    DRESS Syndrome Secondary to Carbamazepine Therapy Presenting with Bilateral Acute Anterior Uveitis and Angle Closure Glaucoma

    Purpose: Drug reaction with eosinophilia and systemic symptoms (DRESS) syndrome is a rare, lifethreatening multi-system adverse drug reaction characterized by febrile skin rash, hematologic abnormalities, and involvement of internal organs. We report a case of DRESS syndrome in a child presenting with primary ophthalmic manifestations. Case Report: An 11-year-old boy presented with severe pain and diminished vision in both eyes six weeks after starting carbamazepine therapy for seizure disorder. Ocular examination revealed features of bilateral acute anterior uveitis, acute onset myopia, and angle closure glaucoma secondary to uveal effusion. Additionally, the patient was febrile with a generalized maculopapular rash, and blood investigations revealed eosinophilic leukocytosis. A diagnosis of carbamazepine-induced DRESS syndrome was made, and carbamazepine therapy was discontinued. Treatment with cycloplegics, topical, and systemic steroids resulted in prompt clinical recovery. Conclusion: Ophthalmologists should be aware that hypersensitivity to anticonvulsants, such as carbamazepine, can present with bilateral uveitis and uveal effusion along with systemic symptoms. Prompt diagnosis and treatment can prevent vision loss and life-threatening complications. Patients should be counselled about potential adverse effects of anticonvulsants before therapy

    Evaluation of Bioactive Secondary Metabolites from Ponyfish Associated Bacteria (Photobacterium leiognathi)

    The marine environment continues to surprise us by producing novel bioactive substances with a wide range of benefits for humans. Materials and Methods: Marine bioluminescent bacteria Photobacterium leiognathi was isolated from pony fish, Secutorruconius which was confirmed with microscopic and molecular characterization. The secondary metabolite of the isolated bacteria was extracted with dichloromethane. The chemical fingerprinting of the isolated metabolite was analyzed through TLC, FT-IR, and HPLC. The nature of the compound present in the metabolite was identified in the gas chromatography-mass spectrometry analysis (GC - MS). The isolated extract was investigated for its antibacterial property against 10 human pathogenic bacteria and also its antioxidant activity using different assays such as 1, 1-Diphenyl-2-picrylhydrazyl, Phosphomolybdenum, Metal chelating, Hydroxyl radical scavenging and hydrogen peroxide scavenging activity. Results: The Presence of functional groups including phenols, sugars, and amino acids in the extracts were identified by TLC. Totally, nine peaks were obtained for the crude extract through the FTIR spectrum range of 400 to 4000 cm-1 for the active sample. The DCM extract showed a broad spectrum of antibacterial activity against the six human bacterial pathogens. Secondary metabolites from the bioluminescent bacteria, P. leiognathi, have strong antioxidant properties. These results will be instrumental in developing novel products with biosensors and bio-imaging applications using P. leiognathi.

    Screening of antiangiogenic potential of twenty two marine invertebrate extracts of phylum Mollusca from South East Coast of India

    ABSTRACTObjectiveTo evaluate the antiangiogenic potential of twenty two marine invertebrate species of Phylum Mollusca from south east coast of India.MethodsLive specimens of molluscan species were collected and their methanolic extracts were evaluated for preliminary antiangiogenic activity using the in ovo chick chorio-allantoic membrane assay. The extracts were further evaluated for in vivo antiangiogenic activity using chemical cautery induced corneal neovascularization assay in rats and oxygen induced retinopathy assay in rat pups.ResultsIn the chick chorio-allantoic membrane assay, four methanolic extracts of marine molluscan species viz. Meretrix meretrix, Meretrix casta, Telescopium telescopium and Bursa crumena methanolic extracts exhibited noticeable antiangiogenic activity at the tested concentration of 200 ÎŒg whereby they significantly inhibited the VEGF induced proliferation of new blood vessels. Among these four extracts, the methanolic extract of Meretrix casta exhibited relatively higher degree of antiangiogenic activity with an inhibitiory percentage (64.63%) of the VEGF induced neovascularization followed by the methanolic extracts of Telescopium telescopium (62.02%), Bursa crumena (60.48%) and Meretrix meretrix (47.01%). These four methanolic extracts were further evaluated for in vivo antiangiogenic activity whereby the methanolic extract of Telescopium telescopium exhibited most noticeable inhibition (42.58%) of the corneal neovascularization in rats in comparison to the sham treated group, and also exhibited most noticeable inhibition (31.31%) of the oxygen induced retinal neovascularization in rat pups in comparison to the hyperoxia group that was observed for considerable retinal neovascularization.ConclusionsThe significant antiangiogenic activity evinced by the extract of Telescopium telescopium merits further investigation for ocular neovascular diseases

    Electrochemical performance of plant trace element incorporated silver nanoparticles synthesis from Datura metel L.

    In our report, silver nanoparticles (AgNPs) were prepared from the leaf extract of Datura metel L. via the green synthesis method. Datura metel L. is a herbal medicinal plant from the Solanaceae family. The as‐prepared AgNPs were characterized using UV‐Vis spectrometer, X‐ray Diffraction (XRD), Fourier Transform Infrared (FTIR) spectroscopy and Scanning Electron Microscopy (SEM) with Energy Dispersive X‐ray (EDAX) analysis. The peak appearance of a Surface Plasmon Resonance (SPR) at 415 nm suggested the creation of AgNPs in the UV‐Vis spectrum. The XRD pattern showed the face‐centered cubic crystal structure of AgNPs with organometallic complex phase. Based on the FTIR spectrum, the peaks revealed the existence of biomolecules. SEM images showed the shape of the clastic rocks and the EDAX profile authenticated the presence of Ag and plant trace element. The cyclic voltammetry, Chronopotentiometry, and electrochemical impedance spectroscopy analysis were performed on an as‐prepared Ag electrode. A specific capacitance of 267.59 F/g at 0.5 A/g and a cyclic retention of 83.7% after 5000 charge‐discharge cycles were obtained. Hence, this material could be utilized in supercapacitor energy storage devices

    Increased susceptibility of 129SvEvBrd mice to IgE-Mast cell mediated anaphylaxis

    <p>Abstract</p> <p>Background</p> <p>Experimental analyses have identified strain-dependent factors that regulate susceptibility to anaphylaxis in mice. We assessed the susceptibility of the widely used 129SvEvBrd (also known as 129S5) mouse strain to IgE/mast cell-mediated anaphylaxis as compared to BALB/c. Mice were subjected to passive and oral Ovalbumin [OVA]-induced active anaphylaxis. Tissue mast cell, plasma histamine, total IgE and OVA-specific IgE levels and susceptibility to histamine i.v infusion were assessed. Bone marrow mast cell (BMMC)s were examined for Fc<sub>Δ</sub>RI, c-kit, degranulation efficiency, proliferation, apoptosis and cytokine profile.</p> <p>Results</p> <p>129S5 mice had significantly increased susceptibility to passive and oral OVA-induced active anaphylaxis. Increased susceptibility to anaphylaxis was associated with increased homeostatic mast cell levels but not OVA-specific IgE or IgG<sub>1 </sub>levels. <it>In vitro </it>analyses of BMMCs revealed no difference in Fc<sub>Δ</sub>RI and c-Kit expression, however, 129S5 BMMCs possessed greater proliferative capacity and reduced caspase-3-mediated apoptosis. IgE-BMMC degranulation assays demonstrated no difference in degranulation efficiency. Furthermore, 129S5 mice possessed increased sensitivity to histamine-induced hypothermia.</p> <p>Conclusions</p> <p>We conclude that 129S5 mice have increased susceptibility to anaphylaxis as compared to BALB/c strain and their increased susceptibility was associated with altered mast cell proliferation and homeostatic tissue levels and responsiveness to histamine. Given the wide spread usage of the 129SvEvBrd strain of mice in experimental gene targeting methodology, these data have important implications for studying IgE-reactions in mouse systems.</p

    Approaches in biotechnological applications of natural polymers

    Natural polymers, such as gums and mucilage, are biocompatible, cheap, easily available and non-toxic materials of native origin. These polymers are increasingly preferred over synthetic materials for industrial applications due to their intrinsic properties, as well as they are considered alternative sources of raw materials since they present characteristics of sustainability, biodegradability and biosafety. As definition, gums and mucilages are polysaccharides or complex carbohydrates consisting of one or more monosaccharides or their derivatives linked in bewildering variety of linkages and structures. Natural gums are considered polysaccharides naturally occurring in varieties of plant seeds and exudates, tree or shrub exudates, seaweed extracts, fungi, bacteria, and animal sources. Water-soluble gums, also known as hydrocolloids, are considered exudates and are pathological products; therefore, they do not form a part of cell wall. On the other hand, mucilages are part of cell and physiological products. It is important to highlight that gums represent the largest amounts of polymer materials derived from plants. Gums have enormously large and broad applications in both food and non-food industries, being commonly used as thickening, binding, emulsifying, suspending, stabilizing agents and matrices for drug release in pharmaceutical and cosmetic industries. In the food industry, their gelling properties and the ability to mold edible films and coatings are extensively studied. The use of gums depends on the intrinsic properties that they provide, often at costs below those of synthetic polymers. For upgrading the value of gums, they are being processed into various forms, including the most recent nanomaterials, for various biotechnological applications.
