MNE’s Regional Location Choice - A Comparative Perspective on East Germany, the Czech Republic and Poland
The focus of this article is the empirical identification of factors influencing Foreign Direct Investment (FDI) in transition economies on a regional level (NUTS 2). The analysis is designed as benchmark between three neighboring post-communist regions, i.e. East Germany, the Czech Republic and Poland. Their different transition paths have not only resulted in economic differences. We can also observe today that the importance of pull factors for FDI varies significantly across the regions. This analysis shows that in comparison with Poland and the Czech Republic, East Germany’s major benefit is its purchasing power, its geographical proximity to West European markets, and its modern infrastructure. Furthermore, the analysis suggests that intra-industry linkages such as specialization and agglomeration economies are relevant factors for the location decision of foreign investors. The "Free Voters": A Decent Alternative for Conservatives?
The Free Voters (Freie Wähler) party is becoming a more serious competitor in the German party system. It gained parliamentary representation in the Rhineland Palatine in 2021 and has been governing with the Christian Social Union in Bavaria since 2018. In addition, the Free Voters party now holds two of the 96 German parliamentary member positions in the European Parliament and is, at the federal level, the extraparliamentary opposition party with the greatest number of votes. Nevertheless, there has, to date, been no analysis examining people's motives for voting for the Free Voters or exploring their future openness to voting for the party. This article addresses this oversight by identifying factors, based on population-representative data from the 2021 Bundestag elections, that favour openness and voting for this increasingly influential party. Our analysis shows that a more general dissatisfaction with democratic outputs, a conservative attitude, and low perceptions of spatial distance from the political positions of the Free Voters have significant positive effects.Die Freien Wähler entwickeln sich zu einem ernstzunehmenden Konkurrenten im deutschen Parteiensystem. Sie sind 2021 in den rheinland-pfälzischen Landtag eingezogen und regieren seit 2018 gemeinsam mit der CSU in Bayern. Zudem stellen die Freien Wähler mittlerweile zwei der 96 deutschen Abgeordneten im Europäischen Parlament und sind auf Bundesebene die stimmenstärkste außerparlamentarische Oppositionspartei. Dennoch gibt es bisher keine Analyse, die die Motive der Wähler:innen für die Wahl der Freien Wähler oder eine mögliche zukünftige Bereitschaft, die Partei zu wählen, untersucht. Dieser Artikel behebt dieses Defizit, indem er auf der Grundlage von bevölkerungsrepräsentativen Daten zur Bundestagswahl 2021 Faktoren identifiziert, die die Offenheit und die Stimmabgabe für diese zunehmend einflussreiche Partei begünstigen. Unsere Analyse zeigt, dass eine allgemeinere Unzufriedenheit mit der demokratischen Performanz, eine konservative Grundhaltung sowie eine geringe wahrgenommene räumliche Distanz zu den politischen Positionen der Freien Wähler positive Effekte haben
Selective inactivation of hypomethylating agents by SAMHD1 provides a rationale for therapeutic stratification in AML.
Hypomethylating agents decitabine and azacytidine are regarded as interchangeable in the treatment of acute myeloid leukemia (AML). However, their mechanisms of action remain incompletely understood, and predictive biomarkers for HMA efficacy are lacking. Here, we show that the bioactive metabolite decitabine triphosphate, but not azacytidine triphosphate, functions as activator and substrate of the triphosphohydrolase SAMHD1 and is subject to SAMHD1-mediated inactivation. Retrospective immunohistochemical analysis of bone marrow specimens from AML patients at diagnosis revealed that SAMHD1 expression in leukemic cells inversely correlates with clinical response to decitabine, but not to azacytidine. SAMHD1 ablation increases the antileukemic activity of decitabine in AML cell lines, primary leukemic blasts, and xenograft models. AML cells acquire resistance to decitabine partly by SAMHD1 up-regulation. Together, our data suggest that SAMHD1 is a biomarker for the stratified use of hypomethylating agents in AML patients and a potential target for the treatment of decitabine-resistant leukemia

factors, one of which is the use of an appropriate microarray surface for the
immobilisation of either protein or antibody sample. For optimisation of the
technology an evaluation of the properties of 8 optimised surfaces was
conducted in the context of it use in both, protein and antibody microarray
technology. The comparison was performed under the same conditions as earlier
studies with HSA and polyclonal anti-fibrinogen antibodies as model
interaction partners. The functionality of all new slide coatings was
demonstrated and the mean signal to spotted concentration ratio as well as the
detection limits and the coefficients of variation were evaluated. Moreover,
new concepts for slide coatings such as dendrimer and PEG-epoxy slides were
assessed and improved qualities of such slide surfaces were observed. Optimal
slide coatings for antibody and protein chips were proposed and the
requirements for both technologies were discussed. Based on these
optimisations, a new technology for the multiplex analysis for compounds on
microarrays was developed, since current solutions, such as conventional
microarrays, microfluidic systems and chips bearing microwells lack the
possibility to screen several analytes on different immobilised binders at a
time or require complicated liquid handling, surface modifications, or
additional equipment. In the new approach, an immunoassay was performed using
standard surfaces and machinery already available for microarray technology
for multiplex analysis on a standard microscope slide without the requirement
for wells or tubes to separate the samples. The new multiple spotting
technique (MIST) comprises immobilisation of a binder onto a surface and
subsequent spotting of the second compound, comprised in a hygroscopic buffer,
on the same spot, on top of the immobilised binder. It was shown, that the
analytes bind their ligands immediately, within the confined space of separate
droplets on the chip surface, thereby eliminating the need for extra
incubation time. Furthermore, the feasibility of the new technique was
illustrated by spotting dilution rows of proteins or monoclonal and polyclonal
antibodies on top of their immobilised interaction partners with a sensitivity
of as little as 400 zeptomoles (240,000 molecules) of analyte. Specificity and
compatibility to commonly used techniques such as total incubation was
displayed by specific recognition of a protein by an equimolar mixture of
antibodies and subsequent detection by a total incubation with labelled
secondary antibodies. The same approach was used to facilitate the
identification of high-affinity binders from a great variety of phage display-
derived recombinant antibodies. While the selection process for phage
displayed antibody fragments itself has been automated, the bottleneck was
shifted further downstream to the identification of monoclonal binders
obtained from the selections. Using the new approach recombinant antibody
fragments were expressed in a single inoculation and expression step and
subjected without purification directly to their immobilised antigens. Since
immobilisation of the scFv�s was not possible so far due to loss of
functionality, the multiplex assay represents a major improvement and allows
automated high-throughput screening of phage-display derived selections with
comparable sensitivity to the commonly used ELISA. Moreover, manual
interaction steps were minimised and the technique was streamlined to be
accessible within the automated selection procedure. ...Die Qualität von Protein- und Antiköper-Mikroarrays ist von vielen Faktoren
abhängig. Ein wichtiger Faktor hierbei ist die Oberflächenchemie, welche
spezifisch die jeweiligen Proteine oder Antiköper an die Oberfläche bindet.
Zur Optimierung der Mikroarray-Technologie wurde ein Vergleich von acht für
Protein und Antikörper Mikroarray optimierte Oberflächenchemien mit HSA und
polyklonalen anti-Fibrinogen Antikörpern als Modellprotein bzw.
Modellantikörper unter de ngleichen Bedingungen wie frühere Studien
durchgeführt. Die Funktionalität aller Oberflächen konnte gezeigt werden, und
das Verhältnis von Signal zu aufgebrachter Konzentration, sowie die
Detektionslimits und Variations-Koeffizienten wurden analysiert. Weiterhin
wurden neue Konzepte zur Herstellung der Oberflächenkopplungschemie wie PEG-
Epoxy-Oberflächen und Dendrimer-Oberflächen untersucht und verbesserte
Eigenschaften festgestellt. Optimale Oberflächenkopplungschemien wurden
erläutert und im Hinblick auf die Anforderungen von Protein- und Antikörper-
Mikroarrays diskutiert. Auf der Basis der Oberflächenoptimierung wurde
weiterhin eine neue Technologie zur multiplexen Analyse von Substanzen auf
Mikroarrays entwickelt, da es mit konventionellen Mikroarrays,
mikrofluidischen Systemen und Nanowell-Chips entweder nicht möglich ist,
gleichzeitig mehrere Analyte auf verschiedenen Bindern zu testen, oder weil
hierfür komplizierte Apparaturen zur Leitung des Flüssigkeitsstroms oder zur
Herstellung der Nanowells benötigt werden. In dem neuen Ansatz wurden
multiplexe Immunoassays auf Mikroarray-Oberflächen ohne Vertiefungen mit Hilfe
von konventionellen Spotting-Robotern durchgeführt. Das Verfahren, benannt
�Multiple Spotting Technique� (MIST), umfasst hierbei die Immobilisierung des
Binders auf einer Oberfläche und den anschließenden Transfer des Analyten mit
Hilfe der gleichen Roboter auf die gleiche Stelle des Mikroarrays und somit
auf den immobiliserten Binder. Da der Analyt sich hierbei in einem
hygroskopischen Puffer befindet, entsteht ein Mikroreaktionsvolumen, in
welchem der Analyt ohne zusätzliche Inkubationszeiten vom Binder gebunden
wurde. Die Durchführbarkeit des Konzepts konnte durch den Transfer von
Verdünnungsreihen monoklonaler und polyklonaler Antikörper auf deren
immobiliserte Antigene und die anschließende Detektion der spezifischen
Antikörper-Antigen Komplexe gezeigt werden. Hierbei wurden Detektionslimits
von 400 Zeptomol (240.000 Antikörpermoleküle) gemessen. Die Spezifität, sowie
die Kompatibilität zu gängigen Mikroarray-Techniken, wie der Gesamtinkubation,
wurden durch die Aufbringung einer äquimolaren Antikörpermischung auf ein
Antigen und anschließenden Gesamtinkubation mit markierten Sekundärantikörpern
gezeigt, welches zur spezifischen Bildung von Antikörper-Antigen-Komplexen
führte. Der gleiche Ansatz wurde auch zur Untersuchung von rekombinanten
Antikörperpools von �Phage Display�-Selektionen angewandt. Während die
eigentliche Selektion von rekombinanten Antikörpern mittlerweile automatisiert
wurde, stellt die anschließende Identifikation positiver Binder bis heute noch
einen großen Arbeitsaufwand dar. In dem neuen Ansatz werden die
Antikörperfragmente-produzierenden Zellen in einem einzigen Inokulations- und
Induktionsschritt im 384er Maßstab kultiviert und anschließend mit Hilfe der
Multiple Spotting Technique auf die Antigene aufgebracht. Gleichzeitig wurden
alle Antikörperfragmente auf Kreuzreaktivität mit Milchpulver untersucht,
wodurch unspezifische Binder identifiziert werden konnten. Ein entscheidender
Vorteil der MIST war hierbei, dass zusätzliche Inkubationszeiten durch die
eingeschränkten Diffusionsmöglichkeiten innerhalb der Reaktionseinheiten
ausgelassen werden konnten. Eine solche Untersuchung mit Hilfe von Mikroarrays
war bisher nicht möglich, da die scFv's ohne Funktionsverlust nicht
immobilisiert werden konnten. Der Vergleich des neuen Verfahrens mit ELISA
zeigte, dass der MIST-basierte Ansatz schneller vergleichbare Sensitivitäten
mit geringerem Antigenverbrauch lieferte. Weiterhin kann der neuartige Ansatz
in die bestehende Automatisierung integriert werden, so dass eine schnellere
Produktion von spezifischen rekombinanten Antikörpern möglich ist. ..
Next generation of protein microarray support materials: Evaluation for protein and antibody microarray applications
The performance of protein and antibody microarrays is dependent on various factors, one of which is the use of an appropriate microarray surface for the immobilisation of either protein or antibody samples. We have investigated the properties of seven new surfaces in the context of both protein and antibody microarray technology. We have demonstrated the functionality of all new slide coatings and investigated the mean signal to spotted concentration ratio, determined detection limits and calculated coefficients of variation. Moreover, new concepts for slide coatings such as dendrimer and poly(ethylene glycol)-epoxy slides were evaluated and improved qualities of novel slide surfaces were observed. Optimal slide coatings for antibody and protein chips were proposed and the requirements for both technologies were discussed
