The problem is complex because it has to be viewed from all points of view and not only as an isolated system in the body, having aspects of feedback on the whole physiopathology belonging to the human body. The highly circulating system needs permanent maintenance. Self-maintenance is done through various physiological mechanisms tightly linked to each other, including the lymphatic, digestive, renal, lung, nervous, glandular system
 It is not possible to completely separate the physiology of a system from the other adjacent systems because they all work synergistically, being permanently controlled by the central and peripheral nervous system

    Iron Metabolism in the Tumor Microenvironment-Implications for Anti-Cancer Immune Response

    New insights into the field of iron metabolism within the tumor microenvironment have been uncovered in recent years. Iron promotes the production of reactive oxygen species, which may either trigger ferroptosis cell death or contribute to malignant transformation. Once transformed, cancer cells divert tumor-infiltrating immune cells to satisfy their iron demand, thus affecting the tumor immunosurveillance. In this review, we highlight how the bioavailability of this metal shapes complex metabolic pathways within the tumor microenvironment and how this affects both tumor-associated macrophages and tumor-infiltrating lymphocytes functions. Furthermore, we discuss the potentials as well as the current clinical controversies surrounding the use of iron metabolism as a target for new anticancer treatments in two opposed conditions: i) the "hot" tumors, which are usually enriched in immune cells infiltration and are extremely rich in iron availability within the microenvironment, and ii) the "cold" tumors, which are often very poor in immune cells, mainly due to immune exclusion

    Biomechanically tunable nano-silica/p-hema structural hydrogels for bone scaffolding

    Innovative tissue engineering biomimetic hydrogels based on hydrophilic polymers have been investigated for their physical and mechanical properties. 5% to 25% by volume loading PHEMA-nanosilica glassy hybrid samples were equilibrated at 37◩C in aqueous physiological isotonic and hypotonic saline solutions (0.15 and 0.05 M NaCl) simulating two limiting possible compositions of physiological extracellular fluids. The glassy and hydrated hybrid materials were characterized by both dynamo-mechanical properties and equilibrium absorptions in the two physiological-like aqueous solutions. The mechanical and morphological modifications occurring in the samples have been described. The 5% volume nanosilica loading hybrid nanocomposite composition showed mechanical characteristics in the dry and hydrated states that were comparable to those of cortical bone and articular cartilage, respectively, and then chosen for further sorption kinetics characterization. Sorption and swelling kinetics were monitored up to equilibrium. Changes in water activities and osmotic pressures in the water-hybrid systems equilibrated at the two limiting solute molarities of the physiological solutions have been related to the observed anomalous sorption modes using the Flory-Huggins interaction parameter approach. The bulk modulus of the dry and glassy PHEMA-5% nanosilica hybrid at 37◩C has been observed to be comparable with the values of the osmotic pressures generated from the sorption of isotonic and hypotonic solutions. The anomalous sorption modes and swelling rates are coherent with the difference between osmotic swelling pressures and hybrid glassy nano-composite bulk modulus: the lower the differences the higher the swelling rate and equilibrium solution uptakes. Bone tissue engineering benefits of the use of tuneable biomimetic scaffold biomaterials that can be “designed” to act as biocompatible and biomechanically active hybrid interfaces are discussed

    We calculate the inclusive production of charged hadrons in \pbp collisions to next-to-leading order (NLO) in the QCD improved parton model using a new set of NLO fragmentation functions for charged pions and kaons. We predict transverse-momentum distributions and compare them with experimental data from the CERN S\pbpS Collider and the Fermilab Tevatron.Comment: the file containing the figures has been replaced: we correct a mistake in the uuencoding procedure and we give the real Fig 4 instead of the spurious one which was accidentally included in the previous file. the text is unchanged

    PO-032 The knock-down of ferritin heavy subunit induces xenobiotic-resistance in k562 cells through the activation of nf-kb pathway

    Introduction The transcriptional factor NF-ÎșB, composed by five subunits (RelA/p65, c-Rel, RelB, p50, p52), is largely involved in many facets of cellular physiology such as innate and adaptive immunity as well as inflammation. In addition, NF-kB play a central role in cancer cell survival and chemoresistance partly by its implication in cross-talks with redox-regulating proteins. Ferritin is the major iron storage protein; it is composed by a variable assembly of Heavy (FHC) and Light (FLC) subunits. FHC, in particular, has been widely demonstrated to be devoted in iron uptake and release thus controlling the redox homeostasis. Material and methods K562 erythroleukemia cells were stably silenced for FHC by using the shRNA method. Then FHC reconstitution was achieved by transient transfection of a FHC specific expression vector. ROS were determined by incubating cells with the redox-sensitive probe 2'−7'-DCF. NAC was used to inhibit ROS production. MTT assay was performed to analyse cell viability. Increasing concentrations of Doxorubicin, ranging from 0 to 5 ”M, were used to treat K562 cells. Results and discussions The results of this study highlighted that FHC amounts negatively affect NF-kB activation in K562 cells. FHC silencing was accompanied by an increased expression of the nuclear NF-kB subunit p65, FHC rescue determined nuclear p65 decrease. FHC silencing is responsible for intracellular ROS production and ROS are implicated in NF-kB pathway. To elucidate the relationship between ROS amount and nuclear p65 content, we determined ROS amounts in our in vitro model and evaluated p65 nuclear expression after treatment with the ROS scavenger NAC. First, we observed that, as expected, ROS levels increased upon FHC silencing and return to basal levels upon NAC treatment. Interestingly, NAC was also able to decrease nuclear p65 amount in FHC-silenced K562 cells. Considering the effect of NF-kB activated pathway on cell survival, we analysed the effect of FHC silencing-mediated p65 increase in K562 cells upon treatment with increasing doses of Doxorubicin. Cell viability assay highlighted that FHC-silencing was accompanied by an increased resistance to the drug with an IC 50 about doubled compared to that of the K562 control cells at each the time points. This resistance of FHC-silenced cells was reverted upon NF-kB inhibitor transfection. Conclusion FHC silencing induced NF-kB activation in K562 cells through the modulation of intracellular ROS content. This regulatory axis can be used to modulate K562 chemoresistance

    The next-to-leading order forward jet vertex in the small-cone approximation

    We consider within QCD collinear factorization the process p+p to jet + jet +X, where two forward high-pTp_T jets are produced with a large separation in rapidity Δy\Delta y (Mueller-Navelet jets). In this case the (calculable) hard part of the reaction receives large higher-order corrections ∌αsn(Δy)n\sim \alpha^n_s (\Delta y)^n, which can be accounted for in the BFKL approach. In particular, we calculate in the next-to-leading order the impact factor (vertex) for the production of a forward high-pTp_T jet, in the approximation of small aperture of the jet cone in the pseudorapidity-azimuthal angle plane.     Biomechanically inspired shape memory effect machines driven by muscle like acting NiTi alloys

    The research shows a bioinspired approach to be adopted to design of systems based on Shape Memory Alloys (SMAs), a class of Smart Materials that has in common with muscles the capability to react to an impulse (thermal for SMAs) with a contraction. The biomechanically inspired machine that is discussed in the paper refers to the antagonistic muscles pairs, which belongs to the Skeletal Muscles and are normally arranged in opposition so that as one group of muscles contract another group relaxes or lengthens. The study proposes a model, a solution not only to design a specific application, but also to provide an approach to be used for a wide range of adaptive applications (switchable windows, smart shadow systems, parking and urban shelters, etc.), where the shape changes in response to different external stimuli. The use of antagonist pairs mechanism provides a solution for better optimized systems based on SMAs where the main and proven advantages are: Easier and faster change of shape, lower need of energy for system operation, lower cost for SMA training and no problem of overheating

    Glassy amorphous metal injection molded induced morphological defects

    Melt rheology in injection molded metastable supercooled liquid metal of Zr44-Ti11-Cu10-Ni10-Be25 alloy may induce selective crystallizations. High mobility Be, Cu and Ni atoms have been observed to differently crystallize in bulk metal glassy supercooled liquids. Here, we analyze the result of morphological microscopic observation conduct on Bulk Metallic Glass (BMG) with composition of a commercial liquid metal alloy (LM001B). The injection molded plate has been supplied by “Liquid Metals Technologies Inc, Ca USA” and manufactured using an Engel injection molding machine operating at 1050-1100°C; the observed sample then has been cut by water jet. FEI Scios Dual-Beam has carried out the microscopic observation. Particularly, through a cross section, we observe the presence of crystalline phases on the short-range order. We investigate the presence of short-range order clusters, their distribution and the effect that they could cause on the alloys’ behaviors and properties

    Collinear Subtractions in Hadroproduction of Heavy Quarks

    We present a detailed discussion of the collinear subtraction terms needed to establish a massive variable-flavour-number scheme for the one-particle inclusive production of heavy quarks in hadronic collisions. The subtraction terms are computed by convoluting appropriate partonic cross sections with perturbative parton distribution and fragmentation functions relying on the method of mass factorization. We find (with one minor exception) complete agreement with the subtraction terms obtained in a previous publication by comparing the zero-mass limit of a fixed-order calculation with the genuine massles results in the MSbar scheme. This presentation will be useful for extending the massive variable-flavour-number scheme to other processes.Comment: 29 pages, 17 figures include
