    The importance of hole concentration in establishing carrier-mediated ferromagnetism in Mn doped Ge

    In the present work, we have prepared Mn-doped Ge using different annealing approaches after Mn ion implantation, and obtained samples with hole concentrations ranging from 10^18 to 2.1x10^20 cm^-3, the latter being the highest reported so far. Based on the magnetotransport properties of Mn doped Ge, we argue that the hole concentration is a decisive parameter in establishing carrier-mediated ferromagnetism in magnetic Ge.Comment: 7 pages, 3 figure

    Reconciling the interests of the economic diversification participants in a single-industry town

    Рассматриваются проблемы согласования интересов участников процесса диверсификации экономики моногорода в процессе управления развитием моногорода на основе реализации стратегии диверсификации. Целью исследования является разработка инструментов гармонизации интересов участников процесса диверсификации экономики моногорода на основе разработки модели полного согласования интересов и сопоставления ее с моделью отдельно взятого моногорода. Методы исследования: системный подход, анализ и синтез, построение множеств согласования интересов с применением диаграммы Эйлера-Венна, методы комбинаторики, построение экономических показателей и рядов динамики, графический метод представления результатов и построения иерархии согласования интересов. Результаты исследования. Представлена модель полного согласования интересов различных групп лиц, в которой отражены сферы возможного пересечения интересов, а также иерархия числа согласуемых интересов. Предложены основные группы участников процесса диверсификации моногорода, а также разграничено понятие участника и роли (функции) участника в данном процессе. Обосновано и определено число множеств в зависимости от числа участников, чьи интересы одновременно согласуются, на примере моногорода Калтан Кемеровской области. На основе предложенной модели построена модель согласования интересов на примере моногорода Калтана. Полученная модель имеет трапециевидную форму, что свидетельствует о достаточно небольшом числе общих согласуемых интересов. Автор приходит к выводу, что в моногороде сформирован треугольник интересов «администрация города - малый бизнес в роли инвестора - вуз», представляющий основу в иерархии согласования интересов моногорода Калтан. Центральное положение при согласовании интересов с прочими участниками занимает исключительно администрация города. На основе данных выводов внесены предложения по направлениям дальнейшего согласования интересов на примере рассматриваемого моногорода.The article explores the problems of reconciling the interests of single-industry towns, a need that arises from the entire process of industry development management through a diversification strategy. The aim of this study is to develop tools to harmonize the interests of the single-industry towns, based on a model of full harmonization with the model of an individual single-industry town. Methods of the research: system approach, analysis and synthesis, building a set of harmonized interests with the application of the Eulerian-Venn diagram, combinatorics methods, construction of economic indicators and series of dynamics, graphical method of presentation of results and building the hierarchy of interests. Results of the study. The paper introduces the model for fully reconciling the interests of different groups of persons, reflecting the areas of possible intersecting interests and the hierarchy of the number of interests involved. The article proposes the main groups of participants in the diversification process and the differentially concept of participant and the role (function) of the participant in the diversification process. The author justifies and estimates the number of sets depending on the number of participants whose interests are concurrently reconciled in Kaltan case study. The proposed model is used for Kaltan, Kemerovo region case study. The result ing model is of a trapezoidal form, which indicates that there are quite a few common consenting interests. The author concludes that a triangle of interests «city administration - small business as investor - university» is formed. This triangle represents the highest level of harmonization of interests in the hierarchy. The city administration is in central position in reconciling interests with other participants. By the example of the considered single-industry town and based on these findings the suggestion on further harmonization of interests was made

    Light-cone distribution amplitudes of octet baryons from lattice QCD

    We present lattice QCD results for the wave function normalization constants and the first moments of the distribution amplitudes for the lowest-lying baryon octet. The analysis is based on a large number of Nf=2+1N_f=2+1 ensembles comprising multiple trajectories in the quark mass plane including physical pion (and kaon) masses, large volumes, and, most importantly, five different lattice spacings down to a=0.039fma=0.039\,\mathrm{fm}. This allows us to perform a controlled extrapolation to the continuum and infinite volume limits by a simultaneous fit to all available data. We demonstrate that the formerly observed violation of flavor symmetry breaking constraints can, indeed, be attributed to discretization effects that vanish in the continuum limit

    Molecular monitoring of minimal residual disease in two patients with MLL-rearranged acute myeloid leukemia and haploidentical transplantation after relapse

    This report describes the clinical courses of two acute myeloid leukemia patients. Both had MLL translocations, the first a t(10;11)(p11.2;q23) with MLL-AF10 and the second a t(11;19)(q23;p13.1) with MLL-ELL fusion. They achieved a clinical remission under conventional chemotherapy but relapsed shortly after end of therapy. Both had a history of invasive mycoses (one had possible pulmonary mycosis, one systemic candidiasis). Because no HLA-identical donor was available, a haploidentical transplantation was performed in both cases. Using a specially designed PCR method for the assessment of minimal residual disease (MRD), based on the quantitative detection of the individual chromosomal breakpoint in the MLL gene, all patients achieved complete and persistent molecular remission after transplantation. The immune reconstitution after transplantation is described in terms of total CD3+/CD4+, CD3+/CD8+, CD19+, and CD16+/CD56+ cell numbers over time. The KIR and HLA genotypes of donors and recipients are reported and the possibility of a KIR-mediated alloreactivity is discussed. This report illustrates that haploidentical transplantation may offer a chance of cure without chronic graft-versus-host disease in situations where no suitable HLA-identical donor is available even in a high-risk setting and shows the value of MRD monitoring in the pre- and posttransplant setting

    Prenatal human skin expresses the antimicrobial peptide RNase 7

    Antimicrobial peptides and proteins (AMPs) play important roles in skin immune defense due to their capacity to inhibit growth of microbes. During intrauterine life, the skin immune system has to acquire the prerequisites to protect the newborn from infection in the hostile environment after birth, which includes the production of skin AMPs. The aim of this study was to analyze the expression of RNase 7, HBD-2/3 and psoriasin during human skin development, thus, providing a deeper insight about the maturity of a fundamental component of the innate immune system. We found low RNase 7 expression levels in the periderm but no expression of HBD-2/3 and psoriasin in first trimester human skin using immunohistochemistry. At the end of the second trimester, RNase 7 is expressed weakly in all epidermal layers with a marked signal in the stratum corneum. HBD-3 and psoriasin are focally expressed while HBD-2 is not detectable. Analysis of supernatants from cultured prenatal skin cells showed that in contrast to adult control, RNase 7 and psoriasin are not found in prenatal skin, suggesting that AMPs are detectable but are not secreted. This study shows the differential expression of AMPs in developing, non-perturbed human prenatal skin. It is conceivable that the combined expression of RNase 7, HBD-3 and psoriasin in fetal skin constitutes a developmental program to exert a broad spectrum of antimicrobial activity to maintain sterility in the amniotic cavity

    Human Dermis Harbors Distinct Mesenchymal Stromal Cell Subsets

    Multipotent mesenchymal stromal cells (MSCs) are found in a variety of adult tissues including human dermis. These MSCs are morphologically similar to bone marrow–derived MSCs, but are of unclear phenotype. To shed light on the characteristics of human dermal MSCs, this study was designed to identify and isolate dermal MSCs by a specific marker expression profile, and subsequently rate their mesenchymal differentiation potential. Immunohistochemical staining showed that MSC markers CD73/CD90/CD105, as well as CD271 and SSEA-4, are expressed on dermal cells in situ. Flow cytometric analysis revealed a phenotype similar to bone marrow–derived MSCs. Human dermal cells isolated by plastic adherence had a lower differentiation capacity as compared with bone marrow–derived MSCs. To distinguish dermal MSCs from differentiated fibroblasts, we immunoselected CD271+ and SSEA-4+ cells from adherent dermal cells and investigated their mesenchymal differentiation capacity. This revealed that cells with increased adipogenic, osteogenic, and chondrogenic potential were enriched in the dermal CD271+ population. The differentiation potential of dermal SSEA-4+ cells, in contrast, appeared to be limited to adipogenesis. These results indicate that specific cell populations with variable mesenchymal differentiation potential can be isolated from human dermis.     Culture expansion of CAR T cells results in aberrant DNA methylation that is associated with adverse clinical outcome

    Chimeric antigen receptor (CAR) T cells provide new perspectives for treatment of hematological malignancies. Manufacturing of these cellular products includes culture expansion procedures, which may affect cellular integrity and therapeutic outcome. In this study, we investigated culture-associated epigenetic changes in CAR T cells and found continuous gain of DNAm, particularly within genes that are relevant for T cell function. Hypermethylation in many genes, such as TCF7, RUNX1, and TOX, was reflected by transcriptional downregulation. 332 CG dinucleotides (CpGs) showed an almost linear gain in methylation with cell culture time, albeit neighboring CpGs were not coherently regulated on the same DNA strands. An epigenetic signature based on 14 of these culture-associated CpGs predicted cell culture time across various culture conditions. Notably, even in CAR T cell products of similar culture time higher DNAm levels at these CpGs were associated with significantly reduced long-term survival post transfusion. Our data demonstrate that cell culture expansion of CAR T cells evokes DNA hypermethylation at specific sites in the genome and the signature may also reflect loss of potential in CAR T cell products.
