The role of residual antigen and of the alarmin IL-33 in shaping the antiviral immune response
The aims of the thesis were threefold:
Firstly, we were interested in studying the potency of residual LCMV antigen-depots in transiently CD4 T cell depleted mice for the induction of an antiviral B cell response.
Secondly, we studied whether a heterologous arenavirus vector prime-boost was beneficial for the establishment of an effective tumor-specific CD8 T cell response.
Thirdly, we aimed to define the role of the IL-33 ST2 axis for the formation of an anti-viral CD8 T cell response in the context of a chronic viral infection
Um novo método para simular o estado hidrológico do solo em condições naturais
Micro, macro and mesofauna in the soil often respond to fluctuating environmental conditions, resulting in changes of abundance and community structure. Effects of changing soil parameters are normally determined with samples taken in the field and brought to the laboratory, i.e. where natural environmental conditions may not apply. We devised a method (STAFD – soil tubes for artificial flood and drought), which simulates the hydrological state of soil in situ using implanted cores. Control tubes were compared with treatment tubes in which floods of 15, 30, 60 and 90 days, and droughts of 60, 90 and 120 days were simulated in the field. Flooding and drought were found to reduce number of individuals in all soil faunal groups, but the response to drought was slower and not in proportion to the expected decrease of the water content. The results of the simulated floods in particular show the value of the STAFD method for the investigation of such extreme events in natural habitats.As micro, macro e mesofaunas no solo respondem frequentemente à variação das condições ambientais, o que resulta em alterações na abundância e na estrutura da comunidade. Os efeitos das alterações nos parâmetros do solo são normalmente determinados com amostras recolhidas no campo e trazidas ao laboratório, ou seja, onde as condições ambientais naturais podem não ser aplicáveis. Criamos um método (STAFD – tubos de amostra de solo para inundações e secas artificiais), que simula o estado hidrológico do solo in situ com núcleos implantados. As amostras de controle foram comparadas com amostras de tratamentos em que foram simuladas inundações de 15, 30, 60 e 90 dias e secas de 60, 90 e 120 dias no campo. Verificou-se que as inundações e a seca reduziram a quantidade de individuos em todos os grupos de fauna de solo, mas a resposta à seca foi mais lenta e não proporcional à redução prevista no teor de água. Os resultados das inundações simuladas demonstram, em especial, o valor do método STAFD para a investigação desse tipo de eventos extremos nos habitats naturais
Supplemental prophylactic intervention for chemotherapy-induced nausea and emesis (spice) trial: Protocol for a multi-centre double-blind placebo-controlled randomized trial
Aim:
There is significant recent interest in the role of ginger root (Zingiber officinale) as an adjuvant therapy for chemotherapy‐induced nausea. The supplemental prophylactic intervention for chemotherapy‐induced nausea and emesis (SPICE) trial aims to assess the efficacy by reduced incidence and severity of chemotherapy‐induced nausea and vomiting, and enhanced quality of life, safety and cost effectiveness of a standardised adjuvant ginger root supplement in adults undergoing single‐day moderate‐to‐highly emetogenic chemotherapy.
Methods:
Multisite, double‐blind, placebo‐controlled randomised trial with two parallel arms and 1:1 allocation. The target sample size is n = 300. The intervention comprises four capsules of ginger root (totalling 60 mg of active gingerols/day), commencing the day of chemotherapy and continuing for five days during chemotherapy cycles 1 to 3. The primary outcome is chemotherapy‐induced nausea‐related quality of life. Secondary outcomes include nutrition status; anticipatory, acute and delayed nausea and vomiting; fatigue; depression and anxiety; global quality of life; health service use and costs; adverse events; and adherence.
Results:
During the five‐month recruitment period from October 2017 to April 2018 at site A only, a total of n = 33 participants (n = 18 female) have been enrolled in the SPICE trial. Recruitment is expected to commence at Site B in May 2018.
Conclusions:
The trial is designed to meet research gaps and could provide evidence to recommend specific dosing regimens as an adjuvant for chemotherapy‐induced nausea and vomiting prevention and management.No Full Tex
The role of positive psychology construct in employee coping
The objectives of this study were to: (1) determine the relationship between locus of control (measured by the Locus of Control Inventory), work engagement (measured by the Utrecht Work Engagement Scale) and sense of coherence (measured by the Orientation to Life Questionnaire); (2) determine whether coping and non-coping individuals (measured by a coping scale) differ significantly regarding these variables, and (3) the strategies they employ to deal with change in their lives (determined by means of structured interviews).
A survey design, and correlational and inferential statistical procedures were used to achieve the quantitative research objectives. Qualitative content analysis was used to analyse and interpret the research data obtained from the open-ended questions on coping strategies. A convenience sample (n = 69) of individuals employed in the manufacturing sector participated in the study. The quantitative results revealed a significant relationship between the three variables and that copers and non-copers differ significantly regarding their coping. The qualitative data analysis indicated that copers and non-copers differ in terms of the coping strategies they employ to deal with change.
The study represents original research which contributes new knowledge and insights for organisational employee wellness practices aimed at helping employees cope more positively with organisational change in the South African organisational context.Industrial and Organisational PsychologyM.A. (Industrial and Organisational Psychology
Predicting Runoff Risks By Digital Soil Mapping
Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq)Digital soil mapping (DSM) permits continuous mapping soil types and properties through raster formats considering variation within soil class, in contrast to the traditional mapping that only considers spatial variation of soils at the boundaries of delineated polygons. The objective of this study was to compare the performance of SoLIM (Soil Land Inference Model) for two sets of environmental variables on digital mapping of saturated hydraulic conductivity and solum depth (A + B horizons) and to apply the best model on runoff risk evaluation. The study was done in the Posses watershed, MG, Brazil, and SoLIM was applied for the following sets of co-variables: 1) terrain attributes (AT): slope, plan curvature, elevation and topographic wetness index. 2) Geomorphons and terrain attributes (GEOM): slope, plan curvature, elevation and topographic wetness index combined with geomorphons. The most precise methodology was applied to predict runoff areas risk through the Wetness Index based on contribution area, solum depth, and saturated hydraulic conductivity. GEOM was the best set of co-variables for both properties, so this was the DSM model used to predict the runoff risk. The runoff risk showed that the critical months are from November to March. PPARgamma activation attenuates T-lymphocyte-dependent inflammation of adipose tissue and development of insulin resistance in obese mice
<p>Abstract</p> <p>Background</p> <p>Inflammation of adipose tissue (AT) has been recently accepted as a first step towards obesity-mediated insulin resistance. We could previously show that mice fed with high fat diet (HFD) develop systemic insulin resistance (IR) and glucose intolerance (GI) associated with CD4-positive T-lymphocyte infiltration into visceral AT. These T-lymphocytes, when enriched in AT, participate in the development of fat tissue inflammation and subsequent recruitment of proinflammatory macrophages. The aim of this work was to elucidate the action of the insulin sensitizing PPARgamma on T-lymphocyte infiltration during development of IR, and comparison of the PPARgamma-mediated anti-inflammatory effects of rosiglitazone and telmisartan in diet-induced obesity model (DIO-model) in mice.</p> <p>Methods</p> <p>In order to investigate the molecular mechanisms underlying early development of systemic insulin resistance and glucose intolerance male C57BL/6J mice were fed with high fat diet (HFD) for 10-weeks in parallel to the pharmacological intervention with rosiglitazone, telmisartan, or vehicle.</p> <p>Results</p> <p>Both rosiglitazone and telmisartan were able to reduce T-lymphocyte infiltration into AT analyzed by quantitative analysis of the T-cell marker CD3gamma and the chemokine SDF1alpha. Subsequently, both PPARgamma agonists were able to attenuate macrophage infiltration into AT, measured by the reduction of MCP1 and F4/80 expression. In parallel to the reduction of AT-inflammation, ligand-activated PPARgamma improved diet-induced IR and GI.</p> <p>Conclusion</p> <p>Together the present study demonstrates a close connection between PPARgamma-mediated anti-inflammation in AT and systemic improvement of glucose metabolism identifying T-lymphocytes as one cellular mediator of PPARgamma´s action.</p
