    The low mass population of the Vela OB2 association from Gaia

    The first Gaia Data Release presents an opportunity to characterise the low-mass population of OB associations, providing larger statistical samples and better constraints on the formation and evolution of clusters and associations. Using previously known low mass members in a small region of Vela OB2 we have designed selection criteria that combine Gaia and 2MASS photometry, independently of any astrometric information, to identify low-mass pre-main-sequence (PMS) stars over the wider association area. Our method picks out the known clusters of young stars around Velorum and NGC-2547, but also identifies other over-densities that may represent previously unknown clusters. There are clear differences in the spatial distributions of the low-mass and the high-mass OB populations, suggesting either that the structure and dynamics of these populations has evolved separately or that the initial mass function can vary considerably on small scales within a single association

Copyright @ 2009 Oxford University Press.Background - Invasive aspergillosis (IA) is the most common cause of death associated with fungal infection in the developed world. Historically, susceptibility to IA has been associated with prolonged neutropenia; however, IA has now become a major problem in patients on calcineurin inhibitors and allogenic hematopoetic stem cell transplant patients following engraftment. These observations suggest complex cellular mechanisms govern immunity to IA. Methods - To characterize the key early events that govern outcome from infection with Aspergillus fumigatus we performed a comparative immunochip microarray analysis of the pulmonary transcriptional response to IA between cyclophosphamide-treated mice and immunocompetent mice at 24 h after infection. Results - We demonstrate that death due to infection is associated with a failure to generate an incremental interferon-γ response, increased levels of interleukin-5 and interleukin-17a transcript, coordinated expression of a network of tumor necrosis factor–α-related genes, and increased levels of tumor necrosis factor–α. In contrast, clearance of infection is associated with increased expression of a number genes encoding proteins involved in innate pathogen clearance, as well as apoptosis and control of inflammation. Conclusion - This first organ-level immune response transcriptional analysis for IA has enabled us to gain new insights into the mechanisms that govern fungal immunity in the lung.The BBSRC, CGD Research Trust, and the MRC

    The dynamics of the γ Vel cluster and nearby Vela OB2 association

    The kinematics of low-mass stars in nearby OB associations can provide clues about their origins and evolution. Combining the precise positions, proper motions, and parallaxes given in the second Gaia Data Release with radial-velocity measurements obtained with the Hermes spectrograph at the Anglo-Australian Telescope, we have an opportunity to study in detail the kinematics of low-mass stars belonging to the nearby γ Vel cluster and the Vela OB2 association it is projected against. The presence of lithium is used to confirm the youth of our targets. We separate our sample into the cluster and association populations based on the Gaia-ESO Survey membership probabilities their parallaxes, and kinematics. We find strong evidence for expansion in the OB association population with at least 4σ significance along all three axes, though the expansion is notably anisotropic. We discuss these results in the context of cluster and association dispersal theories

    The use of steroids and thalidomide in the management of Erythema Nodosumn Leprosum; 17 years at the Hospital for Tropical Diseases, London

    Objectives: Prednisolone and thalidomide are commonly used in the management of erythema nodosum leprosum (ENL) and bring relief to patients with this condition worldwide. However, both ENL and its treatments can cause significant morbidity. This study describes the spectrum of ENL seen at The Hospital for Tropical Diseases, London (HTD), the use of steroids and thalidomide in its management and the complications of their use. Study Design: We conducted a retrospective audit of patients diagnosed with ENL between 1996 and 2013. Data were obtained from hospital records including severity and length of disease, together with treatments received and adverse effects. Results: Between 1996 and 2013, 30 patients were diagnosed with ENL. The median bacillary index (BI) at diagnosis was 4·65, higher than in previous studies. Most patients developed ENL during leprosy treatment (67%) and had chronic ENL (57%). The median length of ENL was 60 months (range 9 – 192); patients with BI . 4·5 had significantly longer duration of disease. 87% patients received prednisolone for median nine months; 35% developed adverse effects including diabetes and hypertension. 87% patients received thalidomide for median 16 months; 65% complained of side effects. There were no pregnancies or venous thromboembolisms. 77% patients stopped prednisolone within two months of starting thalidomide. There were no deaths in our cohort. Conclusion: We describe the clinical course of ENL in a non-endemic country with access to thalidomide and prednisolone. ENL may last far longer than previously described and has significant impact on a patient’s health. In the UK, thalidomide is essential as a steroid-sparing agent, to prevent the adverse effects and mortality of long-term steroids which have been documented elsewhere.</p

    Spatial patterns of the tau pathology in progressive supranuclear palsy

    Progressive supranuclear palsy (PSP) is characterized neuropathologically by neuronal loss, gliosis, and the presence of tau-immunoreactive neuronal and glial cell inclusions affecting subcortical and some cortical regions. The objectives of this study were to determine (1) the spatial patterns of the tau-immunoreactive pathology, viz., neurofibrillary tangles (NFT), oligodendroglial inclusions (GI), tufted astrocytes (TA), and Alzheimer's disease-type neuritic plaques (NP) in PSP and (2) to investigate the spatial correlations between the histological features. Post-mortem material of cortical and subcortical regions of eight PSP cases was studied. Spatial pattern analysis was applied to the NFT, GI, TA, NP, abnormally enlarged neurons (EN), surviving neurons, and glial cells. NFT, GI, and TA were distributed either at random or in regularly distributed clusters. The EN and NP were mainly randomly distributed. Clustering of NFT and EN was more frequent in the cortex and subcortical regions, respectively. Variations in NFT density were not spatially correlated with the densities of either GI or TA, but were positively correlated with the densities of EN and surviving neurons in some regions. (1) NFT were the most widespread tau-immunoreactive pathology in PSP being distributed randomly in subcortical regions and in regular clusters in cortical regions, (2) GI and TA were more localized and exhibited a regular pattern of clustering in subcortical regions, and (3) neuronal and glial cell pathologies were not spatially correlated. © 2012 Springer-Verlag

    Synthesis, Structure and Stereodynamics of Atropisomeric <em>N</em>‑Chloroamides

    Atropisomeric N-chloroamides were efficiently accessed by electrophilic halogenation of ortho-substituted secondary anilides. The stereodynamics of atropisomerism in these novel scaffolds was interrogated by detailed experimental and computational studies, revealing that racemization is correlated with amide isomerization. The stereoelectronic nature of the amide was shown to significantly influence racemization rates, with potentially important implications for other C-N atropisomeric scaffolds

    Multiplexed microsphere diagnostic tools in gene expression applications: factors and futures

    Microarrays have received significant attention in recent years as scientists have firstly identified factors that can produce reduced confidence in gene expression data obtained on these platforms, and secondly sought to establish laboratory practices and a set of standards by which data are reported with integrity. Microsphere-based assays represent a new generation of diagnostics in this field capable of providing substantial quantitative and qualitative information from gene expression profiling. However, for gene expression profiling, this type of platform is still in the demonstration phase, with issues arising from comparative studies in the literature not yet identified. It is desirable to identify potential parameters that are established as important in controlling the information derived from microsphere-based hybridizations to quantify gene expression. As these evolve, a standard set of parameters will be established that are required to be provided when data are submitted for publication. Here we initiate this process by identifying a number of parameters we have found to be important in microsphere-based assays designed for the quantification of low abundant genes which are variable between studies

    The influence of rs53576 polymorphism in the oxytocin receptor (OXTR) gene on empathy in healthy adults by subtype and ethnicity: a systematic review and meta-analysis

    Empathy is essential for navigating complex social environments. Prior work has shown associations between rs53576, a single nucleotide polymorphism (SNP) located in the oxytocin receptor gene (OXTR), and generalized empathy. We undertook a systematic review and meta-analysis to assess the effects of rs53576 on subdomains of empathy, specifically cognitive empathy (CE) and affective empathy (AE), in healthy adults. Twenty cohorts of 8933 participants aged 18-98 were identified, including data from the Sydney Memory and Ageing Study, a cohort of older community adults. Meta-analyses found G homozygotes had greater generalized empathic abilities only in young to middle-aged adults. While meta-analyses of empathy subdomains yielded no significant overall effects, there were differential effects based on ethnicity. G homozygotes were associated with greater CE abilities in Asian cohorts (standardized mean difference; SMD: 0.09 [2.8·10-3-0.18]), and greater AE performance in European cohorts [SMD: 0.12 (0.04-0.21)]. The current literature highlights a need for further work that distinguishes between genetic and ethnocultural effects and explores effects of advanced age on this relationship

    The structure and 3D kinematics of vela OB2

    The kinematics of stars in OB associations can provide insights into their formation, dynamical evolution, and eventual fate. The low-mass stellar content of OB associations are sufficiently numerous as to provide a detailed sampling of their kinematic properties, however spectroscopy is required to confirm the youth of individual stars and to get 3D kinematics. In this paper, we present and analyse results from a large spectroscopic survey of Vela OB2 conducted using 2dF/HERMES on the AAT. This spectroscopy is used to confirm the youth of candidate young stars and determine radial velocities, which are combined with proper motions and parallaxes from Gaia to measure 3D positions and velocities. We identify multiple separate kinematic groups in the region, for which we measure velocity dispersions and infer their virial states. We measure expansion rates for all these groups and find strong evidence for anisotropic expansion in the Vela OB2 association of at least 11σ significance in all three dimensions, as well as some evidence for expansion in the γ Vel and P Puppis clusters. We trace back the motions of these groups into the past and find that the open cluster NGC 2547 is an interloper in the Vela OB2 region and actually formed >100 pc away from the association. We conclude that Vela OB2 must have formed with considerable spatial and kinematic substructure over a time-scale of ∼10 Myr, with clear temporal substructure within the association, but no clear evidence for an age gradient

    Time-Delay Interferometry

    Equal-arm interferometric detectors of gravitational radiation allow phase measurements many orders of magnitude below the intrinsic phase stability of the laser injecting light into their arms. This is because the noise in the laser light is common to both arms, experiencing exactly the same delay, and thus cancels when it is differenced at the photo detector. In this situation, much lower level secondary noises then set overall performance. If, however, the two arms have different lengths (as will necessarily be the case with space-borne interferometers), the laser noise experiences different delays in the two arms and will hence not directly cancel at the detector. In order to solve this problem, a technique involving heterodyne interferometry with unequal arm lengths and independent phase-difference readouts has been proposed. It relies on properly time-shifting and linearly combining independent Doppler measurements, and for this reason it has been called Time-Delay Interferometry (or TDI). This article provides an overview of the theory and mathematical foundations of TDI as it will be implemented by the forthcoming space-based interferometers such as the Laser Interferometer Space Antenna (LISA) mission. We have purposely left out from this first version of our ``Living Review'' article on TDI all the results of more practical and experimental nature, as well as all the aspects of TDI that the data analysts will need to account for when analyzing the LISA TDI data combinations.
