    Optimal Transient Real-Time Engine-Generator Control in the Series-Hybrid Vehicle

    We study the dynamic engine-generator optimal control problem with a goal of minimizing fuel consumption while delivering a requested average electrical power. By using an infinite-horizon formulation and explicitly minimizing fuel consumption, we avoid issues inherent with penalty-based and finite-horizon problems. The solution to the optimal control problem, found using dynamic programming and the successive approximation method, can be expressed as instantaneous non-linear state-feedback. This allows for trivial real-time control, typically requiring 10–20 CPU instructions per control period, a few bytes of RAM, and 5–20 KiB of nonvolatile memory. Simulation results for a passenger vehicle indicate a fuel consumption improvement in the region of 5–7% during the transient phase when compared with the class of controllers found in the industry. Bench-tests, where the optimal controller is executed in native hardware, show an improvement of 3.7%, primarily limited by unmodeled dynamics. Our specific choice of problem formulation, a guaranteed globally optimal solution, and trivial real-time control resolve many of the limitations with the current state of optimal engine-generator controllers

    An attempt to estimate out-of-plane lung nodule elongation in tomosynthesis images

    In chest tomosynthesis (TS) the most commonly used reconstruction methods are based on Filtered Back Projection (FBP) algorithms. Due to the limited angular range of x-ray projections, FBP reconstructed data is typically associated with a low spatial resolution in the out-of-plane dimension. Lung nodule measures that depend on depth information such as 3D shape and volume are therefore difficult to estimate. In this paper the relation between features from FBP reconstructed lung nodules and the true out-of-plane nodule elongation is investigated and a method for estimating the out-of-plane nodule elongation is proposed. In order to study these relations a number of steps that include simulation of spheroidal-shaped nodules, insertion into synthetic data volumes, construction of TS-projections and FBP-reconstruction were performed. In addition, the same procedure was used to simulate nodules and insert them into clinical chest TS projection data. The reconstructed nodule data was then investigated with respect to in-plane diameter, out-of-plane elongation, and attenuation coefficient. It was found that the voxel value in each nodule increased linearly with nodule elongation, for nodules with a constant attenuation coefficient. Similarly, the voxel value increased linearly with in-plane diameter. These observations indicate the possibility to predict the nodule elongation from the reconstructed voxel intensity values. Such a method would represent a quantitative approach to chest tomosynthesis that may be useful in future work on volume and growth rate estimation of lung nodules

    Rebalancing of mitochondrial homeostasis through an NAD+-SIRT1 pathway preserves intestinal barrier function in severe malnutrition.

    BACKGROUND: The intestine of children with severe malnutrition (SM) shows structural and functional changes that are linked to increased infection and mortality. SM dysregulates the tryptophan-kynurenine pathway, which may impact processes such as SIRT1- and mTORC1-mediated autophagy and mitochondrial homeostasis. Using a mouse and organoid model of SM, we studied the repercussions of these dysregulations on malnutrition enteropathy and the protective capacity of maintaining autophagy activity and mitochondrial health. METHODS: SM was induced through feeding male weanling C57BL/6 mice a low protein diet (LPD) for 14-days. Mice were either treated with the NAD +-precursor, nicotinamide; an mTORC1-inhibitor, rapamycin; a SIRT1-activator, resveratrol; or SIRT1-inhibitor, EX-527. Malnutrition enteropathy was induced in enteric organoids through amino-acid deprivation. Features of and pathways to malnutrition enteropathy were examined, including paracellular permeability, nutrient absorption, and autophagic, mitochondrial, and reactive-oxygen-species (ROS) abnormalities. FINDINGS: LPD-feeding and ensuing low-tryptophan availability led to villus atrophy, nutrient malabsorption, and intestinal barrier dysfunction. In LPD-fed mice, nicotinamide-supplementation was linked to SIRT1-mediated activation of mitophagy, which reduced damaged mitochondria, and improved intestinal barrier function. Inhibition of mTORC1 reduced intestinal barrier dysfunction and nutrient malabsorption. Findings were validated and extended using an organoid model, demonstrating that resolution of mitochondrial ROS resolved barrier dysfunction. INTERPRETATION: Malnutrition enteropathy arises from a dysregulation of the SIRT1 and mTORC1 pathways, leading to disrupted autophagy, mitochondrial homeostasis, and ROS. Whether nicotinamide-supplementation in children with SM could ameliorate malnutrition enteropathy should be explored in clinical trials.     Contrast-agent-based perfusion MRI code repository and testing framework: ISMRM Open Science Initiative for Perfusion Imaging (OSIPI)

    Purpose Software has a substantial impact on quantitative perfusion MRI values. The lack of generally accepted implementations, code sharing and transparent testing reduces reproducibility, hindering the use of perfusion MRI in clinical trials. To address these issues, the ISMRM Open Science Initiative for Perfusion Imaging (OSIPI) aimed to establish a community-led, centralized repository for sharing open-source code for processing contrast-based perfusion imaging, incorporating an open-source testing framework. Methods A repository was established on the OSIPI GitHub website. Python was chosen as the target software language. Calls for code contributions were made to OSIPI members, the ISMRM Perfusion Study Group, and publicly via OSIPI websites. An automated unit-testing framework was implemented to evaluate the output of code contributions, including visual representation of the results. Results The repository hosts 86 implementations of perfusion processing steps contributed by 12 individuals or teams. These cover all core aspects of DCE- and DSC-MRI processing, including multiple implementations of the same functionality. Tests were developed for 52 implementations, covering five analysis steps. For T1 mapping, signal-to-concentration conversion and population AIF functions, different implementations resulted in near-identical output values. For the five pharmacokinetic models tested (Tofts, extended Tofts-Kety, Patlak, two-compartment exchange, and two-compartment uptake), differences in output parameters were observed between contributions. Conclusions The OSIPI DCE-DSC code repository represents a novel community-led model for code sharing and testing. The repository facilitates the re-use of existing code and the benchmarking of new code, promoting enhanced reproducibility in quantitative perfusion imaging

    The rise of inconspicuous consumption

    Ever since Veblen and Simmel, luxury has been synonymous with conspicuous consumption. In this conceptual paper we demonstrate the rise of inconspicuous consumption via a wide-ranging synthesis of the literature. We attribute this rise to the signalling ability of traditional luxury goods being diluted, a preference for not standing out as ostentatious during times of economic hardship, and an increased desire for sophistication and subtlety in design in order to further distinguish oneself for a narrow group of peers. We decouple the constructs of luxury and conspicuousness, which allows us to reconceptualise the signalling quality of brands and the construct of luxury. This also has implications for understanding consumer behaviour practices such as counterfeiting and suggests that consumption trends in emerging markets may take a different path from the past
