    Association between steroid hormone receptors and PSA gene expression in breast cancer cell lines

    The prostate specific antigen (PSA) gene is a member of the human kallikrein gene family and is known that to be tightly regulated by androgens in the male prostate The presence of PSA is stronglyassociated with presence of steroid hormone receptors. The aim of this research was to show differential expression and association between steroid hormone receptors and PSA gene expression in breast cancer cell lines. The cell lines investigated were steroid receptor-negative breast carcinoma cell lines BT-20 and HBT-100 and the steroid hormone receptor-positive breast carcinoma cell lines BT- 474, ZR-75-1, MDA-MB-453 and MFM-233. Others include BG-1 (ovarian), MFE-296 (endometrium), HBT-161 (ovarian), HBT-173 (ovarian), A-427 (lung), SK-MES-1(lung), NCI-H460(lung),MIA PaCa (pancreas), and Colo320 (Colon) cell lines. All cell lines were cultured as confluent and then harvested. Stimulationtest was carried out using steroid compound. The concentration of estrogen receptor (ER) and progesterone receptor (PR) in all tumor cell lines were measured with the abbot enzyme immunoassay kit. Among cell lines tested, only steroid receptor positive cells were able to produce PSA at the protein and the mRNA level in response to stimulatory effects of steroid hormones. Our data suggest that the expression of the PSA gene in different cancer cell lines may depends on the presence of steroidhormone receptors

    Increased serum IL-6 level time-dependently regulates hyperalgesia and spinal mu opioid receptor expression during CFA-induced arthritis

    Interleukin (IL)-6 is known to cause pro- and anti-inflammatory effects during different stages of inflammation. Recent therapeutic investigations have focused on treatment of various inflammatory disorders with anti-cytokine substances. As a result, the aim of this study was to further elucidate the influence of IL-6 in hyperalgesia and edema during different stages of Complete Freund’s Adjuvant (CFA)-induced arthritis (AA) in male Wistar rats. AA was induced by a single subcutaneous injection of CFA into the rats’ hindpaw. Anti-IL-6 was administered either daily or weekly during the 21 days of study. Spinal mu opioid receptor (mOR) expression was detected by Western blotting. Daily and weekly treatment with an anti-IL-6 antibody significantly decreased paw edema in the AA group compared to the AA control group. Additionally, daily and weekly anti-IL-6 administration significantly reduced hyperalgesia on day 7 in the AA group compared to the AA control group; however, there were significant increases in hyperalgesia in the antibody-treated group on days 14 and 21 compared to the AA control group. IL-6 antibody-induced increases in hyperalgesia on the 14th and 21st days after CFA injection correlated with a time-dependent, significant reduction in spinal mOR expression during anti-IL-6 treatment. Our study confirmed the important time-dependent relationship between serum IL-6 levels and hyperalgesia during AA. These results suggest that the stages of inflammation in AA must be considered for anti-hyperalgesic and anti-inflammatory interventions via anti-IL-6 antibody treatment

    N-acetylcysteine compared to metformin, improves the expression profile of growth differentiation factor-9 and receptor tyrosine kinase c-kit in the oocytes of patients with polycystic ovarian syndrome

    Background: Paracrine disruption of growth factors in women with polycystic ovarian syndrome (PCOS) results in production of low quality oocyte, especially following ovulation induction. The aim of this study was to investigate the effects of metformin (MET), N-acetylcysteine (NAC) and their combination on the hormonal levels and expression profile of GDF-9, BMP-15 and c-kit, as hallmarks of oocyte quality, in PCOS patients. Materials and Methods: This prospective randomized, double-blind, placebo controlled trial aims to study the effects of MET, NAC and their combination (MET+NAC) on expression of GDF-9, BMP-15 and c-kit mRNA in oocytes [10 at the germinal vesicle (GV) stage, 10 at the MI stage, and 10 at the MII stage from per group] derived following ovulation induction in PCOS. Treatment was carried out for six weeks, starting on the third day of previous cycle until oocyte aspiration. The expression of GDF9, BMP15 and c-kit were determined by quantitative real time polymerase chain reaction (RT-qPCR) and western blot analysis. Data were analyzed with one-way ANOVA. Results: The follicular fluid (FF) level of c-kit protein significantly decreased in the NAC group compared to the other groups. Significant correlations were observed between the FF soluble c-kit protein with FF volume, androstenedione and estradiol. The GDF-9 expression in unfertilized mature oocytes were significantly higher in the NAC group compared to the other groups (P<0.001). Similar difference was not observed between the MET, NAC+MET and control groups. The c-kit expression in unfertilized mature oocytes were significantly lower in the NAC group compared to the other groups (P<0.001). Similar difference was not observed between the MET, NAC+MET and control groups (Registration number: IRCT201204159476N1).     Intranasal administration of endometrial mesenchymal stem cells as a suitable approach for Parkinson�s disease therapy

    This study aimed to investigate the therapeutic effects of intranasal administration of human endometrium-derived stem cells (HEDSCs) in the mouse model of Parkinson�s disease (PD). Thirty days after intrastriatal injection of 6-OHDA, HEDSCs were administrated intranasally in three doses (104, 5 � 104 and 105 cells µl�1). During 120 days after stem cell administration, behavioral tests were examined. Then the mice were sacrificed and the fresh section of the substantia nigra pars compacta (SNpc) was used for detection of HEDSCs-GFP labeled by fluorescence microscopy method. In addition, immunohistochemistry was used to assay GFP, human neural Nestin, and tyrosine hydroxylase (TH) markers in the fixed brain tissue at the SNpc. Our data revealed that behavioral parameters were significantly improved after cell therapy. Fluorescence microscopy assay in fresh tissue and GFP analysis in fixed tissue were showed that the HEDSCs-GFP labeled migrated to SNpc. The data from immunohistochemistry revealed that the Nestin as a differential neuronal biomarker was expressed in SNpc. Also, TH as a dopaminergic neuron marker significantly increased after HEDSCs therapy in an optimized dose 5 � 104 cells µl�1. Our results suggest that intranasal administration of HEDSCs improve the PD symptoms in the mouse model of PD dose-dependent manner as a noninvasive method. © 2019, Springer Nature B.V

    Effect of High-Density Packing Recycled Aggregate on Concrete Strength Properties

    As many residential homes and structures were destroyed or badly damaged because of the military battle in Iraq, it is critical to recycle construction debris widely in the rebuilding and decoration of buildings and infrastructure. This battle resulted in the buildup of massive construction debris, and recycling this waste allows for a cleaner environment. Recycling the construction waste of various installations is an urgent need to decrease the consumption of natural materials and landfills of construction waste, and as a result reduce the environmental pollution. Therefore, this study is a local focus on using concrete debris to obtain high packing density recycled coarse and fine aggregates in various fractions of (0.16 mm - 10 mm) by selecting high-density packing materials that was developed by Kharkhadin A.N in laboratories of Belgorod State Technical University in Russia to preparing of reference mix from ordinary density recycled aggregate. The new concrete samples for two concrete mixtures were prepared, to identify and study the important specifications. Models of cubes and standard prisms were prepared to evaluate the compressive strength and the splitting tensile strength. Also, the modulus of rupture and the unit weight were conducted. The results indicated an increase in the concrete’s mechanical properties using the high-packing density recycled aggregate. The obtained compressive strength, splitting tensile strength and modulus of rupture were (49) MPa, (3.6) MPa, and (7.1) MPa compared with a reference mix (39) MPa, (3.3) MPa and (6.3) MPa, respectively. The reference mix corresponding properties were (39) MPa, (3.3) MPa, and (6.3) MPa, respectively. Also the values of an oven-dry density were (2340) kg/m3 compared with the reference mix (2260kg/m³). These results proved that increasing the packing density of recycled aggregate enhanced the concrete’s strength properties

    On the emergent Semantic Web and overlooked issues

    The emergent Semantic Web, despite being in its infancy, has already received a lotof attention from academia and industry. This resulted in an abundance of prototype systems and discussion most of which are centred around the underlying infrastructure. However, when we critically review the work done to date we realise that there is little discussion with respect to the vision of the Semantic Web. In particular, there is an observed dearth of discussion on how to deliver knowledge sharing in an environment such as the Semantic Web in effective and efficient manners. There are a lot of overlooked issues, associated with agents and trust to hidden assumptions made with respect to knowledge representation and robust reasoning in a distributed environment. These issues could potentially hinder further development if not considered at the early stages of designing Semantic Web systems. In this perspectives paper, we aim to help engineers and practitioners of the Semantic Web by raising awareness of these issues

    Id1 regulates angiogenesis through transcriptional repression of thrombospondin-1

    AbstractId proteins are helix-loop-helix transcription factors that regulate tumor angiogenesis. In order to identify downstream effectors of Id1 involved in the regulation of angiogenesis, we performed PCR-select subtractive hybridization on wild-type and Id1 knockout mouse embryo fibroblasts (MEFs). Here we demonstrate that thrombospondin-1 (TSP-1), a potent inhibitor of angiogenesis, is a target of transcriptional repression by Id1. We also show that Id1-null MEFs secrete an inhibitor of endothelial cell migration, which is completely inactivated by depletion of TSP-1. Furthermore, in vivo studies revealed decreased neovascularization in matrigel assays in Id1-null mice compared to their wild-type littermates. This decrease was completely reversed by a TSP-1 neutralizing antibody. We conclude that TSP-1 is a major target for Id1 effects on angiogenesis

    On the Refractive Index of Ageing Dispersions of Laponite

    Aqueous dispersion of Laponite at low ionic concentration is of interest since it undergoes structural evolution with respect to time, which is usually termed as ageing. In this work we study the refractive index behavior as a function of ageing time, concentration and temperature. We observed that the extended Lorenz-Lorentz equation fitted the refractive index dependence on concentration and temperature very well. The refractive index did not show any dependence on ageing time. However, the dependence of refractive index on concentration showed a marked change as the system underwent transition from an isotropic to a biphasic state. The slope of the refractive index-density data is remarkably close to that of water at all Laponite concentrations. In the context of transport phenomena, optical measurements such as interferometry can exploit the water-like behavior of Laponite dispersions.Comment: 13 pages, 3 figures, to appear in Applied Clay Scienc

    The Ecm11-Gmc2 complex promotes synaptonemal complex formation through assembly of transverse filaments in budding yeast

    During meiosis, homologous chromosomes pair at close proximity to form the synaptonemal complex (SC). This association is mediated by transverse filament proteins that hold the axes of homologous chromosomes together along their entire length. Transverse filament proteins are highly aggregative and can form an aberrant aggregate called the polycomplex that is unassociated with chromosomes. Here, we show that the Ecm11-Gmc2 complex is a novel SC component, functioning to facilitate assembly of the yeast transverse filament protein, Zip1. Ecm11 and Gmc2 initially localize to the synapsis initiation sites, then throughout the synapsed regions of paired homologous chromosomes. The absence of either Ecm11 or Gmc2 substantially compromises the chromosomal assembly of Zip1 as well as polycomplex formation, indicating that the complex is required for extensive Zip1 polymerization. We also show that Ecm11 is SUMOylated in a Gmc2-dependent manner. Remarkably, in the unSUMOylatable ecm11 mutant, assembly of chromosomal Zip1 remained compromised while polycomplex formation became frequent. We propose that the Ecm11-Gmc2 complex facilitates the assembly of Zip1 and that SUMOylation of Ecm11 is critical for ensuring chromosomal assembly of Zip1, thus suppressing polycomplex formation

    DNA end resection by Dna2–Sgs1–RPA and its stimulation by Top3–Rmi1 and Mre11–Rad50–Xrs2

    The repair of DNA double-strand breaks (DSBs) by homologous recombination requires processing of broken ends. For repair to start, the DSB must first be resected to generate a 3′-single-stranded DNA (ssDNA) overhang, which becomes a substrate for the DNA strand exchange protein, Rad51 (ref. 1). Genetic studies have implicated a multitude of proteins in the process, including helicases, nucleases and topoisomerases. Here we biochemically reconstitute elements of the resection process and reveal that it requires the nuclease Dna2, the RecQ-family helicase Sgs1 and the ssDNA-binding protein replication protein-A (RPA). We establish that Dna2, Sgs1 and RPA constitute a minimal protein complex capable of DNA resection in vitro. Sgs1 helicase unwinds the DNA to produce an intermediate that is digested by Dna2, and RPA stimulates DNA unwinding by Sgs1 in a species-specific manner. Interestingly, RPA is also required both to direct Dna2 nucleolytic activity to the 5′-terminated strand of the DNA break and to inhibit 3′ to 5′ degradation by Dna2, actions that generate and protect the 3′-ssDNA overhang, respectively. In addition to this core machinery, we establish that both the topoisomerase 3 (Top3) and Rmi1 complex and the Mre11–Rad50–Xrs2 complex (MRX) have important roles as stimulatory components. Stimulation of end resection by the Top3–Rmi1 heterodimer and the MRX proteins is by complex formation with Sgs1 (refs 5, 6), which unexpectedly stimulates DNA unwinding. We suggest that Top3–Rmi1 and MRX are important for recruitment of the Sgs1–Dna2 complex to DSBs. Our experiments provide a mechanistic framework for understanding the initial steps of recombinational DNA repair in eukaryotes
