    The on-axis view of GRB 170817A

    The peculiar short gamma-ray burst GRB 170817A has been firmly associated to the gravitational wave event GW170817, which has been unaninmously interpreted as due to the coalescence of a double neutron star binary. The unprecedented behaviour of the non-thermal afterglow led to a debate about its nature, which was eventually settled by high-resolution VLBI observations, which strongly support the off-axis structured jet scenario. Using information on the jet structure derived from multi-wavelength fitting of the afterglow emission and of the apparent VLBI image centroid motion, we compute the appearance of a GRB 170817A-like jet as seen by an on-axis observer and we compare it to the previously observed population of SGRB afterglows and prompt emission events. We find that the intrinsic properties of the GRB 170817A jet are representative of a typical event in the SGRB population, hinting at a quasi-universal jet structure. The diversity in the SGRB afterglow population could therefore be ascribed in large part to extrinsic (redshift, density of the surrounding medium, viewing angle) rather than intrinsic properties.     RSA, CMJ, Leger, 10m sprint responses to Pre-season training in semi-Professional Soccer Players

    Aim: The aim of this study was to analyze RSA, CMJ, Leger, 10m sprint responses to Pre-season Training in Semi-Professional Soccer Players (SPSP). Considering that numerous studies1,2 highlighted the combination of high levels of physical, technical and tactical skills during a soccer match, the cure of physical training pose a particular attention on training load that generally increases up to 2.4 times during the pre-season period compared with the in-season3. Methods: Six SPSP (age: 23±7yr; BMI: 23.3±1.8) were requested to perform aerobic training over an 8-week period on alternate days with the functional strength training sessions and sprint training drills as prescribed by the coaches and strength and conditioning staff. Repeated Sprint Ability (RSA, Total Time –TT- and percentage of fatigue index -%FI), Leger, 10m sprint and Counter Mouvement Jump (CMJ) tests, were performed before and after pre-season soccer training. ANOVA for repeated measures was conducted to assess differences (p<0.05) with respect to pre seasonal training. Correlation was calculated between the percentage of variation (Δ) of each test. Results: Compared to the values recorded before the pre-season, improvement of Leger (3%) and %FI (17.6%) and a deterioration of TT (10%), 10m sprint (0.2%) and CMJ (2.4%) were found. In addition, we have found a main effect between before and after pre-seasonal training in TT (F(1,4)=60.2; p=0.001) and Leger (F(1,5)=25; p<0.005). ΔCMJ showed very large correlation with ΔLeger (r=-0.88) and nearly perfect with Δ%FI (r=0.93); while ΔLeger was largely correlated with Δ%FI (r=-0.69). Conclusions: Given that the cure of the physical preparation is considered as an important element in order to influence the final soccer game result, this study want to be useful information for the coach in order to maximize the best physical condition of the whole team relative to the beginning of the regular season

    Reversible two-step unfolding of heme-human serum albumin: A 1H-NMR relaxometric and circular dichroism study

    Human serum albumin (HSA) participates in heme scavenging, the bound heme turning out to be a reactivity center and a powerful spectroscopic probe. Here, the reversible unfolding of heme-HSA has been investigated by H-1-NMR relaxometry, circular dichroism, and absorption spectroscopy. In the presence of 6 equiv of myristate ( thus fully saturating all available fatty acid binding sites in serum heme-albumin), 1.0 M guanidinium chloride induces some unfolding of heme-HSA, leading to the formation of a folding intermediate; this species is characterized by increased relaxivity and enhanced dichroism signal in the Soret region, suggesting a more compact heme pocket conformation. Heme binds to the folding intermediate with K-d = (1.2 +/- 0.1) x 10(-6) M. In the absence of myristate, the conformation of the folding intermediate state is destabilized and heme binding is weakened [K-d = (3.4 +/- 0.1) x 10(-5) M]. Further addition of guanidinium chloride ( up to 5 M) brings about the usual denaturation process. In conclusion, myristate protects HSA from unfolding, stabilizing a folding intermediate state in equilibrium with the native and the fully unfolded protein, envisaging a two-step unfolding pathway for heme-HSA in the presence of myristate

    Ibuprofen modulates allosterically NO dissociation from ferrous nitrosylated human serum heme-albumin by binding to three sites

    Human serum albumin (HSA) is a monomeric allosteric protein. Here, the effect of ibuprofen on denitrosylation kinetics (k(off)) and spectroscopic properties of HSA-heme-Fe(II)-NO is reported. The k(off) value increases from (1.4 +/- 0.2) x 10(-4) s(-1), in the absence of the drug, to (9.5 +/- 1.2) x 10(-3) s(-1), in the presence of 1.0 x 10(-2) M ibuprofen, at pH 7.0 and 10.0 degrees C. From the dependence of k(off) on the drug concentration, values of the dissociation equilibrium constant for ibuprofen binding to to HSA-heme-Fe(II)-NO (K-1 = (3.1 +/- 0.4) x 10(-7) M, K-2= (1.7 +/- 0.2) x 10(-4) M. and K-3 (2.2 +/- .2) x 10(-3) M) were determined. The K-3 value corresponds to the HSA-heme-Fe(II)-NO determined by monitoring drug-dependent absorbance spectroscopic changes (H = (2.6 +/- 0.3) x 10(-3) M). Present data indicate that ibuprofen binds to the FA3-FA4 cleft (Sudlow's site II), to the FA6 site, and possibly to the FA2 pocket, inducing the hexa-coordination of HSA-heme-Fe(II)-NO and triggering the heme-ligand dissociation kinetics.     Growth and glycemic control in children with type 1 diabetes and asymptomatic celiac disease treated with a gluten -free diet for 1 year

    To compare growth and glycemic control in children with type 1 diabetes and silent celiac disease treated with a gluten-free diet for 1 year with those of similar age and gender with type 1 diabetes but without celiac disease, 16 type 1 diabetes patients with silent celiac disease were enrolled and each celiac disease-positive case was matched for age, sex, and duration of diabetes with two type 1 diabetes controls with negative serologic markers of celiac disease. All 16 children with positive celiac disease serology had histologic features consistent with celiac disease despite the absence of symptoms. The mean growth and metabolic control values were similar between children with type 1 diabetes and celiac disease and those with type 1 diabetes but without celiac disease. This study seems to suggest that the early diagnosis of celiac disease and initiation of a gluten-free diet may prevent further deterioration in the nutritional status of children with type 1 diabetes and celiac disease and may reduce the prospect of celiac disease complications without any impact on type 1 diabetes control

    The rise and fall of the high-energy afterglow emission of GRB 180720B

    The Gamma Ray Burst (GRB) 180720B is one of the brightest events detected by the Fermi satellite and the first GRB detected by the H.E.S.S. telescope above 100 GeV. We analyse the Fermi (GBM and LAT) and Swift (XRT and BAT) data and describe the evolution of the burst spectral energy distribution in the 0.5 keV - 10 GeV energy range over the first 500 seconds of emission. We reveal a smooth transition from the prompt phase, dominated by synchrotron emission in a moderately fast cooling regime, to the afterglow phase whose emission has been observed from the radio to the GeV energy range. The LAT (0.1 - 100 GeV) light curve initially rises (FLAT∝t2.4F_{\rm LAT}\propto t^{2.4}), peaks at ∌\sim78 s, and falls steeply (FLAT∝t−2.2F_{\rm LAT}\propto t^{-2.2}) afterwards. The peak, which we interpret as the onset of the fireball deceleration, allows us to estimate the bulk Lorentz factor Γ0∌150 (300)\Gamma_{0}\sim 150 \ (300) under the assumption of a wind-like (homogeneous) circum-burst medium density. We derive a flux upper limit in the LAT energy range at the time of H.E.S.S. detection, but this does not allow us to unveil the nature of the high energy component observed by H.E.S.S. We fit the prompt spectrum with a physical model of synchrotron emission from a non-thermal population of electrons. The 0 - 35 s spectrum after its EF(E)E F(E) peak (at 1 - 2 MeV) is a steep power law extending to hundreds of MeV. We derive a steep slope of the injected electron energy distribution N(Îł)∝γ−5N(\gamma)\propto \gamma^{-5}. Our fit parameters point towards a very low magnetic field (Bâ€Č∌1B'\sim 1 G) in the emission region.Comment: 10 pages, 6 figures, submitted to A&

    Ebolavirus and Marburgvirus (belonging to the Filoviridae family) emerged four decades ago and cause epidemics of haemorrhagic fever with high case-fatality rates. The genome of filoviruses encodes seven proteins. No significant homology is observed between filovirus proteins and any known macromolecule. Moreover, Marburgvirus and Ebolavirus show significant differences in protein homology. The natural maintenance cycle of filoviruses is unknown, the natural reservoir, the mode of transmission, the epidemic disease generation, and temporal dynamics are unclear. Lastly, Ebolavirus and Marburgvirus are considered as potential biological weapons. Vaccine appears the unique therapeutic frontier.     The key role played by charge in the interaction of cytochrome c with cardiolipin

    Cytochrome c undergoes structural variations upon binding of cardiolipin, one of the phospholipids constituting the mitochondrial membrane. Although several mechanisms governing cytochrome c/cardiolipin (cyt c/CL) recognition have been proposed, the interpretation of the process remains, at least in part, unknown. To better define the steps characterizing the cyt c-CL interaction, the role of Lys72 and Lys73, two residues thought to be important in the protein/lipid binding interaction, were recently investigated by mutagenesis. The substitution of the two (positively charged) Lys residues with Asn revealed that such mutations cancel the CL-dependent peroxidase activity of cyt c; furthermore, CL does not interact with the Lys72Asn mutant. In the present paper, we extend our study to the Lys → Arg mutants to investigate the influence exerted by the charge possessed by the residues located at positions 72 and 73 on the cyt c/CL interaction. On the basis of the present work a number of overall conclusions can be drawn: (i) position 72 must be occupied by a positively charged residue to assure cyt c/CL recognition; (ii) the Arg residues located at positions 72 and 73 permit cyt c to react with CL; (iii) the replacement of Lys72 with Arg weakens the second (low-affinity) binding transition; (iv) the Lys73Arg mutation strongly increases the peroxidase activity of the CL-bound protein

    Spectral index-flux relation for investigating the origins of steep decay in Îł-ray bursts

    γ-ray bursts (GRBs) are short-lived transients releasing a large amount of energy (10 − 10 erg) in the keV-MeV energy range. GRBs are thought to originate from internal dissipation of the energy carried by ultra-relativistic jets launched by the remnant of a massive star’s death or a compact binary coalescence. While thousands of GRBs have been observed over the last thirty years, we still have an incomplete understanding of where and how the radiation is generated in the jet. Here we show a relation between the spectral index and the flux found by investigating the X-ray tails of bright GRB pulses via time-resolved spectral analysis. This relation is incompatible with the long standing scenario which invokes the delayed arrival of photons from high-latitude parts of the jet. While the alternative scenarios cannot be firmly excluded, the adiabatic cooling of the emitting particles is the most plausible explanation for the discovered relation, suggesting a proton-synchrotron origin of the GRB emission.The research leading to these results has received funding from the European Union’s Horizon 2020 Programme under the AHEAD2020 project (Grant agreement n. 871158).
