primers Designed For Amplifying TCF7L2 Gen

    Diabetes Mellitus (DM) is a disease characterized by high glucose levels because of the lack of insulin hormone in the body. Prevalence of Type-2 Diabetes Mellitus is associated with the variant of the TCF7L2 gene polymorphism, or SNP. The purpose of this study was to construct a specific primer to amplify the TCF7L2 gene fragment and their capabilities to detect the targeted area. This research was a descriptive study that design the primers using Geneious software version 5.4.7. The study was conducted at the Laboratory of Genetics and Biotechnology FMIPA Universitas Negeri Padang. Based on this research, a 10qF forward primer (NG_012631.1) and 10qR reverse primers (NG_012631.1) has successfully designed. The primers can amplify the targeted area of the TCF7L2 gene with 1208 bp length. The amplified gene region has many variants that are strictly related to the appearance of Type 2 Diabetes Mellitus

    THE EFFECT OF BROMELAIN MICROCAPSUL FORMULATION ON LEUKOCYTE AND TNF-α LEVEL IN MALE WHITE MICE INDUCED BY H5N1 VACCINE

    Objective: Bromelain is a sulfhydryl proteolytic enzyme extracted from the pineapple plant (Ananas comosus. L), which has various activities, including as an immunomodulator. Microencapsulation of bromelain is a process by which a layer surrounds bromelain to produce microcapsules to increase its activity. This research intends to see the effect of bromelain microcapsule formulation on total leukocytes count, leukocyte percentage, and the levels of TNF-α in male white mice exposed to the H5N1 Vaccine. Methods: Experimental animals were divided into three groups, specifically a negative control group given Na CMC 0.5%, the comparison group given 200 mg/kgBW bromelain enzyme, and the test group given 200 mg/kgBW bromelain microcapsules for seven days orally. On the eighth day, the total amount and the percentage of leukocytes and the levels of TNF-α were counted. The data were analyzed by two-way ANOVA and Duncan’s multiple range test (p<0.05). Results: The study showed that the administration of 200 mg/kgBW bromelain microcapsule group significantly reduced total leukocyte count and increased the segmented neutrophil compared to the bromelain group (p<0.05). However, there was no significant correlation between the two groups in reducing monocyte, lymphocyte, eosinophil, and TNF-α levels (p>0.05). Conclusion: It can be concluded that providing bromelain microcapsules can reduce the total amount of leukocytes and increase the segmented neutrophil in male white mice exposed to the H5N1 Vaccine

    One of the factors influencing the level of drug usage is the level of knowledge. Knowledge is an essential aspect in shaping concrete actions. In society, various issues related to drug usage often arise due to a lack of understanding and incorrect behaviours regarding drug usage. Educating the public about the selection and rational use of drugs is compulsary cause of that. This activity was carried out on May 7, 2023, at Jondul Housing, Padang City. The education method employed was a lecture delivered to 35 participating mothers. This method has proven effective for all segments of society, regardless of their educational background. The general public showed high enthusiasm in participating and actively engaged in interactions, asking questions, and discussing with the speaker. The conclusion drawn from this activity is that education and outreach enhanced the public knowledge about the rational use of drugs

    Malondialdehyde and TNF-α lowering effects of purified gambier (Uncaria gambir Roxb.) in diabetic rats

    Background: Malondialdehyde (MDA) is one of a dominat marker in oxidative stress condition, and when inflammation occurred tumor necrosis factor- α (TNF-α) played a significant influence in the propagation this process. Purified gambier (Uncaria gambier Roxb.) contained 90% catechin which is proven to have antioxidant activity and may prevent unwanted inflammatory responses during diabetic state. Objective: The objective of this research was to assess how purified gambier affected plasma MDA and TNF- α levels in alloxan-induced diabetic rats. Material and methods: In this study, 35 rats were used. Alloxan 120 mg/kg BW intraperitoneal injection was administered to induce diabetes conditions in rats. All animals were divided into 5 groups, diabetic control group treated with vehicle, positive control group treated with glibenclamide dose 0.45 mg/kg BW), and treatment groups treated with purified gambier dose of 2.5; 5 and 10 mg/kg BW. All animals were treated respectively for 14 days. Plasma MDA and TNF- α levels were measured on day 3, and 14. Results: Two-way ANOVA was applied to analyze all of the data, these findings suggested that purified gambier has antioxidant-related anti-inflammation actions. possesses blood sugar-lowering activity (p<0.05). The plasma MDA and TNF- α level of treatment group were significantly reduced (p<0.05) compared to diabetes control group. Conclusion: These results depicted that at doses of 2.5–10 mg/kg BW, purified gambier has antioxidant-associated anti-inflammation effects when given for 14 days on diabetic rat model by reducing plasma levels MDA and TNF-α
