    Textile based dye-sensitized solar cells with natural dyes

    Natural dyes extracted from hibiscus petals, elderberries and mallow flowers were investigated in dye-sensitized solar cells. Two approaches were followed: 1. Hybrid glass/fabric cells with titanium dioxide on glass as working electrode and a textile counter electrode; 2. hybrid fabric/glass cells with zinc oxide as working electrode on textile and a glass counter electrode. The zinc oxide layer on cotton was prepared by electroless deposition whereas the titanium dioxide coated glass electrodes were obtained directly from the manufacturer. In both cases the redox couple consisted of iodine / triiodide and the counter electrode was based on an electrically conductive fabric

    Incidence of HIV-related anal cancer remains increased despite long-term combined antiretroviral treatment: results from the french hospital database on HIV.

    PURPOSE: To study recent trends in the incidence of anal cancer in HIV-infected patients receiving long-term combined antiretroviral treatment (cART) compared with the general population. PATIENTS AND METHODS: From the French Hospital Database on HIV, we identified 263 cases of invasive anal squamous cell carcinoma confirmed histologically between 1992 and 2008. We compared incidence rates of anal cancer across four calendar periods: 1992-1996 (pre-cART period), 1997-2000 (early cART period), and 2001-2004 and 2005-2008 (recent cART periods). Standardized incidence ratios (SIRs) were calculated by using general population incidence data from the French Network of Cancer Registries. RESULTS: In HIV-infected patients, the hazard ratio (HR) in the cART periods versus the pre-cART period was 2.5 (95% CI, 1.28 to 4.98). No difference was observed across the cART calendar periods (HR, 0.9; 95% CI, 0.6 to 1.3). In 2005-2008, HIV-infected patients compared with the general population had an excess risk of anal cancer, with SIRs of 109.8 (95% CI, 84.6 to 140.3), 49.2 (95% CI, 33.2 to 70.3), and 13.1 (95% CI, 6.8 to 22.8) for men who have sex with men (MSM), other men, and women, respectively. Among patients with CD4 cell counts above 500/ÎźL for at least 2 years, SIRs were 67.5 (95% CI, 41.2 to 104.3) when the CD4 nadir was less than 200/ÎźL for more than 2 years and 24.5 (95% CI, 17.1 to 34.1) when the CD4 nadir was more than 200/ÎźL. CONCLUSION: Relative to that in the general population, the risk of anal cancer in HIV-infected patients is still extremely high, even in patients with high current CD4 cell counts. cART appears to have no preventive effect on anal cancer, particularly in MSM

    Stimulus Prediction and Postural Reaction: Phase-Specific Modulation of Soleus H-Reflexes Is Related to Changes in Joint Kinematics and Segmental Strategy in Perturbed Upright Stance

    Anticipation determines the timing and efficiency of human motor performance. This study aimed to evaluate the effect of stimulus anticipation on proactive (prior to the event) and reactive (after the event) postural adjustments in response to perturbations. Postural set was manipulated by providing either (i) predictable, (ii) unpredictable, or (iii) cheated perturbations which require balance corrections to maintain postural stability. In 29 subjects, a protocol of anterior and posterior perturbations was applied for the conditions (i–iii). Center of pressure (COP) displacement, ankle, knee, and hip joint kinematics and electromyographic activity (EMG) of the soleus (SOL) and tibialis anterior (TA) muscles were recorded prior (PRE) and after posterior perturbations. SOL H-reflexes at the peak of the short-, medium- ,and long-latency responses (SLR, MLR, LLR) were assessed. For conditions (i to iii) EMG activity and COP differed prior to perturbation onset (p < 0.05). After perturbation, results demonstrated a progressively increased H-reflex amplitude in the MLR and LLR (p < 0.05), delayed muscle activities (p < 0.05), and shifted activation patterns, with muscles of the proximal segment being more involved in the compensatory postural response (p < 0.05). COP displacements and ankle, knee, and hip joint deflections progressively increased (p < 0.05). Neuromechanical coupling showed positive correlations for the anticipation-induced changes in EMG activity and H-reflex amplitude with that of COP displacement (p < 0.05). In conclusion, proactive and reactive postural responses indicated setting dependent modulations of segmental and phasic muscle activation. A shift to proximal muscle groups and facilitated late reflex responses compensating for cheated or unpredicted perturbations was found to recover a safe body equilibrium. In consideration of the phase-specific adaptation and its interrelationship to the kinematics, it suggested that changes in stimulus prediction challenged the central nervous system to appropriately counteract the higher postural challenges. The outcomes of this experiment are of functional relevance for experimental and training settings involving perturbation stimuli. These findings provide fundamental information of the mechanisms underlying postural adjustments in response to external perturbations

    LMNA Co-Regulated Gene Expression as a Suitable Readout after Precise Gene Correction

    LMNA-related muscular dystrophy is an autosomal-dominant progressive disorder caused by mutations in LMNA. LMNA missense mutations are becoming correctable with CRISPR/Cas9-derived tools. Evaluating the functional recovery of LMNA after gene editing bears challenges as there is no reported direct loss of function of lamin A/C proteins in patient-derived cells. The proteins encoded by LMNA are lamins A/C, important ubiquitous nuclear envelope proteins but absent in pluripotent stem cells. We induced lamin A/C expression in induced pluripotent stem cells (iPSCs) of two patients with LMNA-related muscular dystrophy, NM_170707.4 (LMNA): c.1366A > G, p.(Asn456Asp) and c.1494G > T, p.(Trp498Cys), using a short three-day, serum-induced differentiation protocol and analyzed expression profiles of co-regulated genes, examples being COL1A2 and S100A6. We then performed precise gene editing of LMNA c.1366A > G using the near-PAMless (PAM: protospacer-adjacent motif) cytosine base editor. We show that the mutation can be repaired to 100% efficiency in individual iPSC clones. The fast differentiation protocol provided a functional readout and demonstrated increased lamin A/C expression as well as normalized expression of co-regulated genes. Collectively, our findings demonstrate the power of CRISPR/Cas9-mediated gene correction and effective outcome measures in a disease with, so far, little perspective on therapies

    Governance and stakeholder perspectives of managed re-alignment: Adapting to sea level rise in the Inner Forth estuary, Scotland

    With climate change, coastal areas are faced with unprecedented sea level rise and flooding, raising questions as to how societies will choose to adapt. One option is to strengthen existing sea walls to maintain current land uses; however, scientists, policy-makers and conservationists increasingly see the benefits of managed realignment, which is a nature-based coastal adaptation that involves the conversion of reclaimed farmland back to wetlands, allowing periodic local flooding in designated areas to reduce the risk of flooding downstream. We interviewed 16 local organisations, landowners and farmers and held workshops with 109 citizens living the Inner Forth estuary in eastern Scotland, to examine how managed realignment is supported by stakeholder attitudes and their engagement. Most of the farmers we interviewed prefer strengthened sea walls, to maintain their livelihoods and agricultural heritage. Citizens and local organisations were mainly supportive of managed realignment, because it provided wildlife and flood regulation benefits. However, we identified several barriers that could present obstacles to implementing managed realignment, for example, uncertainty whether it would support their principles of economic and rational decision-making. Our findings suggest that the local capacity to cope with rising sea levels is limited by lack of engagement with all relevant stakeholder groups, the limited scope of existing stakeholder partnerships and poor short-term funding prospects of landscape partnerships that would facilitate collaboration and discussion. We suggest that including citizens, landowners, farmers and industries would strengthen existing stakeholder deliberation and collaboration, and support the Inner Forth’s transition towards a more sustainable future shoreline

    Community Seismic Network

    The article describes the design of the Community Seismic Network, which is a dense open seismic network based on low cost sensors. The inputs are from sensors hosted by volunteers from the community by direct connection to their personal computers, or through sensors built into mobile devices. The server is cloud-based for robustness and to dynamically handle the load of impulsive earthquake events. The main product of the network is a map of peak acceleration, delivered within seconds of the ground shaking. The lateral variations in the level of shaking will be valuable to first responders, and the waveform information from a dense network will allow detailed mapping of the rupture process. Sensors in buildings may be useful for monitoring the state-of-health of the structure after major shaking

    Hintergrund und Ziel der Arbeit: Informationen zur Mundgesundheit der Bevölkerung sind wichtig für die Einschätzung von (vermeidbarer) Krankheitslast, für die Einschätzung und Planung von Gesundheitsressourcen und -kosten sowie für die Beurteilung gesundheitlicher Ungleichheiten. Ziel dieser Arbeit ist es, erstmals auf Datenbasis einer bundesweit repräsentativen Stichprobe für die erwachsene Bevölkerung in Deutschland die selbstwahrgenommene Mundgesundheit einschließlich assoziierter Faktoren zu untersuchen. Material und Methoden: Datenbasis ist die Studie Gesundheit in Deutschland aktuell (GEDA 2019/2020-EHIS, n = 22.708 ab 18 Jahre). Im telefonischen Interview wurden die Teilnehmenden gefragt, wie sie den Zustand ihrer Zähne und ihres Zahnfleischs beschreiben würden – „sehr gut“, „gut“, „mittelmäßig“, „schlecht“, „sehr schlecht“. Soziodemografische, verhaltensbezogene und zahnmedizinische Merkmale werden als assoziierte Faktoren untersucht. Ausgewiesen werden Prävalenzen und Ergebnisse multivariater binär-logistischer Regressionen (Odds Ratios, OR). Ergebnisse und Diskussion: 71,4 % der Befragten schätzten ihre Mundgesundheit als sehr gut oder gut ein, 28,6 % als mittelmäßig bis sehr schlecht. Die wichtigsten assoziierten Faktoren waren Beeinträchtigungen beim Kauen und Beißen (OR 4,0), ein unerfüllter zahnmedizinischer Versorgungsbedarf (OR 2,3), männliches Geschlecht (OR 1,5) und ein nicht täglicher Obst- und Gemüsekonsum (OR 1,2), bei Männern zudem eine niedrige Bildung (OR 2,1), das tägliche Rauchen (OR 1,6) und eine nicht jährliche zahnmedizinische Inanspruchnahme (OR 1,4). Aus den Ergebnissen lassen sich Ansatzpunkte zur Förderung der Mundgesundheit ableiten.Background and aim of the work: Information on the oral health of the population is important for the assessment of (preventable) disease burden, for the estimation and planning of health resources and costs, and for the evaluation of health inequalities. The aim of this work is to investigate for the first time self-perceived oral health, including associated factors, based on data from a nationally representative sample for the adult population in Germany. Material and methods: The data basis is the nationwide German Health Update study (GEDA 2019/2020-EHIS, n = 22,708 aged 18 years and older). In the telephone interview, the participants were asked how they would describe the state of their teeth and gums – “very good,” “good,” “fair,” “bad,” or “very bad.” Sociodemographic, behavioral, and dental characteristics are examined as associated factors. Prevalences and results of multivariate binary logistic regressions (odds ratios, OR) are reported. Results and discussion: Of the participants, 71.4% perceived their oral health as very good or good, 28.6% as fair to very bad. Difficulty in chewing and biting on hard foods (OR 4.0), unmet dental care needs (OR 2.3), male gender (OR 1.5), and not consuming fruits and vegetables daily (OR 1.2) were the most important associated factors for fair to very bad self-perceived oral health; for men, low education (OR 2,1), daily smoking (OR 1.6) and not receiving dental care annually (OR 1.4) were also important.     Endophilin, Lamellipodin, and Mena cooperate to regulate F-actin-dependent EGF-receptor endocytosis

    The epidermal growth factor receptor (EGFR) plays an essential role during development and diseases including cancer. Lamellipodin (Lpd) is known to control lamellipodia protrusion by regulating actin filament elongation via Ena/VASP proteins. However, it is unknown whether this mechanism supports endocytosis of the EGFR. Here, we have identified a novel role for Lpd and Mena in clathrin-mediated endocytosis (CME) of the EGFR. We have discovered that endogenous Lpd is in a complex with the EGFR and Lpd and Mena knockdown impairs EGFR endocytosis. Conversely, overexpressing Lpd substantially increases the EGFR uptake in an F-actin-dependent manner, suggesting that F-actin polymerization is limiting for EGFR uptake. Furthermore, we found that Lpd directly interacts with endophilin, a BAR domain containing protein implicated in vesicle fission. We identified a role for endophilin in EGFR endocytosis, which is mediated by Lpd. Consistently, Lpd localizes to clathrin-coated pits (CCPs) just before vesicle scission and regulates vesicle scission. Our findings suggest a novel mechanism in which Lpd mediates EGFR endocytosis via Mena downstream of endophilin
