    Characterizing the Oldenburg ā€˜Butter Shaleā€™ from the Upper Ordovician (Katian) Waynesville Formation along the Cincinnati Arch, USA

    The Upper Ordovician (Katian) strata of the Cincinnati Arch contain numerous mudstone units known locally as ā€˜butter shalesā€™ or ā€˜trilobite shalesā€™. Most of these deposits are heavily collected for their excellently-preserved trilobites. The Oldenburg Butter Shale, however, is a previously-undescribed mudstone package from the Waynesville Formation, known only from limited exposure near Oldenburg, Indiana. The Oldenburg Shale is a 2 m-thick mudstone package with minor beds of shelly packstones, and calcisiltite-filled gutter casts. It contains abundant articulated trilobites. The mudstone portion contains illite, chlorite, quartz, calcite and traces of dolomite and pyrite. In outcrop, the shale exhibits no obvious bedding and breaks conchoidally. When cut and polished, the mudstone shows a mottled fabric, containing Lingulichnus and Chondrites trace fossils. The shelly units contain brachiopods, gastropods, and bryozoans. The gutter casts are 20 ā€“ 30 cm wide, display hummocky stratification, and contain Lingulichnus. Faunally, the Oldenburg is very unlike surrounding Waynesville strata. Instead of being dominated by brachiopods as is typical, the Oldenburg fauna consists of abundant bivalves (Modiolopsis, Ambonychia, and Caritodens), lingulid brachiopods, and the trilobites (Isotelus, and Flexicalymene, and rare Amphilichas in the upper 30 cm). Articulate brachiopods are represented in the shale to a limited extent by the genera Zygospira and Platystrophia. The shale also contains bryozoans, orthoconic cephalopods, rare crinoids and conulariids. Conodonts and scolecodonts are a major component of the microfauna. Taphonomy of the fossils, together with sedimentological features, indicates that this butter shale accumulated rapidly as a series of episodes of distal storm-generated mud and silt flows. Towards the top of the mudstone is a horizon of small concretions, about 7 cm wide. Overlying the butter shale is the pyrite crusted surface of the Mid-Richmondian Unconformity which removes the Oldenburg shale in most other locations. The concretions present at the top of the shale are the likely product of the prolonged sediment starvation accompanying this unconformity

    Taphonomic Comparisons of Two Laurentian Upper Ordovician Epeiric Sea ā€œSmall Shelly Faunasā€

    The Elgin Member of the Upper Ordovician (Katian) Maquoketa Formation of Iowa contains phosphorite beds consisting of millimeter-scale phosphatic fossils, primarily steinkerns. Similar beds occur in the coeval strata of the classic Cincinnatian Series around the Cincinnati, Ohio area. Initial sampling of the phosphate-rich beds of the Maquoketa allows comparison between the faunal composition and taphonomy of these beds and collections from the more extensively sampled Cincinnatian strata. We isolated these fossils by dissolution of bulk samples in acetic acid and examined the same strata in thin section to study the fossils in context. The Maquoketa diminutive phosphatized fossils have been interpreted as evidence of dwarfed faunas indicative of environmental stress, such as anoxia, which may have also contributed to phosphogenesis. An alternative explanation for the small size is that phosphogenesis was size-selective and that phosphatic particles were concentrated by reworking as less-durable shell material was destroyed. These hypotheses can be tested by examining the fauna for ā€œnormalā€ sized elements. Insoluble residue from sampled phosphate-rich strata in both field areas yields abundant molluscan steinkerns, as well as crinoid columnals, conodonts, scolecodonts, bryozoan zooecia steinkerns and other fossils associated with a normal marine fauna. In Cincinnatian occurrences, the composition of the phosphatic assemblages is variable but is a reflection of the variability of faunal composition seen in these strata rather than an indication of an unusual fauna associated with extreme conditions; most are associated with diverse marine assemblages. Insoluble residues from both areas yield steinkerns that precipitated in small pores within larger skeletons. This phenomenon can be seen in thin section, where phosphate is present within certain parts of the larger preserved skeletons. The maximum size of the steinkerns of the Maquoketa is larger than those of most Cincinnatian occurrences, although size is variable in Cincinnatian occurrences. In Cincinnatian strata the abundance of small phosphatic fossils correlates with evidence for reworking; heavily reworked beds yield the most residue. Examined in thin section, the sampled strata of the Maquoketa appear to be heavily reworked and represent an extreme endmember of this concentration of durable phosphatic material. Detailed examination using an SEM and associated XRF elemental mapping reveals that the phosphatic steinkerns of both localities are very similar in their taphonomy. Both consist of botryoidal growths of carbonate fluorapatite (CFA). The botryoidal growth appears to have nucleated on the walls of the original shell, first forming a lining of variable thickness. Some steinkerns have secondary botryoidal growths on the outside of the steinkern indicating continued precipitation of CFA after destruction of the original shell. This secondary precipitation suggests that reworking played a role not only in concentrating the phosphatic material but also in encouraging continued precipitation of CFA. The size of the available pore space appears to have played a role in encouraging the precipitation of CFA. In thin section the CFA is limited to smaller parts of larger shells, such as the apices of gastropods and did not precipitate on the inside of the larger, more open spaces within the shell. Many of the phosphate-filled spaces are also sediment-filled, suggesting that subdivision of the larger space into smaller pores enhanced the precipitation of CFA. The difference in the maximum size of the steinkern achieved in the different assemblages suggests that geochemical factors affected size limits. The most distinctive aspect of phosphate-rich Ordovician strata of mid-Laurentia is the degree of reworking that concentrated the durable small fossils. Details of taphonomy also suggest that phosphate precipitation was an iterative process enhanced by reworking, and that small pore spaces enhanced this mineralization, thus selectively preserving certain sizes and parts of the larger fauna

    Fish-Derived Omega-3 Fatty Acids and Prostate Cancer: A Systematic Review.

    BACKGROUND: The use of natural health products in prostate cancer (PrCa) is high despite a lack of evidence with respect to safety and efficacy. Fish-derived omega-3 fatty acids possess anti-inflammatory effects and preclinical data suggest a protective effect on PrCa incidence and progression; however, human studies have yielded conflicting results. METHODS: A search of OVID MEDLINE, Pre-MEDLINE, Embase, and the Allied and Complementary Medicine Database (AMED) was completed for human interventional or observational data assessing the safety and efficacy of fish-derived omega-3 fatty acids in the incidence and progression of PrCa. RESULTS: Of 1776 citations screened, 54 publications reporting on 44 studies were included for review and analysis: 4 reports of 3 randomized controlled trials, 1 nonrandomized clinical trial, 20 reports of 14 cohort studies, 26 reports of 23 case-control studies, and 3 case-cohort studies. The interventional studies using fish oil supplements in patients with PrCa showed no impact on prostate-specific antigen levels; however, 2 studies showed a decrease in inflammatory or other cancer markers. A small number of mild adverse events were reported and interactions with other interventions were not assessed. Cohort and case-control studies assessing the relationship between dietary fish intake and the risk of PrCa were equivocal. Cohort studies assessing the risk of PrCa mortality suggested an association between higher intake of fish and decreased risk of prostate cancer-related death. CONCLUSIONS: Current evidence is insufficient to suggest a relationship between fish-derived omega-3 fatty acid and risk of PrCa. An association between higher omega-3 intake and decreased PrCa mortality may be present but more research is needed. More intervention trials or observational studies with precisely measured exposure are needed to assess the impact of fish oil supplements and dietary fish-derived omega-3 fatty acid intake on safety, PrCa incidence, treatment, and progression

    Calibrating Water Depths of Ordovician Communities: Lithological and Ecological Controls on Depositional Gradients in Upper Ordovician Strata of Southern Ohio and North-Central Kentucky, USA

    Limestone and shale facies of the Upper Ordovician Grant Lake Formation (Katian: Cincinnatian, Maysvillian) are well exposed in the Cincinnati Arch region of southern Ohio and north-central Kentucky, USA. These rocks record a gradual change in lithofacies and biofacies along a gently northward-sloping ramp. This gradient spans very shallow, olive-gray, platy, laminated dolostones with sparse ostracodes in the south to offshore, nodular, phosphatic, brachiopod-rich limestones and marls in the north. This study uses facies analysis in outcrop to determine paleoenvironmental parameters, particularly those related to water depth (e.g., position of the photic zone and shoreline, relative degree of environmental energy). Within a tightly correlated stratigraphic interval (the Mount Auburn and Straight Creek members of the Grant Lake Formation and the Terrill Member of the Ashlock Formation), we document the occurrence of paleoenvironmental indicators, including desiccation cracks and light-depth indicators, such as red and green algal fossils and oncolites. This permitted recognition of a ramp with an average gradient of 10.20 cm water depth per horizontal kilometer. Thus, shallow subtidal (.lagoonal.) deposits in the upramp portion fall within the 1.5.6 m depth range, cross-bedded grainstones representing shoal-type environments fall within the 6.18 m depth range and subtidal, shell-rich deposits in the downramp portion fall within the 20.30 m depth range. These estimates match interpretations of depth independently derived from faunal and sedimentologic evidence that previously suggested a gentle ramp gradient and contribute to ongoing and future high-resolution paleontologic and stratigraphic studies of the Cincinnati Arch region

    Diet and Anxiety: A Scoping Review.

    Anxiety disorders are the most common group of mental disorders. There is mounting evidence demonstrating the importance of nutrition in the development and progression of mental disorders such as depression; however, less is known about the role of nutrition in anxiety disorders. This scoping review sought to systematically map the existing literature on anxiety disorders and nutrition in order to identify associations between dietary factors and anxiety symptoms or disorder prevalence as well as identify gaps and opportunities for further research. The review followed established methodological approaches for scoping reviews. Due to the large volume of results, an online program (Abstrackr) with artificial intelligence features was used. Studies reporting an association between a dietary constituent and anxiety symptoms or disorders were counted and presented in figures. A total of 55,914 unique results were identified. After a full-text review, 1541 articles met criteria for inclusion. Analysis revealed an association between less anxiety and more fruits and vegetables, omega-3 fatty acids, "healthy" dietary patterns, caloric restriction, breakfast consumption, ketogenic diet, broad-spectrum micronutrient supplementation, zinc, magnesium and selenium, probiotics, and a range of phytochemicals. Analysis revealed an association between higher levels of anxiety and high-fat diet, inadequate tryptophan and dietary protein, high intake of sugar and refined carbohydrates, and "unhealthy" dietary patterns. Results are limited by a large percentage of animal and observational studies. Only 10% of intervention studies involved participants with anxiety disorders, limiting the applicability of the findings. High quality intervention studies involving participants with anxiety disorders are warranted

    Cosmetics Alter Biologically-Based Factors of Beauty: Evidence from Facial Contrast

    The use of cosmetics by women seems to consistently increase their attractiveness. What factors of attractiveness do cosmetics alter to achieve this? Facial contrast is a known cue to sexual dimorphism and youth, and cosmetics exaggerate sexual dimorphisms in facial contrast. Here, we demonstrate that the luminance contrast pattern of the eyes and eyebrows is consistently sexually dimorphic across a large sample of faces, with females possessing lower brow contrasts than males, and greater eye contrast than males. Red-green and yellow-blue color contrasts were not found to differ consistently between the sexes. We also show that women use cosmetics not only to exaggerate sexual dimorphisms of brow and eye contrasts, but also to increase contrasts that decline with age. These findings refine the notion of facial contrast, and demonstrate how cosmetics can increase attractiveness by manipulating factors of beauty associated with facial contrast

    A systematic review on the effects of Echinacea supplementation on cytokine levels: Is there a role in COVID-19?

    COVID-19 is the respiratory illness caused by the novel coronavirus, SARS-CoV-2. Cytokine storm appears to be a factor in COVID-19 mortality. Echinacea species have been used historically for immune modulation. A previous rapid review suggested that Echinacea supplementation may decrease the levels of pro-inflammatory cytokines involved in cytokine storm. The objective of the present systematic review was to identify all research that has assessed changes in levels of cytokines relevant to cytokine storm in response to administration of Echinacea supplementation. The following databases were searched: Medline (Ovid), AMED (Ovid), CINAHL (EBSCO), EMBASE (Ovid). Title and abstract screening, full text screening, and data extraction were completed in duplicate using a piloted extraction template. Risk of bias assessment was completed. Qualitative analysis was used to assess for trends in cytokine level changes. The search identified 279 unique publications. After full text screening, 105 studies met criteria for inclusion including 13 human studies, 24 animal studies, and 71 in vitro or ex vivo studies. The data suggest that Echinacea supplementation may be associated with a decrease in the pro-inflammatory cytokines IL-6, IL-8, and TNF, as well as an increase in the anti-inflammatory cytokine IL-10. The risk of bias in the included studies was generally high. While there is currently no substantive research on the therapeutic effects of Echinacea in the management of either cytokine storm or COVID-19, the present evidence related to the herb's impact on cytokine levels suggests that further research may be warranted in the form of a clinical trial involving patients with COVID-19

    Multi-platform Approach for Microbial Biomarker Identification Using Borrelia burgdorferi as a Model

    The identification of microbial biomarkers is critical for the diagnosis of a disease early during infection. However, the identification of reliable biomarkers is often hampered by a low concentration of microbes or biomarkers within host fluids or tissues. We have outlined a multi-platform strategy to assess microbial biomarkers that can be consistently detected in host samples, using Borrelia burgdorferi, the causative agent of Lyme disease, as an example. Key aspects of the strategy include the selection of a macaque model of human disease, in vivo Microbial Antigen Discovery (InMAD), and proteomic methods that include microbial biomarker enrichment within samples to identify secreted proteins circulating during infection. Using the described strategy, we have identified 6 biomarkers from multiple samples. In addition, the temporal antibody response to select bacterial antigens was mapped. By integrating biomarkers identified from early infection with temporal patterns of expression, the described platform allows for the data driven selection of diagnostic targets

    Development of a prototype lateral flow immunoassay (LFI) for the rapid diagnosis of melioidosis.

    Burkholderia pseudomallei is a soil-dwelling bacterium and the causative agent of melioidosis. Isolation of B. pseudomallei from clinical samples is the "gold standard" for the diagnosis of melioidosis; results can take 3-7 days to produce. Alternatively, antibody-based tests have low specificity due to a high percentage of seropositive individuals in endemic areas. There is a clear need to develop a rapid point-of-care antigen detection assay for the diagnosis of melioidosis. Previously, we employed In vivo Microbial Antigen Discovery (InMAD) to identify potential B. pseudomallei diagnostic biomarkers. The B. pseudomallei capsular polysaccharide (CPS) and numerous protein antigens were identified as potential candidates. Here, we describe the development of a diagnostic immunoassay based on the detection of CPS. Following production of a CPS-specific monoclonal antibody (mAb), an antigen-capture immunoassay was developed to determine the concentration of CPS within a panel of melioidosis patient serum and urine samples. The same mAb was used to produce a prototype Active Melioidosis Detect Lateral Flow Immunoassay (AMD LFI); the limit of detection of the LFI for CPS is comparable to the antigen-capture immunoassay (āˆ¼0.2 ng/ml). The analytical reactivity (inclusivity) of the AMD LFI was 98.7% (76/77) when tested against a large panel of B. pseudomallei isolates. Analytical specificity (cross-reactivity) testing determined that 97.2% of B. pseudomallei near neighbor species (35/36) were not reactive. The non-reactive B. pseudomallei strain and the reactive near neighbor strain can be explained through genetic sequence analysis. Importantly, we show the AMD LFI is capable of detecting CPS in a variety of patient samples. The LFI is currently being evaluated in Thailand and Australia; the focus is to optimize and validate testing procedures on melioidosis patient samples prior to initiation of a large, multisite pre-clinical evaluation

    Identification of Circulating Bacterial Antigens by In Vivo Microbial Antigen Discovery

    Detection of microbial antigens in clinical samples can lead to rapid diagnosis of an infection and administration of appropriate therapeutics. A major barrier in diagnostics development is determining which of the potentially hundreds or thousands of antigens produced by a microbe are actually present in patient samples in detectable amounts against a background of innumerable host proteins. In this report, we describe a strategy, termed in vivo microbial antigen discovery (InMAD), that we used to identify circulating bacterial antigens. This technique starts with ā€œInMAD serum,ā€ which is filtered serum that has been harvested from BALB/c mice infected with a bacterial pathogen. The InMAD serum, which is free of whole bacterial cells, is used to immunize syngeneic BALB/c mice. The resulting ā€œInMAD immune serumā€ contains antibodies specific for the soluble microbial antigens present in sera from the infected mice. The InMAD immune serum is then used to probe blots of bacterial lysates or bacterial proteome arrays. Bacterial antigens that are reactive with the InMAD immune serum are precisely the antigens to target in an antigen immunoassay. By employing InMAD, we identified multiple circulating antigens that are secreted or shed during infection using Burkholderia pseudomallei and Francisella tularensis as model organisms. Potential diagnostic targets identified by the InMAD approach included bacterial proteins, capsular polysaccharide, and lipopolysaccharide. The InMAD technique makes no assumptions other than immunogenicity and has the potential to be a broad discovery platform to identify diagnostic targets from microbial pathogens
