    Evaluation of Variation Attributable to Lab and Technician for Measurements of Beef Carcass Traits Made Using Ultrasound

    National cattle genetic evaluations assume technician and imaging lab do not contribute to phenotypic variation when measuring carcass traits by ultrasound. The objective of this study was to estimate variance components of ultrasound carcass measurements, specifically variance contributed by ultrasound technician and imaging laboratory. Accounting for technician and imaging lab variation may increase accuracy of genetic predictions for carcass traits. Ultrasound carcass predictions for ribeye area (REA), percent intramuscular fat (IMF), and backfat (BF) were provided by the American Angus Association (AAA; n=281,982 animals), American Hereford Association (AHA; n=49,602), and American Simmental Association (ASA; n=59,576) for a total of 391,160 animals. Data provided by each association included ultrasound carcass measurements, contemporary group, technician ID, imaging lab, and a three-generation pedigree for each animal with ultrasound measurements. Variance contributed by additive genetics, technician, and contemporary group on ultrasound carcass measurements were estimated for each breed separately. Because technician and lab were confounded, the contribution of lab to ultrasound carcass variation could not be separated from technician. Therefore, we assessed whether lab contributed to ultrasound carcass variation by estimating genetic correlations between laboratories for ribeye area, percent intramuscular fat, and backfat. This analysis separated carcass traits by laboratory and treated measurements interpreted by each lab as a separate trait. Technician explained 12-27%, 4-23%, and 4-27% of variance for IMF, BF, and REA respectively across all three breeds. On average, the variance contributed by technician was greater than the variance contributed by additive genetics but less than that explained by contemporary group. Genetic correlations between labs across breeds ranged from 0.79 to 0.95 for IMF, 0.64 to 0.94 for BF and 0.78 to 0.98 for REA. Technician contributed to variance in ultrasound measurements, but high genetic correlations between labs suggest lab plays a lesser role in contribution of variance to ultrasound measurements

    Landmarks in vaginal mesh development: polypropylene mesh for treatment of SUI and POP

    Vaginal meshes used in the treatment of stress urinary incontinence (SUI) and pelvic organ prolapse (POP) have produced highly variable outcomes, causing life-changing complications in some patients while providing others with effective, minimally invasive treatments. The risk:benefit ratio when using vaginal meshes is a complex issue in which a combination of several factors, including the inherent incompatibility of the mesh material with some applications in pelvic reconstructive surgeries and the lack of appropriate regulatory approval processes at the time of the premarket clearance of these products, have contributed to the occurrence of complications caused by vaginal mesh. Surgical mesh used in hernia repair has evolved over many years, from metal implants to knitted polymer meshes that were adopted for use in the pelvic floor for treatment of POP and SUI. The evolution of the material and textile properties of the surgical mesh was guided by clinical feedback from hernia repair procedures, which were also being modified to obtain the best outcomes with use of the mesh. Current evidence shows how surgical mesh fails biomechanically when used in the pelvic floor and materials with improved performance can be developed using modern material processing and tissue engineering techniques

    The occurrence of ionospheric signatures of plasmaspheric plumes over different longitudinal sectors

    Plasmaspheric plumes have ionospheric signatures and are observed as storm-enhanced density (SED) in global positioning system (GPS) total electron content (TEC). These ionospheric signatures have been primarily observed over the American sector and in a few limited examples over the European sector. This study examines the longitudinal occurrence frequency of plasmaspheric plumes. We analyzed all images from the Imager for Magnetopause-to-Aurora Global Exploration (IMAGE) Extreme Ultraviolet Imager (EUV) databases for the first half of 2001 and identified a total of 31 distinct plume intervals observed during different storm events. Out of the total IMAGE EUV plumes that we identified, 12 were projected over North America, 10 over Asia, and the remaining 9 were over Europe and the Atlantic Ocean. Using ground-based GPS TEC from MIT\u27s Madrigal database, we searched for corresponding SED/TEC plumes at different longitudinal sector and found 12 ionospheric SED plume signatures over North America, 4 over Europe, and 2 over Asia. This indicates that the observation probability of an ionospheric SED plume when a plasmaspheric plume is seen is 100% in the American sector, 50% in the European sector, and 20% in the Asian sector. This could be due to the fact that the plumes may be either positioned beyond the limit of the ground-based GPS field of view, which happens mainly when there is less plasmaspheric erosion, or are too weak to be detected by the sparse number of GPS receivers over Asia. The in situ plasma densities from the available coincident defense metrological satellite program (DMSP) satellites were also used to study the characteristics of SED/TEC plume at DMSP orbiting altitude (i.e., ∼870 km).     Functional outcomes in early (T1/T2) supraglottic cancer: a systematic review

    ObjectivesOrgan preserving surgery (OPS) and radiotherapy (RT) are both accepted treatment options for early stage supraglottic cancer (SGC). Radiation has supplanted surgery in most cases, because of the perception that surgery results in poorer functional outcomes. However, evidence suggests that OPS with a neck dissection may be associated with improved survival. Our objective was to conduct a systematic review of the literature to compare functional outcomes of OPS and RT for early SGC.MethodsWe searched Medline, EMBASE and Cochrane Central Register of Controlled Trials to identify studies. Studies were included if they reported functional outcomes on 10 or more patients with early stage SGC treated with radiation or OPS, including open partial laryngectomy, transoral laser microsurgery (TLM) or transoral robotic surgery (TORS). Two reviewers independently screened articles for relevance using pre-determined criteria.ResultsFrom 7720 references, we included 10 articles (n=640 patients). 50% (n=320) of patients were treated with surgery. Three head-to-head RT versus OPS papers were included, however different outcome measures were used for each group. Intractable aspiration management (including total laryngectomy or permanent tracheostomy) following OPS was reported in five papers representing 186 patients; the definitive intractable aspiration management rate was 2.6% (95% CI 1.0-6.8%). Four papers reported permanent G-tube rate for the surgical group (n=198), calculating a rate of 5.3% (95% CI 2.6-10.5%), this was not reported for the RT group in any papers. One study reported quality of life. Two studies reported objective voice measures.ConclusionsThis systematic review revealed a paucity of objective measures and significant data heterogeneity, rendering the comparison of functional outcomes following OPS versus RT for early SGC limited. Future research should include objective measures of functional outcomes including laryngectomy rate, g-tube rate, tracheostomy dependence, quality of life, and voice quality measures

    Lessons from Archives: Strategies for Collecting Sociocultural Data in Machine Learning

    A growing body of work shows that many problems in fairness, accountability, transparency, and ethics in machine learning systems are rooted in decisions surrounding the data collection and annotation process. In spite of its fundamental nature however, data collection remains an overlooked part of the machine learning (ML) pipeline. In this paper, we argue that a new specialization should be formed within ML that is focused on methodologies for data collection and annotation: efforts that require institutional frameworks and procedures. Specifically for sociocultural data, parallels can be drawn from archives and libraries. Archives are the longest standing communal effort to gather human information and archive scholars have already developed the language and procedures to address and discuss many challenges pertaining to data collection such as consent, power, inclusivity, transparency, and ethics & privacy. We discuss these five key approaches in document collection practices in archives that can inform data collection in sociocultural ML.     Assessment of the angiogenic potential of 2-deoxy-D-ribose using a novel in vitro 3D dynamic model in comparison with established in vitro assays

    Angiogenesis is a highly ordered physiological process regulated by the interaction of endothelial cells with an extensive variety of growth factors, extracellular matrix components and mechanical stimuli. One of the most important challenges in tissue engineering is the rapid neovascularization of constructs to ensure their survival after transplantation. To achieve this, the use of pro-angiogenic agents is a widely accepted approach. The study of angiogenesis has gained momentum over the last two decades. Although there are various in vitro, ex vivo, and in vivo angiogenesis models that enable testing of newly discovered pro-angiogenic agents, the problem with researching angiogenesis is the choice of the most appropriate assay. In vivo assays are the most representative and reliable models, but they are expensive, time-consuming and can cause ethical concerns whereas in vitro assays are relatively inexpensive, practical, and reproducible, but they are usually lack of enabling the study of more than one aspect of angiogenesis, and they do not fully represent the complexity of physiological angiogenesis. Therefore, there is a need for the development of an angiogenesis model that allows the study of angiogenesis under physiologically more relevant, dynamic conditions without causing ethical concerns. Accordingly, in this study, we developed 3D in vitro dynamic angiogenesis model, and we tested the angiogenic potential of 2-deoxy-D-ribose (2dDR) in comparison with vascular endothelial growth factor (VEGF) using newly developed in vitro 3D dynamic model and well-established in vitro models. Our results obtained using conventional in vitro assays demonstrated that 2dDR promoted proliferation, migration and tube formation of human aortic endothelial cells (HAECs) in a dose-dependent manner. Then, the angiogenic activity of 2dDR was further assessed using the newly developed 3D in vitro model, which enabled the monitoring of cell proliferation and infiltration simultaneously under dynamic conditions. Our results showed that the administration of 2dDR and VEGF significantly enhanced the outgrowth of HAECs and the cellular density under either static or dynamic conditions

    Ferrimagnetism in sputtered MnxCoGe thin films

    Investigations into the magnetic properties of sputtered MnxCoGe films in the range 0.8 <= x <= 2.5 uncovered ferrimagnetic order, unlike the ferromagnetic order reported in bulk samples. These films formed hexagonal Ni2In-type structures in all measured compositions. While the Curie temperatures of the films are comparable to those of hexagonal bulk MnCoGe, here is a reduction in the magnetization of the MnxCoGe film relative to bulk MnCoGe, and a magnetization compensation point is observed in the x < 1 samples. To understand the behavior, we calculated the magnetic moments of Mn-antisite defects in MnCoGe with density-function theory calculations. Models constructed from the calculation suggest that films become ferrimagnetic due to the presence of Mn on the Co and Ge sites. In the x < 1 samples, these defects arose from the disorder in the films, whereas for x > 1, the excess Mn was driven onto the antisites and produced ferrimagnetic order.Comment: 8 pages, 7 figure

    Decellularised extracellular matrix decorated PCL PolyHIPE scaffolds for enhanced cellular activity, integration and angiogenesis

    Wound healing involves a complex series of events where cell–cell and cell-extracellular matrix (ECM) interactions play a key role. Wounding can be simple, such as the loss of the epithelial integrity, or deeper and more complex, reaching to subcutaneous tissues, including blood vessels, muscles and nerves. Rapid neovascularisation of the wounded area is crucial for wound healing as it has a key role in supplying oxygen and nutrients during the highly demanding proliferative phase and transmigration of inflammatory cells to the wound area. One approach to circumvent delayed neovascularisation is the exogenous use of pro-angiogenic factors, which is expensive, highly dose-dependent, and the delivery of them requires a very well-controlled system to avoid leaky, highly permeable and haemorrhagic blood vessel formation. In this study, we decorated polycaprolactone (PCL)-based polymerised high internal phase emulsion (PolyHIPE) scaffolds with fibroblast-derived ECM to assess fibroblast, endothelial cell and keratinocyte activity in vitro and angiogenesis in ex ovo chick chorioallantoic membrane (CAM) assays. Our results showed that the inclusion of ECM in the scaffolds increased the metabolic activity of three types of cells that play a key role in wound healing and stimulated angiogenesis in ex ovo CAM assays over 7 days. Herein, we demonstrated that fibroblast-ECM functionalised PCL PolyHIPE scaffolds appear to have great potential to be used as an active wound dressing to promote angiogenesis and wound healing

    An evidence map of psychosocial interventions for the earliest stages of bipolar disorder.

    Depression, schizophrenia, and bipolar disorder are three of the four most burdensome problems in people aged under 25 years. In psychosis and depression, psychological interventions are effective, low-risk, and high-benefit approaches for patients at high risk of first-episode or early-onset disorders. We review the use of psychological interventions for early-stage bipolar disorder in patients aged 15-25 years. Because previous systematic reviews had struggled to identify information about this emerging sphere of research, we used evidence mapping to help us identify the extent, distribution, and methodological quality of evidence because the gold standard approaches were only slightly informative or appropriate. This strategy identified 29 studies in three target groups: ten studies in populations at high risk for bipolar disorder, five studies in patients with a first episode, and 14 studies in patients with early-onset bipolar disorder. Of the 20 completed studies, eight studies were randomised trials, but only two had sample sizes of more than 100 individuals. The main interventions used were family, cognitive behavioural, and interpersonal therapies. Only behavioural family therapies were tested across all of our three target groups. Although the available interventions were well adapted to the level of maturity and social environment of young people, few interventions target specific developmental psychological or physiological processes (eg, ruminative response style or delayed sleep phase), or offer detailed strategies for the management of substance use or physical health
