While the accuracy has done by looking at the percent recovery (PK, % recovery). In this study, the verification method analysis of the water analyzes method content in an artificial feed is carried out gravimetrically based on the SNI method. The result of this study obtained that the analysis of water quality has qualified ripitabilty, reproducibility, and accuracy because RSD analysis obtained the smaller value than 2/3 of RD Horwitz

    Diversity and Abundance of Phytoplankton in Coastal Areas in Kendari Southeast Sulawesi Indonesia

    Phytoplankton is a microscopic algae that can be found in almost all aquatic habitats. Phytoplankton have an important ecological role in the waters, as the basis of the food chain and as a producer of oxygen, where their composition and abundance are strongly influenced by various environmental factors. This study aims to determine the community structure of phytoplankton including the diversity, abundance and dominance of phytoplankton found in various locations in coastal waters in Kendari, Southeast Sulawesi. Sampling was carried out in several coastal waters in Southeast Sulawesi including Nambo, Bokori, Batu Gong, Toronipa and Tanjung Tiram beaches. Plankton sampling was carried out by filtering about 100 L of seawater using a plankton net with a mesh size of 20 μm. The filtered plankton samples were then preserved in 4% formalin for further observation in the laboratory. Water quality measurements of temperature, salinity, brightness and pH were carried out in situ while nitrate, phosphate and ammonia were carried out ex-situ. The results showed that there were about 30 species of phytoplankton found where the most species were found in Bokori Island (22 species) followed by Tanjung Tiram beach (21 species), Toronipa beach (19 species) and both Nambo and Batu Gong beaches with 17 species. The highest abundance of phytoplankton was found on Bokori Island (563 individuals/L) and the lowest abundance was found on Batu Gong beach (237 individuals/L). The highest diversity index value was found at Toronipa Beach (2.72) while the lowest diversity index value was found at Batu Gong Beach (2.13). For the dominance index value, the highest value was obtained at Batu Gong Beach (0.18) and the lowest value was obtained at Toronipa Beach (0.08). The difference in the structure of the phytoplankton community found in different locations is due to variation of the environmental conditions. Keywords

    Verification method is required to generate data analysis with accurate and precise value corresponding to the standrad method. This study aims to verify fat analysis method by using direct extraction method. The study was conducted in Laboratorium Pengujian, FPIK, Universitas Halu Oleo. Fat analysis method verified was direct extraction method or soxhlet method. The results showed that all precision values from experiments I-VIII, consisting of repeatability and reproductibility, have RSD values (0.03 – 0.14) lower than 2/3 RSD Horwitz (1.99). The accuracy results (% Recovery) indicated similarly RSD, lower than 2/3 RSD Horwitz (1.33). Based on these results, it can be concluded that fat analysis method used in Laboratorium Pengujian, meet the pecise and accuray standard, and generates valid data and accounted for

    Growth, biomass and lipid productivity of a newly isolated tropical marine diatom, Skeletonema sp. UHO29, under different light intensities

    Light is one of the important factors affecting growth and biochemical composition of microalgae. The aim of this study was to determine growth, biomass and lipid productivity of a newly isolated marine diatom, Skeletonema sp.UHO29 under different light intensities. The Skeletonema sp. was cultured in 300 mL conical flasks containing 150 mL of f/2 medium under three different light intensities (28, 60 and 110 μmol photons m-2 s-1 ) with three replications. The cultures were incubated under ambient room temperatures, and a cycle of 12 hours light and 12 hours dark, in batch modes for two weeks. The results showed that the highest specific growth rate (0.56 d-1) was achieved at the highest light intensity (110 μmol photons m-2 s-1) and the lowest (0.11 d-1) was obtained at the lowest light intensity (28 μmol photons m-2 s-1). The highest biomass yield (0.62 g L-1) and biomass productivity (0.34 g L-1 d-1) was achieved at the highest light intensity. However, the highest lipid yield (0.175 g L-1), lipid content (28.78% ash-free dry weight) and lipid productivity (0.067 g L-1 d-1) were achieved at medium light intensity. The alga prefers high light intensity for higher growth rate and biomass productivity but it produces more lipids at medium light intensity. This study indicated that the Skeletonema sp.UHO29 is a potential species for outdoor mass cultivation and is a promising microalgal strain for use as a biodiesel feedstock due to its high growth rate, high biomass and lipid productivity

    Pendampingan Pengelolaan Perpustakaan di Madrasah Ibtidaiyah Nahdlatul Ulama II Tembilahan Hulu

    Perpustakaan merupakan salah satu sarana belajar siswa disekolah, tanpa pengelolaan yang baik, perpustakaan kurang memiliki peran terhadap peningkatan prestasi belajar siswa. Salah satu  kegiatan pengelolaan perpustakaan yang menyebabkan kurangnya peran dalam meningkatkan prestasi siswa adalah peserta didik  tidak memiliki kartu anggota perpustakaan, perpustakaan belum memiliki pengelola khusus sehinga belum memiliki visi, misi dan struktur organisasi, terbatasnya jenis buku dan belum memiliki katalogisasi buku. Tujuan dilakukannya kegiatan pendampingan pengelolaan perpustakaan di MI NU II Tembilahan Hulu adalah (1) Meningkatkan pengelolaan perpustakaan di MI NU II Tembilahan Hulu. (2) menumbuhkan minat membaca pada anak sejak dini di MI NU II Tembilahan Hulu. Metode yang digunakan adalah metode pendampingan dengan teknik observasi, diskusi, workshop, dan gotong-royong. Hasil kegiatan menunjukkan adanya peningkatan pengelolaan perpustakaan, perpustakaan sudah memiliki struktur organisasi perpustakaan, memiliki visi dan misi perpustakaan, memiliki katalogisasi buku, masing-masing peserta didik sudah memiliki kartu anggota perpustakaan, serta memiliki ruang membaca yang menarik minat membaca peserta didik. Dengan pendampingan pengelolaan perpustakaan di MI Nahdhatul Ulama II pengelolaan perpustakaan di MI NU II menjadi lebih baik dan dapat meningkatkan minat membaca peserta didik

    Outdoor cultivation of newly isolated tropical marine microalgae Nannochloropsis sp. UHO3 and Skeletonema sp. UHO29 in raceway ponds for biodiesel feedstock production

    The microalgae Nannochloropsis sp. UHO3 and Skeletonema sp. UHO29 are newly isolated marine microalgae from coastal areas in Kendari, Southeast Sulawesi, Indonesia. Indoor studies of the microalgae showed potential as biodiesel feedstock due to their high growth rates, high biomass and lipid productivity. However, for commercial uses, especially for biodiesel feedstock, the species should have the ability to perform well under real outdoor conditions. This study aimed to determine the growth, biomass, and lipid productivity of the Nannochloropsis sp. UHO3 and Skeletonema sp. UHO29 in outdoor raceway ponds. The species were cultured using 1 m2 raceway ponds at 30 cm depth. The two species were cultured using water-based sea media enriched with f/2 nutrients for Skeletonema sp. and Walne media for Nannochloropsis sp. Cultures were initially operated in batch mode until they reached the stationary phase before operating in semi-continuous mode. The culture duration was about three months. Cell counting was done every two days, while sampling for biomass (dry weight - DW, and ashfree dry weight - AFDW) and lipids were done every four days. The results showed that the two species of microalgae could grow well in outdoor raceway ponds. The specific growth rate of the Nannochloropsis sp. UHO3 ranged from 0.105 to 0.447 d-1 while the specific growth rate of Skeletonema sp. UHO29 ranged from 0.127 to 0.457 d-1. Nannochloropsis sp. UHO3 and Skeletonema sp. UHO29 have high lipid content ranging from 15 to 44% AFDW and from 14 to 53% AFDW, respectively. The biomass productivity of Nannochloropsis sp. UHO3 and Skeletonema sp. UHO29 range between 10-122 g m-2 d-1 and 13-107 g m-2 d-1 respectively, while the lipid productivity value for Nannochloropsis sp. UHO3 ranges from 4 to 31 g m-2 d-1 and for Skeletonema sp. UHO29 range from 5 to 25 g m-2 d-1. The results of this study indicate that the two microalgae species are potentially developed as biodiesel feedstocks due to their fast growth rate, high lipid content, high biomass, and lipid productivity under outdoor conditions
