    Urinary levels of enniatin B and its phase I metabolites: First human pilot biomonitoring study

    Enniatins (Enns) are mycotoxins produced by Fusarium spp. and are widely distributed contaminants of cereals and derivate products. Among the different identified enniatins, Enn B is the most relevant analogue in cereals in Europe. Therefore, the aim of this study was to investigate for the first time the occurrence of Enn B and Enn B phase I metabolites in 300 human urine samples throughout an ultrahigh-performance liquid chromatography high resolution mass spectrometry (UHPLC-Q-Orbitrap HRMS) methodology. Three different sample preparation procedures were evaluated and salting-out liquid-liquid extraction showed satisfactory validation results. Enn B was quantified in 83.7% of samples ranging from 0.006 to 0.391 ng/mL (average content: 0.016 ng/mL). In line with the in vitro observations with human liver microsomes, in the here analyzed samples the Enn B monooxygenated, N-demethylated and dioxygenated metabolites were tentatively found in 87.7%, 96.3% and 6.7% of samples. The data of this pilot biomonitoring survey indicate a frequent intake of enniatins in Italy, supporting further toxicological studies to provide better basis for understanding their potential effects in humans

    Target analysis and retrospective screening of mycotoxins and pharmacologically active substances in milk using an ultra-high-performance liquid chromatography/high-resolution mass spectrometry approach

    Milk is a nutritious food suitable for infants and adults, and it plays an important role in the human diet. However, it may also be a vehicle for food contaminants. In this report, we developed a method using ultra-high-performance liquid chromatography coupled with high-resolution mass spectrometry (UHPLC-Q-Exactive Orbitrap HRMS; Thermo Fisher Scientific, Waltham, MA) for simultaneous identification of target pharmacologically active substances and mycotoxins in milk. We also used the Q-Orbitrap operating in full scan mode to identify other possible drugs and microbial metabolites that occurred in samples. Fifty-six commercially available milk samples from the Italian market were analyzed. Investigated analytes were extracted using a QuEChERS (quick, easy, cheap, effective, rugged, and safe) approach. Method detection and quantification limits and performance criteria set by European regulations were fulfilled. Pharmacologically active substances were detected in 49% of samples (range 0.007–4.53 ng/mL), including nontarget mycotoxins. Retrospective analysis allowed us to identify other antibiotics and pharmacologically active substances, as well as nonregulated fungal/bacterial metabolites at a relatively high incidence. From the obtained values, the need for continuous monitoring of contaminants in the milk production chain is clear. This is the first study to assess the presence of pharmacologically active substances, mycotoxins, and other microbial metabolites in Italian milk samples using the UHPLC-Q-Orbitrap HRMS system

    Multi-Mycotoxin Analysis in Durum Wheat Pasta by  Liquid Chromatography Coupled to Quadrupole  Orbitrap Mass Spectrometry

    A simple and rapid multi-mycotoxin method for the determination of 17 mycotoxins  simultaneously  is  described  in  the  present  survey  on  durum  and  soft  wheat  pasta  samples.  Mycotoxins included in the study were those mainly reported in cereal samples: ochratoxin-A  (OTA), aflatoxin B1 (AFB1), zearalenone (ZON), deoxynivalenol (DON), 3-and 15-acetyl-deoxynivalenol  (3-AcDON and 15-AcDON), nivalenol (NIV), neosolaniol (NEO), fusarenon-X, (FUS-X), T-2 toxin  (T-2) and HT-2 toxin (HT-2), fumonisin B1 and B2 (FB1 and FB2), and four emerging mycotoxins:  three  enniatins  (ENA,  ENA1,  and  ENB),  and  beauvericin  (BEA).  Twenty-nine  samples  were  analyzed to provide an overview on mycotoxin presence: 27 samples of durum wheat pasta, and  two samples of baby food. Analytical results concluded that trichothecenes showed the highest  incidence, mainly DON, NIV, and HT-2 toxin, followed by ZON and ENB, while NEO, FUS-X, OTA,  AFB1, and FUM were not detected in any sample. The highest contents corresponded to ENB and  ranged from 91.15μg/kg to 710.90 μg/kg

    Analysis of Polyphenolic Compounds in Water-Based Extracts of Vicia faba L.: A Potential Innovative Source of Nutraceutical Ingredients

    The water-based extract of broad bean hulls contains several bioactive molecules, including polyphenols well-known to exert antioxidant activity, which could justify its use in nutraceutical formulations. Hence, the current investigation aimed to establish the polyphenolic profile of water-based extracts from broad bean hulls through UHPLC-Q-Orbitrap HRMS analysis. The findings highlighted that p-coumaric acid, chlorogenic acid, and epicatechin were the most common compounds found in the tested extracts, being quantified at a mean concentration of 42.1, 32.6, and 31.2 mg/100 g, respectively. Moreover, broad bean hull extracts were encapsulated into a nutraceutical formulation, after which the antioxidant properties and the bioaccessibility of phenolic compounds during the simulated gastrointestinal (GI) process were investigated and compared with the digested non-encapsulated extract. The data highlighted that following the GI process, the capsules were able to preserve active compounds from the adverse effects of digestion, resulting in a greater antioxidant capacity and polyphenol bioaccessibility in the duodenal and colonic phases, compared with the non-encapsulated extract. Our results showed that the water extract from broad bean hulls may be considered a valuable source of natural polyphenolic compounds; in addition, the use of a gastric-resistant capsule could be a suitable alternative to transport these bioactive compounds to the target tissues

    Endocrine and molecular investigations in a cohort of 25 adolescent males with prominent/persistent pubertal gynecomastia

    Pubertal gynecomastia is a common condition observed in up to 65% of adolescent males. It is usually idiopathic and tends to regress within 1–2 years. In this descriptive cross-sectional study, we investigated 25 adolescent males with prominent (>B3) and/or persistent (>2 years) pubertal gynecomastia (P/PPG) to determine whether a hormonal/genetic defect might underline this condition. Endocrine investigation revealed the absence of hormonal disturbance for 18 boys (72%). Three patients presented Klinefelter syndrome and three a partial androgen insensitivity syndrome (PAIS) as a result of p.Ala646Asp and p.Ala45Gly mutations of the androgen receptor gene. The last patient showed a 17α-hydroxylase/17,20-lyase deficiency as a result of a compound heterozygous mutation of the CYP17A1 gene leading to p.Pro35Thr(P35T) and p.Arg239Stop(R239X) in the P450c17 protein. Enzymatic activity was analyzed: the mutant protein bearing the premature stop codon R239X showed a complete loss of 17α-hydroxylase and 17,20-lyase activity. The mutant P35T seemed to retain 15–20% of 17α-hydroxylase and about 8–10% of 17,20-lyase activity. This work demonstrates that P/PPG had an endocrine/genetic cause in 28% of our cases. PAIS may be expressed only by isolated gynecomastia as well as by 17α-hydroxylase/17,20-lyase deficiency. Isolated P/PPG is not always a ‘physiological’ condition and should thus be investigated through adequate endocrine and genetic investigations, even though larger studies are needed to better determine the real prevalence of genetic defects in such patients

    An Innovative Olive Pâté with Nutraceutical Properties

    Food plays a central role in health, especially through consumption of plant-derived foods. Functional foods, supplements, and nutraceuticals are increasingly entering the market to respond to consumer demand for healthy products. They are foods, supplements, and ingredients which offer health benefits beyond the standard nutritional value. Some benefits are associated with phenolic compounds and phytochemicals with antioxidant properties. An olive pâté (OP) was added with antioxidants derived from olive mill wastewater (OMWW) to obtain a functional product rich in phenolic compounds. The olive pâté is produced from the ground olive pericarp, which shows an excellent natural antioxidant content. The OMWW is a waste product from oil processing, which is also rich in phenolic compounds. The result was a product rich in trans-resveratrol, OH tyrosol, and tyrosol in concentrations such as satisfying the European community’s claims regarding the possible antioxidant action on plasma lipids with excellent shelf-life stability. The total phenolic content was assayed by a colorimetric method, the antioxidant activity by the ABTS [(2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)] test, the phenolic profile by Q Exactive Orbitrap LC-MS/MS. The shelf-life stability was confirmed by yeast, molds, and total microbial count, pH, and water activity determinations, and the best pasteurization parameters were determined.     A major glacial-interglacial change in aeolian dust composition inferred from Rare Earth Elements in Antarctic ice

    We present the first Rare Earth Elements (REE) concentration record determined in 294 sections of an Antarctic ice core (EPICA Dome C), covering a period from 2.9 to 33.7 kyr BP. REE allow a detailed quantitative evaluation of aeolian dust composition because of the large number of variables (i.e. 14 elements). REE concentrations match the particulate dust concentration profile over this period and show a homogeneous crustal-like composition during the last glacial stage (LGS), with only a slight enrichment in medium REE. This signature is consistent with the persistent fallout of a mixture of dust from heterogeneous sources located in different areas or within the same region (e.g. South America). Starting at ∼15 kyr BP, there was a major change in dust composition, the variable character of which persisted throughout the Holocene. This varying signature may highlight the alternation of single dust contributions from different sources during the Holocene. We observe that the frequent changes in REE composition at the onset of the Holocene (10-13.5 kyr BP) are linked to dust size and in turn to wind strength and/or the path of the atmospheric trajectory. This may indicate that atmospheric circulation dictated the composition of the dust fallout to East Antarctica at that time. Although the dust concentrations remained fairly low, a notable return towards more glacial dust characteristics is recorded between 7.5 and 8.3 kyr BP. This happened concomitantly with a widespread cold event around 8 kyr BP that was 400-600 years long and suggests a moderate reactivation of the dust emission from the same potential source areas of the LGS

    The fraction and kinematics of broad absorption line quasars across cosmic time

    Luminous quasars are powerful targets to investigate the role of feedback from supermassive black-holes (BHs) in regulating the growth phases of BHs themselves and of their host galaxies, up to the highest redshifts. Here we investigate the cosmic evolution of the occurrence and kinematics of BH-driven outflows, as traced by broad absorption line (BAL) features, due to the C IV ionic transition. We exploit a sample of 1935 quasars quasars at z=2.16.6z=2.1-6.6 with bolometric luminosity log(Lbol/L_{\rm bol}/erg s1)46.5^{-1})\gtrsim46.5, drawn from the Sloan Digital Sky Survey and from the X-shooter legacy survey of Quasars at Reionisation (XQR-30). We consider rest-frame optical bright quasars to minimise observational biases due to quasar selection criteria. We apply a homogeneous BAL identification analysis, based on employing composite template spectra to estimate the quasar intrinsic emission. We find a BAL quasar fraction close to 20\% at z24z\sim2-4, while it increases to almost 50\% at z6z\sim6. The velocity and width of the BAL features also increase at z4.5z\gtrsim4.5. We exclude that the redshift evolution of the BAL properties is due to differences in terms of quasar luminosity and accretion rate. These results suggest significant BH feedback occurring in the 1 Gyr old Universe, likely affecting the growth of BHs and, possibly, of their host galaxies, as supported by models of early BH and galaxy evolution.Comment: Accepted for publication in Ap

    The physics and astrophysics of X-ray outflows from Active Galactic Nuclei

    The highly energetic outflows from Active Galactic Nuclei detected in X-rays are one of the most powerful mechanisms by which the central supermassive black hole (SMBH) interacts with the host galaxy. The last two decades of high resolution X-ray spectroscopy with XMM and Chandra have improved our understanding of the nature of these outflowing ionized absorbers and we are now poised to take the next giant leap with higher spectral resolution and higher throughput observatories to understand the physics and impact of these outflows on the host galaxy gas. The future studies on X-ray outflows not only have the potential to unravel some of the currently outstanding puzzles in astronomy, such as the physical basis behind the MBHσ-\sigma relation, the cooling flow problem in intra-cluster medium (ICM), and the evolution of the quasar luminosity function across cosmic timescales, but also provide rare insights into the dynamics and nature of matter in the immediate vicinity of the SMBH. Higher spectral resolution (0.5\le 0.5 eV at 11 keV) observations will be required to identify individual absorption lines and study the asymmetries and shifts in the line profiles revealing important information about outflow structures and their impact. Higher effective area (1000cm2\ge 1000 \rm \,cm^{2}) will be required to study the outflows in distant quasars, particularly at the quasar peak era (redshift 1z31\le z\le 3) when the AGN population was the brightest. Thus, it is imperative that we develop next generation X-ray telescopes with high spectral resolution and high throughput for unveiling the properties and impact of highly energetic X-ray outflows. A simultaneous high resolution UV + X-ray mission will encompass the crucial AGN ionizing continuum, and also characterize the simultaneous detections of UV and X-ray outflows, which map different spatial scales along the line of sight.Comment: A Science White Paper submitted to the Astro2020 Decadal Surve

Using twitter data, how short term travelers, such as tourists, can be recognized and how their travel pattern can be analyzed. Study of finding tourist dynamics such as arriving and outgoing of tourist, sum of trips, sum of days and night spent, number of unique visitors, country of residence, main destination, secondary destination, transits pass through, repeat visits are achieved using CDR (call detail records) and DDR (data detail records)
