    Gbp2 interacts with THO/TREX through a novel type of RRM domain

    Metazoan SR and SR-like proteins are important regulatory factors in RNA splicing, export, translation and RNA decay. We determined the NMR structures and nucleic acid interaction modes of Gbp2 and Hrb1, two paralogous budding yeast proteins with similarities to mammalian SR proteins. Gbp2 RRM1 and RRM2 recognise preferentially RNAs containing the core motif GGUG. Sequence selectivity resides in a non-canonical interface in RRM2 that is highly related to the SRSF1 pseudoRRM. The atypical Gbp2/Hrb1 C-terminal RRM domains (RRM3) do not interact with RNA/DNA, likely because of their novel N-terminal extensions that block the canonical RNA binding interface. Instead, we discovered that RRM3 is crucial for interaction with the THO/TREX complex and identified key residues essential for this interaction. Moreover, Gbp2 interacts genetically with Tho2 as the double deletion shows a synthetic phenotype and preventing Gbp2 interaction with the THO/TREX complex partly supresses gene expression defect associated with inactivation of the latter complex. These findings provide structural and functional insights into the contribution of SR-like proteins in the post-transcriptional control of gene expression.Peer Reviewe

    Factors associated with health related quality of life after a hip or knee total replacement, according to a gender approach

    Objective: To identify the factors associated with Health Related Quality of Life (HRQL), and Symptoms and Physical Functional Disability (S&PFD) after a Hip or Knee Total Replacement, according to a gender approach. A longitudinal cohort study design is performed at “La Merced” (Osuna. Sevilla) and “Torrecárdenas” (Almería) Hospitals, via telephone interview since October 2004 to November 2006. The study included all people who underwent a hip or knee total replacement. The dependent variables were HRQL and S&PFD, both measured via short versions of the COOP/WONCA and WOMAC questionnaires.Results: 311 people were included, who were mostly women (82,63%) and 70 years old (average). The hip and knee replacement obtained an statistically significant improvement on HRQL (20,5%) and S&PFD (28,2%), with no gender difference. Five variables were associated to HRQL for Women: HRQL previous to surgery, hospital, local infection as a complication, autonomy for daily living, and marital status. All of them were associated to men, except the last two ones. About S&PFD, six variables were associated for women: HRQL and S&PFD previous to surgery, hospital, local infection as a complication, body mass index and marital status. All of them were associated to men, except the last two ones.Factors associated to improve on HRQL and S&PFD in male population are few, though they find a great explanation of the improvement. Different situation arise on female population because more factors obtained a substantially inferior explanation

    Magnetic structure and properties of the honeycomb antiferromagnet [Na(OH2)3]Mn(NCS)3

    We report the magnetic structure and properties of a thiocyanate-based honeycomb magnet [Na(OH2)3]Mn(NCS)3 which crystallises in the unusual low-symmetry trigonal space group P3̄. Magnetic measurements on powder samples show this material is an antiferromagnet (ordering temperature TN,mag = 18.1(6) K) and can be described by nearest neighbour antiferromagnetic interactions J = −11.07(4) K. A method for growing neutron-diffraction sized single crystals (>10 mm3) is demonstrated. Low temperature neutron single crystal diffraction shows that the compound adopts the collinear antiferromagnetic structure with TN,neut = 18.94(7) K, magnetic space group P3̄′. Low temperature second-harmonic generation (SHG) measurements provide no evidence of breaking of the centre of symmetry

    The tumor suppressor p53 consists of four 393-residue chains, each of which has two natively unfolded (N- and C-terminal) and two folded (core and tetramerization) domains. Their structural organization is poorly characterized as the protein tends to aggregate, has defied crystallization, and is at the limits of NMR studies. We first stabilized the protein by mutation to make it more suitable for extended study and then acquired NMR spectra on full-length protein and various combinations of shorter domain constructs. The NMR spectrum ((15)N,(1)H transverse relaxation optimized spectroscopy) of full-length p53 was close to that expected from the sum of the spectra of isolated individual domains. However, patterns of changes in chemical shifts revealed unexpected interactions between the core domains. We used the NMR data as constraints in docking algorithms and found a previously uncharacterized self-complementary surface for the association of core domains into dimers within the tetrameric complex. Binding to DNA requires about a 70° rotation to break those subunit interactions and form the known protein:protein interface in the p53–DNA complex. We verified the interactions by the effects of mutation on DNA binding.
