    Molecular and Cellular Mechanisms for Microbial Discrimination in Sponges

    Sponges (Porifera) are basal metazoans that feed on and establish symbiotic associations with microbes. How sponges discriminate between bacteria to digest as food, incorporate as symbionts, or eliminate as pathogens remains unknown. This thesis aimed to study mechanisms for microbial discrimination in sponges. Mechanisms likely to modulate sponge-microbe interactions are introduced (Chapter 1). The host transcriptomic response upon seawater and symbiont microbial consortia encounter was characterized by RNASeq in two sponges (Chapter 2). Aplysina aerophoba showed little differential gene expression and no participation of receptors, whereas Dysidea avara responded by regulating NLRs suggesting that microbial discrimination is driven by the repertoire of immune genes as well as to what degree they are induced. On the cellular level, an in-vivo phagocytosis assay was established in Halichondria panicea combining incubations with microalgae, bacteria, and latex beads with cell dissociation and fluorescence-activated cell sorting (FACS) to quantify particle incorporation into sponge cells (Chapter 3). After 30 min particles were predominantly incorporated into choanocyte-like cells and appeared to be translocated to archaeocyte-like cells after 60 min. Lastly, the established assay was combined with proteomic analysis to investigate H. panicea’s phagocytic response upon exposure to a “native” and a “foreign” Vibrio isolate (Chapter 4). Vibrio incorporation into sponge cells was indiscriminate, but the distribution of vibrios into different cell types differed between isolates. Phagocytic-related proteins were in a higher abundance in the foreign vs. the native treatment. These results indicate that bacterial discrimination in H. panicea occurs after internalization leading to differences in the processing of foreign vs native vibrio types

    Using Student Ambassadors to Relay Themes from Changing the Conversation in Engineering First Year Seminars

    This paper describes the efforts at a large mid-Atlantic university to integrate themes from Changing the Conversation into First Year Seminars. Changing the Conversation, a 2008 book by the National Academy of Engineering, found that both male and female students were more attracted to messages describing engineering in terms relating to societal impact, such as the phrases, Engineering makes a world of difference and Engineering is essential to our health, happiness, and safety. Although the research was conducted with younger students, the potential for using these themes in the undergraduate curricula could have the potential to impact persistence in engineering, especially for female students or those from other underrepresented groups. The purpose of the initiative described in the paper, which uses engineering students from a group called the Engineering Ambassadors to relay these messages in freshmen level courses, is to impact student perceptions of engineering and to provide information to students that will be critical in making career decisions. In the Fall of 2011, a pilot program was launched in two sections of a Chemical Engineering First Year Seminar. Engineering Ambassadors made four separate visits to each section, focusing on the following topics: 1) An overview of College of Engineering Majors, 2) Options within Chemical Engineering, 3) Student experiences in the College of Engineering, and 4) How to be a successful engineering student. Woven through each presentation were themes from Changing the Conversation, focusing on how engineers are essential to health, happiness and safety. The students were mentored by a faculty member whose background is in Communication. The quality of student presentations was high, utilizing the assertion-evidence method of slide design. Data was collected to determine whether the following project objectives were met: 1) Students in the First Year Seminars will have a greater understanding of the possible careers in engineering as well as the engineering majors; and 2) Students will be more likely to define engineering in terms associated with health, happiness, and safety. The data showed that the students had a very positive reaction to the Engineering Ambassador visits, although a larger sample size would be necessary to more clearly understand the impact

    An analysis of the evaluation practices of employer-sponsored training in the financial services industry

    Instructional evaluation is essential for assessing the effectiveness of learning events. In today's economy, corporations are under great pressure to reduce expenses, and training budgets often feel the effects. The closure of in-house training programs, combined with the reduction of training budgets, could be seen as evidence of training professionals' inability to prove their worth in terms of organizational benefit. To solidify their value to an organization, training departments must assess the effectiveness of their programs and provide evidence that they are supporting the organization's goals. The purpose of this study was to determine how employer-sponsored training is evaluated in the financial services industry by firms affiliated with DALBAR, Inc., using Kirkpatrick's four-level evaluation model as a framework. The total population for this study consisted of all financial services organizations providing education and training. The target population was limited to the financial services organizations that subscribe to the services provided by DALBAR, Inc. Affiliation with DALBAR was considered an indicator of interest in raising standards of excellence within the financial services industry. Therefore, DALBAR affiliation was believed to represent organizations whose training personnel had current knowledge of industry practices and thereby would report higher usage of evaluation than the total population of other entities. Data was collected using a modified survey instrument. Patterns, trends, models and methods of training evaluation among these financial services organizations were examined. Finally, barriers to implementation of training evaluation were identified and explored. Recommendations for practice include increasing training department staff members' knowledge of evaluation theories and techniques, as well striving to make training evaluation a priority for the organization as a whole. Among the recommendations for future research is the execution of a qualitative study to be conducted through in-depth interviews with selected respondents to explore in greater detail the relationships between organizational characteristics and the implementation of higher levels of evaluation

    Inicialmente se debe aclarar que el presente ensayo es un abordaje crítico de una investigación realizada dentro del Diplomado en Construcción de Redes Sociales hacia la OSP Corporación Artística y Social Skené. Esta OSP funciona como la formación de disciplinas artísticas, como la dramaturgia y las artes escénicas y su finalidad es impartir ese conocimiento que el director de la corporación ha adquirido con los años y lograr inspirar a muchos jóvenes y así mismo mantener vivo el amor por el arte. La corporación social y artística Skené ha atravesado por muchas etapas las cuales han dejado grandes huellas y vacíos dentro de su actividad y su proyección, uno de ellos fue la pandemia ya que no lograron generar tanta conexión con los demás estudiantes que habían en ese entonces y algunos desertaron, para este momento su nivel de conocimiento ante la comunidad era paupérrimo, costaba un poco salir a recorrer escenarios pues no eran reconocidos, esta fue la oportunidad que se vio para iniciar una estrategia comunicativa y social con esta OSP la investigación y entender cómo se podía ayudarlos y también como se podían acercar más a la comunidad.Initially, it should be clarified that this essay is a critical approach to an investigation carried out within the Diploma in Construction of Social Networks towards the OSP Corporación Artística y Social Skené. This OSP works as the training of artistic disciplines, such as dramaturgy and performing arts and its purpose is to impart that knowledge that the director of the corporation has acquired over the years and to inspire many young people and likewise keep alive the love for the art. The Skené social and artistic corporation has gone through many stages which have left great traces and gaps within its activity and its projection, one of them was the pandemic since they did not manage to generate as much connection with the other students that there were at that time and some deserted, for this moment their level of knowledge before the community was very poor, it was a bit difficult to go out to explore the scenarios because they were not recognized, this was the opportunity that was seen to start a communication and social strategy with this OSP the investigation and understand how you could help them and also how they could get closer to the community

    En el ambiente laboral, los factores que influyen en la Unidad de Cuidados Intensivos pueden afectar la salud física y mental de los trabajadores de ahí la importancia de su identificación y control. Así mismo, estos pueden influir en la satisfacción laboral de los empleados. Este estudio tiene como objetivo determinar los factores que influyen en la satisfacción laboral del personal de enfermería del Servicio de la Unidad de Cuidados Intensivos Pediátricos de una clínica de Lima. El estudio es de enfoque cuantitativo, descriptivo observacional, diseño no experimental de corte transversal que permitirá la medición y cuantificación de la variable de estudio. La muestra estará compuesta por personal de enfermería 55 licenciados en Enfermería. Para la recolección de datos se utilizará el instrumento de la Escala de Satisfacción Laboral. Así mismo, se realizó el análisis con el coeficiente de validez V de Aiken obteniéndose un valor de 0.91. También, se realizó el análisis de la confiabilidad del instrumento obteniéndose un resultado 0.99, según la prueba estadística de Alfa de Cronbach.     En el Perú existen tres niveles de gobierno: central (ministerios), regional (gobiernos regionales) y local (municipalidades provinciales y distritales), los cuales se encargan de comprar bienes y servicios y contratar obras con la finalidad de crear, sobre todo, infraestructura, que permita una mejora en la calidad de vida de su población. Lamentablemente, muchas de esas compras realizadas en el Perú se han visto marcadas sea por actos de corrupción que han llevado a cabo coludidos las autoridades estatales junto a malos empresarios carentes de ética empresarial. De igual forma, la desidia, desinterés, incapacidad y desmotivación de los funcionarios públicos ha llevado a que no se efectúe adecuadamente las labores de control, supervision y fiscalización de las obras públicas generando que éstas sean entregadas sin cumplir los estandares de calidad exigidos y empiecen a deteriorarse rápidamente, perjudicando los ingresos fiscales y -lo que es peor- las expectativas y necesidades de la población. A través del presente trabajo de investigación, se busca analizar esta preocupación, esta problemática que afecta tanto a la misma Municipalidad Distrital de Ventanilla como a su población entera. De la solución que se plantee dependerá que las obras convocadas por la administración municipal se concreten adecuada y acertadamente en beneficio de la población más necesitada, principalmente, de los asentamientos humanos apostados a lo largo y ancho del Distrito de Ventanilla
