    Assessing the convergent validity between the automated emotion recognition software Noldus FaceReader 7 and Facial Action Coding System Scoring

    This study validates automated emotion and action unit (AU) coding applying FaceReader 7 to a dataset of standardized facial expressions of six basic emotions (Standardized and Motivated Facial Expressions of Emotion). Percentages of correctly and falsely classified expressions are reported. The validity of coding AUs is provided by correlations between the automated analysis and manual Facial Action Coding System (FACS) scoring for 20 AUs. On average 80% of the emotional facial expressions are correctly classified. The overall validity of coding AUs is moderate with the highest validity indicators for AUs 1, 5, 9, 17 and 27. These results are compared to the performance of FaceReader 6 in previous research, with our results yielding comparable validity coefficients. Practical implications and limitations of the automated method are discussed

    A Viable Business Model for Innovations with Digital Healthcare Applications in Germany

    Germany established laws to quickly introduce digital innovations in healthcare by forcing statutory insurances to reimburse companies producing digital applications. This could enhance the well-being of patients. For example, an application in psychotherapy can cut the waiting time for a psychotherapist in Germany. However, such enhancements will reach the patients only if companies offering digital applications have a viable business model to survive. Our analysis of the business model of a company offering a recognized digital application shows that such business models are not easy to develop. The analysis is transferrable to other countries, if they establish similar laws. First, we describe the legal framework for digital healthcare applications set up in Germany. We also describe the conditions that must be met for such an application to be recognized by the relevant body so that statutory insurances must pay for it. This is followed by a discussion of the reimbursement amount. Then, we develop the business model of a producer of a specific digital healthcare application. Although the possibility of reimbursement for accepted applications constitutes a competitive advantage, underestimating costs from the approval process and marketing may be dangerous.     Shrinkage vectors in flowable bulk-fill and conventional composites: bulk versus incremental application

    OBJECTIVES Sufficient depth of cure allows bulk-fill composites to be placed with a 4-mm thickness. This study investigated bulk versus incremental application methods by visualizing shrinkage vectors in flowable bulk-fill and conventional composites. MATERIALS AND METHODS Cylindrical cavities (diameter = 6 mm, depth = 4 mm) were prepared in 24 teeth and then etched and bonded with OptiBond FL (Kerr, Italy). The composites were mixed with 2 wt% radiolucent glass beads. In one group, smart dentin replacement (SDR, Dentsply) was applied in bulk "SDR-bulk" (n = 8). In two groups, SDR and Tetric EvoFlow (Ivoclar Vivadent) were applied in two 2-mm-thick increments: "SDR-incrementa" and "EvoFlow-incremental". Each material application was scanned with a micro-CT before and after light-curing (40 s, 1100 mW/cm2), and the shrinkage vectors were computed via image segmentation. Thereafter, linear polymerization shrinkage, shrinkage stress and gelation time were measured (n = 10). RESULTS The greatest shrinkage vectors were found in "SDR-bulk" and "SDR-increment2," and the smallest were found in "SDR-increment1-covered" and "EvoFlow-increment1-covered." Shrinkage away from and toward the cavity floor was greatest in “SDR-bulk“ and “EvoFlow-increment2", respectively. The mean values of the shrinkage vectors were significantly different between groups (one-way ANOVA, Tamhane's T2 test, p < 0.05). The linear polymerization shrinkage and shrinkage stress were greatest in Tetric EvoFlow, and the gelation time was greatest in "SDR-bulk". CONCLUSIONS The bulk application method had greater values of shrinkage vectors and a higher debonding tendency at the cavity floor.     Exploring societal preferences for energy sufficiency measures in Switzerland

    Many countries are facing a challenging transition towards more sustainable energy systems, which produce more renewables and consume less energy. The latter goal can only be achieved through a combination of efficiency measures and changes in people’s lifestyles and routine behaviours (i.e. sufficiency). While research has shown that acceptance of technical efficiency is relatively high, there is a lack of research on societal preferences for sufficiency measures. However, this is an important prerequisite for designing successful interventions to change behaviour. This paper analyses societal preferences for different energy-related behaviours in Switzerland. We use an online choice-based conjoint analysis (N=150) to examine preferences for behaviours with high technical potentials for energy demand reduction in the following domains: mobility, heating and food. Each domain comprises different attributes across three levels of sufficiency. Respondents were confronted with trade-off situations evoked through different fictional lifestyles that comprised different combinations of attribute levels. Through a series of trade-off decisions, participants were asked to choose their preferred lifestyle. The results revealed that a vegetarian diet was considered the most critical issue that respondents were unwilling to trade off, followed by distance to workplace and means of transportation. The highest willingness to trade off was found for adjustments in room temperature, holiday travel behaviours, and living space. Participants’ preferences for the most energy-sufficient lifestyles were rather low. However, the study showed that there were lifestyles with substantive energy-saving potentials that were well accepted among respondents. Our study results suggest that the success of energy-sufficiency interventions might depend strongly on the targeted behaviour. We speculate that they may face strong resistance (e.g., vegetarian diet). Thus, it seems promising to promote well-balanced lifestyles, rather than extremely energy-sufficient lifestyles, as potential role models for sufficiency

    Comparative Multi-Epitope-Ligand-Cartography reveals essential immunological alterations in Barrett's metaplasia and esophageal adenocarcinoma

    <p>Abstract</p> <p>Background</p> <p>Barrett's esophagus (BE) is caused by gastroesophageal reflux with consecutive mucosal inflammation, predisposing patients to the development of esophageal adenocarcinoma (EAC). We investigated changes in T cell-related mucosal combinatorial molecular protein patterns in both diseases using the novel Multi-Epitope-Ligand-Cartography, a unique robotic whole-cell imaging technology that simultaneously visualizes dozens of proteins in structurally intact tissues and correlates cellular localization of proteins with function.</p> <p>Results</p> <p>Biopsies were taken during endoscopy from BE, EAC, and normal control tissue, and proteomic microscopy was performed on 32 different epitopes. When the significance level was set to p < 0.0005 and the search depth to five antibody combinations, controls and BE can be differentiated by 63, controls and EAC by 3222, and BE from EAC by 1521 distinct protein combinations.</p> <p>For example, the number of activated apoptotic naĂŻve and memory T cells was significantly increased only in BE, whereas the number of activated apoptotic helper and regulatory T cells was significantly elevated in BE and EAC. In contrast, the number of activated apoptotic cytotoxic T cells was significantly elevated only in EAC. Confirming different pathways in BE and EAC, the number of T lymphocytes with p53 expression and downregulation of bcl2 expression (CD3<sup>+</sup>p53<sup>+</sup>Bcl2<sup>-</sup>NfkB<sup>-</sup>) was significantly increased in EAC compared to BE and controls. Interestingly, the number of precursor T cells (CD7<sup>+</sup>) was significantly elevated only in EAC. These cells lack Bax and caspase-8, suggesting impaired apoptosis in the early stages of T cell differentiation.</p> <p>Conclusion</p> <p>Proteomic analysis showed for the first time that proteins, which are critically involved in the mucosal immune system of the esophagus, are distinctly expressed in BE and EAC, whereas others are comparably altered in both diseases, suggesting that many pathogenic events might be shared by both diseases. Topological proteomic analysis, therefore, helps us to understand the different pathogenic events in the underlying disease pathways.</p
